Effect of Soil Amendment for Controlling Fusarium Wilt of Sesame Caused by Fusarium oxysporum f. sp. vasinfectum (참깨 시들음병(Fusarium oxysporum f. sp. vasinfectum) 방제에 대한 토양 첨가제의 효과)

  • In order to find out formulation and effect of soil amendment on Fusarium wilt of sesame caused by Fusarium oxysporum f. sp. vasinfectum, the study was conducted during the last two years of 1992 to 1993. Among 14 chemicals (1%, w/w) added to soil including CaO individually, Al2(SO4)3, Alum, and CaO suppressed mycelial growth and conidial germination of F.oxysporum f. sp. vasinfectum. CaCl2 suppressed mycelial growth only, while glycerine, KCl, K2 HPO4, and triple superphosphate suppressed conidial germination. Suppression rate was ranged from 21 to 100% on mycelial growth. The 8 chemicals were finally selected. Among the 4 organic compounds, composted pine bark showed definite suppression on mycelial growth and conidial germination of the fungus, whereas milled alfalfa leaves was only effective on conidial germination of Fusarium wilt pathogen. The antagonist Trichoderma harzianum grew well in the soil medium amended with the composted pine bark and chemicals mixture (CPM) amendment (1%, w/w) and suppressed mycelial growth of the fungus effectively. In pot test, Fusarium wilt of sesame was completely controlled by CPM amendment.

Purification and Properties of Cyclodextrin Glucanotrnsferase Synthesizing $2-O-{\alpha}-D-Glucopyranosyl{\;}_{L}-Ascorbic$ Acid from Paenibacillus sp. JB-13

  • A Gram-positive bacterium (strain JB-13) that was isolated from soil as a producer of cyclodextrin glucanotransferase (CGTase) [EC 2.4.1.19] was identified as Panibacillus sp. JB-13. This CGTase could catalyze the transglucosylation reaction from soluble starch to L-ascorbic acid (AA). A main product formed by this enzyme with ${\alpha}-glucosidase$ was identified as $2-O-{\alpha}-D-glucopyranosyl{\;}_{L}-ascorbic$ acid (AA-2G) by the HPLC profile and the elemental analysis. CGTase was purified to homogeneity using ammonium sulfate fractionation, ion-exchange chromatography on DEAE-Seohadex A-50, and gel chromatography on Sephacryl S-200HR. The molecular weight was determined to be 66,000 by both gel chromatography and SDS-PAGE. The isoelectric point of the purified enzyme was 5.3. The optimum pH and temperature was PH 7.0 and $45^{\circ}C$ respectively. The enzyme was stable in the range of pH 6-9 and at temperatures of $75{\circ}C$ or less in the presence of 15 mM ${CaCl_2}.\;{Hg^2+},\;{Mn^+2},{Ag^+},\;and\;{Cu^2+}$ all strongly inhibited the enzyme's activity.

Optimization of Production of Pigment from Monascus sp. in Liquid Culture (액체배양에 의한 홍국색소 생산의 최적배양조건)

  • The optimal conditions for Monascus pigments production of Monascus sp. KM 1001, pigment overproducing mutant, in submerged culture was investigated. The optimal medium for the production of pigment from KM 1001 mutant is determined to be composed of 4% rice powder, 0.15% Bacto-peptone, 0.1% glycine, 0.01% $FeSO_{4}{\cdot}7H_{2}O,\;0.1%\;MgSO_{4}{\cdot}7H_{2}O,\;0.25%\;KH_{2}PO_{4},\;pH4.5$. On optimal conditions,10.0 g/L of the cell mass was obtained at $30^{\circ}C$ for 5 days. Yellow, orange and red pigment of Monascus sp. KM 1001 were produced 3.25 units, 1.59 units and 0.88 units in extracellular part, and 84.96 units, 78.84 units and 91.80 units in intracellular part, respectively.

Isolation and Optimal Producing Conditions of Broad Spectrum Antibiotics from Streptomyces sp. Y-88 (광범위 항생물질을 생산하는 Streptomyces sp. Y-88의 분리 및 생산 최적 조건)

  • In order to isolate antibiotic producing microorganisms, several actinomycetes were isolated from soil samples. The aerial hyphae of Y-88 strain were gray in color with tree types. Under the microscopic examination, the Y-88 isolate formed a spiral aerial spore mass with a smooth surface and a rectiflexibilis type of spore chain. Y-88 utilized glucose, fructose, arabinose, and sucrose, but not rhamnose, raffinose, mannitol, or inositol. In addition, Y-88 produced melanin on the tyrosine agar and the strain could utilize L-valine, L-phenylalanine, and L-hydroxyproline. Based on these results and the cultural and physiological characteristics described in the Bergey's Manual, the Actinomycetes, Y-88, was identificated as a Streptomyces species. The optimum medium conditions for this antibiotic producing Streptomyces sp. Y-88 was 1.6% soluble starch, 0.6% glucose, 0.6% beef extract, 0.01% $K_2HPO_4$, 0.01% $MgSO_4$ $7H_2O$, and 0.01% $ZnSO_4$ $7H_2O$ at an initial pH of 4.0, and 25$^{\circ}C$.

Rheological Properties of Citrus Pectin Solutions (감귤류 펙틴 용액의 리올리지 특성)

  • The steady shear and small amplitude oscillatory dynamic rheological properties of citrus pectin $([\eta]=3.75\;dL/g)$ were characterized for a wide range of pectin concentrations $({\sim}6%)$. The typical power-law flow was observed above 2.0% concentration, and the shear rate dependence of viscosity increased with pectin concentration. The transition from dilute to concentrated regime, determined from the double logarithmic plot of ${\eta_{sp.o}}\;vs\;C[\eta]$, occurred at a critical coil overlap parameter $C^{*}[\eta]\approx4.0$, at which ${\eta_{sp.o}}$ corresponded to approximately 10.0. The slopes of ${\eta_{sp.o}}\;vs\;C[\eta]$, at $C[\eta]\;at\;C[\eta]C^{*}[\eta]$were 1.1 and 4.5, respectively. The steady viscosity $(\eta)$ displayed a good superposition at ${\eta}/{\eta}_o\;vs\;{\gamma}/{\gamma}_{0.8}$ relation with an exception of high concentration (6%), which arised from the significant deviation of flow behavior index (n values of $\eta_{a}=K\gamma^{n-1}$) at high concentration. Dynamic measurements showed that the loss modulus $(G^{\prime\prime})$ was much higher than the storage modulus $(G^\prime)$for all concentrations studied, indicating predominant viscoelastic liquid-like behavior of pectin solutions. The frequency dependence of $G^\prime$ was higher than that of $G^\prime\prime$ at the same concentration, whose trend was more pronounced with decreasing pectin concentration. The shear viscosity $(\eta)$ was almost identical to the complex viscosity $(\eta^{*})$ at low concentration, following the Cox-Merz rule, but they became increasingly different at high concentration.

  • A bacterial strain producing intracellular phytase was isolated from cultivable soil near cowsheds and identified as a fluorescent Pseudomonas sp. BUN1. The BUN1 phytase, partially purified by cation and anion exchange chromatography, exhibited its optimal activity at $40^{\circ}C$ and pH 5.5. As for substrate specificity, it was very specific for phytate and showed little activity on other phosphorylated conjugates. Its activity was greatly inhibited by metal ions such as $Cu^{2+}$, $Cd^{2+}$, and $Zn^{2+}$. Addition of corn starch to PSM (phytasesynthetic medium) [0.5% sodium phytate, 0.5% $(NH_4)_2SO_4$, 0.5% KCl, 0.01% $MgSO_4\cdot7H_2O$, 0.01% $CaCl_2\cdot2H_2O$, 0.01% NaCl, 0.001% $FeSO_4\cdot7H_2O$, 0.001% $MnSO_4\cdot4H_2O$; pH 6.5] for the phytase production significantly induced its enzyme activity in comparison with other carbon sources tested.

Effects of Abdominal Breathing Practice on Oxygen Saturation and Pulserate for Insomnia in Middle-aged Women (복식호흡 수련이 중년여성의 불면증 산소포화도와 맥박에 미치는 영향)

  • Background: Studies on the effect of abdominal or thoracic breathing therapy on sleep or blood oxygen concentration are still scarce. Purpose: This study was to examine the effect on blood oxygen saturation and pulse variability, changes in the severity of insomnia, changes in wakefulness before sleep, and dysfunctional beliefs and attitudes toward the Korean version of sleep in women in their 50s after healing with abdominal breathing and thoracic breathing. We investigated the effect. Methods: Subjects were investigated before and after the change of breathing (breathing) therapy for 12 weeks, 3 times a week, and 36 breaths perweek. Results: It wa evaluated respiratory healing as having no significance in the pulse rate change. However, oxygen saturation was significant in the experimental group, increasing to 93.60 SpO2% before the respiratory rally and 96.5 SpO2% after respiratory recovery (p < .002). In addition, the insomnia severity scale and dysfunctional beliefs about sleep significantly decreased after respiratory rally than before (p < .000). Conclusions: It evaluated that respiratory therapy for the subjects is beneficial to health as it is effective for insomnia, pulse, and oxygen saturation.

Separation and Identification of Galactosylmanno-oligosaccharides from Hydrolyzate of Brown Copra Meal by Trichoderma β-Mannanase

  • Three kinds of oligosaccharides were obtained from the hydrolysate of brown copra meal galactomannan by a purified extracellular ${\beta}$-mannanase from Trichoderma sp. These oligosaccharides were identified as Man-Man, ${Gal^2}{Man_3}(6^2 mono-O-{\alpha}-D-galactopyranosyl-4-O-{\beta}-D-mannotriose)$, and ${Gal^2}{Man_6}(6^2-mono-O-{\alpha}-D-galactopyranosyl-4-O-{\beta}-D-mannohexaose)$, where Gal- and Man-represent ${\alpha}$-1,6-D-galactosidic and ${\beta}$-1,4-mannosidic linkages, respectively. The mode of action of ${\beta}$-mannanase on brown copra meal galactomannan is described on the basis of the structure of these oligosaccharides.

Isolation and Characterization of Cholesterol Degradation Bacteria from Korea Traditional Salt Fermented Flat Fish (가자미 식해로부터 콜레스테를 분해세균의 분리 및 특성)

  • In order to develop the Production and application of cholesterol oxidase, a cholesterol degradation bacteria which produces a remarkable amount of extracellular cholesterol oxidase has been isolated from Korea traditional salt fermented flat fish. The isolated strain was identified as a strain of Bacil1us sp. based on its morphological, physiological characteristics and cellular fatty acid compositions. Experiments were carried out to optimize the condition of cholesterol oxidase production using Bacillus sp. SFF34. Bacillus sp. SFF34 was shown to give the maximum yield of cholesterol oxidase in the medium containing 2.0% glucose, 0.5% yeast extract, 0.02% MgSO$_4$$.$7H$_2$O, 0.025% K$_2$HPO$_4$, 0.15% NH$_4$NO$_3$ and 0.2% cholesterol. The optimum culture conditions, temperature, initial pH and agitation speed were 30$^{\circ}C$, 7.0 and 150rpm respectively. The enzyme production reached a maximum level at 24 hrs of cultivation(2.42 U/ml).

Growth Characteristics and Optimal Culture Conditions of Bacterial Strains Degrading Ethylene Glycol and Terephthalic Acid in Polyester Weight Loss Wastewater (Polyester 감량폐수 중에 존재하는 Ethylene Glycol과 Terephthalic Acid를 분해하는 Bacteria 균주들의 성장특성과 최적 배양조건)

  • Strains degrading ethylene glycol(EG) and terephthalic acid(TPA) were isolated from water systems, and identified as Pseudomonas sp. They were named as Pseudomonas sp. EAW for EG and as Pseudomonas sp. TS2 for TPA. The optimal culture conditions of temperature, pH and nitrogen source were found to be $35^{\circ}C$, 7.5 and ammonium sulfate, respectively. The growth of strains and removal efficiency was slightly promoted by trace elements such as niacin and biotin in case of EG, and by trace elements such as $Na_2MoO_4{\cdot}2H_2O$ and thiamin i case of TPA. With increasing inoculation sloe for batch culture, the removal efficiency of EG by the strain EAW was conspicuously increased, while the removal efficiency of TPA by the strain TS2 was not changed as much as that of EG. The growth rate of the strain EAW was much more decreased than that of the strain TS2 in the enrichment medium, as the frequency of repeated-batch culture in the rich-medium increased. in case of real wastewater, growth rate and removal efficiencies of EG and TPA were lower than those in the enrichment medium. $COD_{Mn}\;and\;COD_{Cr}$ removal efficiencies after 48 hrs batch culture in real wastewater were 89% and 93%, respectively. The specific growth rate was inhibited when the initial concentration of EG or TPA was more than 25g/L.
