New records of nine ciliates (Protozoa: Ciliophora) from Korea: Brief descriptions and remarks

  • Based on morphological observation, we identified nine ciliates previously unrecorded in Korea, Euplotidium itoi Ito, 1958, Anteholosticha verrucosa (Foissner & Schade, 2000) Berger, 2008, Holostichides dumonti Foissner, 2000, Amphisiella australis Blatterer & Foissner, 1988, Terricirra matsusakai Berger & Foissner, 1989, Uroleptoides binucleatus binucleatus Hemberger, 1985, Uroleptoides magnigranulosus (Foissner, 1988) Berger, 2008, Leptopharynx costatus gonohymen Omar & Foissner, 2012, Pseudochilodonopsis mutabilis Foissner, 1981, from three sites in Korea, which were isolated from three environmental samples, consisting of one seawater and two soil samples. These ciliates were assigned to three classes, five orders, five families, and eight genera. In this study, we provide photographs and brief descriptions and remarks of the nine species.

  • Four bacterial strains able to degrade dichlorobenzene as the sole source of carbon and energy were isolated from soil by selective enrichment culture, and among them, one isolation was the best in the cell growth and identified as Pseudomonas sp. DCB3 by its morphology and physiological properties. Cell growth dramatically increased in a minimal medium containing 500ppm of dichlorobenzene was not detected any more at 72 hours after cultivation. The optimal temperature and initial pH for the growth of the isolated strain were 30$\circ$C and 7.0, respectively. Cell growth was increased by supplementing $(NH_2)_2CO$ and 50 ppm yeast extract as additional nutrients. Therefore, it was suggested that Pseudomonas sp. DCB3 could be effectively used for the biological treatment of wastewater containing dichlorobenzene.

New Record of Ciliates, Blepharisma undulans and B. steini (Ciliophora: Heterotrichida: Blepharismidae) from Korea

  • Two ciliate species of Blepharisma were collected from the riverine wetland and mountain peak soil in Korea, Blepharisma undulans Stein, 1867 and B. steini Kahl, 1932, which are reported for the first time from Korea. The description was based on the observations of live and protargol impregnation specimens. Diagnostics of these species (Blepharisma undulans vs. B. steini) are as follows. Body length/width ratio in vivo about 6/1 vs. 2/1, shaped in spindle vs. pyriform to ellipsoid and colored in pinkish vs. reddish. Macronuclei 2 vs. 1, with 4-9 vs. 4-7 spherical micronuclei. Adoral zone of membranelles extends from anterior end to 1/3 vs. 1/2-2/3 of body length, with 47-56 vs. 33-45 adoral membranelles. The numbers of somatic kineties are 22-27 vs. 18-22.

A new record of Fissidens protonemaecola Sakurai (Fissidentaceae, Bryophyta) in Korea

  • An unrecorded Fissidens species, Fissidens protonemaecola Sakurai (Fissidentaceae, Bryophyta), was newly discovered in Korea during an investigation bryophyte flora. In this study, we examined the morphology of F. protonemaecola in Korea. This species was found in soil along a train through a shaded backslope of Mt. Bukbyeongsan (elev. 80 m) on Geojedo Island and on Mt. Gogeunsan (Oreum; elev. 280 m) on Jejudo Island, Korea. This species has dwarfish bud-like shoots scattered on persistent protonemata. An important feature of the protonemata is a diagnostic character of F. protonemaecola that clearly distinguishes it from other Fissidens species. Its Korean name is given as "Cho-rok-sil-bong-hwang-i-kki," inferred from the characteristic protonemata of the species.

Redescription of Gonostomum algicola and G. gonostomoida (Ciliophora: Spirotrichea: Sporadotrichida) Unknown from Korea

  • Two rare ciliates from soil under the oak tree and mosses in the top of mountain in Korea were identified as Gonostomum algicola Gellert, 1942 and G. gonostomoida (Hemberger, 1985), respectively. There is little information on their morphological features, therefore their detailed redescriptions are needed. The description was based on the observation of living and protargol impregnated specimens, and biometric analysis. Their diagnostic characteristics are as follows. Gonostomum algicola; $88-113\times30-40{\mu}m$ in vivo, colourless cortical granules, 20-31 adoral membranelles, two fronto-terminal cirri, five fronto-ventral cirri, no mid-ventral cirri, two transverse cirri, two to three micronuceli. Gonostomum gonostomoida; $60-121\times21-40{\mu}m$ in vivo, no cortical granules, 27-34 adoral membranelles, no fronto-terminal cirri, two fronto-ventral cirral rows with each row bearing three cirri, midventral cirral row with 11-14 cirri, two to three transverse cirri, one to six micronuceli. So far, total three species within the genus Gonostomum have been recorded from Korea by the present study.

A report of 10 unrecorded bacterial species of Korea, isolated from agricultural soil in 2022

  • In 2022, research for native prokaryotic species in Korea reported 10 unrecorded bacterial strains affiliated to phyla Actinomycetota, Bacillota, and Pseudomonadota. The strains formed monophyletic clades with the most closely related species (with ≥98.7% sequence similarity) in the 16S rRNA gene sequencing. Among them, four species of the phylum Actinomycetota, two species of the phylum Bacillota, and four species of the phylum Pseudomonadota have not been reported in Korea, suggesting unrecorded species in Korea. Information on strains such as Gram staining reaction, colony and cell morphology, biochemical characteristics, and isolation sources were provided in the species description.

높은 토양 EC에서 자란 오이묘 조직의 행태적 변화 (Morphological Changes of Tissue in Cucumber Seedlings Grown in High Soil EC)

  • 토양 EC가 다른 조건에서 생장시킨 오이묘의 잎과 생장점에 대한 형태적 변화를 관찰하였다. EC가 $1.0dS{\cdot}m^{-1}$에서 $3.0dS{\cdot}m^{-1}$까지 높아질수록 잎 표면의 모용의 수가 많아지고 길이가 길어졌으나 뒷면의 기공형태와 수는 변화가 없었다. EC $1.5dS{\cdot}m^{-1}$에서 자란 오이묘의 표피세포는 대부분 액포가 차지하고 있었드나 EC $3.0dS{\cdot}m^{-1}$에서 자란 것은 핵, 미토콘드리아, 엽록체와 기타 미세구조가 대부분의 공간을 채우고 있었다. 잎의 해면유조직세포는 EC $1.5dS{\cdot}m^{-1}$에서는 세포는 크고 엽록체가 적었으나 EC $3.0dS{\cdot}m^{-1}$에서는 세포와 액포의 크기가 적었고 상대적으로 엽록체함량이 많았다. 잎의 두께는 EC가 증가할수록 얇아졌고 EC $1.5-2.0dS{\cdot}m^{-1}$에서는 책상조직세포가 아주 진한 색을 띄었다. EC $3.5dS{\cdot}m^{-1}$ 이상에서는 생장점 조직의 분화가 정상적으로 일어나지 않았다.

심층토에 있어서 탈질화에 의한 $N_2 O$ 방출의 평가 (Review of Nitrous Oxide Emission by Denitrification in Subsurface Soil Environment)

  • 현재 지구상에서 농업에 기인하여 배출되는 $N_2$O의 80% 정도가 지구의 온난화 뿐만 아니라 오존층 파괴에까지 영향을 미친다. 토양에서 수분함량 등과 관련한 유기태 탄소는 지하수면의 계절적 변화에 따라 탈질화를 결정하는 주요 요인이 되기도 하며 심층토의 탈질화 활동은 토양내 유기물을 분해하여 유기태 질소를 일시적으로 토양에 축적시키기도 한다. 그리고 토양의 관리방법, 폐기물의 토양처리, 질소질 비료의 시용 등이 $N_2$O 증가에 결정적 요인이 되기도 한다. 그러나 이러한 효과의 정도는 거의 알려져 있지 않을 뿐만 아니라 질산화나 탈질화와 같은 상반되는 과정과 제한 요소와 관련하여 범용적으로 적용할 수 있는 $N_2$O 방출을 예측하는 측정 계수와 같은 연구는 매우 미미한 상태이다. 그러므로 농업토양에서 비료와 유기물 시용 둥에 의해 발생하는 토양의 $N_2$O배출을 효율적으로 관리하기 위하여 심층토에서의 동적 역학적 $N_2$O 배출 측정과 관리 방법을 개발하여야 한다.

전라남도 나주시 동강면 일대 황토(풍화잔류토)의 토양학적 및 광물학적 특성 연구 (Pedological and Mineralogical Characterizations of Hwangto (Yellow Residual Soils), Naju, Jeollanam-do, Korea)

  • 본 연구는 전라남도 나주지역 황토의 토양학적 및 광물학적 특성을 확인하고 주변 모암과의 상관관계를 통해 그 기원 및 형성과정을 알아보고자 하였다. 연구지역은 전라남도 나주시 동강면 장동리이며, 토양단면(약 150 cm 깊이)의 황토를 깊이별로 상부, 중부, 하부층으로 나누어 토양학적 특성(색, pH, 입도분리)과 광물학적 특성(광물조성, 입자의 크기, 모양, 화학조성)을 연구하였다. 모암시료는 박편제작 및 현미경관찰을 통해 구성광물을 기재하고 황토의 구성광물과 연관성을 알아보았다. 연구 결과 황토는 pH 4.3~5.1 범위를 갖는 산성토이며 미사와 점토가 주로 구성된(약 95%) 미사질양토와 미사질식양토였다. 모래와 미사의 주 구성광물은 석영, 운모, 장석이며 점토는 침철석, 수산화층간 버미큘라이트, 일라이트, 카올리나이트, 할로이사이트, 질석과 소량의 석영이 포함되어 있었다. 모래와 미사의 SEM-EDX 분석을 통해 구성광물의 형태를 확인한 결과, 풍화작용으로 인해 부식되어 표면이 거칠고 산화철로 피복되어 있는 장석이 관찰되었고 하부층으로 갈수록 그 양은 증가했다. 점토는 TEM 분석을 통해 다양한 형태의 층상규산염광물이 확인되었으며, 상부에서 하부층으로 갈수록 침철석의 양이 증가했는데 이는 상부 층에서 용탈된 산화철이 하부층으로 이동되어 집적된 것으로 사료된다. 황토의 모암으로 사료되는 주변의 암석은 석영, 사장석, 흑운모, 녹니석 등으로 이루어진 화강반암이었다. 즉, 화강암류의 모암에서 장석과 운모 등이 풍화작용을 받아 일라이트, 질석, 수산화층간버미큘라이트 및 카올리나이트로 변하였으며 침철석은 흑운모 풍화에 의해 형성된 것으로서, 본 연구지역의 황토는 오랜 기간 풍화작용에 의해 형성된 풍화잔류토로 판단된다.

L-글루타민산 생산균 Brevibacterium lactofermentum의 Bacteriophag에 관한 연구 (Studies on the Bacteriophages of Brevibacterium lactofermentum)

  • Many industrial processes those employ bacteria are subjected to phage infestations. In L-glutamic acid fermentions using acetic acid, the phage infestations of the organisms have been recently recognized. In efforts to elucidate the sources of phage contamination involved in the abnormal fermentation, a series of study was conducted to isolate the phages both from the contents of abnormally fermented tanks and the soil or sewage samples from the surroundings of a fermentation factory, to define major charateristics of the phage isolates, and finally to determine the correlation between the phage isolates and temperate phages originating from the miscellaneous bacterial species isolated from the soil or sewage samples. The results are summarized as follows; 1) All phages were isolated from the irregular fermentation tanks and soil or sewage samples, and they were designated as phage PR-1, PR-2, PR-3, PR-4, PR-5, PR-6, and PR-7, in the order of isolation. These PR-series phages were proved to be highly specific for the variant strains of Br. lactofermentum only, namely, phage PR-1 and PR-2 for Br. lactofermentum No. 468-5 and phage PR-3~PR-7 for Br. lactofemrentum No. 2256. By cross-neutralization test, the 7 phagescould be subdivided into 3 groups, i. e., phage PR-I and PR-2 the first, phage PR-3, PR-4, PR-5, PR-6 the second, and the phage PR-7 the third. 2) The 7 phages were virulent under the experimental conditions. They produced plaques with clear and relatively sharp margins without distinct halo. The mean sizes of plaques were 1.5mm in diameter for phage PR-1 and PR-2, and 1. Omm for phages PR-3~PR-7. Double layer technique modified by Hongo and described by Adams, was applied to assay of the PR-series phages. The factors influencing the plaques were as follows;young age cells of host bacteria cultured for 3-6 hours represented the largest number and size, optimum was pH 7.0, incubation temperature was $30^{\circ}C$, and agar concentration and amount of overlayer medium were 0.6% and 0.2ml, respectively. 3) PR-series phages were stable in 0.05M tris buffer and 0.1M ammonium acetate buffer solution. The addition of $5{\times}10^{-3}M$ magnesium ion effectively increased the stability. Thermostability experiments indicated that PR-series phages were stable at the teinperture between $50^{\circ}{\sim}55^{\circ}C$ in nutrient medium, $45^{\circ}{\sim}50^{\circ}C$ in buffer solution. However, the phages mere completely inactivated at 603C and 65$^{\circ}$C within 10 minutes. The phages were stable at the range of pH6~9 in nutrient medium and of pH 8-9 in buffer solution, respectively. Exposure of the phages to UV for 25, 60 and 100 seconds resulted in the complete loss of infectivily, respectively. 4) Electron microscopy showed that PR-series phage particles exhibited rather similar morphology, differing in the size All of PR-series phages had a multilateral head and had a simple long tiil about three to five times long as compared with head. By the size, phage PR-1 and PR-2, PR-3, PR-4, PR-5, and PR-6 and PR-7 were classified into same groups, respectively. The head and tail size of phage PR-1, PR-5, PR-5(T) and PR-7 were 85nm, 74nm and 235nm and 350mm, and 72nm and 210nm, respectively. 5) Nucleic acids of PR-series phages were double stranded DNA. The G+C contents of phage PR-1, PR-5 and PR-7 were 56.1, 52.9 and 53.7, respectively. The values of G+C contents derived from the $T_m$ were in agreement with the chemically determined values. 6) PR-series phages effectively adsorbed on their host bacteria at the rate of more than 90% during 5 min. K value for phage PR-1, PR-5 and PR-7 were calculated to be $6{\times}10^9 ml$ per minute, respectiveky. The pH of the medium did effect adsorption rate, but both temperature and age of host cells did not. Generally, optimum adsorption condition of phages seemed to be almost same as optimum growth conditions of host bacteria. 7) In one-step growth experiments, the latent periods at $30^{\circ}C$ for PR-1, and PR-7 were about 70, 50 and 55 min, respectively. The corresponding average burst size was 200, 70 and 90, respectively. Lpsis period according to the multiplicity of infection and a phage series. In case of m. o. i. 100, strain No. 2256 (PR-5) and No. 468-5(PR-1) failed to grow and turbidity decreased after 50 and 70min, respectively. 8) In the lysate of a plaque purified phage PR-5 infected bacteria, there observed 2 types ofphage particles, i. e., phage PR-5 and PR-5 (T) of similar morphology but differing at the length of phage tail, and phage tail like particles. The phage taillike particles could be divided into 4 types by the length. Induction experiments of Br. lactofermentum with UV irradiation, mitomycin C or bacitracin treatment produced neither phage PR-5 (T) or phage tail-like particles. 9) No lysis occured when the growth of 7 strains of miscellaneous bacteria, isolated from soil and sewage samples, were inoculated with either phage PR-5 (T) or phage tail-like particles the inoculation of phage PR-5 pellet resulted in the growth inhibition of the orgainsms in the spot test. The lysates obtained from 3 miscellaneous soil derived bacteria following mitomycin C treatment the growth of Br. lactofermentum, but did not lyze the bacterium.
