Comparative Studies on Methods of Extracting Polyacetylene Compounds from White Ginseng (인삼 Polyacetylene 성분의 추출방법 비교연구)

  • The amounts of panaxynol and panaxydol, which are major polyacetylone compounds In white ginseng were determined by capillary-GC (FID), and the extraction efficiencies when using varictus extraction solvents (petroleum ether, dichloromethane, ether, ethyl acetate, acetone, acetonitrilr and methanol) and various extraction methods (shaking, Soxhlet and reflux) were compared . The GC column was SPB-1 fused silica capillary (0.25 mm id x30 m, Supelco), and the column oven temperature was programmed to rise from $200^{\circ}C$ to $300^{\circ}C$ at the rate of $4^{\circ}C$ per minute. The extraction efficiencies for panaxynol and panaxydol according to extraction souvtints were the highest in methanol and decreased in the order of dichloromethane, acetone, ether, ethyl acetate, acetonitrile and petroleum ether. The extracttion efficiencies for panaxynol and panaxydol according to extraction methods were the highest for reflux and the lowest for shaking, and those with Soxhlet were almost equal to those for reflux. The analytical amounts of panaxynol and panaxydol obtained by reflux with methanol %mere 4.2 and 6.4 mg/g, respectittely in white ginseng.

  • Two extraction methods for total petroleum hydrocarbon (TPH) from contaminated soil were evaluated. The soil used for this study was sandy loam. Diesel oil was selected as representative petroleum hydrocarbons and was spiked at 100, 10,000, 50,000mg TPH/kg dry soil. Percentage recovery of TPH by shaking method was higher compared to Soxhlet extraction. At extraction time of 2 hours and sample to solvent ratio of 1 : 5, the highest percentage recovery was obtained. In this condition, percentage recovery of TPH in soil contaminated with 100mg/kg and 50,000mg/kg as TPH was 95.9% and 95.5%, respectively The volume of solvent lost by volatilization in shaking method was relatively small compared to Soxhlet extraction.

Measurement of Lipid Content of Compost in the fermentation Process using Near-Infrared Spectroscopy

  • Near infrared spectroscopy (NIRS) was applied to determination of the lipid content of compost during compost fermentation of tofu(soybean-curd) refuse. The reflected rays in the wavelength range between 800 and 2500 nm were measured at 2 nm intervals. The absorption of lipid observed at 4 wavelengths, 1208, 1712, 2312 and 2352 nm on the second derivative spectra. To formulate a calibration equation, a multiple linear regression analysis was carried out between the near-infrared spectral data and on the lipid content in the calibration sample set (sample number, n=60) obtained using a Soxhlet extraction method. The calibration equation for prediction of lipid, the value of the multiple correlation coefficient (R) was 0.975 when using the wavelengths of 1208 and 1712nm. To validate the calibration equation obtained, the lipid content in the validation sample set (n=35) not used for formulating the calibration equation were calculated using the calibration equations, and compared with the values obtained using the Soxhlet extraction method. Good agreement were observed between the results of the Soxhlet extraction method and those values of the NIRS method. The simple correlation coefficient (r) and standard error of prediction (SEP) were 0.964 and 0.815 %, respectively. Then, the NIRS method was applied to a compost fermentation in which the time course the lipid content were measured and good results were obtained. The study indicates that NIRS is a useful method for process management of the compost fermentation of tofu refuse.

  • A simultaneous and rapid gas chromatographic determination of organophosphorus, organo-chlorine, carbamate, and pyrethroid pesticides in polished rice was developed. The pesticides were extracted from samples with acetone by automated soxhlet apparatus and this extract was evaporated to dryness. The residue was dissolved in hexane, the solvent was applied to a Sep-Pak florisil catridge, was eluted with 50% ethyl acetate in n-hexane, and was injected to dual GC-ECD/NPD system. This simple method affords a high recovery of hydrophilic pesticides, allows rapid analysis, and is cheap to perform. Except for dichlorvos and captan, recoveries of 46 pesticides were over 60%.

  • An analytical method for simultaneous and rapid determination of organophosphorus, organochlorine, carbamate, and pyrethroid pesticides in polished rice was developed. The analysis is performed by gas chromatograph with mass selective detector (GC/MSD) in selected ion monitoring (SIM) mode. Pesticides were extracted from samples with acetone by automated Soxhlet apparatus and this extract was evaporated to dryness for the analysis. The residue was dissolved in hexane, followed by a treatment with a Sep-Pak florisil catridge. Pesticides were positively confirmed by GC/MS, retention times, and ion ratios. This analytical method allows a rapid, reliable, and a good recovery of hydrophilic pesticides. Except far captafol, captan, dichlofluanid and dichlorvos, recoveries of 42 pesticides were over 70%.

Analytical Method for Dioxin and Organo-Chlorinated Compounds: (Ⅱ) Comparison and Extraction Methods of Dioxins from XAD-2 Adsorbent

  • Supercritical fluid extraction (SFE), ultrasonic extraction (USE), and accelerated solvent extraction (ASE) were compared with the well known Soxhlet extraction for the extraction of polychlorinated biphenyls (PCBs) and polychlorinated dibenzo-p-dioxins(PCDDs) from the XAD-2 resin which was used to adsorb PCDDs in the atmosphere. XAD-2 resin spiked with five PCDDs was chosen as a sample. The optimum conditions for the extraction of PCDDs by SFE were turned out to be the use of CO2 modified with 10% toluene at 100 ℃ and 350 atm, with 5 min static extraction followed by 20 min dynamic extraction. SFE gave a good extraction rate with good reproducibility for PCDDs ranging from 68 to 98%. The ultrasonic extraction of PCDDs from XAD-2 was investigated and compared with other extractions. A probe type method was compared with a bath type. Two extraction solvents, toluene and acetone were compared with their mixture. The use of their mixture in probe type, with 9 minutes of extraction time, was found to be the optimum condition. The average recovery of the five PCDDs for USE was 82-93%. Accelerated solvent extraction (ASE) with a liquid solvent, a new technique for sample preparation, was performed under elevated temperatures and pressures. The effect of tem-perature on the efficiency of ASE was investigated. The extraction time for a 10 g sample was less than 15 min, when the organic solvent was n-hexaneacetone mixture (1 : 1, v/v). Using ASE, the average recoveries of five PCDDs ranged from 90 to 103%. SFE, USE, and ASE were faster and less laborious than Soxhlet extraction. The former three methods required less solvent than Soxhlet extraction. SFE required no concentration of the solvent extracts. SFE and ASE failed to perform simultaneous parallel extractions because of instrumental limitations.

Optimization of the whole extract of Zarawand Mudaharaj (Aristolochia rotunda L.) root by Response Surface Methodology (RSM)

  • The chemical constitution of a drug has been accepted as an important basis for pharmacological action in Unani medicine. Various dosage forms have been developed on this concept, such as decoctions (Joshanda), infusions (Khesanda), extract (Rub / Usara), and syrup. Zarawand Mudaharaj (ZM.) / Aristolochia rotunda L. root was subjected to extraction process using Soxhlet's apparatus by using Response Surface Methodology (RSM) to design the number of random runs of the extracts with variation in the factors of temperature, the concentration of ethanol in water, time for extraction, for optimizing and maximizing the yield concentration. The data obtained, was analyzed with regression equation and ANOVA two-way summary to interpret the interaction of the factors for yield maximization. Minitab version 18 was used to design and analyze data. Validation of the optimum conditions for maximum yield of the whole extract of ZM. Root was carried out by re-run of the extract using the optimized conditions. The maximum yield percentage thus obtained using RSM was 20.87% whereas using these optimum conditions 21.35 % yield was obtained thereby validating the method. The association between the response functions and the process variables was identified by a three-factor recorded Box-Behnken design. In the present study RSM is used because itis a cheap and affordable method to optimize maximum yield percentage which may be reliably used by researchers. The study set in the surface conditions for ZM. root extraction by the Soxhlet apparatus for maximizing the yield percentage.

Evaluation of analytical method for polybrominated diphenyl ethers (PBDEs) in manufactured products waste (제품폐기물 중 폴리브롬화에테르류 (PBDEs) 분석방법 비교 연구)

  • In this study, soxhlet extraction and dissolution-precipitation method are used for separating analyte from polymer materials. In soxhlet extraction efficiency test, it has been found that the DCM, Toluene, THF, and mixtures Acetone/Hexane (1:4, v/v) gave good extraction efficiency, while the use of the ethyl ether, acetone/ethyl ether (1:4, v/v), acetone/hexane (1:1, v/v), DCM/hexane (1:1, v/v) resulted in significantly lower values. In case of dissolution-precipitation method, there is no considerable difference with used different dissolving solvent. The elution amount of multi layer silica and Florisil column were determined with hexane 250 mL and 70 mL of hexane, respectively. Range of PBDE in real waste plastics was N.D.~1,028 ppm.

A Study on the Extraction of Monasil PCA using Liquid CO2 (액체 이산화탄소 이용한 Monasil PCA 추출에 대한 연구)

  • Poly(acrylic acid) (PAA) microspheres is one of the widely-used polymeric materials for the bio-field application and the electric materials. For the synthesis of PAA microspheres, the polymerization technique using surfactants is applied. After the synthesis, the purification and separation processes are required for the removal of surfactant. When general organic solvents were used, many problems, such as huge amount of waste solvent, additional separation processes, and the possibility of residual media, were occurred. Thus, High-pressure Soxhlet extraction using liquid $CO_2$ was developed to solve these problems. In this study, High-pressure Soxhlet extraction of the synthesized PAA microspheres using liquid $CO_2$ was conducted for the removal of Monasil PCA which is used for the dispersion polymerization of acrylic acid in compressed liquid Dimethyl ether (DME). The morphology of the extracted PAA particles was checked by field emission scanning electron microscopy (FE-SEM) and the residual concentration of Monasil PCA was analyzed by inductively coupled plasma - Optical Emission Spectrometer (ICP-OES). For studying the effect of the solvent effect, Soxhlet extraction was conducted using n-hexane, liquid DME, and liquid $CO_2$. In case of n-hexane, some extracted PAA microspheres were produced. However, deformation was also occurred due to the high thermal energy of n-hexane vapor. Liquid DME could not remove Monasil PCA. When using liquid $CO_2$, the extracted PAA microspheres which were free for the residual solvent were produced without deformation. For finding the optimum operating condition, high-pressure Soxhlet extraction was conducted for 8 hours with changing the temperature of reboiler and condenser. When the extractor temperature is $19.6{\pm}0.2^{\circ}C$ and the pressure is $51.5{\pm}0.5$ bar, the best removal efficiency was obtained.
