Taxonomic Studies of Cellulose Decomposing Fungi Imperfecti (섬유소 분해능을 가진 불완전 균류의 분류)

  • Twenty-one strains isolated, cellulose decomposing fungi, were identified on the basis of morphological, physiological and biochemical properties as Acremonium sp., Aspergillus sp., Chaetomium sp., Chrysonilla sp., Doratomyces sp., Fusarium sp., Gliomastix sp., Penicillium sp., Trichoderma sp., Varicosporium sp. and Verticillium sp.. The optimum tempeture for growth was in the range of $20-30^{\circ}C$. Most of the isolated stains utilized all tested carbon sources, and scarcely utilized urea as a nitrogen source. Only the strain No.2 had high activity of cellulase.

An Action Mechanism of Substance P on the Tracheal Smooth Muscle Contraction in Rabbits (토끼 기관의 평활근 수축에 미치는 substance P 의 작용기전)

  • Substance P[SP] has been known to be a peptide which may be plays a role as a neurotransmitter in central nervous system as well as peripheral autonomic nervous system. It has been reported that SP was widely distributed in the nerve of the tracheal smooth muscle and induced the muscle contraction. However, definite action mechanism of SP in the tracheal smooth muscle was not clear, yet. Thus, present experiment was performed to elucidate an effect of substance P and an action mechanism on contraction of the smooth muscle in rabbits. In order to find a neural mechanism to the effect of SP on the tracheal smooth muscle contraction, atropine sulfate, tetrodotoxin, propranol and phentolamine were administered at 10 min before the addition of SP. Otherwise,to find effect of SP antagonists on the action of SP, [D-Pro2, D-Try7,9]SP, [D-Arg1, D-Pro2, D-Trp7,9, Leu11]SP and [D-Pro4, D-Trp7,9]SP were administered as a same fashion. These following results were obtained. 1] SP induced contraction of the tracheal smooth muscle under resting condition and the contraction was increased dose-dependently. 2] Cholinergic blocker[atropine], neural blocker[tetrodotoxin] and adrenergic blocker[propranol and phentolamine] didn`t have an effect on the contractile response. 3] Three SP antagonists inhibited the contractile response. 4] Isoproterenol relaxed the contraction induced by SP. The above results suggested that SP induced contraction of the tracheal smooth muscle directly act to the smooth muscle in rabbits. The autonomic nervous system did not seem to participate in the SP action.

Construction of Schizosaccharomyces pombe spThp1 Null Mutants and its Characterization (분열효모 Schizosaccharomyces pombe에서 spThp1 유전자 결실돌연변이의 제조와 특성 조사)

  • The sp%pl null mutant was constructed to study the function of fission yeast Schizosaccharomyces pombe spThp1, which is homologous to budding yeast Saccharomyces cerevisiae THP1. Tetrad analysis showed that the spThp1 is not essential for vegetative growth. The spThp1 null mutant also showed no massive poly(A)+ RNA export defect. However, spThp1 null is genetically associated with spMex67 null. These results suggest that spThp1 is involved in mRNA export out of the nucleus.

  • A new limnephilid caddisfly, Nothopsyche bilobata n. sp., is described. The female adult of Apatania martima Ivanov and Levanidova and the larva of Ecclisomyia kamtshatica (Martynov) is newly recorded from Korea. Three undetermined species, Nemotaulius sp. A, Nemotaulius sp. B, and Nothopsyche sp. A, are additionally described.

A Mycological Survey in Indoor Environments (건물(建物) 환경(環境)의 진균(眞菌) 분포(分布) 조사(調査))

  • This paper was concerned with the investigation on the species of fungi detected in indoor environments. Mycological survey on the wall of buildings and houses in Seoul was performed from December 26, 1983 to February 11, 1984. The collected fungi with the sterilized cotton rod were caltivated and isolated in the Potato-Detrose Agar medium with chloramphenicol. The fungi detected in hospital were Cladosporium sp., Penicillium sp., Alternaria sp., Chaetomium sp. and others. The fungi detected in household were Penicillium sp., Alternaria sp., Aspergillus sp., Cladosporium sp., Rhizopus sp., Trichoderma sp. and others. The kitchen of household was more contaminated by fungi than the living room and others. In hospital, it was much contaminated by the same species of the aboves on the wall of lobby and refectory. As the above results were recognized, the higher humidity and the more poor ventilation, the more species and amount of fungi were contaminated. Further studies on the identification of pathogenic fungi should be continued. The proper regulation for the hygienic maintenance of indoor environment in household, hospital and other public buildings should be recommended as well.

Characterization of PAH (Polycyclic Aromatic Hydrocarbon)-Degrading Bacteria Isolated from Commercial Gasoline (상용 휘발유로부터 분리한 다환 방향족 탄화수소(PAH) 분해 세균의 특성)

  • BACKGROUND: Recent studies have described the importance of bacteria that can degrade polycyclic aromatic hydrocarbons (PAHs). Here we screened bacterial isolates from commercial gasoline for PAH degraders and characterized their ability to degrade PAHs, lipids and proteins as well as their enantioselective epoxide hydrolase activity, salt tolerance, and seawater survival. METHODS AND RESULTS: One hundred two bacteria isolates from commercial gasoline were screened for PAH degraders by adding selected PAHs on to the surface of agar plates by the sublimation method. A clear zone was found only around the colonies of PAH degraders, which accounted for 13 isolates. These were identified as belonging to Bacillus sp., Brevibacterium sp., Micrococcus sp., Corynebacterium sp., Arthrobacter sp., and Gordonia sp. based on 16S rRNA sequences. Six isolates belonging to Corynebacterium sp., 3 of Micrococcus sp., Arthrobacter sp. S49, and Gordonia sp. H37 were lipid degraders. Arthrobacter sp. S49 was the only isolate showing high proteolytic activity. Among the PAH-degrading bacteria, Arthrobacter sp. S49, Brevibacterium sp. S47, Corynebacterium sp. SK20, and Gordonia sp. H37 showed enantioselective epoxide hydrolase activity with biocatalytic resolution of racemic styrene oxide. Among these, highest enantioselective hydrolysis activity was seen in Gordonia sp. H37. An intrinsic resistance to kanamycin was observed in most of the isolates and Corynebacterium sp. SK20 showed resistance to additional antibiotics such as tetracycline, ampicillin, and penicillin. CONCLUSION: Of the 13 PAH-degraders isolated from commercial gasoline, Arthrobacter sp. S49 showed the highest lipid and protein degrading activity along with high active epoxide hydrolase activity, which was the highest in Gordonia sp. H37. Our results suggest that bacteria from commercial gasoline may have the potential to degrade PAHs, lipids, and proteins, and may possess enantioselective epoxide hydrolase activity, high salt tolerance, and growth potential in seawater.

Species Identification of Tripitaka Koreana (팔만대장경판(八萬大藏經板)의 수종(樹種))

  • Tripitaka Koreana was made during Koryo Dynasty from 1236 to 1251 A.D. Buddhist scriptures were engraved on 81.340 wooden plates. This study was made to identify species in these archaeological wooden plates using light and scanning electron microscopes. The results were as follows more than 62% of whole specimens investigated was Prunus sp., which was used in wooden plate 64% and wooden plate edge 56%, Pyrus sp. was used 13% of whole specimens and 31% of wooden plates. Therefore, 75% of whole Tripitaka Koreana was made by these two species. Acer sp., Betula sp., Machilus sp., Cornus sp., Daphnilhyllum sp., Prunus sp. were also identified but extreamly rare. Especially Machilus sp. and Daphnilhyllum sp. originally distributed in subtropical zone of west and south coast in Korea were identified, suggested strongly another engraving place of Tripitaka Koreana, Namhae bunsadogam.

Selection and Bacterialization into Rootzone of the Various Plant Growth Promoting Rhizobacteria in Peatmoss Compost on the Early Growth of Cucumber and Tomato Plug Seedlings (수종의 식물생장촉진 근권미생물의 분리 및 근권처리가 오이와 토마토 플러그묘의 초기생장에 미치는 영향)

  • Azospirillum sp., photosynthetic bacteria(Rhodopseudomonas sp.) and Pseudomonas sp. were separated and screened from soil and soilless culture, and identificated. The antifungal activities against root-infected pathogens and plant growth promoting effects of the cultured solution of the starins(5.0$\times$105 cells/$m\ell$) in the peatmoss compost on the early growth of cucumber and tomato seedling were investigated. Azospirillum sp. and Pseudomonas sp. showed a antifungal activities against Fusarium sp., Pythium sp. and Rhizoctonia sp in thed ranges of 51.0% to 72.0% on potato dextrose agar medium, however photosynthetic bacteria had not antifungal activities. When cultured solution of Azospirillum sp., photosynthetic bacterial and Pseudomonas sp. were bacterialized by mixing with peatmoss compost, early growth of cucumber and tomato in terms of plant height, number of leaves, leaf area, root length, fresh anf dry weight of leaf, stem and root were promoted, especially photosynthetic bacteria had a the best plant growth promting activities.

Six New Agelas Species (Demospongiae: Agelasida: Agelasidae) from Kosrae Island, The Federated States of Micronesia

  • This paper describes six new species of sponges in the genus Agelas from Kosrae Island, The Federated States of Micronesia. Most Agelasid sponges are known from only tropical regions. All the new Agelas species; A. fragum n. sp., A. kosrae n. sp., A. purpurea n. sp., A. bakusi n. sp., A. vansoesti n. sp. and A. incrustans n. sp. are compared with other valid species that were studied. Six new species differ from the other species by morphology, growth form, skeletal fibres, habitats and spicule size. Agelas fragum n. sp. is characterized by its tuberculate surface and primary fibres with brush-like spicules. Agelas kosrae n. sp. is differs in skeletal structure and have tertiary fibres. Agelas purpurea n. sp. is characterized by primary, secondary and tertiary fibres are all cored with spicules. Agelas bakusi n. sp. is similar to Agelas clathrodes in shape, but differs in the primary fibres. Agelas vansoesti n. sp. is characterized by having acanthostrongyles. Agelas incrustans n. sp. is distinguished by its encrusting and not cavernous interior.

Using $CuSO_4$ for Preventing Algae Attachment on the Sedimentation Basin of Industrial Water Treatment Plant (공업용수 생산 정수장의 침전지 조류 부착방지를 위한 황산동 투입)

  • Existing states of attached algae in the sedimentation basin was observed during late april to early october, and the attached algae were visible 7 days after of cleaning the trough and the communities of algae became larger with increasing the operation periods. Attached algae community included bluegreen algae (Oscillatoria sp.), diatom (Synedra sp.,) and green algae (Mougeotia sp., Oedogonium sp.) and suspended diatom (Stephanodiscus sp.) as well. Diatom (Cymbella sp., Navicula sp., Synedra sp. and Stephanodiscus sp.), green algae(Mougeotia sp. and Cosmarium sp.) and blue-green algae (Anabaena sp.) were detected in the effluent of sedimentation basin. The chlorophyll-a (chl-a)concentrations of algae community on a square centimeter after 14 and 28 days were distinctively different depended on the copper sulphate treatment. The concentration of chl-a were $4{\mu}g/L/cm^2$ and $19{\mu}g/L/cm^2$ for the copper sulphate treated water and $59{\mu}g/L/cm^2$ and $147{\mu}g/L/cm^2$ for the untreated water. Diatom algae fragments were observed in red-brownish sediments on the bottom of industrial water distribution basin and degraded blue-green and green algae formated organic sediments combined with oxidized iron.
