Eutrophication and Freshwater Red-tide Algae on Early Impoundment Stage of Jeolgol Reservoir in the Paikryeong Island, West Sea of South Korea (백령도 절골저수지의 부영양화와 담수적조)

  • A systematic water quality survey was conducted in August, 2005 for a drinking water supply reservoir (the Jeolgol reseuoir located in an island), which is at an early stage of impoundment, to investigate the causes of water color deterioration of the reservoir and the clogging of filter beds of a water treatment plant. The reservoir shape was simple and its average depth was 5.5 m, increasing from upreservoir toward the downreservoir end near the dam. Dissolved oxygen (DO) and chloropllyll-a (chi-a) showed a large variation while water temperature had a smaller range. Transparency ranged from 0.6 to 0.9 m (average 0.7 m). The average value of turbidity was 9.3 NTU, ranging from 8.0 12.1 NTU. The transparency and the turbidity appear to be affected by a combination of biological and non-biological factors. The poor transparency was explained by an increase of inorganic colloids and algal bloom in the reservoir. The blockage of the filter bed was attributed to the oversupply of phytoplanktons from the reservoir. The range and the average concentration of chi-a within the reservoir were 31.6 258.9 μgL1, 123.6 μgL1 for the upper layer, and 17.0 37.4 μgL1, 26.5 μgL1 for the bottom layer, respectively. A predominant species contributing the algal bloom was Dinophyceae, Peridinium bipes f. occultatum. The distribution of Peridinium spp. was correlated with chi-a concentrations. The standing crop of phytoplankton was highest in the upreservoir with 8.5×103cellsmL1 and it decreased toward the downresevoir. Synedra of Bacillariophyceae and Microcystis aeruginosa of Cyanophyceae appeared to contribute to the algal bloom, although they are not dominated. It is mostly likely that sloped farmlands located in the watershed of the reservoir caused water quality problems because they may contain a significant amount of the nutrients originated from fertilizers. In addition, the aerators installed in the reservoir and a shortage of the inflowing water may be related to the poor water quality. A long-term monitoring and an integrated management plan for the water quality of the watersheds and the reservoir may be required to improve the water quality of the reservoir.

Studies on the Reproductive Cycle of Damselfish, Chromis notatus (Temminck et Schlegel) (자리돔의 생식주기에 관한 연구)

  • Annual reproductive cycle of the Damselfish, Chromis notatus collected monthly at the four coastal areas around Chejudo, Korea are studied on the bases of histological observations of gonadal tissue and various quantitative variables including gonadosomatic index (GSI), fatness, egg diameter composition and the first maturity. The ovary consisted of a pair of saccular structure with many ovarian sacs. Oogonia proliferated along the germinal epithelium of the ovarian sac. Young oocytes with basophilic cytoplasm showed several nucleoli along the nuclear membrane. When the oocytes reached about 450μm in diameter, nucleus migrate toward the animal pole, nuclear membrane disappeared and most of cytoplasm were filled with yolk materials and oil drops. After ovulation, residual follicle and growing oocytes remaining in the ovarian sacs degenerated. But early young oocytes without follicle layer were not degenerated, and growing continuously till the next year. The testis consisted of a pair of lobular structures in the right and left were united in the posterior seminal vesicle. Cortex of testis was composed of many sperm ducts connected with lobuli. GSI began to increase from March, starting season of longer day length and higher water temperature, and reached the maximum value between June and August. It began to decrease from September with the lowest value appearing between October and February without any evident variation. The annual reproductive cycle could be devided into five successive stage : growing(April to Many), mature(May to August), ripe and spent (June to August) and recovery and resting stage(September to March). The spawning peak occurred from June to August. According to the frequency distribution of egg diameter, Chromis notatus was a polycyclic species to spawn twice or more in a spawning season. Fatness, correlated with gonadal phases, was remarkably decreased by spawning. Percentage of the first maturity . in femate and male fish ranging from 7.0 to 7.9 cm were 50% and from 9.0 to 9.9 cm in total length 100%.

Population density of potato virus vectors In the Kwanghwal Area, Kimje-gun, Cholla-Pukto, on the western coast (씨감자 생산을 위한 매개 진딧물 조사 - 전북 김제군 광활면의 진딧물 분포상 -)

  • Present system of seed potato production in Korea has several weak points and consequently has difficulties in covering annual shortage of 60,000 tons of seed potatoes. The author has an opinion that this so called 'High land system' of seed potato production adopted by the Government should be replaced by the 'Coastal area system' which is proposed by the author and has many advantages over present 'High land system'(2). In coastal areas where enormous acreage of rice paddies are spread, mostly around the villages. the primary host plants of the vectors are found. Therefore, the only source of aphid vectors are limited to the villages. The farmer's houses scattered more sparsely also have minor importance. In the previous paper(2), the author reported that the aphid vector populations were lower in the coastalareas than at Taegwanryong where the Alpine Experiment Station for the production. of seed potatoes is located. However, the number to vectors at Okku showed rather high density, where the trap was placed at the distance of 200 m from a village where peach and Hibiscus trees, the primary hosts of Myzus pesrsicae and Aphis gossypii were grown. To clarify the flight distance from the source of the aphid vectors, a trial was carried out in the Kwanghwal area, Kimje-gun, Cholla-pukto. on the western coast. 13 traps were placed at four directions and the distances between the traps were 250 m. (Fig. I) The traps 'Were operated from June 21 to October 31. The results are shown in Table 1. A total of some 70 species of aphids were found, including 5 speceis of potato virus vectors. The vectors are as follows: I. Myzus persicae (Sulzer) 2. Aphis gossypii Glover 3. Aulacorthum solani (Kaltenbach) 4. Lipaphis erysimi (Kaltenbach) 5. Macrosiphoniella sanborni (Gillette) Out of a total of 12,797 aphids, 5,187(48%) vectors were found. The trap catches at the 13 locations are shown in Fig. 2 and the numbers of the vectors at each location for each vector, except Macro-siphoniella sanborni. of which only a single individual was caught, are shown in Fig. 3-6. Number of vectors at C (3,279) (Centre of the village) is considerably higher than that at Suwon (763); however, EI. SI. WI and NI. where the distanecs from Care 250 m, showed lower numbers of vectors than that at Taegwanryong (347). The number of vectors at NI was rather than at the other 3 locations at the distance of 250m from the village. This was because C was in the southern part of the village. Consequently NI was much closer to the village than the other 3 locations of the same distance from C. Numbers of catches of the most important vector. Myzus persicae, are shown in Fig.3. The distribution pattern is typical except S2andW3, where several farmer's houses were found. If only the rice paddies were found in these locations. the numbers of the vectors would be small as the distances increase. Numbers of catches of the other 3 vectors are shown in Fig. 4-6. From these results. the author has drawn the following conclusions: 1. The aphid vector sources at the rice paddy belt in the western coast are the villages. 2. The vector densities at the locations where the distances are 250 m from the centre of the village are lower than that at Taegwanryong. 3. The vector densities become gradually lower as the distances from the centre of village increase. However. depending on the host plant situation at each location, the vector densities are variable. These minor sources of aphid vectors may be eliminated so that seed potatoes can be grown. 4. Thus. under the direction of specialists, fields suitable for seed potato production can be found in the coastal areas.

Induction of Phase I, II and III Drug Metabolism/Transport by Xenobiotics

  • Drug metabolizing enzymes (DMEs) play central roles in the metabolism, elimination and detoxification of xenobiotics and drugs introduced into the human body. Most of the tissues and organs in our body are well equipped with diverse and various DMEs including phase I, phase II metabolizing enzymes and phase III transporters, which are present in abundance either at the basal unstimulated level, and/or are inducible at elevated level after exposure to xenobiotics. Recently, many important advances have been made in the mechanisms that regulate the expression of these drug metabolism genes. Various nuclear receptors including the aryl hydrocarbon receptor (AhR), orphan nuclear receptors, and nuclear factor-erythoroid 2 p45-related factor 2 (Nrf2) have been shown to be the key mediators of drug-induced changes in phase I, phase II metabolizing enzymes as well as phase III transporters involved in efflux mechanisms. For instance, the expression of CYP1 genes can be induced by AhR, which dimerizes with the AhR nuclear translocator (Arnt) , in response to many polycyclic aromatic hydrocarbon (PAHs). Similarly, the steroid family of orphan nuclear receptors, the constitutive androstane receptor (CAR) and pregnane X receptor (PXR), both heterodimerize with the ret-inoid X receptor (RXR), are shown to transcriptionally activate the promoters of CYP2B and CYP3A gene expression by xenobiotics such as phenobarbital-like compounds (CAR) and dexamethasone and rifampin-type of agents (PXR). The peroxisome proliferator activated receptor (PPAR), which is one of the first characterized members of the nuclear hormone receptor, also dimerizes with RXR and has been shown to be activated by lipid lowering agent fib rate-type of compounds leading to transcriptional activation of the promoters on CYP4A gene. CYP7A was recognized as the first target gene of the liver X receptor (LXR), in which the elimination of cholesterol depends on CYP7A. Farnesoid X receptor (FXR) was identified as a bile acid receptor, and its activation results in the inhibition of hepatic acid biosynthesis and increased transport of bile acids from intestinal lumen to the liver, and CYP7A is one of its target genes. The transcriptional activation by these receptors upon binding to the promoters located at the 5-flanking region of these GYP genes generally leads to the induction of their mRNA gene expression. The physiological and the pharmacological implications of common partner of RXR for CAR, PXR, PPAR, LXR and FXR receptors largely remain unknown and are under intense investigations. For the phase II DMEs, phase II gene inducers such as the phenolic compounds butylated hydroxyanisol (BHA), tert-butylhydroquinone (tBHQ), green tea polyphenol (GTP), (-)-epigallocatechin-3-gallate (EGCG) and the isothiocyanates (PEITC, sul­foraphane) generally appear to be electrophiles. They generally possess electrophilic-medi­ated stress response, resulting in the activation of bZIP transcription factors Nrf2 which dimerizes with Mafs and binds to the antioxidant/electrophile response element (ARE/EpRE) promoter, which is located in many phase II DMEs as well as many cellular defensive enzymes such as heme oxygenase-1 (HO-1), with the subsequent induction of the expression of these genes. Phase III transporters, for example, P-glycoprotein (P-gp), multidrug resistance-associated proteins (MRPs), and organic anion transporting polypeptide 2 (OATP2) are expressed in many tissues such as the liver, intestine, kidney, and brain, and play crucial roles in drug absorption, distribution, and excretion. The orphan nuclear receptors PXR and GAR have been shown to be involved in the regulation of these transporters. Along with phase I and phase II enzyme induction, pretreatment with several kinds of inducers has been shown to alter the expression of phase III transporters, and alter the excretion of xenobiotics, which implies that phase III transporters may also be similarly regulated in a coordinated fashion, and provides an important mean to protect the body from xenobiotics insults. It appears that in general, exposure to phase I, phase II and phase III gene inducers may trigger cellular 'stress' response leading to the increase in their gene expression, which ultimately enhance the elimination and clearance of these xenobiotics and/or other 'cellular stresses' including harmful reactive intermediates such as reactive oxygen species (ROS), so that the body will remove the 'stress' expeditiously. Consequently, this homeostatic response of the body plays a central role in the protection of the body against 'environmental' insults such as those elicited by exposure to xenobiotics.

Taxonomy of the infraspecific taxa of Adonis amurensis Regel et Radde (Ranunculaceae) (복수초(미나리아재비과)의 종내분류군에 대한 분류)

  • The infraspecific taxa of Adonis amurensis Regel et Radde are currently known to include one subspecies (subsp. nanus Y. Lee), two varieties (var. ramosa Makino, and var. dissectipetalis Y. Lee) and two forms (for. argentatus Y. Lee, and for. viridescensicalyx Y. Lee). Among them, the remaining three taxa except for subsp. nanus and var. dissectipetalis are unified with A. amurensis. Therefore, we examined the morphological characteristics and geographical distribution of A. amurensis in effort to elucidate the taxonomic status of subsp. nanus and var. dissectipetalis. After the subsp. nanus samples had been transplanted from their type locality, i.e. highland at more than 1,000 m above sea level, to a lowland habitat, we measured the samples' morphological characteristics. The range of values we recorded of the plant height and floral diameter from the subsp. nanus samples were within the parameters used to characterize A. amurensis. Accordingly, subsp. nanus has been characterized as an ecophene of A. amurensis. It is considered as a synonym of A. amurensis. Among the populations of A. amurensis (sensu Y.Lee, author), those with highly dissected petals at their apices have previously been described as var. dissectipetalis. However, this characteristic has been observed together with obtuse or rounded petal apices not only in one population but also in a single individual. However, populations with these characteristics belong to A. pseudoamurensis due to the simultaneous development of leaves and flowers, branched stems, acute leaf apices, and petals that are shorter than sepals. Therefore, we suggest that A. amurensis var. dissectipetalis be unified with A. pseudoamurensis. Very useful characteristics for distinguishing A. amurensis from its relative species are anthesis, the presence or absence of branches, the development of scales into normal leaves, the relative length of the sepals and petals, and the position of the stipules. A. amurensis is geographically restricted to E. Asia, including E. Siberia, and especially on the Korean Peninsula, it is distributed throughout the northern provinces of Gyeonggi province and in Gangwon province, Gyeongbuk province (Mt. Bohyeon-san) and Jeonbuk province (Mt Jeoksang-san) along Baekdudaegan mountain.

Predicting the Goshawk's habitat area using Species Distribution Modeling: Case Study area Chungcheongbuk-do, South Korea (종분포모형을 이용한 참매의 서식지 예측 -충청북도를 대상으로-)

  • This research aims at identifying the goshawk's possible and replaceable breeding ground by using the MaxEnt prediction model which has so far been insufficiently used in Korea, and providing evidence to expand possible protection areas for the goshawk's breeding for the future. The field research identified 10 goshawk's nests, and 23 appearance points confirmed during the 3rd round of environmental research were used for analysis. 4 geomorphic, 3 environmental, 7 distance, and 9 weather factors were used as model variables. The final environmental variables were selected through non-parametric verification between appearance and non-appearance coordinates identified by random sampling. The final predictive model (MaxEnt) was structured using 10 factors related to breeding ground and 7 factors related to appearance area selected by statistics verification. According to the results of the study, the factor that affected breeding point structure model the most was temperature seasonality, followed by distance from mixforest, density-class on the forest map and relief energy. The factor that affected appearance point structure model the most was temperature seasonality, followed by distance from rivers and ponds, distance from agricultural land and gradient. The nature of the goshawk's breeding environment and habit to breed inside forests were reflected in this modeling that targets breeding points. The northern central area which is about 189.5km2(2.55 %) is expected to be suitable breeding ground. Large cities such as Cheongju and Chungju are located in the southern part of Chungcheongbuk-do whereas the northern part of Chungcheongbuk-do has evenly distributed forests and farmlands, which helps goshawks have a scope of influence and food source to breed. Appearance point modeling predicted an area of 3,071km2(41.38 %) showing a wider ranging habitat than that of the breeding point modeling due to some limitations such as limited moving observation and non-consideration of seasonal changes. When targeting the breeding points, a specific predictive area can be deduced but it is difficult to check the points of nests and it is impossible to reflect the goshawk's behavioral area. On the other hand, when targeting appearance points, a wider ranging area can be covered but it is less accurate compared to predictive breeding point since simple movements and constant use status are not reflected. However, with these results, the goshawk's habitat can be predicted with reasonable accuracy. In particular, it is necessary to apply precise predictive breeding area data based on habitat modeling results when enforcing an environmental evaluation or establishing a development plan.

Analysis of Molecular Epidemiological Properties of Staphylococcus aureus Isolates from Domestic Animals and Human Patients by PCR (Polymerase Chain Reaction을 활용한 국내 동물과 사람환자에서 분리한 Staphylococcus aureus 분리주의 분자역학적 특성분석)

  • This study was conducted to analyze the molecular epidemiological properties and to select the most efficient and reliable PCR method on 116 of Staphylococcus aureus (S. aureus) isolates from Korean cattle, black goat, pig, dog, chicken, mouse and also human clinical cases from hospital. The distribution patterns of SSG [species specific genes; coagulase (coa), protein A (spa), nuclease (nuc) and aroA (RsaI) gene] were analyzed by PCR method. Among the SSGs, the nuc-gene was found in all strains (100%) tested and followed by coa-gene (87.9%), spa-gene (91.4%) and aroA-gene (26.7%), in order. The genetic subtyping by RFLP method was performed on the coa [AluI] and aroA-gene [RsaI] PCR products. The mecA-gene PCR and PCR-RFLP techniques were chosen to detect and verify of MRSA strains. Only the human strains (12.1%) were detected the positive mecA-gene products (533 bp), which were divided into two specific bands [201 & 332 bp] by HhaI enzyme digestion. On coa-gene and spa-gene typing, coa-gene was typed with ten kinds of genotype and coa-3 type were determined as the most predominant genotype, while spa-gene was divided into eleven kinds of genotype and also spa-7 type were selected the most prevalent genotype based on their genetic variations. On the aroA and coa-gene subtyping by PCR-RFLP, aroA-gene products were discriminated with only seven types of genotype, while coa-gene products were further divided into an eleven genotype, respectively. In comparison of SID values of five PCR based typing methods, the coa-PCR-RFLP (SID0.894) was evaluated the most efficient and reliable tools and followed by coa-PCR (SID0.883) and aroA-PCR-RFLP (SID0.462), in order. In conclusion, we could determined that the coa-PCR-RFLP method was the most suitable genetic analysis tool for S. aureus and MRSA strains from domestic animals and humans.

Numerical and Experimental Study on the Coal Reaction in an Entrained Flow Gasifier (습식분류층 석탄가스화기 수치해석 및 실험적 연구)

  • The numerical modeling of a coal gasification reaction occurring in an entrained flow coal gasifier is presented in this study. The purposes of this study are to develop a reliable evaluation method of coal gasifier not only for the basic design but also further system operation optimization using a CFD(Computational Fluid Dynamics) method. The coal gasification reaction consists of a series of reaction processes such as water evaporation, coal devolatilization, heterogeneous char reactions, and coal-off gaseous reaction in two-phase, turbulent and radiation participating media. Both numerical and experimental studies are made for the 1.0 ton/day entrained flow coal gasifier installed in the Korea Institute of Energy Research (KIER). The comprehensive computer program in this study is made basically using commercial CFD program by implementing several subroutines necessary for gasification process, which include Eddy-Breakup model together with the harmonic mean approach for turbulent reaction. Further Lagrangian approach in particle trajectory is adopted with the consideration of turbulent effect caused by the non-linearity of drag force, etc. The program developed is successfully evaluated against experimental data such as profiles of temperature and gaseous species concentration together with the cold gas efficiency. Further intensive investigation has been made in terms of the size distribution of pulverized coal particle, the slurry concentration, and the design parameters of gasifier. These parameters considered in this study are compared and evaluated each other through the calculated syngas production rate and cold gas efficiency, appearing to directly affect gasification performance. Considering the complexity of entrained coal gasification, even if the results of this study looks physically reasonable and consistent in parametric study, more efforts of elaborating modeling together with the systematic evaluation against experimental data are necessary for the development of an reliable design tool using CFD method.

Taxonomic reconsideration of the Philadelphus schrenkii complex (고광나무분류군(Philadelphus schrenkii complex)의 실체에 대한 형태 고찰)

  • The Philadelphus schrenkii complex including P. schrenkii var. schrenkii, P. schrenkii var. jackii, P. schrenkii var. mandshuricus, P. tenuifolius, P. pekinensis, P. seoulensis, P. lasiogynus, and P. scaber, is distributed in China, Korea, japan, and far eastern Russia. Their taxonomic delimitations are obscure because quantitative characters, such as presence/absence and density of hair on leaf, petiole, style, disc, calyx-tube, and pedicel, are highly variable with no apparent characteristics that consistently distinguish from each other within the group. Thus, these morphological characters are reexamined using univariate analyses. P. pekinenesis is distinct from other taxa in having glabrous disk, calyx, pedicel, and leaf blade. In addition, the number of flowers in an inflorescence [(5)7-9(11) vs 5-7 in other taxa], smaller calyx tube [2.5-3 mm vs. (2.5)3-4(5.9) in other taxa], and slightly divided styles help distinguish P. pekinenesis from other related taxa in Korea. However, P. schrenkii var. schrenkii, P. schrenkii var. jackii, P. schrenkii var. mandshuricus, and P. lasiogynus show continuous variation in the pubescence of disk, calyx, pedicel, and leaf blade, and this may reflect a lack of divergence among them. Particular emphases are placed on P. tenuifolius and P. seoulensis, because of the high degree of morphological intergradation with respect to the degree of pubescence in disk, calyx, pedicel, and leaf blade and their inconsistent circumscription across various systematic treatments. P. scaber, distributed in the southern Korea, has often been treated as an independent species having exfoliated branchlets, distinct serrated leaf margin, and recurved styles. However, this current study suggests that P. scaber should be recognized as a form of P. tenuifolius because these distinguishing characters are continuous between P. tenuifolius and P. scaber. In geographical distribution, P. pekinensis is mainly distributed in the southern part of Korea, while P. tenuifoilus and P. schrenkii are commonly found all over the country.

  • Four natural Quercus stands in Kwangju, Kyonggi-Do, of which ages ranging from 32 to 38 years old, were studied to compare their growth, biomass and net production. Ten 10m×10m quadrats were set up and ten sample trees were harvested for dimension analysis in each stand. The largest mean DBH and height were shown by Q. acutissima stand, and followed by Q. variabilis stand, Q. mongolica stand, and Q. dentata stand in descending order. Tree density was the highest at Q. variabilis stand, and followed by Q. dentata stand, Q. mongolica stand, and Q. acutissima stand in descending order. Biomass was the largest at Q. acutissima stand(122.73t/ha), and followed by Q. variabilis stand(87.03t/ha), Q. mongolica stand(72.14t/ha), and Q. dentata stand(38.56t/ha) in descending order. Net production was the greatest at Q. mongolica stand(7.49t/ha/yr.), and followed by Q. variabilis stand(6.47t/ha/yr.), Q. acutissima stand(6.06t/ha/yr.), and Q. dentata stand(3.52t/ha/yr.) in descending order. The highest net assimilation ratio was exhibited by Q. acutissima stand (3.275), and followed by Q. variabilis stand(2.898), Q. mongolica stand(2.888), and Q. dentata stand (1.840) in descending order. The difference in net assimilation ratio and net production among four stands was caused by differences in their leaf biomass. The difference in net production and biomass among four stands was due to that in the distribution of net production among stems, branches and leaves.
