Anti-Inflammatory Effects and Cytoprotective Effects of Smilacis Chinae Radix (토복령의 항염증 및 세포보호 효과에 미치는 영향)

  • Smilacis Chinae Radix has been used as an anti-inflammatory agent. This study was performed to anti-inflammatory and MAP kinase signaling pathway in vitro. Experimental studies were obtained by measuring the Cytotoxicity, production of NO, PGE2, TNF-$\alpha$ and protein level of catalase, SOD, MAP kinase, The results were summarized as follows: Smilacis Chinae Radix was not cytotoxic effects against Raw264.7 and HEK293 cells. Concentration of $100{\mu}g/m{\ell}$ Smilacis Chinae Radix inhibited the production of NO in the Raw264.7 cell stimulated with LPS. All concentrations of Smilacis Chinae Radix not significantly inhibited the production of PGE2 in the Raw264.7 cell stimulated with LPS. All concentrations of Smilacis Chinae Radix did not inhibit the production of TNF-$\alpha$ in the Raw264.7 cell stimulated with LPS. Smilacis Chinae Radix has not effect of blocking NF-${\kappa}B$ into nucleus in LPS-induced macrophage Raw264.7 cell. Smilacis Chinae Radix has the effect of Cytoprotection through activation of ERK and inhibition of p38 and JNK. Accordingly the results show Smilacis Chinae Radix could induce anti-inflammation and Cytoprotection effects against In vitro, but it needs more research on the precise mechanism of such effects.

  • Smilacis Chinae Radix is root of Smilax china L.. It has been known as a korean folk medicine for anti-inflammatory, anti-microbial diuresis, detoxification and relieving enteropathy as diarrhea. Reactive oxygen species(ROS) are continuously produced at a high rate as a by-product obtained in the aerobic metabolism. A major portion of living organisms has defense system as superoxide dismutase or catalase against damage produced by ROS. Several lines of evidence provided that ROS appears to cause to develop aging and various diseases. In this study, we have investigated the antioxidant activities of Smilacis Chinae Radix in order to screen the antioxidant substances from natural products. As a result, EtOAc extract of Smilacis Chinae Radix exhibited potent antioxidant effect on various antioxidant experiment. The major components of antioxidant activity were isolated from EtOAc extract of Smilacis Chinae Radix. Their structure of compounds were identified as quercetin and (-)-epicatechin by spectroscopic evidence, respectively.

Analysis on Antioxidant Activity and Agronomic Characteristics of Extract from Smilacis Chinae Radix

  • The Smilacis chinae Radix refers to the root of Smilax chinae L distributed in mountain and filed of Korea, and it is a vine shrub in the Lilaceae family, called Berchemia berchemiaefolia, and is referred to as Smilacis chinae Radix in it's a natural medicine name. Antibacterial, inflammatory, and antioxidant activity were studied in Smilacis chinae Radix. In this study, biological activities such as antioxidant (DPPH, ABTs, TPC), cytotoxicity, wrinkle improvement, and whitening improvement to increase the utilization value of Smilacis chinae Radix and identify the botanical value. Therefore, we tried to explore the applicability of Smilacis chinae Radix as a functional cosmetic material. Smilacis chinae Radix (SCR) was dried and extracted with ethanol. In order to measure the biological activity of the SCR, antioxidant activity, inhibition activities of collagenase, tyrosinase and cell viability were measured. The DPPH (1,1-diphenyl-2-picryl hydrazyl) radical scavenging activity in the extract with a concentration of 400㎍/mL is 91.22% ± 0.41%%. ABTs (2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity in the extract with a concentration of 400㎍/mL is 99.60% ± 0.03%. Total polyphenol contents (TPC) are 0.203 ± 0.05 mg GAE/mg Ext when SCR was lmg/mL. And the Cell viability for HaCaT derived human keratinocyte and Raw264.7, a mouse-derived macrophage was determined using the MTT assay. When cell was treated with 100㎍/mL of SCR, HaCaT cell showed cell viability of 78.09 ± 0.1% and Raw264.7 cell showed cell viability of 91.88 ± 0.42%. From the above results, we have shown the possibility that the CSR have antioxidant ability, inhibition activity of collagenase and tyrosinase and cell safety ability which can be useful in a functional cosmetic material.

A Case Report for a Toxic Liver Injury Caused by Voluntary Administration of Smilacis Chinae Radix (청미래덩굴 뿌리 복용으로 발생한 독성간염환자 1예의 치료보고)

  • Drug-induced liver injury (DILI) is a common cause of acute hepatitis. Regarding the risk of DILI from herbal preparations, there are controversial issues such as exaggerated reports straying far from the truth and lack of discrimination between herbal drugs and folk remedies or food supplements. This study reported one case of a patient with DILI caused by Smilacis Chinae Radix and cured in an Oriental hospital. Smilacis Chinae Radix has been used as an anti-inflammatory, anti-microbial, detoxification and anticancer compound, as a folk remedy. This report would provide helpful information for management of DILI by folk remedies or herbal drugs.

Isolation and Quantitative Determination of Dioscin from Smilacis Chinae Radix (토복령(土茯令, Smilacis Chinae Radix)으로부터 Dioscin의 분리 및 함량분석)

  • The methods for isolation and quantitative determination of dioscin, a spirostanol triglycoside, from the roots of Smilax china have been developed. Isolation of dioscin was achieved by silica gel and RP-18 chromatography. The HPLC method used for quantitative determination of dioscin enabled the standardization of the crude drug. It suggests that the content of dioscin in Smilax china should be above 0.01%.
