Mineralogical Evolution of Non-Andic Soils, Jeju Island (제주도 Non-Andic 토양의 광물학적 진화)

  • While about 80% of Jeju soils are classified as Andisols, the soils derived from volcanic ash in Dangsanbong are not Andisols. There is a significant difference of precipitation in localities of Jeju island. The study area is characterized by the lowest amount of annual rainfall in Jeju Island, and by the layered silicates as dominant solid phase in clay fraction. The purpose of this study was to characterize the mineralogy of the non-Andie soils in detail, especially hydroxy-interlayered silicates. Two major soil horizons are recognized in the soil profile developed in the Dangsanbong area, which can be designated as A and C. The soil pH($H_{2}0$), ranges from 6.6 to 7.3 increasing with depth, is higher than that of typical Andisols(pH<6.0). While the pH(NaF), ranges from 9.49 to 9.81, indicates that significant amount of amorphous phases might be present as exchanging complexes. It is estimated to about 1.542.88 wt% by using chemical selective dissolution. The organic content of surface horizon is about 2 wt%. This soil are composed of quartz, feldspar and olivine as major constituents with minor of silicate clays. Quartz is frequently observed in A and distinctly decreases in its amount with depth, while olivine is dominant phase in C and rarely observed in A. In the <0.2$\mu\textrm{m}$ size fraction, smectite and kaolinite/smectite interstratification are dominant with minor of illite. The amounts of smectite decrease with depth, while the amounts of kaolinite/smecite interstratification increase with depth, which indicates the trend of mineral transformation with increasing the degree of weathering. The proportion of kaolinite in kaolinite/smectite interstratification is about 85%, and is not changed significantly through the profile. In the 2-0.2$\mu\textrm{m}$size fraction, vermiculite, smectite, illite and kaolinite are major components with minor of chlorite. Most of chlorite are interstratified with smectite. Chlorite which is not interstratified with smectite occurs only in surface horizon. The proportion of the chlorite in the chlorite/smectite interstratification is 59-70(%) and increases with depth. Hydroxy-interlayered vermiculite(HIV) with hydroxy-Fe/AI in their interlayers occurs in both A and C horizon. The amounts of hydroxy-Fe/AI decrease with depth. Hydroxy-interlayered smectite(HIS) of which interlayers might be composed of hydroxy-Mg/Al occurs only in C horizon. As the results of mineralogical investigation for the soil profile in the study area, clay minerals might be changed and evolved through the following weathering sequences: 1) Smectite Kaolinite, HIS, Vermiculite, 2) Vermiculite HIV Chlorite.

Effect of Germination on the Quality and Amino acid Composition of Soymilk (대두발아(大豆發芽)가 대두유(大豆乳)의 품질(品質) 및 아미노산(酸) 조성(組成)에 미치는 영향)

  • Changes in the quality and amino acid composition of soymilk prepared from soaked and germinated soybeans were investigated. Soybeans were soaked in water for 3 hrs and germinated at $18{\pm}1^{\circ}C$ for 5 days followed by water extraction at room temperature, and then the soymilk was boiled for 30 min. The initial yields of total solid and protein after soaking were 80.7% and 88.6%, respectively and decreased slowly during germination. A slow decrease in lipids and a rapid reduction in total sugar content were found during germination. The change in protein fraction content of soymilk showed an initial increase followed by a gradual decrease. The intrinsic viscosity increased rapidly after 3 days of germination to maximum value at 4th day, then decreased. The amino acid composition of protein fraction of soymilk showed little change while that of nonprotein fraction changed significantly. After 4 days of germination, aspartic acid and alanine increased more than twice, and methionine and tyrosine decreased to half of their initial composition in soymilk prepared from soaked soybean.

  • Background: NF-$\kappa$B is a characteristic transcriptional factor whose functional activity is determined by post-translational modification of protein and subsequent change of subcellular localization. The involvement of the NF-$\kappa$B family of the transcription factors in the control of such vital cellular functions as immune response, acute phase reaction, replication of certain viruses and development and differentiation of cells has been clearly documented in many previous studies. Several recent observations have suggested that the NF-$\kappa$B might also be involved in the carcinogenesis of some hematological and solid tumors. Investigating the possibility that members of the NF-$\kappa$B family participate in the molecular control of malignant cell transformation could provide invaluable information on both molecular pathogenesis and cancer-related gene therapy. Method: To determine the expression patterns and functional roles of NF-$\kappa$B family transcription factors in human lung cancer cell lines NCI-H792, NCI-H709, NCI-H226 and NCI-H157 were analysed by western blot, using their respective antibodies. The nuclear and the cytoplasmic fraction of protein extract of these cell lines were subsequently obtained and NF-$\kappa$B expression in each fraction was again determined by western blot analysis. The type of NF-$\kappa$B complex present in the cells was determined by immunoprecipitation. To detect the binding ability of cell-line nuclear extracts to the KB consensus oligonucleotide, electrophoretic mobility shift assay(EMSA) was performed. Results: In the cultured human lung cancer cell lines tested, transcription factors of the NF-$\kappa$B family, namely the p50 and p65 subunit were expressed and localized in the nuclear fraction of the cellular extract by western blot analysis and immunocytochemistry. Immunoprecipitation assay showed that in the cell, the p50 and p65 subunits made NF-$\kappa$B complex. Finally it was shown by Electrophoretic Mobility Shift Assay(EMSA) that nuclear extracts of lung cancer cell lines are able to bind to NF-$\kappa$B consensus DNA sequences. Conclusion: These data suggest that in human lung cancer cell lines the NF-$\kappa$B p50/p65 complex might be activated. and strengthen the hypothesis that NF-$\kappa$B family transcription factors might be involved in the carcinogenesis of human lung cancer.

Evaluation of Treatment Efficiencies of Pollutants under Different Pollutant Fractions in Activated Sludge-Constructed Wetland System for Treating Piggery Wastewater (활성슬러지-인공습지 공법에 의한 돈사폐수처리시스템에서 오염물질의 분획별 정화효율 평가)

  • BACKGROUND: To design and develop a constructed wetland for effective livestock wastewater treatment, it is necessary to understand the removal mechanisms of various types of pollutants in constructed wetlands. The objective of this study was to evaluate the treatment efficiency of pollutants under different types of fraction in constructed wetland system for treating piggery wastewater. METHODS AND RESULTS: The piggery wastewater treatment plant that consisted of activated sludge tank, aerobic and anaerobic beds was constructed. The concentration of COD(Chemical oxygen demand) in effluent by fraction was 71.5 mg/L for soluble COD, 142 mg/L insoluble COD. The concentration of SS(Suspended solid) in effluent by existing form was 102 mg/L for volatile SS, 15.5 mg/L for fixed SS. The concentration of T-N(Total nitrogen) and T-P(Total phosphorus) in effluent by existing form were 12.8 mg/L and 3.05 mg/L for dissolved form, 35.0 mg/L and 1.93 mg/L for suspended form. The removal efficiencies of COD, SS, T-N and T-P in hot season(summer and autumn) were higher than those in cold season(spring and winter). The removal efficiencies of COD, SS, T-N and T-P in effluent were 98.0, 99.0, 98.2 and 99.2% for all seasons, respectively. CONCLUSION(S): The results indicated that removal types of pollutants were insoluble COD(ICOD), volatile SS(VSS), dissolved T-N(DTN) and dissolved T-P(DTP) in constructed wetlands for treating piggery wastewater.

Determination of Petroleum Aromatic Hydrocarbons in Seawater Using Headspace Solid-Phase Microextraction Coupled to Gas Chromatography/Mass Spectrometry (HS-SPME-GC/MS를 이용한 해수 내 유류계 방향족탄화수소 분석법)

  • The headspace solid-phase microextraction (HS-SPME) followed by gas chromatography/mass spectrometry procedure has been developed for the simultaneous determination of petroleum aromatic hydrocarbons such as benzene, toluene, ethylbenzene and xylene isomers (BTEX) and polycyclic aromatic hydrocarbons (PAHs) in seawater. The advantages of SPME compared to traditional methods of sample preparation are ease of operation, reuse of fiber, portable system, minimal contamination and loss of the sample during transport and storage. SPME fiber, extraction time, temperature, stirring speed, and GC desorption time were key extraction parameters considered in this study. Among three kinds of SPME fibers, i.e., PDMS ($100{\mu}m$), CAR/PDMS ($75{\mu}m$), and PDMS/DVB ($65{\mu}m$), a $65{\mu}m$ PDMS/DVB fiber showed the most optimal extraction efficiencies covering molecular weight ranging from 78 to 202. Other extraction parameters were set up using $65{\mu}m$ PDMS/DVB. The final optimized extraction conditions were extraction time (60 min), extraction temperature (50), stirring speed (750 rpm) and GC desorption time (3 min). When applied to artificially contaminated seawater like water accommodated fraction, our optimized HS-SPME-GC/MS showed comparable performances with other conventional method. The proposed protocol can be an attractive alternative to analysis of BTEX and PAHs in seawater.

Safety and Anticancer Effects of Platycodon grandiflorum Extracts (도라지 추출물의 안전성 및 항암 효과)

  • This study investigated the antimutagenic and anticancer effects of Platycodon grandiflorum extract (PGE) and its fractions against carcinogenic N-nitrosodimethylamine (NDMA) and genotoxicity. The Ames Salmonella mutagenicity test employing histidine mutants of Salmonella Typhimurium TA98 and TA100 was used to examine the mutagenicity of PGE and its fractions. Bacterial reversion assay with S. Typhimurium TA98 and TA100 did not show a significantly increased number of revertant colonies. The same test was used to examine the ability of PGE and its fractions to prevent acquisition of N-methyl-N'-nitro-N-nitrosoguanidine- and 4-introquino-line-1-oxide-induced mutations. PGE and its fractions inhibited mutagenesis in a dose-dependent manner. Among the fractions, ethyl acetate fraction from PGE (PGEA) exhibited a higher antimutagenic effect than other fractions. PGE and its fractions suppressed the growth of cancer cell lines, including human cervical adenocarcinoma, human hepatocellular carcinoma, human breast adenocarcinoma, human lung carcinoma, and transformed primary human embryonic kidney cells. In addition, we evaluated the antitumor activity of PGEA and its fractions in sacorma-180 solid tumor-bearing mice. In vivo anticancer activity results showed that PGE and its fractions could more effectively suppress tumor growth than the control. PGEA showed higher in vitro and in vivo anticancer effects than PGE and other fractions, and PGEA inhibited NDMA formation. Thus, we showed that PGEA has antimutagenic and anticancer activities, making it a candidate anticancer material under these experimental conditions.

Development of Chemiluminescence Immunoassay(CIA) & ELISA for the Detection of Anti-sperm Antibodies in Male Serum (항정자 항체 검출을 위한 CIA 및 ELISA 개발을 위한 기초 연구)

  • New immunoassay systems for the detection of anti-sperm antibodies were developed. For this, sperm surface protein was purified by the immunoaffinity column prepared by the coupling of rabbit anti-human IgG antibodies to Sepharose-4B. Fraction eluted by tris-HCI buffer containing SDS showed a single band having molecular weight of about 60KD on electrophoresis. Enzyme HRP labelled goat anti-human IgG and chemiluminescence aminobutylethyl-isoluminol(ABEI) labelled rabbit anti-human IgG were used for ELISA and CIA, respectively. These two labelled conjugate bound well with human IgG. When serum dilution curves were made to titrate positive serums, two kinds of curves with steep and sluggish slopes were obtained Serum samples were categorized into 3 groups: positive, weak positive and negative based on slope of curve and O.D. values at 1:160 dilution of serum. When ELISA and CIA were compared to conventional method Kibrick test by the determinations of 62 male serums with different diagnosis, the results of ELISA and CIA agreed well, but both disagreed with that of Kibrick test. This study showed that purified sperm surface antigen can be used to develope solid-phase immunoassay systems such as ELISA and CIA which may eliminate the problems encounted the immobilization of living sperm in other tests.

Thermophilic Anaerobic Biodegradability of Agro-industrial Biomass (농축산바이오매스 고온 혐기성 생분해도 평가)

  • Anaerobic digestion(AD) is the most promising method for treating and recycling of different organic wastes, such as organic fraction of municipal solid waste, household wastes, animal manure, agro-industrial wastes, industrial organic wastes and sewage sludge. During AD, i.e. organic materials are decomposed by anaerobic forming bacteria and fina1ly converted to excellent fertilizer and biogas which is a mixture of carbon dioxide and methane. AD has been one of the leading technologies that can make a large contribution to produce renewable energy and to reduce $CO_2$ and other green-house gas(GHG) emission, it is becoming a key method for both waste treatment and recovery of a renewable fuel and other valuable co-products. Currently some 80% of the world's overall energy supply of about 400 EJ per year in derived from fossil fuels. Nevertheless roughly 10~15% of this demand is covered by biomass resources, making biomass by far the most important renewable energy source used to date. The representative biofuels produced from the biomass are bioethanol, biodiesel and biogas, and currently biogas plays a smaller than other biofuels but steadily growing role. Traditionally anaerobic digestion applied for different biowaste e.g. sewage sludge, manure, other organic wastes treatment and stabilization, biogas has become a well established energy resource. However, the biowaste are fairly limited in respect to the production and utilization as renewable source, but the plant biomass, the so called "energy crops" are used for more biogas production in EU countries and the investigation on the biomethane potential of different crops and plant materials have been carried out. In Korea, with steadily increasing oil prices and improved environmental regulations, since 2005 anaerobic digestion was again stimulated, especially on the biogasification of different biowastes and agro-industrial biomass including "energy crops". This study have been carried out to investigate anaerobic biodegradability by the biochemical methane potential(BMP) test of animal manures, different forage crops i.e. "energy crops", plant and industrial organic wastes in the condition of thermophilic temperature, The biodegradability of animal manure were 63.2% and 58.2% with $315m^3CH_4/tonVS$ of cattle slurry and $370m^3CH_4/tonVS$ of pig slurry in ultimate methane yields. Those of winter forage crops were the range 75% to 87% with ultimate methane yield of $378m^3CH_4/tonVS$ to $450m^3CH_4/tonVS$ and those of summer forage crops were the range 81% to 85% with ultimate methane yield of $392m^3CH_4/tonVS$ to $415m^3CH_4/tonVS$. The forge crops as "energy crops" could be used as good renewable energy source to increase methane production and to improve biodegradability in co-digestion with animal manure or only energy crop digestion.

Oxygen Permeation and Mechanical Properties of La0.6Sr0.4Co0.2Fe0.8O3-δ Membrane with Different Microstructures (미세구조에 따른 La0.6Sr0.4Co0.2Fe0.8O3-δ 분리막의 산소투과 및 기계적 특성)

  • Oxygen permeability and the mechanical properties of mixed ionic-electronic conductive $La_{0.6}Sr_{0.4}Co_{0.2}Fe_{0.8}O_{3-{\delta}}$ perovskite-type membrane, fabricated by solid state reaction, were investigated with regard to microstructure. The microstructure of the membrane was controlled by changing the sintering temperature and holding time. The average grain size and relative density were evaluated as a function of sintering conditions. As the fraction of grain boundary decreased, oxygen permeability showed a tendency to increase. Especially the maximum oxygen flux of 0.37 ml/$cm^2$${\cdot}$min was measured for the specimen sintered at 1300${\circ}C$ for 10 h, which has high density and relatively large grain size. Fracture strength was dependent on the relative density of sintered body, while fracture toughness increased with average grain size.

Gas Hydrate Exploration by using PCS(Pressre Core Sampler): ODP Leg 204 (압력코어를 이용한 가스 하이드레이트 탐사: ODP Leg 204)

  • Natural gas in deep sediment may occur in three phases based on the physical and chemical conditions. If the concentration of gas in pore water is less than the solubility, gas is dissolved. If the concentration of gas is greater than its solubility (water is saturated or supersaturated with gas), gas occurs as a fee gas below the gas hydrate stability Lone (GHSZ) and is present as solid hydrate within the GHSZ. The knowledge of gas concentration in deep sediment appears critical to determine the phase of natural gases and to understand the formation and distribution of gas hydrate. However, reliable data on gas concentration are usually available only from the upper section of marine sediment by the headspace gas technique, which is widely used for sampling of gases from the sediments. The headspace gas technique represents only a fraction of gases present in situ because sediments release most of the gases during recovery and sampling. The PCS (Pressure Core Sampler) is a downhole tool developed to recover a nominal $1{\cal}m$ long, $4.32{\cal}cm$ diameter core containing $1,465cm^3$ of sediment, pore water and gas at in situ pressure up to 68.9 MPa. During Leg 204, the PCS was deployed at 6 Sites. In situ methane gas concentration and distribution of gas hydrate was measured by using PCS tool. Characteristics of methane concentration and distribution is different from site to site. Distribution of gas hydrate in the study area is closely related to characteristics of in situ gas concentration measured by PCS.
