  • In an effort to produce high quality Angelica gigas in the southern area of Korea, a variety of mulching materials were used to prevent withering during the summer. The experimental results are summarized as follows. Soil water content and temperature under various mulching materials were higher than those without mulching, in all the cases studied. Among various mulching materials P. E.(polyethylene) mulching material showed the highest effect. The appearance of weeds was the lowest when using the black P. E. mulching, while growth of weeds was the greatest when using transparent mulching. Growth and development of the plant were better with P. E. mulching than straw mulching or no mulching, allowed for more and bigger leaves. The bolting rate was about 2∼5%, with a slightly larger value under P. E. mulching. Under P. E. mulching, probably due to good physical properties of the soil the number of roots was large, and the length and diameter were good. Active growth of roots under P. E. mulching resulted in a yield increase of the plant. The black P. E. mulching material showed a prominent effect on suppression of weed growth, and the black P. E. material was considered to be the most recommendable among various P. E. mulching materials when there is a lack of labor.

  • In an effort to produce high quality Curcuma aromatica Salisbury in the southern area of Korea, a variety of shade-method was used to prevent withering during the summer. The experimental results are summarized as follows. The effect of shading on emergence of Curcuma aromatica Salisbury rhizome was none or very little. The effect of shading on the growth and development of Curcuma aromatica Salisbury was the highest under a shade rate of 15∼30%. In this case the size of the plant height was the biggest and leaves the largest, thus showing the best condition for plant growth. The growth of rhizome under a shade rate of 15% was more active than that without shading. The weight of rhizome in this case was the largest, and thus showing the best condition for rhizome growth.

  • Lettuce (Lactuca sativa) was transformed with Agrobacterium tumefacience LBA4404 containing human granulocyte macrophage colony stimulating factor (hGM-CSF) gene to produce in cell suspension cultures. Cell suspension culture was established using callus from transgenic lettuce plant. Integration of hGM-CSF gene into plant chromosome was confirmed through genomic PCR and Southern blot analysis. In addition, Northern blot analysis indicated the expression of the introduced hGM-CSF gene in transformed lettuce. The recombinant hGM-CSF was expressed in transgenic cell cultures derived from transgenic plants as a yield of about 149.0 $\mu\textrm{g}$/L in culture filtrate, which was determined by ELISA. These results demonstrated that transformed lettuce cell suspension cultures could be used as a production system of therapeutic proteins such as hGM-CSF.

  • The Specific Plant Species such as N degree (2 species) and V degree (5 species) which is categorized by the Ministry of Environment are appeared as 7 species. II degree species among Endangered species of wild flora is Abeliophyllum distichum, and the Rare and Endangered Species which is categorized by the Korea Forest Service are enlisted as the five taxa. The Bukhansan is one of the phytogeographic ally important areas among national parks of Korea because this region is located in the northern limit of Southern Plant Types, such as Carpinus laxiflora, Cornus kousa, Styrax japonica and Zanthoxylum piperitum. The flora of the area was enlisted as 638 taxa, 103 families, 362 genera, 541 species, 87 varieties, and 10 forms. Naturalized Species was identified as 58 taxa, 16 families, 52 genera, 54 species and 4 varieties. The Ratio of Naturalized and Urbanization Index was 9.09% and 20.21%, respectively.

  • In order to obtain basic informations for selecting early maturing rice varieties which is suitable for early cropping before Alisma plantago in the southern part of Korea. Eleven rice varieties were grown from May to September in 1999∼2001 at Sunchon Youngieon Experiment Field and yield components and yield of plants were investingated. Early maturing rice cv. Jinbubyeo showed higher rough rice yield than any other varieties used in the experiment. It showed high yield components, culm length, panicle length, number of panicles per plant, number of spikelets per panicle and ratio of ripened grains. therefore, it was concluded that Jinbubyeo was the most suitable variety with high yield for the cultivation before Alisma plantago at the southern part of Korea. The heritability of culm length number of spikelets per panicle and rough rice yield were high and that of panicle length number of panicle per plant, ratio of ripened grain and 1,000 grain wt. of milled rice were low. The rough rice yield showed highly significant positive correlations with culm length, panicle length, number of spikelets per panicles and ratio of ripened grains.

  • Since there is no report about more than one gene expression simultaneously in a single mitochondria, this report is very important to novel type of eukaryotic gene expression. In this study, investigated whether mitochondrial expressed gene and GFP that expression in mitochondria of plant expressed to mitochondria. Expression vector (pBin) containing AtBI-1 (mitochondrial expressed gene) and GFP driven by 35S promoter was bombarded by gene gun to leaves and cotyledon of tobacco. Regenerated shoot confirmed expression of AtBI-1 in mitochondria by GFP expression, PCR, and Southern analysis. Successful mitochondria of plant cell transformation in this report implies possible eukaryotic mitochondrial transformation including plants and animals, and moreover two or more gene expression which can be excellent applicable protocols to pharmaceutical field including antibody production.

  • The transport and allocation of photosynthetic assimilate is an important regulatory factor in plant productivity, In order to modify assimilate partitioning in rice, transgenic plants containing a potato sucrose transporter (SuT) gene were developed. Calli derived from rice seeds (Oryza sativa L. cv Dongjin) were cocultured with A. tumefaciens LBA 4404 harboring the SuT gene. Calli were transferred to MS medium supplemented with 50 mg/L hygromycin, 500 mg/L carbenicillin, 2 mg/L kinetin, 0.1 mg/L NAA. After 2 weeks, hygromycin resistant shoots were obtained from the calli on the selection medium. Roots were induced from the putative transgenic shoots on rooting MS medium supplemented with 250 mg/L cabenicillin. Plant regeneration rate from the calli was about 150%. Stable incorporation of the potato SuT gene into rice genomic DNA was confirmed by PCR and Southern blot analysis.

  • The argE gene of E.coli was introduced into #Ilmibyeo# cultivar of rice by Agrobacterium tumefaciens and a large number of transgenic plants were produced. Embryogenic calli were co-cultivated with A. tumefaciens strain AGL1 carrying the plasmid pCAMBIA1300 containing hygromycin resistance(HygR). Transgenic plants showing in vitro resistance to 50mg/L hygromycin were obtained using a selection procedure. Stable integration of argE and HPT genes into chromosomal DNA was proven by southern blot analysis and PCR analysis of genomic isolated from $T_0$ progenies. The fragments of 650 bp(HPT) were detected in transgenic rice lines. The 230 bp(argE) fragments were showed in agarose gel, and detected fragments were matched with size of argE specific primer. The microscopic feature of leaf on scanning electron microscope(SEM) revealed differences between clear and chalky in shape and arrangement of stoma but did not discriminate.
