Optimization of Monochamus alternatus media and culture period for cordycepin production in Cordyceps militaris culture using solid-state fermentation

  • In this study, we investigated the effect of solid culture medium on the production of cordycepin in Cordyceps militaris. The regression equation was expressed as follows: Y1 = 755.3-58.6625X1+4.79432E-14X2-46.6625X3-5.66269E-14X1X2-0.025X1X3+1.62475E-14X2X3-160.6625X12+0.0125X22-206.9625X32, where, Y represents the value of cordycepin content (㎍/g), X1 corresponds to the weight of M. alternatus in solid culture medium (g/bottle), X2 to the water content of the solid culture medium (%), and X3 to the culture period (day). The solid culture medium was optimized using the response surface methodology, and the optimal medium composition was as follows: the weight of M. alternatus in solid culture medium and water content were 16.2% and 100.7% (20.14 mL water/20 g solid culture medium), respectively, with a culture period of 39 days. Under these conditions, the cordycepin content of the fruiting bodies reached 150.0 ㎍/g (actual value). The supplementation of M. alternatus in solid culture for improved cordycepin content of C. militaris seems to be a promising alternative to wild and solid cultivation.

양액재배 농가의 고형배지 선택 기준에 관한 연구 (Solid Culture Medium Selection Criteria for Hydroponics Farm Households)

  • This study aimed to analyze the selection criteria and priority settings for solid culture medium used in hydroponic crop production in farm households. Expert brain storming was carried out to extract solid culture medium selection criteria for hydroponic farming. As a result, 3 criteria of economy (cost), productivity, and environment, and 9 factors were extracted. A questionnaire survey of hydroponic farm households was conducted in Gyeonggi, Gangwon, and Chooncheong provinces. AHP analysis of the hydroponic solid culture medium selection criteria identified productivity as the most important criterion, chosen by 58.7% of the respondents, followed by economy (28.4%) and environment (12.9%). The 9 factors were rated by the respondents in the following decreasing order of importance: 1, crop yield (28.3%); 2, pest occurrence (18.5%); 3, maintenance/management costs (12.0%); 4, convenience of maintenance/management (13.4%); 5, initial investment cost (11.6%); 6, material energy consumption (6.5%); 7, waste recyclability (4.0%); 8, waste disposal costs (3.4%); and 9 environmental emissions (1.81%). These results imply that hydroponic farm households consider cultivation-related quality factors more important than economic factors, such as price of culture medium or installation cost.

Solid-State Fermentation for the Production of Meroparamycin by Streptomyces sp. strain MAR01

  • The antibiotic meroparamycin was produced in the free culture system of Streptomyces sp. strain MAR01. Five solid substrates (rice, wheat bran, Quaker, bread, and ground corn) were screened for their ability to support meroparamycin production in solid-state fermentation. In batch culture, wheat bran recorded the highest antibacterial activity with the lowest residual substrate values. The highest residual substrate values were recorded for both ground corn and Quaker. On the other hand, no antibacterial activity was detected for rice as a solid substrate. The use of the original strength of starch-nitrate medium in the solid-state fermentation gave a lower antibacterial activity compared with the free culture system. Doubling the strength of this medium resulted in the increase in the activity to be equivalent to the free culture. The initial pH (7.0) of the culture medium and 2 ml of spore suspension (1 ml contains $5{\times}10^{9}spores/ml$) were the optima for antibiotic production. The water was the best eluent for the extraction of the antibiotic from the solid-state culture. Ten min was enough time to extract the antibiotic using a mixer, whereas, 60 min was required when shaking was applied. Semicontinuous production of meroparamycin using a percolation method demonstrated a more or less constant antibacterial activity over 4 runs ($450-480{\mu}g/ml$). The semicontinuous production of the antibiotic was monitored in a fixed-bed bioreactor and the maximum activity was attained after the fourth run ($510{\mu}g/ml$) and the overall process continued for 85 days.

  • The growth characteristics and production of color pigments by Monascus strains were investigated during liquid culture, and production of monacolin K in red mold rice was carried out by solid state fermentation. Four different Monascus ruber strains were cultured in potato dextrose yeast extract broth (PDYB) media at $25^{\circ}C$ for 15 days. The high producing strain for red pigment was not corresponded to the strain for yellow pigment. Production of red pigment was high in the strain causing the fast pH change in culture broth. Production of monacolin K in red mold rice by solid state fermentation was influenced by a combination of wet cell weight and spore density in inoculum by liquid culture. Most strains showed the high production of monacolin K in red mold rice, when submerged fermentation was carried out for 5 days as inoculum for solid state fermentation. These results suggest that submerged fermentation period of inoculum have an effect on the production of monacolin K in red mold rice by solid state fermentation, and monacolin K in red mold rice could be increased by controlling the condition of submerged fermentation for inoculum.

Extraction of β-glucosidase from Bagasse Fermented by Mixed Culture under Solid State Fermentation

  • Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.

글라디올러스 'Topaz' 캘러스의 기관형성에 미치는 배양 조건의 영향 (Effects of Culture Conditions on Organogenesis in Gladiolus 'Topaz' Callus)

  • 본 연구는 글라디올러스 캘러스로부터의 재분화 조건을 구명함으로써 캘러스 배양시스템을 확립하는데 기여하고자 하였다. 글라디올러스 캘러스로부터의 유식물체 획득은 캘러스를 2,4-D 무첨가 1/2 MS고체배지에 치상한 후 24시간 일장하에서 15$^{\circ}C$로 배양하였을 때 효과적이었다. 캘러스로부터의 기관형성에 미치는 배양방법의 영향을 검토한 결과, 부정근의 형성은 액체진탕배양이 그리고 부정아의 형성은 액채정치배양이 각각 우수한 것으로 확인되었다. 본 실험의 결과를 통해 글라디올러스 ‘Topaz' 캘러스로부터의 재분화를 위한 최적 배양 조건을 선발하는 한편, 액체진탕배양법에 의해 기관-캘러스 혼합체를 유도할 수 있었으며, 이를 이용하여 기내 유식물체를 대량생산할 수 있는 가능성을 확인할 수 있었다.

Production of Red Pigments by Monascus purpureus in Solid-state Culture

  • To maximize and sustain the productivity of Monascus pigments, various environmental and nutritional parameters, such as the initial moisture content, pH, inoculum size, sample size, and nutrient supplement, that influence pigment production were evaluated in solid-state cultures as follows: initial moisture content, $50\%;$ pH, 6.0; inoculum size $1\;\times\;10^4$ spore cells $(grams\;of\;dry\;solid\;substrate)^{-1};$ sample size, 300 g. All supplementary nutrients (carbon, nitrogen, and mineral sources) added has inhibitory effects on the cell growth and red pigment production. In open tray culture the maximum biomass yield and specific productivity of red pigments were 223 mg DCW $(grams\;of\;initial\;dry\;substrate)^{-1}$ and, $47.6\;OD_{500}\;(DCW\;grams)^{-1}h^h{-1}$ respectively.

Production of Aerial Conidia of Lecanicillium lecanii 41185 by Solid-State Fermentation for Use as a Mycoinsecticide

  • The production of aerial conidia of Lecanicillium lecanii 41185, a highly virulent fungus, by solid-state fermentation was studied for use as a biocontrol agent against aphids. Among several agro-industrial solid media, steamed polished rice was found to produce the highest amount of aerial conidia. The optimal conditions for aerial conidia production were determined to be a 28.5% moisture content in the rice, 25$^{\circ}C$ culture temperature, rice pH of 6.0, 75% ambient relative humidity, 4-dold seeding culture, 0.6% $KNO_3$, and 12 d of culture time. The conidia yield increased from $5.7\times10^9$ conidia/g polished rice to 18.2 $18.2\times10^9$ conidia/g polished rice following application of these optimized conditions.

항고형암제의 활성평가를 위한 in vitro 삼차원 암세포 배양계의 확립 (Establishment of in vitro 3-Dimensional Tumor Model for Evaluation of Anticancer Activity Against Human Solid Tumors)

  • For the efficient determination of activity against solid tumors, an in vitro tumor model that resembles the condition of in vivo solid tumors, is required. The purpose of this study was to establish a rapid culture method and viability assay for an in vitro 3-dimensional tumor model, multicellular spheroid (MCS). Among 12 human cancer cell lines, a few cell lines including DLD-1 (human colorectal carcinoma cells) formed fully compact MCS which was adequate for in vitro viability assay. DLD-1 MCS showed steady growth reaching $700\;{\mu}m$ diameter after 11 day culture. DLD-1 cells grown as MCS showed significant increase in $G_0/G_1$ phase compared to the monolayer cells (73.9% vs 45.7%), but necrotic regions or apoptotic cells were not observed. The cells cultured as MCS showed resistance to 5-FU (10.3 fold higher $IC_{50}$) compared to monolayers, however, tirapazamine (a hypotoxin) showed similar activity in both culture systems. In summary, MCS may be a valid in vitro model for activity screening of anticancer agents against human solid tumors and also exploitable for studying molecular markers of drug resistance in human solid tumors.

Optimization of in Vitro Cultivation of Inonotus Obliquus

  • This study was performed to get the basic information concerned to the optimum culture condition of Inonotus obliquus. Several solid media, PDA, MEA and Czapek-Dox, and three liquid media were adopted for the in vitro cultivation. Some main features of the fungal morphological characteristics under cultivation conditions were observed and described. Preliminary results showed that appearance of the mycelial mat, hyphal size and substrate pigmentation differed according to the media. The PDA medium was the most favorable substrate for the growth on solid culture, followed by MEA and Czapek-Dox media. Concerned to the addition of amino acids, 5 amino acids, such as alanine, alginine, isoleucine, leucine and threonine, enhanced to the mycelial growth. Isoleucine was shown the best fungal growth. An important morphological hyphal structure for the fungus, the setae, was found in abundance and diverse its shape and size. In liquid culture, fresh potato broth was the best growth stimulant of the fungus, followed by Malt extract and potato broth. Addition of yeast extract to the liquid media had improved the biomass, but not laccase production.
