Endocrine Profiles and Blood Chemistry Patterns of Cloned Miniature Pigs in the Post-Puberty Period

  • Although the majority of surviving pigs cloned by somatic cell nuclear transfer (SCNT) appear to be physiologically normal, there is a general lack of detailed hemato-physiologic studies for the period of early adulthood to substantiate this claim. In the present study, we investigated variation in blood chemistry and endocrinological parameters between mesenchymal stem cells (MSCs) derived from cloned and normal age-matched female and male miniature pigs. Cloned females and males showed normal ranges for complete blood count assessments. Biochemical assessments showed that ${\gamma}$-GGT, ALT and cholesterol levels of male and female clones were significantly (P<0.05 or P<0.01, respectively) higher than that of age-matched control miniature pigs. Variations in insulin and IGF-1 were higher in female clones than in male clones and controls. Thus, although female and male cloned miniature pigs may be physiologically similar to normal animals, or at least within normal ranges, a greater degree of physiological and endocrinological variation was found in cloned pigs. The above variation must be taken into account before considering cloned female or male miniature pigs for various biomedical applications.

Effect of Rumen Protected Methionine on Lactational Performance of Dairy Cows

  • Thirty-six Holstein dairy cows were used to evaluate the effect of a rumen protected methionine supplement (RPMet). The cows were divided into two groups of 18 each (control/experimental). The experimental group was given 15 g/d of RPMet (Mepron $^{(R)}$M85, Degussa) from the 4th to the 26th week postpartum. All cows were fed a similar amount of forage including alfalfa silage, corn silage and timothy silage. Concentrate mixture was offered in proportion to the milk yield of each cow. Sufficiency of major metabolizable AAs was checked. Milk yield and milk composition was monitored for each individual cow. A metabolic profile test (MPT) was carried out at the 7th, 11th and 21st week postpartum. Without supplement, both methionine and leucine fell short of the daily requirement. Supplementation with 15 g/d RPMet was calculated to be within a sufficient margin of safety. Milk yield tended to remain higher in the supplemented group than in the controls during supplementation with RPMet. The differences in weekly milk production at the 17th, 18th, 19th and 22nd weeks postpartum were significantly high in the RPMet group (p<0.05). The average 305-d milk yield and the percentages of milk fat, milk protein and solids-not-fat were not affected by the treatment. No differences were observed in either the somatic cell count in the milk or the reproductive status. Judging from MPT, all the cows were in good health during lactation.

Lysozyme Activity in Buffalo Milk: Effect of Lactation Period, Parity, Mastitis, Season in India, pH and Milk Processing Heat Treatment

  • Lysozyme activity in buffalo milk in relation to the period of lactation, parity of animal, weather conditions and udder infections was studied. Effect of storage and heat processing of milk on lysozyme activity was determined. Lysozyme activity was higher in buffalo milk than in cow milk. Buffalo colostrum showed lysozyme activity 5 times of that in mature milk. Lysozyme activity in buffalo milk was not influenced by the parity of animal and the stage of lactation, however, it increased during extreme whether conditions (winter and summer). Lysozyme in both cow and buffalo milk exhibited maximum activity at pH 7.4. Buffalo milk lysozyme was fully stable while the cow milk lysozyme was partly inactivated by pasteurization (low temperature-long time as well as high temperature-short time treatments). Lysozyme in buffalo milk was more stable than in cow milk during storage and heat treatment. A 10 to 50-fold increase in milk lysozyme activity was observed in mastitic cows. An assay of lysozyme activity in milk can be used to diagnose mastitis in cattle but not in buffaloes. Some buffaloes exhibited 1000 fold greater lysozyme activity and moderately raised somatic cell count in milk, but there was no sign of mastitis in these animals. A possible role of milk lysozyme in prevention of mastitis in buffaloes is discussed.

MILK FAT CONTENT AND PRODUCTION PERFORMANCE OF HOLSTEIN DAIRY COWS FED FISH MEAL

  • Performance and production of twenty lactating Holstein cows fed diets containing either soybean meal or fish meal as the primary protein source were compared in a continuous or split feeding scheme. At 1 wk prepartum four groups of five animals were placed on each experimental diet. Animals assigned to the continuous feeding scheme were continued on these diets for 10-wk postpartum. At 4 wk postpartum, the diets for the groups assigned to switching protein sources were changed. These treatments were continued for another 6 wk. Milk production and dietary intakes were recorded daily. Milk constituents were measured every 2 d. Cows weights, rumen fluid samples and jugular blood samples were collected weekly. Data showed no effect of early lactation diet on cow performance or milk characteristics. Overall, compared to the soybean meal diet, the fish meal diet lowered the milk fat percent and increased production of milk per unit of dry matter ingested. No differences were observed for volatile fatty acid content of rumen fluid, blood mineral content, milk protein, somatic cell count, 4%-fat corrected milk, dry matter intake, or body weight.

  • The purpose of this study was to herd management and control of dairy cows by milk components analysis in Gyeongsangnamdo. Milk components analysis were carried out milk yield (MY), milk fat (MF), milk protein (MP), milk urea nitrogen (MUN) and somatic cell count (SCC) but, milk solid (MS), day of non-pregnant condition (DNPC), and days of primipara (DPRI) involved in report. Dairy farms were divided high group, middle group, low group according to the standard records for milk components. Examination records were divided by farm, parity, year, season and month, the number of samples were 28,957. Feeding management practice and the prediction for the risk possibility of productive disease such as reproductive and metabolic disorders by evaluating fat, protein, solids. Determination of MY, MF, MP, MS were Milkoscan 4,000~5,000 Serier (FOSS Electric Co., Copenhagen, Denmark). Correlation coefficient of milk protein (MP) and milk solid (MS) was ascertain r=0.759. SCC was ascertain 372.8$\pm$11.34 (thousand unit) and DNPC was ascertain 155.3$\pm$5.15 (days) in seven parity.

  • This study was carried out to isolate of causative agents from CMT-positive and mean somatic cell count(SCC) $\geq$500,000 cells/ml mastitic milk, and evaluate to antimicrobial susceptibility of isolates in Iksan branch area from January to November, 1996. 1. The CMT-positivity(SCC 500,000 cells/ml) of 610 heads was 36.2% (221), and of 2,373 quarter milks was 16.1% (383). 2. The Gram-positive isolates were 153 strains which was Staphylococcus sp (115), Micrococcus sp (18), Streptococcus sp (10), Listeria monocytogenes (5) and Enterococcus faecalis(5). 3. The Gram-negative isolates were 66 strains including E coli(14), Yersinia sp (13), Shigella sp(8), Enterobacillus sp(8), Cedecea sp(5), Pseudomonas aeruginosa(5), Proteus sp(5), Klebsiella sp(4), Salmonella sp(2), kluyvera ascorbate(1) and Tatumella ptyseos (1). 4. The Gram positive strains of isolates were moderately susceptible to T/s, Cp, Fd, Imp, Aug, Rif, Cft and Va. And the Gram negative strains of Isolates were moderately susceptible to T/s, Cp, Imp, Pi and Ti, In order. 5. Multiple antimicrobial resistant patterns were encountered 62 and 36 from Gram positive and negative isolates, respectively.

젖소 유방염 관리에 따른 세균 및 체세포수 등급 실태 조사 분석 (Analytical studies of bovine mastitis management by standard plate counts(SPC) and somatic cell counts(SCC))

  • 1. The number of average milking cows, clinical forms of mastitis, mastitis-developing cows, and cows killed by mastitis a year were 25.7, 1.8(7%), 6.3(26%), and 2.7(10.1%)heads, respectively. The annual grade changes of standard plate counts(SPC) and somatic cell counts(SCC) showed the grade 1A of SPC diminished sharply from April to August, we think it was due to the lack of proper management in farming season and the grade 3 of SCC indirectly influenced increased in huge during August. 2. The average number of parturitions of farms was 2.3, but 50% of below 1 parturition were 22 farms(31%), 50% of above 3 parturitions were 16(23%) out of 71 farms. According to grades of the number of parturitions of milking cows per each farm, the farms' grades recording 3 parturitions and 50% were little bit excellent. 3. The actual situation research of foremilking CMT revealed 35 out of 74 farmer didn't do CMT Among them(35 out of 74 farmers), 80% did not test thanks to the troublesome process of the CMT. SCC grade 3, among farms who did foremilking CMT once or twice a month and who did not were 29% and 40% respectively and SPC grade 1A were 55% and 9%, respectively. 4. The research of actual situation on milking management let us know 29 farms(39%) did not do lastmilking, 37 farms(49%) usually did overmilking, and 34 farms(46%) did milking for 4 or 5 minutes. Grades according to average requiring times of milking showed SCC grade 1 of farms milking within 7 minutes was 11% and SPC grade 1A was 34%, on the other side, farms milking more than 7 minutes were 0% in SCC grade 1 and 13% in SPC grade 1A. Grades according to the starting time of milking after rubbing teats showed SPC grade 1A of farms starting milking at about 1 minute and over 2 minutes were 50% and 20%, respectively. 5. The research of actual situation on hygienic milking management uncovered 65 farms(88%) were using one towel which was used in washing teats and udders to wash more than 3 to 4 cows, and 53 farms(72%) were using one dried towel to dry udders not for each cow but for more than 3 to 4 cows after washing. Also, on milking turns disclosed 30 farms(40%) were milking cows in the order of incoming without isolation of a dominant group. According to grades of towels used in washing teats and udders, farms using a towel for each cow were 56% and a towel for over 3 cows were 31% in SPC grade 1A. According to using-or-not grades of dried towels after washing udders, farms using a towel for each cow were 79% and a towel for over 3 cows were 21% in SPC grade 1A. 6. Farms doing teat-dipping before milking were 7(10%), not doing teat-dipping after milking, or doing sometimes were 9(12%), and doing right after milking were 57(77%). And farms doing teat-dipping after dry cows and before delivery were 21(28a ). Farms using bethadine as an antiseptic solution were 70(95%), 40 farms(59%) diluted it with water as weak as 5 to 10 times, and on drying cows 64 farms(87%) slowly did it more than 2 days. Grade 1A of SPC of farms doing teat-dipping at every milking was 38%, farms doing occasionally or not was 33%, and farms doing it right after milking was 37% and doing after milking more than 5 cows was 20%. Grade 1A of SPC among farms diluting bethadine 5 times and diluting 5 to 10 times with water were 36% and 33%, respectively, and Grade 3 of SCC were 35% and 32%, respectively. 7. Studies on nonlactating period medical treatment, as the cows were on dry, 54 farms treated with their own hands.73 farms(98%) had bovine mastitis treated for themselves. And on applying medicines against mastitis, 55 farmers chose them on the basis of their own experience, 42 farms(57%) were treated more than 3 days. 41 farms(55%) dumped away the mastitis infected milk separately, 24 farms(32%) were feeding and milking at the same time. 8. Fifty-six farms(76%) always washed and disinfected milking machines after milking. Farms using the milking machines at low, or variable vacuum pressures, or at the vacuum pressure, set at the moment of its installation were 31(42%), and farms that did not know pulsation ratio were 27(37%). Farms changing liners when they were torn 8(11%), 58 farms(78%) said they checked milking system when there were wrong with them, 31 farms(42%) changed milking hoses when they found out problems, and 42 farms(57%) cleaned vacuum and milking systems when they felt dirty. The SPC grade 1A of farms washing and sterilizing milking machines was 38% and farms only washing was 28%.

전남지방 산양유의 성분 및 체세포수의 계절적 변화 (Seasonal variation of goat milk composition and somatic cell count in Jeonnam province)

  • Consistent information on the chemical composition and its seasonal variation of goat udder half milk is limited in Korea. The objective of this study was to analyze the seasonal variation of the chemical composition of goat milk to take establish various parameters into consideration on the pricing of the goat milk. Variations in chemical composition, somatic cell count (SCC) and bacterial count of 1,038 udder half milk samples from 650 heads raised in 7 farms of Jeonnam province were determined by season. Fat, protein, lactose, non-fat solids, milk urea nitrogen (MUN), pH, SCC and bacterial counts were also analyzed. The average composition of the milk was: fat $3.80{\pm}1.36%$, protein $3.23{\pm}0.80%$, lactose $4.39{\pm}0.54%$, total solids $12.18{\pm}1.80%$, non-fat solids $8.38{\pm}0.80%$, and milk urea nitrogen $28.44{\pm}5.00mg/dL$. The average pH was $6.81{\pm}0.24$. The average of SCC and bacterial counts were $2.54{\pm}4.60{\times}10^6cells/mL$ and $1.25{\pm}3.76{\times}10^5CFU/mL$, respectively. Chemical composition, pH, SCC and bacterial counts of dairy goat milk varied widely during the lactation period and by season. The fat concentration was the lowest in spring ($3.39{\pm}1.53%$) and the highest in autumn and winter ($3.98{\pm}1.30%$ and $3.98{\pm}1.48%$). Protein concentration was the lowest during summer ($2.92{\pm}0.48%$) and the highest in winter ($2.92{\pm}0.48%$). Lactose concentration was the lowest in autumn ($4.24{\pm}0.41%$) and the highest in spring ($4.58{\pm}0.35%$). The lowest total solid value was obtained in the spring season ($11.75{\pm}1.80%$) which was then increased in winter ($12.85{\pm}1.96%$). Non-fat solid concentration was the lowest in summer ($8.07{\pm}0.64%$) and the highest in autumn ($8.94{\pm}0.82%$). MUN concentration was the highest in summer ($8.07{\pm}0.64%$), and the pH concentration was the highest in spring at $6.93{\pm}0.27%$. Seasonal variation of SCC and bacterial count were the lowest in spring ($0.94{\pm}1.54{\times}10^6cells/mL$ and $0.22{\pm}0.61{\times}10^5CFU/mL$, respectively) and was the highest in winter ($3.95{\pm}7.14{\times}10^6cells/mL$ and $2.23{\pm}5.54{\times}10^4CFU/mL$, respectively).

Near Infrared Spectroscopy for Diagnosis: Influence of Mammary Gland Inflammation on Cow´s Milk Composition Measurement

  • Nowadays, medical diagnostics is efficiently supported by clinical chemistry and near infrared spectroscopy is becoming a new dimension, which has shown high potential to provide valuable information for diagnosis. The investigation was carried out to study the influence of mammary gland inflammation, called mastitis, on cow´s milk spectra and milk composition measured by near infrared spectroscopy (NIRS). Milk somatic cell counts (SCC) in milk were used as a measure of mammary gland inflammation. Naturally occurred variations with milk composition within lactation and in the process of milking were included in the experimental design of this study. Time series of unhomogenized, raw milk spectral data were collected from 3 cow along morning and evening milking, for 5 consecutive months, within their second lactation. In the time of the trial, the investigated cows had periods with mammary gland inflammation. Transmittance spectra of 258 milk samples were obtained by NIRSystem 6500 spectrophotometer in 1100-2400 nm region. Calibration equations for the examined milk components were developed by PLS regression using 3 different sets of samples: samples with low somatic cell count (SCC), samples with high SCC and combined data set. The NIR calibration and prediction of individual cow´s milk fat, protein, and lactose were highly influenced by the presence of mil samples from animals with mammary gland inflammation in the data set. The best accuracy of prediction (i.e. the lower SEP and the higher correlation coefficient) for fat, protein and lactose was obtained for equations, developed when using only “healthy” samples, with low SCC. The standard error of prediction increased and correlation coefficient decreased significantly when equations for low SCC milk were used to predict examined components in “mastitis” samples with high SCC, and vice versa. Combined data set that included samples from healthy and mastitis animals could be used to build up regression models for screening. Further use of separate model for healthy samples improved milk composition measurement. Regression vectors for NIR mild protein measurement obtained for “healthy” and “mastitic” group were compared and revealed differences in 1390-1450 nm, 1500-1740 nm and 1900-2200 nm regions and thus illustrated post-secretory breakdown of milk proteins by hydrolytic enzymes that occurred with mastitis. For the first time it has been found that monitoring the spectral differences in water bands at 1440 nm and 1912 nm could provide valuable information for inflammation diagnosis.

원유의 이화학적 특성에 미치는 감귤 부산물 TMR 사료 급여의 영향 (Effects of Feeding TMR Feed with Dietary Citrus Byproducts on Chemical Properties of Raw Milk from Holstein Cow's Raw Milk)

  • 본 연구는 감귤 부산물 TMR 사료 급여가 젖소 원유의 품질에 미치는 영향을 검토하기 위하여 실시하였다. 감귤 부산물을 혼합하지 않은 TMR 사료를 급여한 원유를 T0, 그리고 감귤 부산물을 혼합한 TMR 사료를 급여한 원유를 T1이라 하였다. 젖소는 홀스타인종으로 제주도 소재 낙농 목장에서 사양 시험을 하였으며, 1회에 7 마리씩 3반복 실행하였다. T0 및 T1의 산유량은 각각 24.16 및 25.97 kg이고, 체세포수는 66만 및 44만5천 개로 감귤 부산물 급여에 의해 산유량이 증가하고 체세포수가 감소하였다. T0 및 T1 원유의 총균수는 각각 $2.05{\times}10^6$$1.97{\times}10^6$ CFU/mL로 유의적 차이가 없었다. T0 및 T1 원유의 비중은 각각 1.0335 및 1.0342로 비슷하고, 적정 산도는 각각 0.14% 및 0.14%로 같은 수준을 나타내었다. 원유의 유지방 함량은 T1이 4.03%로 낮고, 유단백, 유당, 무지유고형분 및 총 고형분 함량은 현저한 차이를 보이지 않았다. 원유의 휘발성 향기 성분은 감귤 부산물 급여에 의한 유의적 차이를 보이지 않았다. 원유의 항산화력은 감귤 부산물 TMR 사료 급여에 의해 향상되었다.
