A Study on Wine of Yi Dynasty in 1600 (조선시대(朝鮮時代) 술에 관한 분석적(分析的) 고찰(考察) -조선중기(朝鮮中期) 1600 년대(年代)를 중심(中心)으로-)

  • As people know how to brew a wine from fruits and cereals, they continued to develope various wines good to their taste. Korean wines are also ones made from cereals and they have long been eager to improve the delicate taste. They used to drink Takju, raw rice wine, made from nonglutinous rice and Nuruk, a kind of yeast starter. During Koryo Dynasty, Soju a liquor was imported from Won(the Chinese dynasty). Nowadays this traditional folk wine, which had been developed variously and drunk all over the country, is decreasing year after year. The purpose of this study was to review on the wines ; its kinds, raw materials, brewing method, manufacturing utensils, measuring units and devices and the terms for wine making based on 20 documents published in 1600, in the middle of Yi dynesty. The results of review were as follows. 1. There were 121 kinds of wines at that time in Korea. 2. Among the raw materials for wines, major materials were glutinous rice, nonglutinous rice, wheat flour, wheat, mung bean, and black soybean. And minor materials were pepper corn, Lycium chinenisis, cinnamon, pine needles, pine nuts, jujube, mugwort leaves, lotus leaves, pine corn, pine bud, chrysanthemum, pine flowers, honey, Acanthopanox seoultenses, bamboo-root, marrowbone of blak cow, sweet flag, Ciprus noblis, Saurea lappa, honey suckle, Tricho santhes, azalea, the leaves of the paper mulberry, and bark of chungum tree. 3. There were several kinds of wines such as a wine without using Nuruk, a wine made from glutinous rice, nonglutinous rice, or glutinous and nonglutinous rice with flour. 4. There were several brewing methods for wines such as a wine boiled with ring rice cake, a wine brewed with loaves of rice cake, a wine brewed with hard boiled rice, a wine brewed with rice gruel, and a wine brewed with powdered rice gruel. 5. There were 23 kinds of utensils including measuring devices for weight and volume.

The Changes of Side Dishes in "Eumsikdimibang", "Gyuhapchongseo", "Chosunm usangsinsikyorijebub" according to the Current of the Time ("음식디미방","규합총서(閨閤叢書)","조선무쌍신식요이제법(朝鮮無雙新式料理製法)"에 수록된 시대적 흐름에 따른 부식류의 변화)

  • This study examined the changes of side dishes in "Eumsikdimibang", "Gyuhapchongseo", "Chosunmusangsinsikyorijebub" over time. As food ingredients, seaweed, red pepper, salted fish, Korean hot pepper paste have been used in "Gyuhapchongseo". The use of cattle by parts, saltwater fish, Chinese cabbage, powdered red pepper, garlic have increased in"Chosunmusangsinsikyorijebub". Cooking techniques, such as the use of wheat flour, a double boiler by the use of steam, and boiling and then roasting cuisine were mostly used in"Eumsikdimibang", but reduced in"Gyuhapchongseo". In "Gyuhapchongseo", the cooking methods are primarily aimed at a positive visual effect, and slices of dried meat seasoned with spices have increased. In"Chosunmusangsinsikyorijebub", various Tang (Guk), the taste of food changed by controlling the gravy content (Gigimi, Chigye, Chorim), meat mixing oil, vegetable and mushroom together to cook and boiling down the main food ingredients to soak the seasoning were increased. Dog-meat steamed dish using the intestine of dog in "Eumsikdimibang" was changed to small intestine of cattle steamed dish in "Gyuhapchongseo". And seasoned dog meat with choncho in "Eumsikdimibang" influenced on beef tail soup with Korean hot pepper paste in "Gyuhapchongseo", and Yookgyejang soup using Korean hot pepper paste in"Chosunmusangsinsikyorijebub". In steamed young chicken, the stuffing such as soybean paste, choncho, welsh onion, leek and flour in "Eumsikdimibang" was changed to minced beef, welsh onion, dropwort and Shiitake mushroom in"Gyuhapchongseo". The steamed young chicken in "Chosunmusangsinsikyorijebub" with stuffing was added to chicken soup using glutinous rice, and ginseng powder. Now, the chicken soup was changed to Samgyetang with glutinous rice, and ginseng. In "Chosunmusangsinsikyorijebub", various vegetable dishes were cooked with beef.

  • This study was to investigate the effect of 5~20% soybean flout(SBF) and/or 0.5~5% calcium on the quality of bread. pH of bread was increased with increasing an amount of SBF and calcium. pH of bread was higher than that of dough. The scores of specific volume and sensory evaluation for the bread quality were shown higher and increased its relative volume in the group of added both of 5~15%, SBF and 0.5~2%, calcium Endothermic peak for a thermal property of bread was increased with increasing the storage time of bread. The bread staling was progressed rapidly in control and calcium added groups only. with increasing an amount of SBF, L values of bread was decreased, but a and b values were increased. However, L, a and b values were shown no difference in the group of calcium added only. Therefore, the optimum blending ratios of SBF and calcium for the quality of bread were 5~10%, SBF and 0.5~1% calcium, respectively.

Bioconversion of Ginsenosides from Red Ginseng Extract Using Candida allociferrii JNO301 Isolated from Meju

  • Red ginseng (Panax ginseng), a Korean traditional medicinal plant, contains a variety of ginsenosides as major functional components. It is necessary to remove sugar moieties from the major ginsenosides, which have a lower absorption rate into the intestine, to obtain the aglycone form. To screen for microorganisms showing bioconversion activity for ginsenosides from red ginseng, 50 yeast strains were isolated from Korean traditional meju (a starter culture made with soybean and wheat flour for the fermentation of soybean paste). Twenty strains in which a black zone formed around the colony on esculin-yeast malt agar plates were screened first, and among them 5 strains having high ${\beta}$-glucosidase activity on p-nitrophenyl-${\beta}$-D-glucopyranoside as a substrate were then selected. Strain JNO301 was finally chosen as a bioconverting strain in this study on the basis of its high bioconversion activity for red ginseng extract as determined by thin-layer chromatography (TLC) analysis. The selected bioconversion strain was identified as Candida allociferrii JNO301 based on the nucleotide sequence analysis of the 18S rRNA gene. The optimum temperature and pH for the cell growth were $20{\sim}30^{\circ}C$ and pH 5~8, respectively. TLC analysis confirmed that C. allociferrii JNO301 converted ginsenoside Rb1 into Rd and then into F2, Rb2 into compound O, Rc into compound Mc1, and Rf into Rh1. Quantitative analysis using high-performance liquid chromatography showed that bioconversion of red ginseng extract resulted in an increase of 2.73, 3.32, 33.87, 16, and 5.48 fold in the concentration of Rd, F2, compound O, compound Mc1, and Rh1, respectively.

Studies on the Quality of Kochujang Prepared with Grain and Flour of Glutinous Rice (찹쌀과 찹쌀가루가 고추장의 품질에 미치는 영향)

  • Three types of Kochujang(soybean-red pepper paste) were prepared with glutinous rice flour (group A), glutinous rice grain (group B) or the mixture of flour and grain (group C) to investigate some chemical composition, enzyme activities and the texture during aging period. 'There were no remarkable differences in the contents of moisture, crude protein, crude fat, minerals and alcohols, titrable acidity and pH among the tested groups. Amino nitrogen content, acidic protease and saccharogenic amylase activities were measured to be higher values for group A whereas reducing sugar content higher values for group B. All tested groups contained Ca, Cu, K, Mn, Mg, P and Fe, particularly higher amount of K but a trace of Cu, Mn and Fe. All tested groups showed higher content of aspartic acid, glutamic acid, cystine, proline and lower content of histidine and valine 60 days after the preparation. The total free amino acid content was 16.49 mg/g for group A, 13.68 mg/g for group B and 15.84 mg/g for group C. The measurement of texture showed high values of hardness and adhesiveness with group A immediately after the preparation and with group B at the end of aging. Cohesiveness and springiness were high with group A while low with group B throughout aging period.

Distillers' Dried Grain as a Replacement for Plant-derived Sources in the Diet of Juvenile Muddy Loach Misgurnus anguillicaudatus (배합사료 내 식물성 원료 대체원으로 막걸리 부산물이 미꾸리(Misgurnus anguillicaudatus) 치어의 성장 및 체조성에 미치는 영향)

  • We designed a 16-week feeding experiment to test the effects of dietary supplementation with distillers' dried grain (DDG) on the growth, feed utilization, and body composition of juvenile muddy loach Misgurnus anguillicaudatus. Seven isonitrogenous and isocaloric diets were formulated to contain 0% DDG (Control); 10%, 20%, and 30 % DDG from rice (diets RM10, RM20 and RM30); and 10%, 20%, and 30% DDG from rice and wheat flour (diets RWM10, RWM20 and RWM30), respectively. Three replicate groups of juvenile muddy loach averaging $1.5{\pm}0.10$ g were fed one of the diets to satiation twice daily. Survival of juvenile muddy loach fed the RWM20 and RWM30 diets was higher than that of the control group (P<0.05), while weight gain tended to decrease when fish were fed diets containing DDG. Weight gain of fish fed the RWM10 diet was greater than that of fish fed other diets, but the RM30 diet resulted in less weight gain than did the control (P<0.05). The feed and protein efficiency ratios of fish fed the RM10, RM20, RM30, RWM10, RWM20 and RWM30 diets were all similar to the control (P>0.05). Neither daily feed intake nor proximate composition of the whole body was affected by dietary DDG (P>0.05). The compositions of isoleucine, leucine, lysine, threonine, and valine were all elevated in fish fed the RM30, RWM10, RWM20 and RWM30 diets relative to those of other groups (P<0.05). In contrast, methionine + cystine and phenylalanine + tyrosine were all lower in fish fed the RM30, RWM10, RWM20 and RWM30 diets, as compared to those of other groups (P<0.05). Our results suggest that DDG is suitable as a partial replacement for soybean meal and wheat flour, and could be used at a rate of up to 20% for rice, or 30% for rice and wheat flour, for optimum growth performance of juvenile muddy loach.

Effect of Oligosaccharides on Retrogradation of Sulgidduk (올리고당을 첨가한 설기떡의 노화지연에 관한 연구)

  • The effects of four different oligosaccharides with 2, 4, 6% (w/w) (fructo-oligosaccharide, xylo-oligosaccharide, chito-oligosaccharide and soybean-oligosaccharide) on gelatinization and retrogradation of sulgidduks (Korean rice cake) were examined. The amylograph results of rice flour showed that chito-oligosaccharide hastened gelatinization, and delayed retrogradation. Blue value results of chito-oligosaccharide added sulgidduks showed retarded retrogradation during storage (1, 2 and 3 days). Chitooligosaccharide and xylo-oligosaccharide added sulgidduks showed significantly lower hardness during storage. Lightness (L) decreased and redness (a) and yellowness (b) increased with increasing oligosaccharide amounts. In the sensory evaluation of sulgidduks, color of fructo-oligosaccharide added sulgidduks obtained the highest score among oligosaccharide added sulgidduks. During storage, xylo-oligosaccharide and fructo-oligosaccharide added sulgidduks had higher flavor, taste, graininess and overall quality scores than the control. Physicochemical tests showed that chito-oligosaccharide retarded retrogradation, whereas chitooligosaccharide- added sulgidduks had low scores in sensory tests due to aftertaste of chito-oligosaccharide. To improve the sensory quality of chito-oligosaccharide added sulgidduks, mixtures of chito-oligosaccharide with xylo-oligosaccharide and fructooligosaccharide were applied at ratios of 3%:3%, 2%:4% and 1%:5%, respectively. The addition of chito-oligosaccharide and xylo-oligosaccharide at ratios of 2%:4% and 1%:5% to sulgidduks showed relatively high scores in the sensory evaluation retarding retrogradation.

Fed-batch cultivation for cell growth and spore production by probiotic B. polyfermenticus SCD

  • The optimal temperature, pH and aeration rate for spore production by Bacillus polyfermenticus SCD in 500 ml shake flask and 5-1 jar fermenter were found to be $32^{\circ}C$, 7.0 and 1.0 vvm. respectively. When batch culture processes was performed under optimized culture conditions. viable cells were $3.3{\times}10^{10}$ CFU/ml and spore cells were $3.3{\times}10^{10}$ CFU/ml. Fed-batch culture processes were also examined with regard to higer maximum viable cell and spore production. The highe viable cells and spores were obtained in 5-1 jar fermenter at 72 h cultivation time by strategy in an intermediate feeding mode with 60% glucose solution 150 ml and 5% soybean flour solution 150 ml fed to the fermenter twice, and the productivity of spore cells was significantly increased. Finally. volumetric productivity of spore cells on fed-batch culture indicated $9.9{\times}10^8$ CFU/ml/h, which was approximately 2 times higher than batch culture. Thus, fed-batch culture show a promise as an industrial production method.

  • Selection of optima] nutrient sources and cultural methods for liquid spawn culture of Flammulina velutipes were carried out. The optimal temperature and pH for mycelial growth of F. velutipes were $20^{\circ}C$ and 6.0 to 7.5. respectively. In the 250ml ${Delta}$-flask culture. the amount of inoculum and culture period for the optimal mycelial growth of F. velutipes were 3 mycelial disks(diametcr 6mm) and 6 days, respectively. For the mass production of submerged culture. the optimal inoculum amount and aeration rate of F. velutipes were 5%(inoculum vol/medium vol.) and l.0vvm(vol of air/vol. of medium/min), respectively.

Silkworm (Bombyx mori) Response to Differently Formulated Artificial Diets

  • Artificial diet (AD) has shown with different advantages over mulberry leaves (ML). Various types of such feed for silkworm have been developed and proposed. The aims of this study were possibility rearing whole instar of silkworm on AD and to find some economic formulation for rearing silkworm, compare to that ML. Eleven ADs (D$_1$ to D$_{11}$) were prepared with different percentage of mulberry leaf powder, and other ingredients such as soybean meal, cellulose, potato starch, agar, wheat flour, rice bran, etc. Five formulated diets (D$_1$ D$_2$, D$_{5}$, D$_{10}$ and D$_{11}$) appeared to have similar response to that ML (D$_{m}$). There were no significant differences for duration period between ADs, all of which recorded with higher duration than mulberry leaves. Compared to other died used in the present study D$_1$ and D$_{10}$ showed better growth development and survival rate. Larvae fed D$_1$, D$_2$, D$_{5}$, D$_{10}$ and D$_{11}$ grew faster and produced heavier and more useful cocoons.coons.coons.coons.
