A geoscientific research was performed on 12 samples of comb-pattern potteries which were excavated at Daecheonri neolithic site, Korea. The texture and compositions of 12 potteries and surrounding metrix of soil and rocks were compared with the help of petrographic microscope, XRD and REE data. As to the manufacturing techniques thick potteries are caused by the amount and number of coarse grains which are assumed to be added shards when their distributional pattern are considered. DC1, DC3, DC 7 and DC11 samples have clearly oriented textures, and the orientation of vesicles in DC3 and DC11 samples arranged in the same direction with those on the pottery surface. This indicates the use potter's wheel technique in manufacturing potteries. Burning temperature is assumed over $800^{/circ}C$, particularly under reduction environment. As to the source, raw materials of pottery matrix are derived from the bottom of excavation site, or in an extracted outcrop of the northwestern foothill from site, while the shard materials are very similar with those extracted from sandy loams near sites. Finally any use pattern of pottery may control the pottery thickness, shard addition, and matrix selection.
BACKGROUND: Plastic mulches widely used in raised-bed culture mainly to conserve water, control weeds and raise soil temperature. The most widely used plastic mulch colour is black. Reflective red and far-red light can affect on growth and yield of various vegetable crops. Objectives of this study were to investigate the characteristics of reflective light of black and red plastic mulches, and to evaluate the reflective red and far-red light on the quality characteristics and yield of the Oriental melon (Cucumis. Melo L.) and tomato (Lycopersicon esculentum L.). METHODS AND RESULTS: Oriental melon and tomato were cultivated over the reflective red and black plastic mulches in plastic house. Reflected red and far-red light over the red plastic mulch were 2.6 times higher than those of black plastic mulch. Red to F-Red ratio of black plastic mulch, red plastic mulch and sunlight were 1.14, 0.93 and 1.16 respectively. Intensity of reflected red and far-red light over red plastic mulch were highest at surface height of 30 cm. The higher the height of the surface decrease the intensity of far-red light. Accordingly, Red to F-Red ratio were increased. Reflective red plastic mulch increased the weight of fruit and content of sugar in Oriental melon and tomato. CONCLUSION(s): Yield of Oriental melon over reflective red plastic mulch was higher than that of black plastic mulch. These results suggested that reflected red and far-red light over the red plastic mulch affected allocation of photosynthate in growing Oriental melon.
In an attempt to improve the productivity of ${\beta}-galactosidase$ from Bacillus sp. A1, which was isolated from soil and has remarkably higher transgalactosylation activity than lactose hydrolysis activity, a chemical mutation procedure using N-methyl-N'-nitro-N-nitrosoguanidine followed by selection was conducted. The final selection, designated as Bacillus sp. A4442, turned out to show a substantially increased enzyme productivity. Catabolite repression by glucose and lactose requirement as an inducer for the enzyme biosynthesis, which were shown in the parent strain, was markedly diminished; instead it was found out that galactose acts as another inducer. Because pH of medium, one of the most important factors for cell growth as well as enzyme production, is closely related with the sugar concentration during culture, it was kept in the optimum range of $6.5{\sim}7.5$; for this the initial glucose concentration was adjusted to be 0.5% which was thereafter maintained by the controlled pumping-in of lactose using the pH-stat technique. By doing so, we were able to increase the productivity of ${\beta}-galactosidase$ with high transgalactosylation activity up to $44\;unit/m{\ell}-broth$.
An exoinulase-producing bacterium was isolated from soil, and identified as Streptomyces sp. The maximum inulase production was achieved when inulin as carbon source and soybean meal as organic nitrogen source were included in the culture. The exoinulase was considered to be a constitutive enzyme produced not only by inulin but also by soluble starch or glucose. The purified enzyme on DEAE-cellulose and Sephadex G-200 column showed the maximal activity at $pH\;5.5{\sim}6.0$ and $50^{\circ}C$, but lost 65% inulase activity at $50^{\circ}C$ after 1 hour incubation. This exoinulase was activated by $Mn^{+2}$, wherease more that 80% inactivation was observed with $Ag^+$, $Hg^{+2}$ and $Fe^{+3}$. The enzyme was possibly a metalloenzyme in that EDTA and 8-hydroxyquinoline inhibited the enzyme. Km values for inulin (16.51 mM) and sucrose (14.62 mM) were in very close range suggesting mostly equal affinity toward the subatrates. However, the maximum velocity for inulin was 10 times greater than for sucrose.
To remove phosphate accumulated in the soil and water, Acinetobacter lwoffi PO8 possessing a high ability to accumulate phosphate was isolated from a active sludge. Bacterium was cultured in the liquid medium containing $150\;{\mu}g/mL$ of phosphate at $30^{\circ}C$ in different culture conditions to examine intracellular phosphate uptake. The initial pH in the range of $7.5{\sim}8.5$ was effective on the growth and phosphate uptake of the strain. Glycerol and arabinose used as a carbon sources showed 93 and 91% the phsphate uptake, respectively. Among the nitrogen sources, ammonium salt such as $NH_4NO_3$ and $(NH_4)_2SO_4$ was effectively utilized on the phosphate uptake compared with amino compounds. The rate of phosphate uptake of $NH_4NO_3$, and $(NH_4)_2SO_4$, was 95 and 96%, respectively The growth and Phosphate uptake ability in the strain were significantly promoted when metal ions were added in the medium; $Co^{2+}$, however, was not utilized by the strain. The capacity of phosphate uptake was enhanced to $10{\sim}20%$ when arginine, methionine, or lysine was added. Using $^{32}P$ to examine the uptake Pattern of intracellular phosphate, experiment result showed that polyphosphate was largely found in the fraction of intracellular inorganic phosphate of Acinetobacter lwoffi PO8.
Salt accumulation in soils of greenhouse due to the massive application of nitrogen fertilizers causes salt stress on the various crops, a serious problem in domestic agriculture. Since the majority of the salinity is nitrate, the excess nitrate should be removed; therefore, a bacterial strain having high capacity of nitrate uptake and identified as Enterobacter amnigenus GG0461 was isolated from the soils of greenhouse. Optimum conditions for the bacterial growth and nitrate uptake were investigated. GG0461 was able to grow without nitrate; however, nitrate facilitated the growth. The rate of nitrate uptake increased at alkaline pH and both growth and nitrate uptake were maximal at pH 8-9. When the initial pH of culture medium was increased to pH 8 or 9, it was decreased to neutral upon bacterial growth and nitrate uptake. These results imply that the major factor mediating bacterial nitrate uptake is a nitrate/proton antiporter. The fact was supported by the effect of nitrate addition in the absence of nitrate, since the addition of nitrate greatly increased the nitrate uptake and rapidly decreased pH of media.
Gaesung city of North Korea is meaningful because of the development of Gaesung industrial complex as a peace-building project and economical cooperation works between South Korea and North Korea. This article presents the characteristics of natural landscape of Gaesung through geographical names. This research was studied by literature analysis of North Korea, depth interview of North Korean labour and refugees, and second times fieldwork. The main results of this study are as follows. Firstly, the mountain landscape of Geasung revealed on geographical names characterized mountainousness and round shape. Especially, the places where are enclosed by mountain and streams are foggy and rocky. Next the landscape of stream characterizes sandy, unstable, and tidal river. Thirdly the fauna and flora which are reflected on geographical names are hawk, snake, tiger, homed owl, snapping turtle, chestnut tree, pine tree, pear tree, zelkova tree, and willow etcetera. The environmental conditions of habitation of these are similar to the natural environment of Gaesung. Besides, the geographical names related to soil distributed in northern mountain area of Geasung. The place names implicated cliff located on the riverside. In addition to this, the geographical name of 'Bul' implicated plain distributed in the riverside of 'Seoam' which a branch stream of 'Sacheo'. And the place names related rock located in place enclosed by mountain, river, and village. In cnclusion, Gaesung is more important to us as the Gaesung industrial complex and tourism development. But the fundamental research on Gaesung is a few because Gaesung belongs to North Korea. Therefore, this research will be useful for the control and management of Gaesung in the future.
The in vitro plantlets of cassava (Manihot esculenta Crantz cv. MColl 22) could be regenerated from nodal explant cultures in a liquid MS basal medium containing 0.01 mg/L zeatin for 2 weeks. The plantlets of 1.5∼2.5 cm in shoot length were transplanted to a glass bottle containing fine sand and acclimated under non-sterile conditions after excising their intact roots by: 1) prune leaving roots base of 1∼1.5 cm; 2) complete removal of roots; and 3) cutting off the rooting zone. The majority of in vitro-formed intact roots continued growth after transferred to soil, and all of the damaged roots stopped further growth. The plantlets with excised roots began to develop new roots within 7∼10 days after being transferred to a glass bottle, and a few of the pruned roots developed lateral roots from the remaining portion. Pruning and removal of in vitro roots resulted in a high survival rate (>87%), and did not significantly affect ex vitro root regeneration and acclimation, but the plantlets in which the rooting zone had been cut-off showed 73% survival rate. Pruning or removal of in vitro roots before transfer of plantlets is recommended for useful method of commercial micropropagation because of easier handling and high survival rate of plantlets.

Crown gall of stonefruit and nut trees is one of the very few plant diseases subject to efficient biological control. The disease is caused by the soil-inhabiting bacteria Agrobacterium tumefaciens and Agrobacterium rhizogenes and the original control organism was a non-pathogenic isolate of A. rhizogenes strain K84. Control is achieved by dipping planting material in a cell suspension of strain K84 which specifically inhibits pathogenic strains containing a nopaline Ti plasmid. Because the agrocin 84-encoding plasmid (pAgK84) is conjugative, it can be transmitted from the control strain to pathogenic strains which, as a result, become immune to agrocin 84 and cannot be controlled. To prevent this happening, the transfer genes on pAgK84 were located and then largely eliminated by recombinant DNA technology. The resulting construct, strain K1026, is transfer deficient but controls crown gall just as effectively as does strain K84. Field data from Spain confirm that pAgK84 can transfer to pathogenic recipients from strain K84 but not from strain K1026. The latter has been registered in Australia as a pesticide and is the first genetically engineered organism in the world to be released fro commercial use. It is recommended as a replacement for strain K84 to prevent a breakdown in the effectiveness of biological control of crown gall. Several reports indicate that both strains K84 and K1026 sometimes control crown gall pathogens that are resistant to agrocin 84. A possible reason for this is that both strains produce a second antibiotic called 434 which inhibits growth of nearly all isolates of A. rhizogenes, both pathogens and non-pathogens. Crown gall of grapevine is caused by another species, Agrobacterium vitis. It is resistant to agrocin 84 and cannot be controlled by strains K84 or K1026. It is different from other crown gall pathogens in several characteristics, including the fact that, although a rhizosphere coloniser, its also lives systemically in the vascular tissue of grapevine. Pathogen free propagating material can be obtained from tissue culture or, less surely, by heat therapy of dormant cuttings. A number of laboratories are searching for a biocontrol strain that will prevent, or at least delay, reinfection. A non-pathogenic A. vitis strain F/25 from South Africa looks very promising in this regard.

A strain producing strongly fibrinolytic enzyme was isolated from soil and was identified to be Bacillus subtilis by biochemical and physiological characterization. The optimal culture conditions for the production of fibrinolytic enzyme was determined to be 1.0% tryptone, 1.5% soluble starch, 0.5% Peptone, 0.5% NaCl, $(NH_{4})_{3}PO_4.3H_{2}O, and MgSO_{4}.7H_{2}O.$ Initial pH and temperature were pH 8.0 and $30^{\circ}C$ , respectively, The highest enzyme production was observed at 30 hours of cultivation at $30^{\circ}C$ The fibrinolytic enzyme was purified to homogeneity by DEAE Sephadex A-50 ion exchange column chromatography, 70% ammonium sulfate precipitation, Sephadex G-200 and G-75 gel filtration column chromatography. The molecular weight of the purified enzyme was 28,000 as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. A gene encoding the fibrinolytic enzyme was cloned into a plasmid vector pBluescript, transforming E.coli XL-1 Blue. The clone was able to degrade fibrin, This indicated that the gene could encode a fibrinolytic enzyme. The nucleotide sequence of the 2.7 kb insert was determined in both direction. One open reading frame composed of 1023 nucleotides was found to be a potential protein coding region. There was the putative Shine-Dalgano sequence and TATA box upstream of the open reading frame. The homology search data in the genome database showed that both the 2.7 kb insert and 1 kb open reading frame carried no significance in the nucleotide sequence of known fibrinolytic enzyme from Bacillus serovars. The recombinant cell harboring the novel gene involved in fibrinolysis was subjected to protein purification. The molecular mass of the purified fibrinolytic enzyme was determined to be 31864 Dalton, which was highly in accordance with the molecular mass(33 kDa) of the fibrinolytic gene deduced from the insert. The fibrinolytic enzyme was Purified 50.5 folds to homogeneity in overall yield of 10.7% by DEAE Sephadex A-50 ion exchange, 85% ammonium sulfate precipitation, Sephadex G-50, Superdex 75 HR FPLC gel filtration. In conclusion, a novel fibrinolytic gene from Bacillus subtilis was identified and characterized by cloning a genomic library of Bacillus subtilis into pBleuscript. For the soybean fermented by this strain, it is found that there increased assistant protein about 20% compared to the soybean not fermented and increased about 30% according to amino acid analysis and, in particular, essential amino acid increased about 40%. When keeping this fermented soybean powder at room temperature for about 70days, it showed very high stability maintaining almost perfect activity and, therefore, it gave us great suggestion its possibility of development as a new functional food.
