용융법에 의한 수용성 유리의 제조 및 특성 (I) : 용해 특성, 살균 효과 및 세포 독성 (Preparation and Characterization of Water-Soluble Glass through Melting Process (I) : Dissolution Characteristics, Bactericidal Effects and Cytotoxicity)

  • Water-soluble phosphate glasses containing Ag or Cu ion were prepared through melting process. Then the powdered glass samples were dissolved in D.I. water at room temperature with changing the dissolution time. In terms with the glass composition, dissolution characteristics, bactericidal effects and cytotoxicities were investigated. Dissolved amounts increased uniformly with dissolution time, and the dissolution rate was higher for ternary glass than for binary glass and with less metal oxide amount. And the dissolution rate of the glass with Ag ion was higher than that with Cu ion, and the bactericidal effect of the glass with Ag ion was also greater. Solution with more than 25 ppm of Ag was observed to have strong cytotoxicity to L929, and solutions of lower Ag concentration or with Cu seemed to have little cytotoxicity.

수용성 유리를 이용한 조류 생장 억제형 기능성 모르타르의 개발 (Development of a Functional Mortar for Algae Growth Restraining by Using Soluble Glass)

  • 본 연구는 수중 구조물의 수명과 조류의 성장이 관계가 있다는 연구결과에 근거하여 조류의 이상증식 억제가 구조물에 영향을 미칠 수 있다는 것에 초점을 맞추었다. 따라서 본 연구는 조류의 활용을 극대화시키면서 조류로 인한 문제점을 억제할 수 있도록 조류의 생장을 억제할 수 있는 기능성 모르타르 개발을 목적으로 하였다. 조류의 생장을 억제하기 위한 방법으로 금속 ion(Hg+, Ag+, Cu2+, Au+ 등)은 조류의 생장을 억제하는 물질로 오래전부터 사용되어져왔다. 이에 착안하여 수중에 용출되었을 때 유효한 금속 ion의 상태로 장시간에 걸쳐 안정한 수용성 유리를 사용하여 조류의 생장을 억제할 수 있는 기능성 모르타르 시편을 제작하였다. 제작된 모르타르 시편은 시편의 억제 기능성을 평가하기 위해 해수와 담수에 생장하는 일반적인 조류를 배양시킬 수 있는 배지를 각각 제작한 다음 개발된 모르타르 시편을 투입한 후 일정기간이 지난 뒤 배지에 잔류하는 조류의 양을 측정함으로써 개발된 모르타르 기능성을 판단하였다. 조류의 양을 측정하기 위하여 사용된 분석은 용존 유기탄소(dissolved organic carbon)의 양과 엽록소(chlorophyll)의 양을 측정하는 두 가지 분석 결과를 이용하였으며 분석을 통해 나온 결과는 개발된 시편의 조류 억제 기능성에 대하여 정성적인 평가를 하였다. 즉, 용존 유기탄소의 양이 증가하고 엽록소의 양이 감소하면 조류에 대한 생장 억제성이 있는 것으로 판단하였다. 두 가지 분석 결과, 해수와 담수 모두 조류 생장 억제용 모르타르는 억제 기능성이 나타나고 있었다.

Effects of Intraruminal Soluble Glass Bolus on Blood Selenium and Plasma Mineral Level of Grazing Does under Backyard Conditions in Selected Areas in Nueva Ecija, Philippines

  • Soluble glass bolus with selenium (Se), copper (Cu) and cobalt (Co) was administered intraruminally to Philippine grazing does fed under backyard farming conditions to determine its effect on blood mineral status. Forty-five does were dosed with SGB intraruminally every 6 months, whereas 15 were without SGB during this experimental period of 10 months. Blood of does in both treatment groups were collected every other month and blood Se, plasma Cu, Ca, P and Mg were determined in this study. All does did not show clinical Se, Cu, Ca, P or Mg deficiency during this experimental period. Selenium concentration of treated does increased (p<0.01) after beginning of this experiment, whereas the level of control does decreased slowly (p<0.01). Two months after SGB administration, all treated does had higher blood Se than the lower limit of $20{\mu}g/l$ suggested by NRC (1981), whereas some control does had lower blood Se concentration than the lower limit of $20{\mu}g/l$. On the other hand, plasma Cu concentration of treated does started to increase more remarkably than control does 4 months after this experiment had started although the difference was insignificant. There were no significant differences between plasma Ca, P or Mg concentration of does in both treatment groups. Soluble glass bolus had no harmful effects on plasma macro mineral concentrations of grazing does. This study showed that SGB was available as mineral supplement to improve imbalances of selected mineral of grazing goats in the traditional backyard conditions in Luzon Island, Philippines.

Effect of a Copper, Selenium and Cobalt Soluble Glass Bolus Given to Grazing Yaks

  • Two field trials were carried out to evaluate the performance of a soluble glass copper, cobalt and selenium bolus for maintaining adequate levels of the three trace elements in yaks. Forty yaks were used in trial 1 and 60 yaks were used in trial 2. In each trial two commercial soluble glass boluses were administered to half of the yaks. Blood samples were taken from the jugular vein at day 0, 30, 60, 90 in trial 1 and at day 0, 45, 75 and 105 in trial 2. The samples were analysed for copper status (serum caeruloplasmin activity and copper concentration), cobalt status (serum vitamin $B_{12}$ concentration and cobalt concentration), selenium status (erythrocyte glutathione peroxidase activity and selenium concentration) and serum zinc concentration. The erythrocyte glutathione peroxidase activities, serum caeruloplasmin activities and serum vitamin $B_{12}$ concentrations for trial 1 and 2 were all significantly increased for the bolused yaks (p<0.001 or p<0.01) on all sampling days. The bolused yaks had a significantly higher selenium and copper status in serum than the control yaks on all sampling days in trial 1 and 2 (p<0.05 or p<0.01). There were no significant differences in zinc and cobalt concentrations between the bolused yaks and the controls.

  • 은 이온과 구리 이온을 동시에 함유하는 살균용 인산염계 수용성 유리를 용융법으로 제조하였다. 분말상의 유리 시편을 상온의 증류수에 넣어 조성과 시간에 따른 용해 특성을, bulk상 시편에 대해서는 용해 전.후의 표면변화를 각각 관찰하였고, 살균 효과 및 세포독성을 평가하였다. 은 이온:구리 이온=1:1인 조성에서 최대 용해량을 나타내었으며 용해거동은 초기부터 total dissolution이 지배적이었다. 시간의 경과에 따라 용해는 진행이 되지만, 표면에 생성되는 새로운 층은 관찰되지 않았다. Pseudomonas sp, e. coli, staphylococcus aureus, 및 salmonella균에 대한 살균 효과는 용해량의 증가에 따라 증가하여 우수한 살균효과를 나타내었다. L929 세포에 대한 독성 실험 결과, 은 이온의 농도가 10ppm이상에서만 독성을 나타내었다.

��프신의 고정화(固定化)에 관한 연구(硏究) (A Study on Immoblization of Pepsin)

  • ��신을 DEAE-cellulose, CM-cellulose, Sepharose-4B와 porous glass beads을 사용(使用)하여 고정화(固定化)시켰던 바 그 결과(結果)를 요약(要約)하면 다음과 같다. 1. CM cellulose와 DEAE-cellulose의 ��신 흡착량(吸着量)은 건조된 bead 1 gr에 대하여 각각 10 mg와 15 mg이었고 glutaralde을 사용(使用)함으로서 20 mg와 27 mg로 흡착량(吸着量)이 증가(增加)하였고 효소(酵素)의 활력(活力)은 용해성효소(溶解性酵素)에 비해 각각 22%, 24%로 나타났다. 2. Sepharose-4B을 cyanogen bromide을 사용(使用)하여 활성화(活性化)시킨후 glutaraldehyde을 사용(使用)하여 공유결합을 유도하였던 바 건조된 bead 1 gr에 ��신 19 mg가 통합(統合)되었고 효소(酵素)의 활력(活力)은 23%로 나타났다. 3. Porous glass beads의 표면(表面)을 succineamido - propyl glass beads의 형태(形態)로 유도체를 만든후 ��신을 고정화(固定化)시켰을 때 bead 1 gr가 ��신 40 mg을 통합(統合)할 수 있었고 효소(酵素)의 활력(活力)은 용해성효소(溶解性酵素)에 비해 45%로 나타났다.

인산염계 수용성 유리의 용출 및 구조 분석 (Dissolution and Structure Analysis of Phosphate Water Soluble glasses)

  • Potassium-Calcium-Phosphate glasses in range $XCaO\cdot(50-X)K_2O$ \cdot $50P_2$$O^{5}$were investigated. Glass transition temperature(Tg) of prepared glasses were below $520^{\circ}C$, thermal expansion coefficient from $270.3$\times$10^{7}$ to $604.5$\times$10^{7}$/$^{\circ}C$. The structure of XCaO.(50-X)$K_2$O\cdot$50P_2$$O^{5}$ glasses were examined by FT-IR spectroscopy indirectly. As CaO was increased, Ts, Tg, P-O-P bonding strength and chemical durability were increased. Glass surface change was observed with increasing dissolution time using bulk specimen, weight loss and pH change were measured as function of the dissolution time.

Development of a Functional Mortar for Restraining Surface Algal Growth

  • Proliferation of algae on the surface of concrete or mortar in aquatic habitat has a negative impact on maintenance of concrete-based structures. Growth of algae may decrease stability of structure by bio-deterioration. In this study, we developed a functional mortar for restraining bio-deterioration by using $Cu^{2+}$ ion. The mortar contains soluble glass beads made of $Cu^{2+}$ ion, which can dissolve into water slowly. Mortars prepared with different ratio of glass beads (0, 2, 5, 10, and 15%) were placed in a culture medium with algae and incubated over a month period. Water chemistry, chlorophyll-a, and extracellular enzyme activities were measured. The incubation was conducted in both freshwater and seawater conditions, to assess applicability to both aquatic conditions. Overall, mortar with Cu glass exhibited lower chlorophyll-a content, suggesting that the functional mortar reduced algal growth. DOC concentration increased because debris of dead algae increased. Cu glass also decreased phosphatase activity, which is involved in the regeneration of inorganic P from organic moieties. Since, P is often a limiting nutrient for algal production, algal growth may be inhibited. Activities of ${\beta}$-glucosidase and N-acetylglucosaminidase were not significantly affected because carbon and nitrogen mineralization may not be influenced by the Cu glass beads. Our study suggests that functional mortar with Cu glass beads may reduce the growth of algae on the surface, while it has little environmental impact.

소결법에 의한 다공질 결정화유리의 제조 (Preparation of Porous Glass-Ceramics by the Sintering)

  • In manufacturing process of porous glass-ceramics by the filler method, the sintering behaviour of crystallizable glass powder mixed with various salts was studied and also the effects of precipitated crystal phases on the properties of porous glass-ceramics were investigated. Fine-grained crystallizable glass powder was homogeneously mixed with various slat having grain size 100~200 ${\mu}{\textrm}{m}$ and sintered for densification. After washing out the inorganic salt with distilled water, the porous sintered body was heat treated additionly for crystallization. The MgO-Al2O3-SiO2 base glass was used as crystallizable glass powder and the water soluble salts such as K2SO4 and MgSO4 were used as filler. When K2SO4 was used, leucite crystal phase was formed as a result of the ion exchange and porous glass-ceramics which exhibit high temperature resistance and high thermal expansion coefficient of 17$\times$10-6/$^{\circ}C$ could be obtained. On the contrary, when MgSO4 was used, only slight ion exchange is observed and $\mu$-cordierite and $\alpha$-cordierite crystal phases were formed and porous glass-ceramics which exhibit low thermal expansion coefficient schedule were determined with the results of DTA curves, thermal shrinkage curves and XRD patterns analysis. From DTA curves and thermal shrinkage curves, it was found that the sintering densification have been completed at the temperature range of exothermic peak for crystallization. The pore size distributions and pore diameters were measured by mercury porosimeter. The pore diameter of porous glass-ceramics was 10~15 ${\mu}{\textrm}{m}$ when 100~200${\mu}{\textrm}{m}$ grain size of K2SO4 was used and it was 25~30 ${\mu}{\textrm}{m}$ when the same grain size of MgSO4 was used. The porous glass-ceramics K2SO4 used shows bimodal pore size distribution and its porous skeleton structure was ascertained by SEM observation.
