Development of Herbicide Resistant Plant Through Plant Tissue Culture (제초제(除草劑) Butachlor 및 Simetryne에 저항성(抵抗性)인 식물체(植物體) 선발육성(選拔育成))

  • This study was conducted to select and develop herbicide resistant plant through tissue culture. Growth response of seedlings and callis of various rice varieties with Echinochloa species was assessed under the treatment of various rates of butachlor [N-(butoxy methyl)-2-chloro-2', 6'-diethyl acetamide] and simetryne [2,4-bis(ethyl amino)-6-methyl thio-1,3,5-triazine]. Further, succinate dehydrogenase activity was determined in herbicide treated callus to characterize different response of plants to herbicide. Rice variety like Sangpung showed relative resistance in both callus and seedling states against butachlor, indicating maintenance of resistance. However, in the simetryne treatment, the similar response was not observed in callus and seedling state, although there was a great different response among plant materials against simetryne. Rice variety which exhibited resistance in callus and seedling states showed low succinate dehydrogenase inhibition index. Succinate dehydrogenase inhibition index can be used as an important marker characters to differentiate varietal response of plant to herbicide. Rice plant was differentiated from butachlor and simetryne tolerant callus treated at $2.5{\times}10^{-5}$ M is growing under the growth chamber and can be used for resistant source.

Weed Emergence and It's Competition in the Differently Cropped Paddy Fields in Southern Districts (남부(南部)논에서의 수도작기변동(水稻作期變動)에 따른 잡초발생(雜草發生) 및 경합특성(競合特性)에 연구(硏究))

  • To estimate the fundamental situation of weed emergence and it's competition to paddy rice (critical competition period and threshold level) in the southern paddy area in Korea, two rice cultivars (Pungsanbyo and Tongjinbyo) under two cropping periods (Mono- and After-barley-cropping) were experimented. As a result, the dominant weed species (Monochoria vaginalis and Scirpus juncoides Roxb.) and the better emerging conditions (rather at the monocropping and cv. Punsanbyo) were identified. Also, the main components of yield decrease (25-55% of the weed-free check) of both cultivars as affected by weed competition were the number of spikelets, panicle and ripening rate in monocropping, and the ripening rate and grain weight in after-barley-cropping, respectively. Under the situation of combinated dominance with Monochoria v. and Scirpus j. among others, the duration of critical competition periods was enlarged.

Antagonistic Mode of Action of Fenoxaprop-P-ethyl Phytotoxicity with Bentazon (Fenoxaprop-P-ethyl의 제초활성에 대한 Bentazon의 길항작용기구)

  • Antagonistic mode of action of fenoxaprop-P-ethyl [ethyl(R)2-4-{(6-chloro-2-benzoxazolyloxy) phenoxy}propionate] with bentazon was investigated with respect to absorption, translocation, metabolism, and change in target site response of fenoxaprop-P-ethyl using four-leaf stage of rice(Oryza sativa L.) and barnyardgrass [Echinochloa eras-galli (L.) P. Beauv.]. Shoots of rice and barnyardgrass was more sensitive to fenoxaprop-P-ethyl than the roots. More than 90% of fenoxaprop-P-ethyl was absorbed within 6 hours after treatment and 30% of the absorbed was acropetally and basipetally translocated at 24 hours after treatment. Fenoxaprop-P-ethyl was rapidly transformed to its acid form, fenoxaprop(2-[4-(6-chloro-2-benzoxazolyloxy)phenoxy]propionic acid), which was subsequently metabolized to polar conjugates. However, changes in absorption, translocation, and metabolism of fenoxaprop-P-ethyl by bentazon treatment were not found in both species. Background activity of acetyl-CoA carboxylase(ACCase) in rice and barnyardgrass was 26.5 and 23.2nmol/min/mg, respectively. Concentration required to inhibit fifty percent enzyme activity$(I_{50})$ in vitro was 6.5~7.4${\mu}M$ of fenoxaprop-P-ethyl and more than 500${\mu}M$ of bentazon. There were no significant differences in $I_{50}$ value between two treatments of fenoxaprop-P-ethyl alone and its bentazon mixture. However, bentazon reduced ACCase activity in vivo and inhibited electron transport in chloroplast thylakoid. Based on the results obtained, it is concluded that the antagonistic effect of bentazon occurs due not to direct effect on target site of fenoxaprop-P-ethyl, but to indirect involvement in reducing herbicidal activity of fenoxaprop-P-ethyl through physiological disturbances caused by bentazone at whole chloroplast level.

The Allelopathic Effect of Alfalfa residues on Crops and Weeds (잡초와 작물에 대한 알팔파 잔유물의 Allelopathy 효과)

  • This experiment was conducted to test the effect on germination and seedling growth of major industrial crops and weed control potential using alfalfa plant. When dried alfalfa residues were mixed into vermiculite, germination, length of shoot and root of crops, such as Perilla frutescens, Sorghum nevosum, Platycodon grandiflorum and weeds, Digitaria saguinalis, Setaria viridis, Siegesbechia pubescens, Ammaranthus lividus, and Solanum nigrum, were significantly inhibited as the dried residue concentration increased. More than 10% concentration of the dired residue caused 80% germination and growth inhibition. The fresh alfalfa exudation also inhibited the germination and seedling growth of crop, barley, rye, alfalfa, and sesame, and weeds, Echinochloa crus-galli, Siegesbechia viridis, and Portulaca oleracea. The degree of inhibition showed the different response according to the fresh exudation concentration, types of crops and weeds. Generally, as the exudation concentration increased, the germination and seedling growth of crops and weeds inhibited. The exudation of dried residue also exhibited the strong inhibition effect on germination and seedling growth of crops, alfalfa, Platycodon grandiflorum, barley, sesame, rye and weeds, D. sanguinalis, S. pubescens, S. viridis, P. oleracea, E. crus-galli, At the 10% concentrations, S. pubescens, and P. oleracea were not germinated and showed only 15% germination in the S. viridis. From this study, would conclude that alfalfa plant contained water soluble phytotoxic substances which were inhibitory to weeds and crops. This results suggest that alfalfa had some possibility to control some weed species using toxic compounds like natural herbicide.

  • The experiments were conducted in order to find out the response of algae as affected by fertilizer and salinity, and to determine the chemical control method of algae in 1991. The higher the nitrogen fertilizer level, the more the occurrence of a blue-green alga Oscillatoria concerning with soil flakes. An increase in phosphorus fertilizer level stimulated the growth of green alga Hydrodictyon, whereas an increase in nitrogen level caused its abrupt reduction due to the harmful effect at high concentration of nitrogen, The high level of nitrogen fertilizer also reduced the growth of Euglena, although the detrimental effect was less compared with that of Hydrodictyon. A blue-green alga Oscillatoria, a green alga Cladophora and a diatom Navicula were tolerant to salinity. Piperophos/dimethametryn was highly effective in controlling various algae such as a green alga Hydrodictyon, suspended unicellular green algae and Euglena species. A fungicide propineb and copper sulfate also effectively controlled Hydrodictyon.

Herbicidal Activity of Natural Compound Chrysophanic Acid Under the Greenhouse Condition (온실조건에서 천연물질 Chrysophanic Acid의 제초활성)

  • A series of experiments was conducted to investigate the herbicidal activity of natural compound chrysophanic acid under the greenhouse condition in 16 weed species. Chrysophanic acid showed non-selective herbicidal activity. While chrysophanic acid exhibited severe injury by foliar treatment, little or no injury was found by the soil treatment. Among the tested weeds, the most effective activity was found in grass and broad leaf weeds, a lower significant difference in herbicidal activity was found in sedge. At early post-emergence, weeds appeared to be very susceptible to chrysophanic acid with $2,000{\mu}g\;mL^{-1}$. The higher the natural compound concentrations, the lower weed growth. At middle post-emergence, weeds appeared to be very effective to chrysophanic acid with $30,000{\mu}g\;mL^{-1}$.

Fact-finding Survey on Occurrence of Paddy Field Weeds and The Use of Paddy Field Herbicides at Farmer's Level in Korea (논잡초 발생양상 및 논 제초제 사용 실태조사)

  • A fact-finding survey was conducted to obtain the basic information on nation-wide weed distribution and farmer's opinions to weed control strategy in paddy field. Most respondents considered Echinochloa spp., Monochloa vaginalis, Sagittaria trifolia, and Scirpus juncoides etc. as dominate weeds in paddy fields. And herbicide resistant weed species were M. vaginalis, S. juncoides, E. oryzicola and S. pygmaea etc. Purchase guidances for herbicide selection, experience of farmers was 34.0% and dealers recommendation was 33.9% at farmer's level. And paddy field herbicides application frequence were one time 15.6% but twice 75.6%. Preferred herbicides were ranked in the following order: oxadizone 12% EC, butachlor 5% GR, butachlor 33% CS, mefenacet pyrazodulfuron-ethyl 21.42% SC, and oxadiargyl 1.7% EC etc. in paddy fields. And in most preferred foliar herbicide were bentazone MCPA 38.6% SL and bentazone cyhalop-butyl 18.5% ME, 48.5% and 22.3%, respectively.

Rate of Ground Cover and Weed Occurrence in Pepper Cultivation as Affected by Seeding of Ground Covers (고추재배 시 피복작물에 의한 피복율 변화와 잡초발생 양상)

  • This study was conducted to evaluate the effects of seeding of Vicia tetrasperma and V. hirsuta on the rate of ground cover as well as growth and yield of organic pepper plants in 2008 and 2009. All seeds of both species were gathered from Dangjin, Chungnam in late June of 2006 and stored for two year at room temperature. Capsella bursapastoris, Echinoloa crus-galli, Digitaria ciliaris, and Commelina communiswas were observed as main weeds during the winter and summer, respectively, at the field of V. tetrasperma and V. hirsuta. C. bursapastoris did not affect growth and yield of pepper plants. Seeding of V. tetrasperma and V. hirsuta maintained high percentage of ground cover later in the growing season of pepper plants and reduced significant amounts of weed in 2009. V. tetrasperma and V. hirsuta, therefore, produced more pepper yield compared to that of control but similar yield to the conventional system (polyethylene film).

Weed Occurrence in Organic Apple Orchards by Different Cover Crops (유기재배 사과과원의 피복작물별 잡초발생)

  • This study was conducted to select the cover crops in organic apple orchard by estimating the coverage of each cover crops and time-periodic weed occurrence. Seeding of Festuca myuros and Trifolium repens showed high contents of soil organic matter and phosphorus as compared with control. Seeding of Trifolium sp., Hordeum vulgare and Secale cereal reduced relatively occurrence of weed in May and S. cereal among others showed the greatest effect in June. In July, seeding F. arundinaceae and Poa pratensis showed low coverage and a lot of weeds occurred in their field. Fifty one species were occurred as weeds and it needs to removal of high and broadleaf weeds such as Chenopodium ficifolium, C. album var. centrorubru and Humulus japonicus in spring to increase the rate of ground cover. P. pratensis having the lowest cover rate showed the similar dry weight of weeds as compared with control. Trifolium sp. and Persicaria hydropiper had a high correlation with contents of soil organic matter and C. album var. centrorubru had a correlation with contents of soil phosphorus by CCA analysis.

Mesotrione and Seeding Rate Effects to Recover Kentucky Bluegrass Contaminated by Creeping Bentgrass (Creeping bentgrass에 침입된 Kentucky bluegrass 회복을 위한 Mesotrione과 Kentucky bluegrass 덧파종 효과)

  • Creeping bentgrass (Agrostis stolonifera L.) is one of the highest maintained turfgrass but often problematic especially for Kentucky bluegrass fairway. Mesotrione is one of selective herbicide that is firstly registered for corn (Zea mays L.) and provides preemergence and postemergence control of broadleaf and annual grassy weeds. Although mesotrione is effective to eradicate area contaminated by creeping bentgrass, protracted time is required to recover damaged area by rhizome extension of Kentucky bluegrass. Overseeding is typically used to fill bare or damaged areas using appropriate turf species to create a uniform turfgrass surface. The objectives of this study were to evaluate mesotrione and seeding rate effects to recover Kentucky bluegrass contaminated by creeping bentgrass. Six treatments consisted of three mesotrione rates and two Kentucky bluegrass seeding rates. The mesotrione rate were 0, 0.05 and 0.10 m $ml^{-2}$. Two seeding rate of to Kentucky bluegrass 'Midnight' were 15 and 30 g $m^{-2}$. Mesotrione application and Kentucky bluegrass overseeding at the same time is helpful to damage creeping bentgrass but not for establishment of Kentucky bluegrass to refill damaged area. To maximize mesotrione effects, temperature above $20^{\circ}C$ would be recommended based on this study.
