생약복방제의 조성 변화가 부패성 효모 Zygosaccharomyces sp. 의 성장에 미치는 영향 (Effects of Changes in Composition of Herb Extract Product on Growth of Spoilage Yeast, Zygosaccharomyces sp.)

  • The aim of the present study was to investigate effects of food preservative addition and changes in composition of herb extract product on the growth of spoilage yeast, Zygosaccharomyces sp. Herbs such as Panax ginseng, Cinnamomum cassia, Lycium chinense, Zyzyphus juiuba and Jingiber officinale were altogether put into water and essence was extracted at 80oC, and then the extract was concentrated at 75oC. The herb extract product was made by adding vitamins, amino acids and honey to the concentrated herb extract. The amount of gas produced from the herb extract product was increased as inoculated cell number increased but decreased as Brix concentration increased. Gases were produced in small amount when incubation was made at 4oC but large amounts of gases were produced at 25 or 40oC of incubation. The gas production and growth of Zygosaccharomyces sp. were measured after browning reaction was induced by heating at 85oC for 12 hours. It appeared that heating treatment did not induce any significant change in the gas production and growth of the cell. The effects of addition of various sugar to the herb extract produce were also invesigated. Amounts of gas production were in the order of glucose>sucrose>oligosaccharide>stevioside. The viable cell count was measured as 6.0$\times$107 CFU/g when glucose was added to the herb extract product. The viable cell counts were 5.0$\times$106, 3.0$\times$103, and 3.0$\times$102 CFU/g in sucrose, oligosaccharide and stevioside added herb extract product, respectively. The amount of gas production from the herb extract product was remarkably reduced by addition of such food preservatives as sodium benzoate and DF 100. TLC(thin layer chromatography) chromatogram of the herb extract showed stability of the herb extract in the above treatments.

  • Carcinogenesis is a multi-stage process that generally consists of at least three steps; initiation, promotion, and progression. If one of these carcinogenic steps were suppressed or delayed, the cancer could be prevented. Cancer chemoprevention is defined to be inhibition or reversal of the carcinogenic process by the specific chemical agents and is a novel approach to cancer management alternative to conventional chemotherapy. Chlorophylln(CHL), a water-soluble derivative of chlorophyll, containing sodium and copper, has been known to be strong antimutagen in several test systems, but its mechanism of antimutagenic action is unknown. In the present experiment, the possibility of CHL as chemopreventive drugs on 7,12-dimethylbenz[a]anthracene(DMBA)-induced hamster buccal pouch carcinogenesis was investigated by mutagenicity test, carcinogenicity test, and frequency or spectrum of H-ras mutations in the both of DMBA-induced and chlorophylln-pretreated-DMBA induced tumor by polymerase chain reaction and non-isotopic restriction fragment length polymorphism. The treatment of CHL reduced the yields and multiplicity of the 0.5% DMBA-induced tumor, 86% to 62.5% and $3.7{\pm}0.6$ to $1.4{\pm}0.3$, respectively. The occurrence of histidine revertant by $20{\mu}mole$ DMBA was inhibited 25.6 to 81.7% by 1 to $5{\mu}M$ CHL in a dose-dependent manner. The mutation rates of H-ras gene in DMBA-induced and CHL-pretreated-DMBA induced tumor were 96%, 94% of which the most mutations were in codon 12/13. These results suggest that CHL inhibits the carcinogenic action of DMBA by the formation of complex between CHL and DMBA or the inhibition of the activation of DMBA in vivo. But CHL did not affect the mutation rates or its spectrum in already formed tumor.

케나프를 이용한 수초지 제조에 관한 연구 (The Prodoction of Kenaf Hand-Made Paper)

  • Hanji, the korean traditional papers were mostly made from mulberry paper. But the production and demand of hanji have decreased rapidly because mulberry paper yields were insufficient and handworked hanji procedures were complicated. Recently, the researches on hanji were carried out to improve the properties of hanji. Kenaf fibers have been interested as a substitute resource of mulberry paper for hanji production. In this research, Kenai pulps were manufactured with removal methods of lignin or hemicellulose from kenaf fibers and paper mulberry pulps with traditional alkali methods. Kenaf papers, paper mulberry, and kenaf/paper mulberry mixed papers were manufactured with their pulps. The crystallinity, fiber length, color of the pulps and tensile strength, tear strength, water absorption of the papers were investigated. The results were as follow: The removal rates of lignin of chemical retted kenaf fibers with sodium chlorite reaction for 40 minutes were 70% and were higher than 40% of double retted fibers. Paper mulberry pulps has less lignin and hemicellulose than kenaf differently. The crystallinity of paper mulberry pulps were very low with 60%, but kenaf pulps were 90%. The chemical retted CR-40-1 pulps were similar with paper mulberry pulps on fiber length & fibrilation of fibers. Tensile strength of paper mulberry were higher than kenaf papers because of fibrilation of paper mulberry, but tear strength were lower. Tensile strength and tear strength were improved on kenaf/paper mulberry 30/70 mixed papers.

  • For-${\alpha}$-APM을 For-Asp anhydride와 Phe-OMe를 MEK, $CH_3CN$, 물 등의 용매를 사용하여 반응시켜 효율적으로 합성하였다. For-${\alpha}$-APM과 For-${\beta}$-APM의 혼합물로부터 순수한 For-${\alpha}$-APM의 회수는 pH 4.00에서의 연속적인 분별추출로 분리 수거가 가능하였다. $H_2_O2$ 공급원으로 $H_2_O2$/THF, 과탄산소다, $H_2_O2$/HCl/MeOH 등의 다양한 산화계를 사용하여, $H_2_O2$에 의한 산화방법으로 탈포밀반응을 성공적으로 수행하여 높은 수율로 아스파르테임을 얻을 수 있었다. 이 때 HCl이나 TsOH와 같은 산이 $H_2_O2$의 분해를 억제시키고 산화능을 증대시킴으로써 탈포밀 반응의 효율을 높일 수 있었다.

PEMFC용 고분자 전해질 막을 위한 BFBN을 포함한 sulfonated poly(ether sulfone)s의 합성 및 특성 (Preparation and Properties of Sulfonated Poly(ether Sulfone)s Containing BFBN for PEMFC)

  • Sulfonated poly (ether sulfone)s containing BFBN were prepared from 2,3-bis(4-fluorophenyl)-2-butenedinitrile , 4,4-sulfonyldiphenol and sulfonated 4,4'-difluorodiphenylsulfone sodium salt using potassium carbonate, and followed acidification reaction with 1M $H_2SO_4$. BFBN was prepared from 4-fluorobenzylcyanide, $CCl_4$, NaOH using trimethylbenzylammoniumchloride . Sulfonated poly(ether sulfone)s containing BFBN were studied by FT-IR, $^1H$-NMR spectroscopy, and thermo gravimetric analysis (TGA). The water uptake of synthesized S-PBFBN membranes exhibit 31~62% compared with 28% of Nafion 211. The S-PBFBN membranes exhibit proton conductivities ($80^{\circ}C$, RH 100) of 53.8~117.4 mS/cm compared with 137.4 mS/cm of Nafion 211.

황산염에 노출된 구조용 합성섬유와 라텍스 개질 시멘트 모르타르의 부착특성에 미치는 광물질 혼화재의 효과 (Effect of Mineral Admixture on Bond between Structural Synthetic Fiber and Latex Modified Cement Mortar under Sulfate Environments)

  • It has been well known that concrete structures exposed to acid and sulfate environments such as sewer etc. show significant decrease in their durability due to chemical attack. Such deleterious acid and sulfate attacks lead to expansion and cracking in concrete, and thus, eventually result in damage to cement mortar by forming expansive hydration products due to the reaction between cement hydration products and acid and sulfate ions. In this study, the effect of fly ash and blast furnace slag on the bond performances of structural synthetic fiber in latex modified cement mortar under sulfate environments. Fly ash and blast furnace slag contents ranging from 0 % to 20 % are used in the mix proportions. The latex modified cement mortar specimens were immersed in fresh water, 8 % sodium sulfate ($Na_2SO_4$) solutions for 28 and 50 days, respectively. Pullout tests are conducted to measure the bond performance of structural synthetic fiber from latex modified cement mortar after sulfate environments exposure. Test results are found that the incorporation of fly ash and blast furnace slag can effectively enhance the PVA fiber-latex modified cement mortar interfacial bond properties (bond behavior, bond strength and interface toughness) after sulfate environments exposure. The microstructural observation confirms the findings on the interface bond mechanism drawn from the fiber pullout test results under sulfate environments.

Maternal high-fructose intake during pregnancy and lactation induces metabolic syndrome in adult offspring

  • BACKGROUND/OBJECTIVES: Nutritional status and food intake during pregnancy and lactation can affect fetal programming. In the current metabolic syndrome epidemic, high-fructose diets have been strongly implicated. This study investigated the effect of maternal high-fructose intake during pregnancy and lactation on the development of metabolic syndrome in adult offspring. SUBJECTS/METHODS: Drinking water with or without 20% fructose was administered to female C57BL/6J mice over the course of their pregnancy and lactation periods. After weaning, pups ate regular chow. Accu-Chek Performa was used to measure glucose levels, and a tail-cuff method was used to examine systolic blood pressure. Animals were sacrificed at 7 months, their livers were excised, and sections were stained with Oil Red O and hematoxylin and eosin (H&E) staining. Kidneys were collected for gene expression analysis using quantitative real-time Polymerase chain reaction. RESULTS: Adult offspring exposed to maternal high-fructose intake during pregnancy and lactation presented with heavier body weights, fattier livers, and broader areas under the curve in glucose tolerance test values than control offspring. Serum levels of alanine aminotransferase, aspartate aminotransferase, glucose, triglycerides, and total cholesterol and systolic blood pressure in the maternal high-fructose group were higher than that in controls. However, there were no significant differences in mRNA expressions of renin-angiotensin-aldosterone system genes and sodium transporter genes. CONCLUSIONS: These results suggest that maternal high-fructose intake during pregnancy and lactation induces metabolic syndrome with hyperglycemia, hypertension, and dyslipidemia in adult offspring.

NaBH4 가수분해 반응에 의한 무인항공기용 PEMFC 수소공급 (Hydrogen Supply to PEMFC for Unmanned Aero Vehicles Using Hydrolysis Reaction of NaBH4)

  • 무인항공기가 장시간 비행하기 위해서는 배터리보다 고분자전해질 연료전지(PEMFC)가 적합하다. 본 연구에서는 PEMFC에 수소를 공급하는 $NaBH_4$가수분해 반응 시스템에 대해 연구하였다. $NaBH_4$가수분해 반응 시스템의 무게를 감소시키기 위해 수소수율 향상, 응축수 회수, 안정적인 수율 유지 등에 대해 실험하였다. 반응기 압력제어를 함으로써 수소 수율을 3.4% 향상시켰다. 수소를 공랭하여 PEMFC 스택에 공급하는 과정에서 발생하는 응축수를 $NaBH_4$ 저장조에 회수하였다. 이 과정에서 응축수가 $NaBH_4$분말을 용해시켜 보충됨으로써 14%의 무게 감량효과가 있었다. 2.0 L/min의 속도로 수소를 발생시킬 때 Co-P-B촉매 2.0 g을 사용해서 10시간동안 96% 수소수율로 $NaBH_4$가수분해 반응 시스템을 안정적으로 구동하였다.

물유리에서 소달라이트의 합성: 합성모액 조성이 결정화도와 입자 크기에 미치는 영향 (Synthesis of Sodalite from Water Glass: Effect of the Composition of Synthetic Mixtures on Its Crystallinity and Crystallite Size)

  • 물유리로 제조한 알칼리성 실리카-알루미나 합성모액에서 $Na_2O$, $SiO_2$, $H_2O$ 함량이 소달라이트의 결정화도와 입자크기에 미치는 영향을 조사하였다. x=2.5~7.5, y=1.4~3.0, z=140~400 범위에서 한 성분의 함량만 바꾸어 제조한 $x\;Na_2O{\cdot}y\;SiO_2{\cdot}Al_2O_3{\cdot}z\;H_2O$ 조성의 합성모액을 $140^{\circ}C$에서 2일간 수열 반응시켜 소달라이트를 합성하였다. $Na_2O$ 함량이 많아지면 소달라이트의 결정화도가 높아지나 입자는 작아졌다. 합성모액의 $SiO_2/Al_2O_3$ 몰비가 2 근처에서 소달라이트가 생성되었으며, 물의 함량이 달라지면 합성되는 제올라이트의 종류가 달라졌다. 이 합성조건에서는 $SiO_2/Al_2O_3$ 몰비가 2.0 근처이고, $Na_2O$ 함량이 많은 영역에서는 결정화도가 높은 소달라이트가 합성되었으며, 나트륨 이온의 구조유도체로서 효과로 입자 크기는 작아졌다.

물리화학적 공정에 의한 용존성 Humic Acid의 제거 (Removal of Dissolved Humic Acid with Physicochemical Treatment Process)

  • THMs의 전구물질로 잘 알려진 humic acid를 대상으로 오존처리, 응집처리 및 활성탄 흡착처리와 같은 물리화학적 처리공정 도입에 따른 humic acid의 제거특성을 조사하고, 염소주입시 생성 가능한 부산물을 정성적으로 규명하기 위하여 본 연구를 실시하였다. Humic acid에 오존을 주입한 결과 pH가 급격히 감소하였는데, 이는 분자량이 큰 humic acid가 오존에 의하여 $H_2O$$CO_2$로 완전히 산화되지 못하고 중간생성물인 저급 지방산으로 분해된 결과로 판단되었다. 그리고 용존성 humic acid의 응집특성을 조사하기 위하여 PAC 160 mg/L로 응집을 실시한 결과 TOC는 약 25%, $COD_{Cr}$는 24%가 제거되었으나, 색도는 단지 5%만이 제거되어, humic acid에 의해 유발되는 색도는 응집으로 제거하기 어려운 것으로 판단되었다. 그러나 오존처리에서는 95% 이상의 색도가 제거되었으며, 이때 색도는 오존 접촉시간에 대해 1차반응으로 제거되었고, 반응속도상수값 k는 $0.067min^{-1}$로 조사되었다. 활성탄 흡착 실험에서는 오존 전처리를 실시함으로써 활성탄 흡착 효율이 크게 증가함을 확인할 수 있었으며, humic acid에 염소를 주입한 결과 THMs만 검출되었을 뿐 다른 휘발성 미량 유기화합물은 검출되지 않았고, 오존처리를 실시한 오존처리수에서도 알데히드류 및 케톤류와 같은 부산물은 검출되지 않았다.
