Potato Scab Caused by Streptomyces acidiscabies (Streptomyces acidiscabies에 의한 감자 더뎅이병)

  • Bacterial species isolated from common scab lesion on potato (Solanum tuberosum L. cv. Dejima) was identified as Streptomyces acidiscabies. This organism had flexuous spore chains and white spore mass color, produced melanin pigment on tyrosine agar medium but did not produce on peptone agar medium. S. acidiscabies grew on agar medium at pH 4.0, used L-arabinose, D-fructose, D-glucose, D-mannitol, rhamnose, sucrose, D-xylose and meso-inositol except reffinose as carbon sources. It was also susceptible to thallium acetate (10 $\mu\textrm{g}$/ml, 100$\mu\textrm{g}$/ml), phenol (0.1%, wt/vol), streptomycin (20 $\mu\textrm{g}$/ml), and was resistant to 7% NaCl, crystal violet (0.5 $\mu\textrm{g}$/ml), penicillin (10 IU/ml) and oleandomycin (25 $\mu\textrm{g}$/ml, 100 $\mu\textrm{g}$/ml).

Expression Patterns of CaMV 35S Promoter-GUS in Transgenic Poatoes and Their Clonal Progenies

  • Two potato (Solanum tuberosum L.) cultivars were transformed by Agrobacterium tumefaciens harboring cauliflower mosaic virus (CaMV) 35S promoter and $\beta$-glucuronidase (GUS) gene. Expression patterns of the CaMV 35S promoter according to tissue types and developmental stages, and genetic stability of GUS gene were investigated in the clonal progenies of transgenic potatoes. Kanamycin-resistant shoot emerged from tuber disc after 4 weeks of culture, and root was induced 6 weeks after culture on the selection medium. Shooting frequency of cvs. Superior and Dejima were 43% and 27%, respectively. Mature transformants and their clonal progenies showed no phenotypical abnormality. GUS activity was expressed primarily at parenchymatous cells of phloem tissue around the vascular cambium in the stem and root, and higher activity was found at the apical meristem of shoot, root and adventious shoot bud. GUS activity was higher at tubers of young explants than at stored tubers. These facts indicate that expression level of the CaMV 35S promoter differed according to tissue types and developmental stages of the organs. The GUS gene was stably inherited to each clonal progeny and normally expressed.

Influence of Polyethylene Film Mulch and Seedling Types on Growth and tuber Yield of Fall-Grown Potato (폴리에틸렌필름 멀칭 및 종묘종류가 가을감자의 생육과 수량에 미치는 영향)

  • Normal seed tubers and plug seedlings raised from stem cutting and mini-tubers ($10\pm3$ g) were planted or transplanted on August 20,2001 in bare soil or under transparent and black polyethylene film mulches to evaluate the effects of mulch material and seedling type on the growth and tuber yield of fall-grown potato (Solanum tuberosum L. cv. Dejima) in Jeju region. Total tuber number per plant was 4.6 regardless of mulch treatment, and direct planting seed potatoes resulted in the greatest tubers (5.14)/plant and followed by transplanting seedlings raised from cutting and mini-tubers (4.15 and 5.15 tubers/plant). Transparent and black polyethylene film mulches increased total tuber yield by 16 and 8%, respectively, and average tuber weight by 14 and 12%, respectively, compared with no mulch (21.16 t/ha and 72.2 g/tuber). Transplanting plug seedlings raised from cutting resulted in the greatest tuber yield, followed by transplanting plug seedlings raised from mini-tubers and direct planting seed potatoes.

Improvement of Growth of Potato (Solanum tuberosum L. cv. Dejima) Plants at In Vitro and Ex Vitro and Energy Efficiency by Environmental Control with Growth Stage in Photoautotrophic Micropropagation System (광독립영양 기내 미세증식 시스템에서 생육단계별 환경조절을 통한 감자의 기내 및 기외 생육과 에너지 효율 향상)

  • This study was conducted to evaluate the effect of optimized environment conditions with growth stage in photoautotrophic micropropagation on the growth of potato (Solanum tuberosum L. cv. Dejima) plantlets and energy efficiency. Optimum environment conditions at each stage were decided in our previous study. For the evaluation of optimized environment control, potato plantlets were cultured under four different conditions: photoautotrophic optimum conditions of photosynthetic photon flux density (PPFD) and $CO_2$ levels with growth stage (POG), photoautotrophic constant condition with average PPFD and $CO_2$ levels (PCA), photoauototrophic constant condition with maximum PPFD and $CO_2$ levels (PCM), and photomixotrophic conventional condition with 3% sucrose (PMC) as control. As a result, environment control with growth stage (POG) significantly promoted all the growth characteristics such as the number of nodes and unfolded leaves, shoot height, shoot diameter, and fresh and dry weights of potato grown in vitro. In addition, based on dry weight consumed electricity and $CO_2$ were the lowest in POG suggesting the highest energy efficiency among the treatments. After transferring potato plantlets to greenhouse, the plantlets under POG showed vigorous growth, which was pretty similar with those under PMC. The accumulations of dry matter in POG were 4.7 times in vitro and 3.8 times in greenhouse as much as those in the conventional control (PCM). Thus, we concluded that in vitro environment control with growth stage induced vigorous growth of potato plantlets both in vitro and in greenhouse with less energy consumption.

Effects of Tuber Size on Field Growth and Yield of 'Dejima' Potatoes Produced through Wick-based Hydroponics (심지양액재배 '대지'의 씨감자 크기에 따른 포장 생육과 수량)

  • Two field experiments were carried out in 2002 to evaluate effects of the sizes of tubers produced through a wick-based hydroponics on growth and yield of seed potatoes (Solanum tuberosum L. 'Dejima'). Nine sizes of seed tubers (<1, 1~3, 3~5, 5~7, 7~10, 10~20, 20~30, 30~50, and 50~80 g) were used in spring crop. In fall crop, however, tubers except for the 1 g and 50~80 g wereused with tubers 7 and 15 g produced through aeroponics. Emergence rate greatly increased as tuber size was increased up to 10~20 g and then leveled off in both croppings. The number of days from planting to emergence in spring crop was about 34 days regardless of tuber size, while that in fall crop decreased from 34 to 16 days as tuber size increased from 1~3 to 30~50 g. The number of stems per plant, stem length and diameter, and leaf size tended to increase with tuber size. As the seed tuber size increased up to 10~20 g, the number of tubers per plant, average tuber weight, total and seed tuber yields linearly increased and then slowly increased with seed tuber size. Total and seed potato yields were not significantly different between tubers produced through a wick-based hydroponics and an aeroponics when similar sized tubers were planted in the fall cropping.

Effects of Wick Number on Growth and Yield of Seed Potatoes Grown in a Wick-based Hydroponics (심지수의 차이에 따른 심지양액재배 씨감자의 생육 및 수량)

  • A glasshous experiment was carried out in 2002 to evaluate growth differences between two wick types, and to determine the optimum number of wicks in a wick-based hydroponics for production of 'Dejima' seed potatoes (Solanum tuberosum L.). The six minitubers ($7.0{\pm}0.2g$/tuber) produced through aeroponics were planted in a polystyrene box (51 cm long $\times$ 31 cm wide $\times$ 20 cm high) containing growth medium (perlite+peatmoss 1:2, v/v mixtures). Before the boxes were filled with the medium, 6 wicks per box were vertically and 2 to 10 wicks were horizontally installed, respectively, at the bottom of the boxes through holes. Comparing with 6 vertical wick treatment, 6 horizontal wick treatment increased the number of tubers per plant, but decreased average tuber weight. Yield of tubers greater than 5 g (seed potatoes) was not significantly affected by wick type. As the number of the horizontal wicks per box was increased from 2 to 6, the number of seed potatoes increased from 2.8 to 8.0/plant and then leveled off. Average tuber weight increased with increasing the number of the wicks up to 8 and then leveled off. As the number of the wicks per box was increased from 2 to 6, seed potato yield increased from 1.81 to 6.59 kg/$m^2$ and then slowly increased up to 10 wicks per box, reaching 12.9 kg/$m^2$. The results indicate that installing 8 wicks per box ($0.16m^2$) horizontally for the wick culture system in production of 'Dejima' seed potatoes is beneficial considering seed potato yield, cost of wicks, and convenient filling with medium.

Growth and Yield According to Wick Number Under Wick Culture System of Potato Plug Seedlings (감자 플러그묘 심지재배에서 심지수에 따른 상위급종서의 생산성)

  • This study was conducted in 2001 to develop a technique for mass production of superior seed tuber by using a wick culture system as an alternative plan to solve problems in hydroponic culture of potato. Potato (Solanum tuberosum L. cv. Dejima) plug seedings were planted on a styrofoam bed (volume $0.032\textrm{m}^3$) containing two different media (Jeju scoria + peatmoss and perlite + peatmoss, (1:2, v/v) mixture, respectively). Each bed was equipped with various number of wicks (2, 4, 6, 8 and 10 ea/bed). After harvest, the total porosity of both media was 83-85%. Bulk density of Jeju scoria + peatmoss was higher compared to that of perlit + peatmoss. However, the number of wicks did not influence to the bulk density. Conversely, moisture capacity of Jeju scoria + peatmoss was remarkably greater than that of perlite + peatmoss and was increased from 55 to 70% as elevating number of wicks. Although the number of wicks resulted in the areal growth of plants, the total yield and the weight of tuber were highest at the bed equipped with six wicks in both media.

Effects of Foliar Application of Chitosan and Seedling Types on Increasing the Production of Fall-growing Potatoes(Solanum tuberosum) in Jeju Island (Chitosan 처리와 종묘종류가 가을감자의 생산성 향상에 미치는 영향)

  • This study was conducted to investigate the effect of foliar application of Chitosan and seedling types on increasing the production of fall-growing potatoes(Solanum tuberosum) at the height of 250m from the sea in Jeju Island. The experiment main-plots consisted of four Chitosan applications, and subplots had three seedling types as cutting plug seedlings, mine-tuber plug seedling planting plot and seed potatoes planting plot. The foliar application of Chitosan of 2000ppm was carried out 0, 1, 3, 5 times at intervals of ten days after ten days of transplanting. Plant height and top dry weight were significantly more vigorous as three to five application of Chitosan. In the small tubers(under 80g), the number of tubers and tuber yield were relatively increased in the seed potatoes planting plot and mine-tuber plug seedling plot, but the large tubers(over 80g) was higher in the cutting plug seedling planting plot. T-N content in leaves was increased as the number of application of Chitosan was increased. A similar tendency was shown in K, P Ca, Mg and Na. The growth and tuber yield in cutting plug seedlings, mine-tuber plug seedling planting plot and seed potatoes planting plot were effectively increased as three to five foliar applications of Chitosan was increased.

Effects of Temporary pH Reductions of Solution on Tuberization of Potato(Solanum tuberosum L.) in Hydroponics (배양액의 pH저하 처리에 의한 감자소괴경 형성 촉진)

  • The purpose of this study was to evaluate the effect of temporary pH reductions on tuberization of ‘Dejima’potato in aeroponics. The pH and EC of nutrient solution were adjusted to 6.0∼6.5 and 1.2mS/cm, respectively. On 35th day after planting, plants were subjected to pH 3.0, 4.0, 5.0 and 6.5 for 10hrs. After 5 days mini-tuberization was shown in pH 3.0 treatment and was significantly increased up to 20 days. Temporary low pH treatment resulted in the increase of stolen formation and of tuber dry weight. Number of mini-tubers per plant on 90th-day after planting was 72.1, 69.8, 65.2, and 60.3 in pH 3.0, 4.0, 5.0 and 6.5 respectively.

Mass Propagation of Plug Seedling using Stem Cutting and Their Tuber Yield in Potato

  • For the mass production of plug seedlings in cultivar ‘Dejima’ potato (Solanum tuberosum L.) the optimal apical cutting diameter for rooting and rapid multiplication of stem cuttings in hydroponics were determined. In addition, the best planting date was predicted to increase tuber yield of plug seedlings at fall cropping in Cheju-Do, Korea. Days to initial rooting decreased as the cutting diameter was reduced. Plant height, leaf number, root length and root weight per plant were favorable as the cutting diameter was small. The ideal cutting diameter was 1-2 mm in this experiment. In the hydroponic cultures, the Japanese standard (JS) nutrient solution was the most effective for multiplication of stem cuttings. It was able to propagate more than 20 times a month from a single mother plant. Viability of plants, which were derived from plug seedlings using stem cuttings, was excellent when transplanted to the field. The number of tubers and tuber yield in both of the plug seedlings and seed potato planting plots were high when planted on 25 August. The number and yield were reduced when planted on 15 August, 5 September and 15 September. The degree of decrease of tuber yield in the plug seedling planting plot however, was lower than that of seed potatoes when the planting date was late. In the case of small tubers (under 30 g), the number of tubers and tuber yield were evidently increased in the seed potato tuber planting plot; the yield of large tuber (over 80g) in the plug seedling planting plot was higher than that of the seed potato. The total tuber yield per plant in the plug seedling planting plot was less than that of the seed potato; therefore, in order to increase tuber yield it was necessary to increase field plant density.
