Hepatoprotective Activity of Scopoletin, a Constituent of Solanum lyratum

  • Scopoletin (7-hydroxy-6-methoxycoumarin), a coumarin, was isolated from the aerial part of Solanum lyratum Thunb. by the activity-guided fractionation employing carbon te trachloride-intoxicated primary cultured rat hepatocytes as a screening system. Its hepatoprotective activity was first evaluated by measuring the release of glutamic pyruvic transaminase and sorbitol dehydrogenase from carbon tetrachloride-intoxicated rat hepatocytes into the culture medium. Scopoletin significantly reduced the releases of glutamic pyruvic transaminase and sorbitol dehydrogenase from the carbon tetrachloride-intoxicated primary cultured rat hepatocytes by 53% and 58%, respectively, from the toxicity in a dose-dependent manner over concentration ranges of 1mcM to 50mcM. Further studies revealed that at the concentration of 10mcM, scopoletin significantly preserved glutathione content by 50% and the activity of superoxide dismutase by 36% and also inhibited the production of malondialdehyde to the degree as seen in the control.

민간약 "가지"의 생약학적 연구 (Pharmacognostical Studies on the Folk Medicine 'GaJi')

  • Korean folk medicine 'GaJi' has been used to treat a boil, cough, mushroom poisoning and stomach cancer. The botanical origin of the crude drug has never been studied pharmacognostically. To clarify the botanical origin of 'GaJi', the morphological and anatomical characteristics of the stems of Solanum species growing in Korea, i.e. S. japonense Nakai, S. lyratum Thunb., S. melongena L., S. nigrum L. were compared. As a result, it was determined that GaJi was the stem of Solanum melongena L.

  • A new biflavonol glycoside named as solanoflavone (1) was isolated from aerial part of Solanum melongena. The chemical structure was elucidated as isorhamnetin-3-O-$\beta$-D-glucopyranoside-(4'$\to$O$\to$4"')-galangin-3"-O-$\to$-D-glucopyranoside on the basis of physicochemical and spectroscopic methods, including 2D NMR spectral techniques.

  • Solanum lyratum (Solanaceae) has been used as a traditional analgesic, antipyretic and hepatoprotective agents in Korea. In this study, we investigated the hepatoprotective effect of ethylacetate extract of Solanum lyratum (SL) on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of SL (150, 300 mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase levels.   • A phenolic glucoside and two steroidal sapogenins were isolated from the aerial part of Solanum Iyratum (Solanaceae). They were identified as 2-hydroxy-3-methoxybenzoic acid glucose ester (1), ${\Delta}^{3,5}$-deoxytigogenin ((25R)-spirosta-3,5-diene) (2), diosgenin (($3{\beta}$, 25R)-spirost-5-en-3-ol) (3), respectively by several spectroscopic methods including IR, MS and NMR. Compound 1 has not been previously isolated from Solanaceae plants. Compounds 2 and 3 are isolated from S. Iyratum for the first time in the present study.

  • Solanum andigena의 광주성 주기중의 방사성 콜레스테롤의 대사를 연구하였다. 대사산물로서 방사성 솔라닌과 그 가수분해물 솔라니딘을 분리 결정화하였으며, 콜레스테롤은 이 식물체 내에서 형성되는 에스테르화된 형태로 분리하였다. 단일 및 장일식물간의 콜레스테롤을 대사에는 뚜렷한 차이는 찾아 볼 수 없으나. 괴경중의 스테로이드 알카로이드에 콜레스테롤이 관여하고 있는 것으로 논의되었다.

These compounds never previously isolated from this genus.

  • Cell suspension cultures of Solanum mammosum transformed inoculated salicyl alcohol into salicin $(salicyl\;alcohol\;2-0-{\beta}-D-glucopyranoside)$. The highest level of salicin (59.3 mg/flask) in the cells was formed within 3 days after inoculating with salicyl alcohol (50 mg /flask containing 50 ml medium). The glucosylation capability of salicyl alcohol by cell suspension cultures of S. mammosum was relatively higher than that reported previously.

까마중 추출물을 이용한 천연 염색직물의 염색성 및 항균성 (A Study on the Dyeability and Antibiosis of Fabrics Dyed with Solanum Nigrum Extract)

  • The purpose of this study was to identify the utility value with material of natural dyeing and to examine the dyeability and antibiosis of fabrics dyed with Solanum nigrum extract. The results obtained are as follows. First, the color tone of dyed fabrics was tinged with from yellow series to green series, in the chrominance the fabric dyed with Fe mordant was showed the highest value. Second, colorfastness to dyeing showed difference as to mordants, but colorfastness to dyeing except for sunlight colorfastness showed the good colorfastness result on the whole. Third, in the test results of antibiosis, the dyed both cotton and silk fabrics showed the very excellent antibiosis effect for bacteria but showed the insignificant effect for fungus.

Plant Regeneration from Mesophyll Protoplasts Culture of Solanum sisymbriifolium

  • The optimal culture conditions were studied for plant regeneration from mesophyll protoplasts of Solanum sisymbriifolium. Axenic seedlings of S. sisymbriifolium were used as a explant for protoplast culture. Many viable protoplasts were isolated by incubating leaf slices in an enzyme solution containing 0.25% Meicerase and 0.05% Macerozyme for 16 hr at $25^{\circ}C$ without shaking. Protoplast density of $5.0{\times}10^4\;ml^{-1}$ in Kao medium containing 5.0 mg/L NAA, 1.0 mg/L 2,4-D and 1.0 mg/L BA was optimal for colony formation. Most colonies were formed when protoplasts were cultured at $25^{\circ}C$ after initial culture at $30^{\circ}C$ for one week. On the MS agar medium with 1.0 mg/L zeatin, 38.4% of protoplast-derived calli differentiated shoots. These shoots rooted on 1/2MS medium with 5.0 g/L sucrose and 2.5 g/L gellan gum, and developed into whole plants.
