The Antioxidant and Antimicrobial Activity of Solanum nigrum L. Fruit Powder by Extraction Solvent (왕까마중(Solanum nigrum L.) 열매 분말의 추출 용매에 따른 항산화 및 항균 활성)

  • This study was conducted in order to investigate the antioxidant and antimicrobial activity of Solanum nigrum L. fruit powder after undergoing different extraction solvent processes. The total phenolic content of Solanum nigrum L. fruit powder measured a 14.66 GAE mg/g after undergoing ethanol extraction, and the total flavonoid content measured at 201.23 mg CE/g when undergoing ethanol extraction. The ABTS radical scavenging activity was 160.38~209.53 TEAC umol/g, and the DPPH radical scavenging activity was 53.99~90.76 TEAC umol/g, which indicated a higher level of antioxidant power in the ethanol extract as opposed to in the water extract. The FRAP (ferric reducing antioxidant power) of Solanum nigrum L. fruit powder was 115.58~194.58 TEAC umol/g, and B. subtilis KCTC 2189 showed greater antimicrobial activity in the ethanol extract (concentration 200 ug/uL) as opposed to the water extract. Solanum nigrum L. fruit powder revealed differences in antioxidant and antimicrobial activity between the different extraction solvents. In particular, ethanol extract had higher antioxidant and antibacterial activity, meaning it is more favorable for usage as a functional food material.

Development of PCR-based markers for selecting plastid genotypes of Solanum hjertingii (Solanum hjertingii 색소체 유전자형 선발을 위한 PCR 기반 분자마커 개발)

  • The tetraploid Solanum hjertingii, a wild tuber-bearing species from Mexico is a relative of potato, S. tuberosum. The species has been identified as a potential source of resistance to blackening for potato breeding. It does not exhibit enzymatic browning nor blackspot which are physiological disorders. However, due to their sexual incompatibility, somatic hybridization between S. hjertingii and S. tuberosum must be used to introduce various traits from this wild species into potato. After somatic hybridization, molecular markers are essential for selecting fusion products. In this study, the chloroplast genome of S. hjertingii was sequenced by next-generation sequencing technology and compared with those of other Solanum species to develop specific markers for S. hjertingii. The chloroplast genome has a total sequence length of 155,545 bp, and its size, gene content, order and orientation are similar to those of the other Solanum species. Phylogenic analysis including 15 other Solanaceae species grouped S. hjertingii with S. demissum, S. hougasii, and S. stoloniferum. After detailed comparisons of the chloroplast genome sequence with eight other Solanum species, we identified one InDel and seven SNPs specific to S. hjertingii. Based on these, five PCR-based markers were developed for discriminating S. hjertingii from other Solanum species. The results obtained in this study will aid in exploring the evolutionary aspects of Solanum species and accelerating breeding using S. hjertingii.

Solanum elaeagnifolium Cav. (Solanaceae), an unrecorded naturalized species of Korean flora (은빛까마중 (Solanum elaeagnifolium Cav.), 우리나라 미기록 귀화식물)

  • We report a naturalized alien species, Solanum elaeagnifolium Cav. from Chodo Isl., Samsanmeon, Yeosushi, Jeollanam-do province. The native distribution range of the species is the southwestern US and the adjacent region of northern Mexico. The species is related to S. nigrum L., but the species can be distinguished from S. nigrum by the silver green stellate trichome on the surfaces of plants, the elongated leaf shape, and the size and color of the flowers and fruits. Solanum elaeagnifolium Cav. belongs to the Solanum subgenus Leptostemonum. The species is recorded on the invasive species list in the subtropical or temperate regions of many European, Mediterranean, African, South American, Asian, and Australian countries. Therefore, we also need a careful monitoring and prevention strategy for this new invasive species in Korea.

A convergence study of antibacterial effect of solanum nigrum extract on candida albicans (까마중 추출물의 Candida albicans에 대한 항균효과에 대한 융합적 연구)

  • The purpose of this study was to antibacterial effect of solanum nigrum extra in candida albicans. In this study, the antibacterial effects of 70% methanol extract and solanum nigraum were investigated. The content of flavonoids and polyphenol contents in solanum nigrum extraction. The antimicrobial activity was determined by spectrophotometric growth inhibition assay(optical density) and colony forming untis (CFU) for the candida albicans. Total contents were flavonoids and polyphenols of solanum nigrum, the result was $8.23{\pm}0.13mg/g$ and $14.97{\pm}1.58mg/g$, respectively. The CFU results showed lower CFU counts in the experimental groups compared to the control group. And the result of the bacterial viability was decreased in the experimental group. Therefore the extracts of solanum nigrum were found to have antimicrobial activity in candida albicans. From the these results, solanum nigrum extracts have a possibility to be a candidate for natural antibacterial components in dental products.

  • Korean folk medicine Bae Pung Dung has been used to cure a cold and jaundice. The botanical origin of the crude drug has never been studied pharmacognostically. To clarify the botanical origin of Bae Pung Dung, the morphological and anatomical characteristics of the leaves and stems of Solanum species growing in Korea, i.e. S. japonense Nakai, S. lyratum Thunb., S. nigrum L. were compared. As a result, it was determined that Bae Pung Dung was the whole plant body of Solanum lyratum and Solanum japonense.

Effects of solanum macrocarpon (African eggplant) on haematological parameters of wistar rats exposed to urban air pollution

  • Our study investigated the effects of Solanum macrocarpon (African eggplant) on haematological and biochemical parameters of male albino rats exposed to urban air pollution ($O_3$, $PM_{10}$, CO, $SO_2$, $NO_2$ and VOC). Male albino Wistar rats were exposed for 63 days either to urban air pollution without treatment (A); oral supplementation with Solanum macrocarpon given at 36 and 75 mg/kg body weight (BW), representing C and D respectively and exposed to air pollution; oral supplementation with Solanum macrocarpon given at 75 mg/kg BW after exposure to air pollution (B); or kept in animal house without exposure to air pollution (E). Animals exposed to air pollution showed significant alterations in haematological and biochemical parameters signaling that the blood and organs were badly injured. There were significant elevations in white blood cells (WBC) and its indices, reduction in red blood cells (RBC) and significant depletions of non-enzymic antioxidants, total protein and increase in lipid peroxidation values. Solanum macrocarpon supplementation in the feed of animals halted significantly the deleterious effects of air pollution, with co-administration during exposure given better results.

Solanum Nigrum Polysaccharide (SNL) Extract Effects in Transplanted Tumor-bearing Mice - Erythrocyte Membrane Fluidity and Blocking of Functions

  • Background: Solanum nigrum L. has been used in traditional Chinese medicine because of its diuretic and antipyretic effects. The present research concerned effects of crude polysaccharides isolated from Solanum nigrum L. on erythrocyte membranes of tumor-bearing $S_{180}$ and $H_{22}$ in mice. Materials and Methods: Fluorescence-labeled red blood cell membranes were used with DPH fluorescence spectrophotometry to examine erythrocyte membrane fluidity, and colorimetry to determine degree of erythrocyte surface membrane blocking. Extent of reaction by tumor-bearing mice with the enzyme erythrocyte membrane bubble shadow detection of red cell membrane variation in the degree of closure before and after administration. Results: Solanum nigrum polysaccharide could significantly improve the $S_{180}$ and $H_{22}$ tumor-bearing mice erythrocyte membrane fluidity, compared with the control group, the difference was significant (p<0.01), SNL can significantly improve the red blood cell membrane and then $S_{180}$ tumor-bearing mice sealing ability, compared with the negative control group, the difference was significant(p<0.05, p<0.01). $H_{22}$ tumor-bearing mice can increase red cell membrane and then sealing ability, the difference was significant (p<0.05). Solanum nigrum polysaccharide degree of fluidity and blocking two transplanted tumors in mice restored the ability to raise the red cell membrane has a significant effect. Conclusions: Solanum nigrum L.-type mice transplanted tumor can affect the red blood cell membrane fluidity and re-closed, through the red cell membrane of red blood cells to enhance the immune function of the possibility of erythrocyte immunity against tumor formation garland provide experimental basis.

  • The effect of thidiazuron on callus growth and shoot regeneration of Solanumspecies was very positive. Increasing concentrations of thidiazuron stimulated callus growth of Solanum ptycanthum and Solanum nigrum. Shoot regeneration of S. ptycanthum and S. nigrum was greater on medium with thidiazuron than that with IAA and BA. Thidiazuron at $0.5{\mu}M$ or above increased the number of shoots regenerated from S. ptycanthum calli compared to the IAA with BA. High concentrations of thidiazuron $( >I{\mu}M)$ increased the number of shoots than BA or low levels of thidiazuron and IAA or BA The addition of IAA to thidiazuron media reduced S. ptycanthum shoot formation.

  • Protopasts were isolated from cultured cells of potato (Solanum tuberosum L.) tuber tissue. The ability of callus formation from the culture cells was higher in cultivars Dejima and Superior than in Shimabara and Irish Cobbler on Lam's medium. Therefore, the former was used as sources for protoplast isolation. Friable calli were transferred to liquid media and cells in exponential phase were used for protoplast isolation. In both of Dejima and Superior, the yield of protoplasts was high in the enzyme solution of 2% Onozuka cellulase and 1% macerozyme. Also, viability of isolated protoplasts was very good. Thus, it seems that these protoplasts would be applicable to various aims of research.
