Isolation of Antimicrobial Active Substances from Sophorae Radix (고삼에서 항균물질의 분리)

  • Bioassay-directed fractionation of the dired roots of Sophorae Radix led to the isolation of Sophoraflavanone G. Their structures were elucidated using 1H-NMR, 13C-NMR, UV, IR and mass spectral ananlyses. These compounds exhibited a moderate antimicrobial activities against Streptococcus gordonii, Actinobacillus actinomycetemoomitans and Porphylomonas gingivalis.

Effects of Sophorae Radix Fractionations on Cardiovascular Anaphylaxis in Pithed Rats (고삼 전탕액 분획물이 순환기 Anaphylaxis에 미치는 영향)

  • Objectives and Methods : Unlike respiratory anaphylaxis responses, mediators responsible for cardiovascular responses are not clearly elucidated. Main characteristics of cardiovascular anaphylaxis include hypotension and cardiac failure (anhythmia and cardiac contraction failure). In this experiment, the fractionations of Sophorae Radix (SR) were tested for its preventive effects against cardiovascular anaphylaxis in pithed rats. Results : Of the SR fractionations, water fractions, at the concentration of 20 and 60mg/kg, was significantly effective on all the cardiovascular changes in pithed rats. Also, of the cardiovascular changes, depressor response was significantly attenuated by the ethyl acetate (EA) fraction, at the concentration of 60mg/kg. Conclusions : These results suggest that water and EA fractions of the SR fractionations possess anti. anaphylactic effects in pithed rats. Additional research is needed to identify active principles for the observed pharmacological effects.

  • This study was carried out to investigate the effects herbal extracts on the skin inflammatory reactions. Among the herbal ingredients of herbal extracts, ethanol extract of Glycyrrhizae Radix showed potent radical scavenging activity, more than 86$\%$ at a concentration of 0.01$\%$, tested by DPPH(I,1-diphenyl-2-picryl-hyrazyl) method. And ethanol extract of Aurantii Immaturus Fructus inhibited the lipopolysaccharide-induced release of nitric oxide(NO), 99$\%$ at 50㎍/㎖, by the macrophage RAW 246.7 cells. In addition, the ethanol extracts of Sophorae Radix, Glycyrrhizae Radix showed antibacterial activities on Propionibacterium acnes that causes acne, Sophorae Radix, Glycyrrhizae Radix and Aurantii Immaturus Fructus on Corynebacterium xerosis that cause osmidrosis axillae, Glycyrrhizae Radix and Aurantii Immaturus Fructus on Staphylococcus aureus that was reported to cause impetigo and atopic disease. Taken together, I expect that herbal extracts may be used as a drug for treatment on skin inflammation.

Antimicrobial Effects of Sophorae Radix Extracts against Oral Microorganisms (구강미생물에 대한 고삼의 항균효과)

  • Objectives : Sophorae Radix (SR), the dried root of Sophorae Flavescens Aiton, has been used to treat atherosclerosis, arrhythma and skin diseases including scabies and eczema. The present study was examined to evaluate antimicrobial activities of SR extracts against oral microorganism. Methods : Antimicrobial properties of SR extracts were determined by agar diffusion method and minimum inhibitory concentration (MIC) against Streptococcus mutans, Streptococcus sobrinus and Actinomyces viscosus. Analysis of kurarinone from SR extracts was conducted using UPLC (Ultra Performance Liquid Chromatography). Results : The ethanolic extracts of SR showed stronger antimicrobial effect than methanolic extracts, while the aqueous extracts of SR had no activity. In addition, the higher content of kurarinone was found in ethanolic extracts than methanolic extracts. The purified kurarinone from ethanolic extracts showed potent antimicrobial activity with the MIC value of $3.9{\sim}7.8{\mu}g/m{\ell}$. Conclusion : An ethanolic extract of SR showed antimicrobial properties against several oral microorganisms, and kuranrinone contributed to antimicrobial action of SR. Thus, ethanolic extracts of SR or purified kurarinone should be beneficial for the preparation of the useful agent for treating oral disease including anticaries.

Effects of Sophorae Radix Extracts on the Expression ofcell cycle regulatory porteins in Human Gingival Fibroblasts (고삼추출물이 치은섬유아세포의 세포주기 조절단백질 발현에 미치는 영향)

  • Fibroblasts are major cellular components of gingiva and periodontal ligament. They regulate the healing process after surgery or injury. Recently, many natural medicines, whose advantages are less side effects and possibility of long-term use, have been studied for their capacity, their anti-bacterial and anti-inflammatory effects and regenerative potential of periodontal tissues. Sophorae radix have been traditionally used as an anti-bacterial and antiinflammatory drug in oriental medicine. The purpose of present study was to investigate the effects of Sophorae radix extract on cell cycle progression and its molecular mechanism in human gingival fibroblasts. Sophorae radix extracts($100{\mu}g/ml$) notably increased cell proliferation and cell activity in the human gingival fibroblasts as compared to non-supplemented controls. There was an increase in the S phase and a decrease in the G1 phase in $100{\mu}g/ml$ of Sophorae radix extracts group as compared to non-supplemented controls. The level of cyclin E and cdk 2 protein in test group was higher than that of control groups. But that of cyclin D, cdk 4, and cdk 6 was not distinguished from controls. The level of p53 protein in test group was lower than that of controls, whereas that of p21 was not different. The level of pRB protein in test group was higher than that of controls, whereas that of p16 was lower. These results indicate that the increase of cell proliferation by Sophorae radix extracts may be due to the increased expression of cyclin E and cdk 2, and the decreased expression of p53 and p16 in human gingival fibroblasts.

  • 청열조습(淸熱燥濕), 거풍살충(祛風殺蟲)의 효능이 있는 고삼(苦蔘)의 체내 면역활성에 미치는 영향에 대해 실험한 결과 아래와 같은 지견을 얻었다. ICR 생쥐에 고삼(苦蔘) 수침액을 투여한 결과 백혈구수 및 면역장기 변화는 없었다. in vitro실험에서 IgM PFCs에 유의성 있는 영향을 주지 못했고, BSA를 주사한 생쥐에서의 Arthus 반응에도 영향이 없었다. 그러나 지연형 과민반응을 억제하였으며, 비특이적 면역에 관여하는 macrophage의 탐식능을 약간 저하시켰다. 이상의 결과 고삼(苦蔘) 수침액(水浸液)은 세포성 면역을 억제하는 것으로 사료된다.

Effects of Several Herbs on the Blood Vessel (수종 한약재가 혈관에 미치는 영향)

  • Rhizoma Arisaematis, Lignum Akebiae, Rhizoma Zedoariae, Cortex Eucommiae, Folium Perillae, Radix Sophorae Subprostratae, Radixi, Radix Ledeboutriellae, Rhizoma Atractylodis, Herba Ephedrae, Radix Puerariae and Radi Aconitx Bupleuri have been used in Korea for many centuries as a treatment for various disease. The purpose of the present study is to determine the effect of several herbs on norepinephrine(NE) induced blood vessel contraction in rabbits and pigs. Rabbit(2 kg, male) were killed by $CO_2$ exposure and a segment (8-10mm) of each rabbit was cut into equal segments and mounted in a tissue bath. Contractile force was measured with force displacement transducers under 2-3 g loading tension. The dose of norepinephrine(NE) which evoked 50% of maximal response $(ED_{50})$ was obtained from cumulative dose response curves for NE $(10^{-6}{\sim}10^{-3}M)$. Contractions evoked by NE $(ED_{50})$ were inhibited significantly by Rhizoma Arisaematis, Lignum Akebiae, Rhizoma Zedoariae, Cortex Eucommiae, Folium Perillae, Radix Sophorae Subprostratae and Herba Ephedrae in abdominal aorta. Contractions evoked by NE $(ED_{50})$ were inhibited significantly be Lignum Akebiae, Rhizoma Zedoariae, Cortex Eucommiae, Herba Ephedrae, Radix Puerariae and Radix Bupleuri in femoral artery. Contractions evoked by NE $(ED_{50})$ were inhibited significantly by Radix Sophorae Subprostratae, Radix Aconiti and Herba Ephedrae in renal artery. These results indicate that each herb can relax NE induced contraction of rabbit and pig blood vessel selectively, and that this relaxation relates to Gui-Gyung(歸經).

The Experimental Study on Antifungal effects of mixture of Sophorae Subprostratae Radix, Acoti Radix, and Hibisci Syriaci Cortex by the Three Types of Extraction on three species of the dermatophytes. (다양한 山豆根·川烏·木槿皮 혼합물 용액이 足部白癬菌에 미치는 영향)

  • This experimental study was performed to investigate the antifungal effects of mixture of Sophorac Subprostratae Radix, Aconti Radix, and Hibisci Syriaci Cortex by the three types of extraction in the dermatophytes such as Trichophyton rubrum, Trichophyton mentagrophytes, and Epidermophyton floccosum. The following results were obtained by using Minimum Inhibition Concentration(MIC) and Inhibition Zone. 1. MIC on Trichophyton mentagrophytes in Sample S-2, Sample S-3, and Sample-Y were 10${\mu}l$, respectively, showing the highest antifungal effect. 2. MIC on Epidermophyton floccosum in Sample S-2, Sample S-3, and Sample-Y were 10㎕ respectively, showing the highest antifungal effect. 3. MIC on Trichophyton rubrum in Sample S-3, Sample-Y were 10${\mu}l$ respectively, showing the highest antifungal effect. 4. The size of inhibition zone on Trichophyton mentagrophytes for Sample S-3 were 22.3mm in 10${\mu}l$, showing the highest antifungal effect. 5. The size of inhibition zone on Epidermophyton floccosum for Sample S-3 was 34.3mm in 10${\mu}l$, showing the highest antifungal effect. 6. The size of inhibition zone on Trichophyton rubrum for Sample-Y was 26.5mm in 10${\mu}l$, showing the highest antifungal effect. Whereas. in extractions, that of Sample S-3 was 12.5mm in 10${\mu}l$, showing the highest antifungal effect. 7. From the above results, Sample S-3 on Trichophyton mentagrophytes, Epidermophyton floccosum and Sample-Y on Trichophyton rubrum showed the highest antifungal effects. This results suggests that vinegar extraction excels other extraction method using water or ethanol to measure the antifungal effect on dermatophytes. In addition, the result shows that the extract of herbal medicines can be used to tinea pedis and if further study is performed, the use of the extract of herbal medicines will be valuable and beneficial in the clinical medicines. Involvement of Transient Receptor Potential Melastatin 7 Channels in Sophorae Radix-induced Apoptosis in Cancer Cells - Sophorae Radix and TRPM7 -

  • Sophorae Radix (SR) plays a role in a number of physiologic and pharmacologic functions in many organs. Objective: The aim of this study was to clarify the potential role for transient receptor potential melastatin 7 (TRPM7) channels in SR-inhibited growth and survival of AGS and MCF-7 cells, the most common human gastric and breast adenocarcinoma cell lines. Methods: The AGS and the MCF-7 cells were treated with varying concentrations of SR. Analyses of the caspase-3 and - 9 activity, the mitochondrial depolarization and the poly (ADPribose) polymerase (PARP) cleavage were conducted to determine if AGS and MCF-7 cell death occured by apoptosis. TRPM7 channel blockers ($Gd^{3+}$ or 2-APB) and small interfering RNA (siRNA) were used in this study to confirm the role of TRPM7 channels. Furthermore, TRPM7 channels were overexpressed in human embryonic kidney (HEK) 293 cells to identify the role of TRPM7 channels in AGS and MCF-7 cell growth and survival. Results: The addition of SR to a culture medium inhibited AGS and MCF-7 cell growth and survival. Experimental results showed that the caspase-3 and -9 activity, the mitochondrial depolarization, and the degree of PARP cleavage was increased. TRPM7 channel blockade, either by $Gd^{3+}$ or 2-APB or by suppressing TRPM7 expression with small interfering RNA, blocked the SR-induced inhibition of cell growth and survival. Furthermore, TRPM7 channel overexpression in HEK 293 cells exacerbated SR-induced cell death. Conclusions: These findings indicate that SR inhibits the growth and survival of gastric and breast cancer cells due to a blockade of the TRPM7 channel activity. Therefore, TRPM7 channels may play an important role in the survival of patients with gastric and breast cancer.
