Biochemical Adaptation of Pinus pumila on Low Temperature in Mt. Seorak, Korea

  • We tested the hypothesis that alpine plants have special physiological and biochemical mechanisms in addition to their structural adaptation in order to survive under extreme conditions. The photosynthetic organs of Pinus pumila were used to examine the seasonal changes in sugar concentration, antioxidative enzyme, and lipid peroxidation. The concentrations of sucrose, glucose, fructose and reducing sugar were the highest in the leaves in April. But sugar contents in buds and inner barks did not respond sensitively on temperature change. Meanwhile superoxide dismutase (SOD) activity responded sensitively on the change of temperature and SOD in all tissues maintained high activity in April. Meanwhile anthocyanin content increased rapidly in June but the increase of anthocyanin content was not enough to prevent their tissues from the damage by the exposure of high temperature or other stress. In conclusion, under low temperature condition, P. pumila increased the concentration of soluble sugars and SOD activity in their tissues in order to overcome extreme environmental condition. But in summer, these stress defense system against high temperature might be disturbed slightly. This results in the increase of malondialdehyde (MDA) contents in three tissues by lipid peroxidation.

저장 기간에 따른 콜라비 김치의 품질 특성 (Quality Characteristics of Kohlrabi Kimchi during Storage)

  • In the present study, kohlrabi-kimchi was prepared for assessment of changes in sensory qualities and measurement of changes in quality characteristics according to the storage period for the purpose of using kohlrabi as an ingredient of Kimchi. Examination of the physicochemical properties of kohlrabi-kimchi according to the storage period showed that the acidity increased steadily throughout the storage period, while soluble-solids content, pH and hardness decreased as the storage period elapsed. Meanwhile, the L value increased and then it decreased as the storage period elapsed, but, the a value increased and the b value gradually decreased after day 14 of storage. Reducing sugars increased up to day 10 of storage, and then they decreased slowly after day 15 of storage. Measurement of changes in lactic acid bacteria in kohlrabi-kimchi according to the storage period showed bacterial growth showing typical tendencies of Kimchi with a rapid increase on day 5 of storage and a decrease thereafter. Assessment of sensory qualities of kohlrabi-kimchi according to the storage period showed that the highest scores for appearance and sourness, odor, texture, and overall scores were obtained on day 15 of storage. Based on the above results, it was determined that the optimal maturity period of kohlrabi-kimchi is day 15 of storage.

Waterlogging induced oxidative stress and the mortality of the Antarctic plant, Deschampsia antarctica

  • We investigated the mortality and the oxidative damages of Deschampsia antarctica in response to waterlogging stress. In field, we compared the changes in the density of D. antarctica tuft at the two different sites over 3 years. The soil water content at site 2 was 6-fold higher than that of site 1, and the density of D. antarctica tuft decreased significantly by 55.4% at site 2 for 3 years, but there was no significant change at site 1. Experimental results in growth chamber showed that the $H_2O_2$ and malondialdehyde content increased under root-flooding treatment (hypoxic conditions-deficiency of $O_2$), but any significant change was not perceptible under the shoot-flooding treatment (anoxic condition-absence of $O_2$). However, total chlorophyll, soluble sugar, protein content, and phenolic compound decreased under the shoot-flooding treatment. In addition, the catalase activity increased significantly on the 1st day of flooding. These results indicate that hypoxic conditions may lead to the overproduction of reactive oxygen species, and anoxic conditions can deplete primary metabolites such as sugars and protein in the leaf tissues of D. antarctica. Under present warming trend in Antarctic Peninsula, D. antarctica tuft growing near the shoreline might more frequently experience flooding due to glacier melting and inundation of seawater, which can enhance the risk of this plant mortality.

Hydrophilic Extracts of the Bark from Six Pinus Species

  • Pine barks are important biomass resources because they are utilised in the production of pine wood and rosins. However, no chemical study has been conducted on the hydrophilic status of pine barks in Indonesia. This aim of this study is to explore the hydrophilic extracts of the barks from six Pinus species (P. elliotii, P. caribeae, P. oocarpa, P. merkusii P. montezumae, and P. insularis). The hydrophilics of pine barks were analysed using gas chromatography-mass spectrometry. The presence of polyphenol contents in the ethanol extracts obtained from the barks of six Pinus species was determined using the tannin-formaldehyde method, Folin-Cioucalteu assay, and vanillin-HCl assay. The ethanol and hot water soluble extractives derived from inner barks were higher in quantity when compared to those derived from the outer bark samples. The polyphenol measurement showed that the highest value of total phenol content was derived from the outer bark of P. montezumae whereas those of the total phenol and tannin- formaldehyde contents were derived from the inner and outer barks of P. oocarpa. GC-MS analysis revealed that nitrogenous compounds are dominant constituents in the inner and outer barks of the six species, followed by sugars and monophenolics, respectively.

Relationship between Lignin Content and the Durability of Wood Pellets Fabricated Using Larix kaempferi C. Sawdust

  • This work was conducted to examine the relationship between lignin content and the durability of larch (LAR) pellets. LAR sawdust was immersed in tap water (TW), sulfuric acid (AC) and sodium hydroxide (AK) solutions, and then the immersed sawdust was used for pellet fabrication. Klason lignin (KL) content of the immersed LAR, contents of soluble lignin (SL) and monomeric sugars liberated from the immersion of LAR, and durability of LAR pellets were measured. KL content decreased as the concentration of AC and AK solutions increased, but glucose content increased with increase in AC and AK concentration. Durability of wood pellets fabricated using non-immersed LAR sawdust was the highest, followed by those made using TW-, AK- and AC-immersed sawdust. LAR pellets became more durable as the concentration of KL and SL increased, but a significant positive correlation was found only between pellet durability and KL content. Through the fluorescent microscopic observation and SEM-EDX analysis, it was verified that lignin content of non-immersed LAR pellets was higher than that of AC- and AK-immersed LAR pellets. These results suggest that lignin might contribute to an increase in inter-particle bonding in wood pellets.

Time-dependent changes of fruit metabolites studied by 1H NMR

  • The browning phenomenon of fruits can be easily observed when fruits or vegetables (apples, pears, bananas, potatoes, etc.) are cut with a knife and the part turns brown. When this browning occurs, changes in taste, color, and nutrients usually are introduced. The cause of this browning phenomenon has been well studied for a long time, but these studies have mainly focused on preventing deterioration of processed foods during food processing or storage. Resultantly, there are few studies on how much changes in nutrients (saccharides, amino acids, fats, water-soluble low molecular weight ammonium ions, etc.) are caused by browning. The purpose of this study is to determine the change in nutrients during browning using apple as a model fruit. We conducted a comparative study on how much the nutrient fluctuations differ depending on the presence or absence of pretreatment such as the application of heat. All analysis was conducted using 1H NMR. The ANOVA analysis showed that the concentrations of 4 amino acids (alanine, asparagine, isoleucine, and valine), 3 types of sugars (fructose, glucose, and xylose), 1 type of organic acid (lactate) and choline were significantly increased in samples showing browning. In addition, the groups before and after browning were clearly separated using multivariate statistical analysis methods (PCA, PLS-DA), which was greatly contributed by two sugar components (fructose and glucose) present in high concentrations in apples.

Inhibition Effects of Persicaria amphibia (L.) Delarbre on Oxidative DNA Damage via ATM/Chk2/p53 pathway

  • Persicaria amphibia as an England native plant, is a rhizomatous perennial, one of the rather amphibious plants. Its aquatic form contains water-soluble sugars, starch, and protein. P. amphibia have up to 18% tannins in stems and rhizomes. Previous studies have confirmed the anti-inflammatory activity of live bacteria roots, but no studies on bioactivity are known. DNA damage responses (DDRs) pathways are considered a crucial factor affecting the alleviation of cellular damage. The ataxia-telangiectasia mutated and Rad3 related (ATM) and checkpoint kinase 2 (Chk2) pathways are the main pathways of DNA damage response. Also, p53 is a key integrator of cellular response to oxidative DNA damage, contributing repair, or leading transcription including apoptosis. In the present study, we conducted an investigation into the inhibitory effects of P. amphibia on oxidative DNA damage for confirming potential to complementary medicine and therapies. In conclusion, P. amphibia can provide protective effects against double-stranded DNA break (DSB) caused by oxidative DNA damage.

Chemical composition of barley and co-products from barley, corn, and wheat produced in South-East Asia or Australia

  • Objective: A study was conducted to determine the chemical composition of barley and co-products from barley, corn, and wheat produced in South-East Asia or Australia, and to test the hypothesis that production area or production methods can impact the chemical composition of wheat co-products. Methods: Samples included seven barley grains, two malt barley rootlets, one corn gluten feed, one corn gluten meal, one corn bran, eight wheat brans, one wheat mill mix, and four wheat pollards. All samples were analyzed for dry matter, gross energy, nitrogen, amino acids (AA), acid hydrolyzed ether extract, ash, minerals, starch, and insoluble dietary fiber and soluble dietary fiber. Malt barley rootlets and wheat co-products were also analyzed for sugars. Results: Chemical composition of barley, malt barley rootlets, and corn co-products were in general similar across countries. Wheat pollard had greater (p<0.05) concentrations of tryptophan, magnesium, and potassium compared with wheat bran, whereas wheat bran had greater (p<0.05) concentration of copper than wheat pollard. There were no differences in chemical composition between wheat bran produced in Australia and wheat bran produced in Thailand. Conclusion: Intact barley contains more starch, but fewer AA, than grain co-products. There were only few differences in the composition of wheat bran and wheat pollard, indicating that the two ingredients are similar, but with different names. However, corn gluten meal contains more protein and less fiber than corn bran.

뽕나무 내한성에 관한 연구 -특히 지조함유물질과 내한성과의 관계에 대하여- (Studies on Freezing Tolerance of Mulberry, Morus species -Relation between Freezing Tolerance and Some Substances in Mulberry Branches-)

  • 본 연구는 뽕나무 품종별 그리고 추기수확시기 및 방법에 따르는 내한성의 정도와 내한성에 영향을 미치는 지조의 함유성분량과의 관계를 조사하여, 다음과 같은 결과를 얻었다. 1. 뽕나무 품종별 내한성은 용천추우, 자천 강원 3호 및 시평이 가장 강하였고, 노상, 개양서반, 유전 및 구상28호가 중간정도이었으며, 일지뢰, 모상, 구상21호 및 수원상3호가 가장 약한 품종에 속하였다. 2. 추기수확시기 및 방법과 잔엽부위에 따른 동아 50% 치사온도에 의한 내한성 검정결과는 포장에서의 선고장비율과 부의 상관관계(r=-0.5*)를 나타내었다. 뽕나무의 내한성 증대작용은 9월초부터 9월말까지 계속되며, 9월 중순에 그 정도가 가장 커서 이때의 적엽은 내한성을 약화시켰으며, 특히 전적엽 및 수확 후 기부잔엽을 내한성을 크게 약화시키었다. 그러나 적심 처리를 하면 내한성을 약화를 줄일 수 있었다. 3. 품종별 지조의 성분함량과 내한성과의 관계를 보면 조단백질 및 수용성 단백질함량은 부의 상관관계(r=-0.70*)이었으나, 이들에 대한 전탄수화물의 함량비에서는 정의 상관관계(각각 r=0.31 및 r=0.71*)이었으며, 당 및 RNA 함량과도 높은 정의 상관관계(각각 r=0.67* 및 r=0.99**)가 인정되었다. 그러나 건물률, 지방. 전탄수화물함량 및 DNA함량과는 무관하였다. 4. 뽕나무 지조에는 동기에 raffinose, lactose, sucrose, glucose, fructose. arabinose, xylose, ribose (추정) rhamnose등 9종의 당류가 동정되었다. 이들 당류는 내한성과 깊을 관련을 갖는데, 특히 당의 대부분을 차지하는 sucrose, glucose 및 fructose의 함량차가 크게 영향 하였다. 5. 추기수확시기에 따라서는 9월 12일 전적엽구의 지조내 RNA함량의 심한 감소를 제외하고는 고 시기가 늦어지는데 따라서 그 함량이 증가하였다. 수확방법에 따라서는 전반적으로 수확을 하게 되면 RNA함량이 감소되었으나 적심잔엽을 하면 그 감소를 줄일 수 있었다. 수확 후 잔엽부위별로 보면 중상부의 잔엽은 지조내 RNA함량을 증가시켰고, 상부의 말숙엽과 하부의 노령 옆을 잔엽하면 그 함량이 감소되었다. 수확방법에 따른 RNA함량과 내한성과는 꼭 일치하지는 않았으나, 정의 상관관계가 인정되었다.(r=0.51*).

옥수수 수염의 이화학적 특성과 변이 (Physicochemical Characteristics of Corn Silk)

  • 옥수수 수염의 일반적인 생육특성과 함유된 각종 일반성분 및 유리당, 유기산, 아미노산 및 향기성분에 관한 기초자료를 제공하기 위하여 본 시험을 수행하였으며 얻어진 결과를 요약하면 다음과 같다. 1. 옥수수의 수염은 출사 후 3일 이내에 수정이 완료되고 길이 신장이 정지되나 수정되지 않은 수염은 지속적으로 신장하여 출사 후 8일에 23-25cm였다. 2. 출사기에 옥수수 수염의 수분함량은 92-94%, 출사후 30일에는 70-75%였다. 3.옥수수 수염은 엽록소 b의 함량이 높고, 엽록소 a/b의 비율은 사료용 옥수수> 단옥수수> 초당옥수수> 찰옥수수 순이었다. 4. 유리당은 glucose와 fructose가 주종을 이루고, 그 구성비율은 glucose가 55% fructose가 42% 이었으며 sucrose 및 maltose는 4% 미만으로 낮았다. 5. 가용성 고형물 함량은 미수정 수염이 13.7-16.8 Brix %, 수정된 수염은 12.6-13.7 Brix %로 1.1-3.1 Brix % 차이가 있었다. 6. 옥수수 수염의 유기산은 phytic, oxalic, malic, shikimic, glutaric 및 acetic acid가 검출되었는데, 이중 phytic, oxalic 및 glutaric acid가 주요 유기산이었고 유기산의 함량은 사료용 옥수수> 초당옥수수> 찰옥수수> 단옥수수 순으로 높았다. 7. 단백질 함량은 3.36-4.17%로서 출사후 3일에 최대치를 나타내었고, 단백질 함량은 사료용 옥수수> 찰옥수수> 단옥수수> 초당옥수수 순으로 높았다. 8. 아미노산중 serine, glycine 및 threonine 3종은 10% 이상 함유되어있었으며 glycine의 구성비율이 12.3%로 가장 높았고 황(S)함유 아미노산인 methionine과 cystine 함량은 각각 2.1% 및 1.3%로 함량이 낮았다. 9. 향기성분으로 acetaldehyde, ethanol, acetone, DMS, isobutylaldehyde, cis-3-hexanol, 3-hexe-1-ol, acetate, trans-2-hexanol, pentanol 등이 검출되었고, 사료용의 주요 향기성분은 acetaldehyde와 DMS, 찰옥수수는 acetaldehyde, ethanol 및 DMS였다. 10. 옥수수 수염의 길이는 유기산 함량과 r=0.556, 아미노산 함량과는 r=-0.514로 유의성이 인정되었고, 유리당 함량은 유기산 함량과 r=0.703, flavonoid 함량과 r=0.544로 유의한 정의상관이 있었다. 특히, flavonoid 함량은 엽록소 함량과 정의 상관(r=0.523)이 인정되어 엽록소는 flavonoid 함량의 판단지표로 활용될 수 있을 것으로 사료되었다.
