  • As a continuation of the previous studies, a third group of thirty five hot aqueous extracts from natural products were screened for platelet activating factor(PAF) receptor binding antagonistic acitivities using rabbit platelet. The results demonstrate that Arctium lappa is potential source of PAF antagonist.

  • Lampteromyces japonicus is the most popular source of mushroom poisoning in Japan. We isolated poisonous substance, illudin S, 40 years ago, which shower strong anti-tumoric activity. Its analogs are now the most hopeful anti-cancer agent. Clitocybe acromelalgia gives unique poisoning which exhibits symptoms similar to acromelalgia and erythromelalgia. We intended to isolate the toxin causing these poisoning but neuroexciatory compounds, acromelic acids A and B, were isolated. They are now known as the most potent glutamate agonists and good reagents for the study of the glutamate receptor of a neurocell.

  • There are a large number or chemical compounds that are present in a polluted atmosphere and that alone or in combination are important to consider for their potential effect on the respiratory system and impact on athletic performance. A general categorization or description of the level of pollution in terms of the concentration of one or more compounds or by type such as oxidizing compounds is inadequate and misleading. A useful initial categorization of pollutant compounds according to their mechanism of production, primary or secondary, is often made. For health effects, consideraiions of the physical state, gaseous or particulate, and the solublity and reactivity of the pollutant is also important. Pollutant compounds or substances that are emitted directly from a source and that undergo little or no chemical change in the atmosphere from source to receptor are termed primary pollutants.

  • Simple mass balance method was developed to estimate the contribution of two major fugitive dust sources in the Port of Incheon to a nearby residential area in this study. Using the relatively small number of TSP data as well as the data on mass fraction of Fe and organic materials in the sampled dust, our simplified method demonstrated its ability to estimate the contribution of each fugitive source to a specific location including the residential area with relatively reasonable accuracy. It is clear from this simple method can be applied to the situation where two major fugitive dust sources are responsible for the high TSP concentration around the source area and there are clear marker chemicals representing the characteristics of the fugitive dust sources.

Individual Particle Analysis for Developing a Source Profile of Yellow Sands (황사의 오염원분류포 개발을 위한 개별입자분석)

  • To quantitatively estimate mass contribution of long-range transported yellow sand, their sources should be separated independently from various local soil sources having similar elemental compositions. While it is difficult to estimate total mass loadings of pure yellow sand by traditional bulk analysis, it can be clearly solved by an particle-by-particle analysis. To perform this study, two yellow sand samples and three local soil samples were collected by a mini-volume sampler. These samples were three analyzed using a scanning electron microscope(SEM) equipped with an energy dispersive x-ray analyser (EDX) was used to obtain basic chemical information of individual yellow san particles. A total of 19 elements in a single particle were measured to develop a source profile with newly created homogeneous particle classes (HPCs) as chemical variables. The present study showed that the yellow sand samples as well as three local soil samples were characterized with reasonably well created HPCs. Finally the mass fraction of each HPC in each sample was calculated and then compared each other.

  • The purpose of the study was to identify sources of particulate matters statistically and to estimate the mass contribution quantitatively in the Pusan metropolitan area. Then, the study has used the TTFA (target transformation factor analysis) model, a receptor model, to apportion aerosol mass with the raw data of 106 ambient samples characterized by 24 heavy metal variables. The TTFA was extensively applied to generate source profiles and their aerosol mass contributions. Though a couple of sources were not identified, four to seven sources were able to be extracted at 3 different sites (Jang Rim-Dong, Kwang Bok-Dong, and Kwang An-Dong) in Pusan area and finally mass conributions could be calculated.

  • One of the better-characterized transcription factor of E. coli is the cAMP receptor protein(CRP) and the CRP binds cAMP and DNA. The cAMP-CRP complex is involved in regulation of many genes at bacteria. The cAMP-CRP regulatory element represents, in some respects, a global regulatory network. The aim of this work was to study the structure and the mechanisms controlling the expression of CRP in Serratia marcescens. We have been get 5 different clones from Serratia which stimulated the cells to use maltose as a sole carbon source in E. coli TP2139. The crp gene clone, pCKB12, was confirmed by Southern hybridization with E. coli crp gene. The location of the crp gene was determined by construction subclones carrying various portions of pCKB12. To investigate the potential role of CRP in E. coli, lacZ fused plasmids were constructed and investigated the ${\beta}$-galactosidase activity of the fused plasmid. The Serratiamarcescens cAMP receptor protein can substitute the E. coli CRP in transcriptional activation at the lacZ gene. These results suggest that Serratia marcescens cAMP receptor protein complex functions to regulate several promoters in E. coli.

Expressions of Pituitary Adenylate Cyclase-Activating Polypeptide and Its Receptor Gene in the Rat Uterus (흰쥐 자궁에서 Pituitary Adenylate Cyclase-Activating Polypeptide와 수용체 유전자의 발현)

  • The present study was performed to analyze the gene expressions of pituitary adenylate cyclase-activating polypeptide(PACAP) and its receptor in the rat uterus, a candidate for novel extrahypothalamic source and target. The PACAP cDNA fragments corresponding to the common exon region which is found in both the rat hypothalamus and testis were produced from all tissue samples including the rat uterus by reverse transcriptionpolymerase chain reaction (RT-PCR). No PCR product was amplified from the rat hypothalamic, pituitary, ovarian and uterine samples when the 5' primer corresponding to the testis-specific exon 1 region was used, while the predicted size of product was detected from the testis sample. RT-PCR using the uterine RNA and specific primers for the PACAP receptor yielded products with predicted sizes. Transcripts for the rat uterine PACAP receptor were identified as type I isoforms with hip-hop and hip- or hop-type inserts. After pregnant mare's serum gonadotropin (15 IU) treatment of immature rats (day 25), the level of PACAP mRNA was increased in 24 h and 48 h group, and was declined to the lowest in 72 h group. The present study shows the presence of transcripts for PACAP and its receptor isoform in the rat uterus. These finding ssuggest that the uterine PACAP ight act as a novel autocrine and/or paracrine factor via its specific receptors on the reglulation of rat uterine function and physiology during the reproductive cycle.

  • Objective: We hypothesized that Cr source can alter adipogenic-related transcriptional regulations and cell signaling. Therefore, the objective of the study was to evaluate the biological effects of chromium acetate (CrAc) on bovine intramuscular (IM) and subcutaneous (SC) adipose cells. Methods: Bovine preadipocytes isolated from two different adipose tissue depots; IM and SC were used to evaluate the effect of CrAc treatment during differentiation on adipogenic gene expression. Adipocytes were incubated with various doses of CrAc: 0 (differentiation media only, control), 0.1, 1, and 10 μM. Cells were harvested and then analyzed by real-time quantitative polymerase chain reaction in order to measure the quantity of adenosine monophosphate-activated protein kinase-α (AMPK-α), CCAAT enhancer binding protein-β (C/EBPβ), G protein-coupled receptor 41 (GPR41), GPR43, peroxisome proliferator-activated receptor-γ (PPARγ), and stearoyl CoA desaturase (SCD) mRNA relative to ribosomal protein subunit 9 (RPS9). The ratio of phosphorylated-AMPK (pAMPK) to AMPK was determined using a western blot technique in order to determine changing concentration. Results: The high dose (10 μM) of CrAc increased C/EBPβ, in both IM (p = 0.02) and SC (p = 0.02). Expression of PPARγ was upregulated by 10 μM of CrAc in IM but not in SC. Expression of SCD was also increased in both IM and SC with 10 μM of CrAc treatment. Addition of CrAc did not alter gene expression of glucose transporter 4, GPR41, or GPR43 in both IM and SC adipocytes. Addition of CrAc, resulted in a decreased pAMPKα to AMPKα ration (p<0.01) in IM. Conclusion: These data may indicate that Cr source may influence lipid filling in IM adipocytes via inhibitory action of AMPK phosphorylation and upregulating expression of adipogenic genes.

Luminous efficiency's estimation method of transmitted light in lenses (렌즈에서 투과된 광의 시감도 추정 방법)

  • The eye sensitivity in the difference conditions of a light source intensity consists of two functions by the receptor of cone and rod according to a wavelength. We derived a distribution function of $P{\lambda}=A{\cdot}e^{-({\lambda}-{\lambda}_u)^2/2W^2}$ using a respond probability of the receptor of cone-rod for a photon. It was well applied for a CIE eye's sensitivity curve of a wavelength. When there is lens In front of eye, luminous efficiency should be corrected. Transmission light which permeate to depends on absorption wavelength, and relationship of final luminous efficiency's estimation method is expressed by multiplication of luminous efficiency and transmittance intensity of lens. $$Pf({\lambda})=T({\lambda}){\cdot}P({\lambda})$$. The theory was applied to photopic and scopic vision with brown color lens.
