Phosducin (PDC) is a photoreceptor cell-specific protein that is phosphorylated by cyclic nucleotide-dependent protein kinase. PDC and PDC-like proteins (PDCL, PDCL2, and PDCL3) are members of a conserved family of small thioredoxin-like proteins that modulate the ${\beta}$- and ${\gamma}$-subunits of G-proteins. In mammals, Pdc, Pdcl, and Pdcl3 genes show ubiquitous expression; however, Pdcl2 gene expression is limited to the testis and ovary. The aim of the present study was to examine the expression patterns of chicken Pdcl2 (cPdcl2) during testicular and ovarian development. Protein sequence comparisons performed using the CLUSTAL X program revealed that the amino acid sequences and potential phosphorylation sites of cPDCL2 and mammalian PDCL2 proteins were highly conserved. Quantitative real-time PCR analysis revealed that cPdcl2 was differentially expressed in the testis and ovary. Specifically, cPdcl2 expression was detected at low levels in the ovary at all time points. In the testis, cPdcl2 expression was detected at low levels until 5 weeks of age. At 8 weeks of age, however, cPdcl2 showed increased expression levels in the testis. Using in situ hybridization, we detected high levels of cPdcl2 expression in the testis, particularly in the spermatocytes and round spermatids. In summary, our data describe expression patterns of germ cell-specific Pdcl2 during testicular and ovarian development in chickens.
Background: The human protein methyl-transferase DOT1L catalyzes the methylation of histone H3 on lysine 79 (H3K79) at homeobox genes and is also involved in a number of significant processes ranging from gene expression to DNA-damage response and cell cycle progression. Inhibition of DOT1L activity by shRNA or small-molecule inhibitors has been established to prevent proliferation of various MLL-rearranged leukemia cells in vitro, establishing DOT1L an attractive therapeutic target for mixed lineage leukemia (MLL). Most of the drugs currently in use for the MLL treatment are reported to have low efficacy, hence this study focused on various natural compounds which exhibit minimal toxic effects and high efficacy for the target receptor. Materials and Methods: Structures of human protein methyl-transferase DOT1L and natural compound databases were downloaded from various sources. Virtual screening, molecular docking, dynamics simulation and drug likeness studies were performed for those natural compounds to evaluate and analyze their anti-cancer activity. Results: The top five screened compounds possessing good binding affinity were identified as potential high affinity inhibitors against DOT1L's active site. The top ranking molecule amongst the screened ligands had a Glide g-score of -10.940 kcal/mol and Glide e-model score of -86.011 with 5 hydrogen bonds and 12 hydrophobic contacts. This ligand's behaviour also showed consistency during the simulation of protein-ligand complex for 20000 ps, which is indicative of its stability in the receptor pocket. Conclusions: The ligand obtained out of this screening study can be considered as a potential inhibitor for DOT1L and further can be treated as a lead for the drug designing pipeline.
This study was conducted to investigate the dietary and nutritional status supplied by group feeding for blue color and white color in Seoul and Kyunggi area. The results are summarized as follows : 1. Blue color and white color of the average food supply per person per day was 1370.0g. Blue color of the total fool supply was consisted of 35.1% grain, 44.1% vegetables. White color of the total fool supply was consisted of 31.8% grain, 44.0% vegetable. The amount of milk product, fruit and fat supply were very small. 2. Blue color and white color of the ratio of main dish and side dish supplied was 69 : 31, 60.0 : 39.7. 3. Main dish all is rice. Side dish ard kinds of soupes and Jigeai Johngol in the two groups. In the kind of side dish white color much more than blue color. Dessert are scarcely supplied in the two groups. 4. The mean daily supplies of energy and 9 nutrients were above the level of the R.D.A. Blue color and white color of the mean daily amount of energy supply was 2653.0㎉ 3022.6㎉. Blue color and white color of the ratio of carbohydrate, protein, and fat supplied was 68.4%, 18.6%, 13.1%, and 62.1%, 20.9%, 17.0%. 5. We previously observed that Bacillus subtilis spores from sspE mutants presented a lower germination capacity in media containing high salt concentrations (0.9M NaCl). This deficiency was attributed to the absence of SASP-E (gamma-type small-acid-soluble protein), rich in osmocompatible amino acids released by degradation. Herein we observed that, in addition, this mutant spore presented a reduced capacity to use L-alanine as germinant (L-ala pathway), required longer times to germinate in calcium dipicolinate ($Ca^{2+}$-DPA), but germinated well in asparagine, glucose, fructose, and potassium chloride (AGFK pathway). Moreover, mild sonic treatment of mutant spores partially recovered their germination capacity in L-ala. Spore qualities were also altered, since sporulating colonies from the sspE mutant showed a pale brownish color, a higher adherence to agar plates, and lower autofluorescence, properties related to their spore coat content. Furthermore, biochemical analysis showed a reduced partition in hexadecane and a higher content of $Ca^{2+}$-DPA when compared with its isogenic wild-type control. Coat protein preparations showed a different electrophoretic pattern, in particular when detected with antibodies against CotG and CotE. The complementation with a wild-type sspE gene in a plasmid allowed for recovering the wild-type coat phenotype. This is the first report of a direct involvement of SASP-E in the spore coat assembly during the differentiation program of sporulation.
본 연구는 민들레 지상부와 지하부의 혼합 추출에 따라 항산화 활성에 상승 효과가 있는지 확인하기 위해 민들레 지상부와 지하부를 9:1 또는 8:2 중량 비로 혼합한 혼합 추출분말과 지상부, 지하부 단독 추출분말의 생리활성 성분을 분석하고 항산화 활성을 비교하였다. 민들레의 대표적인 성분으로 알려진 luteolin, chicoric acid를 분석한 결과 지상부의 함량이 지하부보다 월등히 높았으며 혼합 추출분말의 함량은 지상부보다 낮고 지하부보다 높았다. HepG2 cell에서의 산화적 스트레스 방어 효과 및 CAT 활성을 확인하여 효소적 항산화 시스템에 대한 영향을 평가한 결과 민들레 지상부와 지하부 간에 CAT 활성의 차이는 없었으나 혼합 추출분말의 활성이 현저히 증가하여 혼합 추출을 통한 상호작용이 영향을 미쳤을 것으로 보였다. DPPH 라디칼 소거활성 및 지질과 산화 억제에 대한 $EC_{50}$ 분석 결과에서도 지상부보다 혼합 추출분말의 항산화능이 유의적으로 높은 것으로 나타났다. 항산화능 평가 항목에 따라 8:2 혼합과 9:1 혼합의 활성에 차이가 있었는데 DPPH 라디칼 소거활성에서는 9:1 혼합의 $EC_{50}$이 더 낮고 지질과산화 억제 활성에서는 8:2 혼합의 $EC_{50}$이 더 낮았다. 이와 같이 민들레 지상부와 지하부를 혼합하여 추출할 때 지상부, 지하부 단독 추출물보다 항산화 활성이 상승한 것은 청미래 덩굴잎 열수추출물과 토복령 열 수추출물을 혼합하였을 때 EDA 및 FRAP와 같은 항산화 활성이 높아진다는 연구 및 로스팅 커피와 홍삼 혼합 추출물의 항산화 효과 평가에서 커피와 홍삼의 함량 조절을 통해 항산화력을 변화시킬 수 있다는 연구에서 2개 이상의 식품 또는 천연물을 혼합할 때 항산화 활성에 상승효과를 나타낸다는 결과와 유사한 것으로서 민들레 지상부와 지하부에 함유된 각기 다른 화합물들의 상호 작용으로 인해 항산화 활성이 상승한 것으로 보였다. 그러나 혼합 추출을 통한 생리활성 물질의 변화 및 상호 작용에 대해서는 추가적인 연구를 통해 확인이 필요하겠다. 본 연구는 기존의 민들레 전초 또는 각 부위별 항산화능을 평가한 연구와는 달리 민들레 지상부와 지하부를 9:1 또는 8:2로 혼합한 추출분말이 단독 추출 분말보다 항산화 활성이 높은 것을 확인함으로써 지상부, 지하부의 혼합을 통해 항산화 활성에 시너지 효과가 있는 것을 확인하였다. 지질과산화 억제 활성, HepG2 cell에서의 산화적 스트레스 방어 효과 및 catalase 활성에서 지상부와 지하부 혼합 비율에 따른 유의적인 차이 없이 모두 지상부보다 높은 활성을 보였고 DPPH 라디칼 소거활성과 같은 총 항산화능 평가에서는 8:2 혼합보다 9:1 혼합 추출분말의 활성이 유의적으로 높으며 수율도 9:1 혼합 추출분말이 8:2 혼합보다 약 1.5배 가량 높게 나타나 9:1 혼합 추출분말이 상업적인 항산화 활성 소재로써의 가치가 더 높을 것으로 기대된다.
Dietary powder from Plantaginis ovatae testa was prepared by mechnical milling/grinding of the outer layer of the seed. The crystalline/surface structures of its powder (100 mesh) were examined, and several physical functionalities including, water capacity, oil holding capacity, emulsion/foam properties and physiological functionality such as in-vitro glucose and bile acid retarding effects were also investigated. Water holding capacity(WHC) of psyllium powder was $33.71{\pm}0.10g$ water retained/g solid at room temperature, whileas oil holding capacity(OHC) for soybean or rice bran oil were about 1.80g oil retained/g solid. These values of WHC and OHC were about 5.6 times higher and 2.8 times lower than those of commercial ${\alpha}$-cellulose, respectively. Changes of pH showed a small effect on WHC, but WHC increased with temperature. Emulsion capacity of 2%(w/v) psyllium was about 60% level of 0.5%(w/v) xanthan gum but emulsion stability after incubation of 24 hours showed about 1.4 times improvement of xanthan gum(0.5%,w/v). Also, psylliume(above 2%, w/v) alone had higher foam capacity than that of xanthan(1.1 times) and especially, 1 or 2% addition of psyllium improved the foam stability of protein solution(1% albumin+0.5% $CaCl_2$) by factor of 3.3 and 6.0 times, respectively. The glucose and bile acid retarding effects of psyllium powder were relatively very excellent suggesting the prevention from diabetes and arteriosclerosis. Especially, psyllium showed the 3.7 and 3.3 times higher effect on in-vitro glucose and bile acid retardation than those of commercial ${\alpha}$-cellulose, respectively.

The complete chloroplast genome sequence of Panax ginseng breeding line 'G07006', showing higher salt tolerance, was confirmed by de novo assembly using whole genome next-generation sequences. The complete chloroplast (CP) genome size is 156,356 bp, including two inverted repeats (IRs) of 52,060 bp, separated by the large single-copy (LSC 86,174 bp) and the small single-copy (SSC 18,122 bp) regions. One hundred fourteen genes were annotated, including 80 protein-coding genes, 30 tRNA genes, and 4 rRNA genes. Among them, 18 sites were duplicated in the inverted repeat regions. By comparative analyses of the previously identified CP genome sequences of nine cultivars of P. ginseng and that of G07006, five useful SNPs were defined in this study. Since three of the five SNPs were cultivar-specific to Chunpoong and Sunhyang, they could be easily used for distinguishing from other ginseng accessions. However, on arranging SNPs according to their gene location, the G07006 genotype was 'GTGGA', which was distinct from other accessions. This complete chloroplast DNA sequence could be conducive to discrimination of the line G07006 (salt-tolerant) and further enhancement of the genetic improvement program for this important medicinal plant.
A survey on feeding practices was conducted with thirty-one cattle farmers belonging to three categories: households without land and no income from agricultural labour (Class 100;10 farms), households without land but deriving considerable income from agricultural labour (Class 101;10 farms), and households with land and without income from agricultural labour (Class 110;11 farms). Information on the types of feeds given of one year. In addition, samples of the feeds offered and refused were collected every two weeks and analysed for dry matter, organic matter (OM), crude protein (CP) and in vitro organic matter digestibility (OMD). Grass was usually cut at an early stage of growth, as such the CP(11.8%) and OMD (62%) were relatively high. All types of rice straw (whole, lower and upper part) and sugarcane forage (tops and leaves) were low in CP (<6 and <8.9%, respectively) and OMD (<45 and <47%, respectively). Rice bran and tofu waste was of much better quality than any other feed. The average number of different feeds in the rations (mean of all farms) was 1.98, with a lower value for class 101 (1.80), than for classes 100 and 110 (2.11 and 2.02, respectively). Of the total amount of OM consumed, 42% was rice straw, 21% grass, 19% maize forage, 10% sugarcane forage, <4% other forages (soya and groundnut straw), 1.3% rice bran and 2.9% tofu waste. The total amount of OM offered varied from <80 $g/kg^{0.75}/d$ in August/September to 1.5 times as much in May (P<0.01). The intake of digestible organic matter (IDOM) for farm class 110 ($37.7g/kg^{0.75}/d$) was significantly (p<0.001) lower than that for classes 100 and 101 (44.1 and $41.3g/kg^{0.75}/d$, respectively). The highest CP/IDOM ratio was recorded for farm class 101 (0.201 as compared to 0.181-0.184).
Preservation of forage crops as silage offers opportunity to avoid the high risk of rain-damaged hay in the humid south-central USA. Recent developments with baled silage or baleage make silage a less expensive option than typical chopped silage. Silage has been important in the region primarily for dairy production, but baleage has become an option for the more extensive beef cattle industry in the region. Silage samples submitted to the Louisiana State University Agricultural Center Forage Quality Lab from 2006 through 2013 were assessed for dry matter (DM) and forage nutritive characteristics of chopped silage and baleage of the different forage types from commercial farms primarily in Louisiana and Mississippi. Of the 1,308 silage samples submitted, 1,065 were annual ryegrass (AR) with small grains (SG), the warm-season annual (WA) grasses, sorghums and pearl millet, and the warm-season perennial (WP) grasses, bermudagrass and bahiagrass, providing the remaining samples. Concentration of DM was used to indicate an effective ensiling opportunity, and AR silage was more frequently within the target DM range than was the WA forage group. The AR samples also indicated a high-quality forage with average crude protein (CP) of 130 g/kg and total digestible nutrient (TDN) near 600 g/kg. The cooler winter weather at harvest apparently complicated harvest of SG silage with chopped SG silage lower in both CP and TDN (104 and 553 g/kg, respectively) than either AR silage or baleage of SG (137 and 624 g/kg for CP and TDN, respectively). The hot, humid summer weather along with large stems and large forage quantities of the WA grasses and the inherently higher fiber concentration of WP grasses at harvest stage indicate that preservation of these forage types as silage will be challenging, although successful commercial silage samples of each forage type and preservation approach were included among samples of silages produced in the region.
The effects of five colored soybean (blue, red, yellow, white, and black) varieties on the quality characteristics of soybean paste (Doenjang) were investigated for proximate composition, salt content, pH, color, organic acid content, amino acid content, and mineral content after 80 days of aging. Soybean paste (Doenjang) samples were divided into TDS (traditional Doenjang prepared with soybean) and TDM (traditional Doenjang prepared with mixed soybeans, kidney bean, black soybean, kind of small bean, sword bean, green bean, red bean, at a radio of 1:1:1:1:1(w/w)). Moisture content was 49.66% for TDM and 49.52% for TDM. Protein and salt contents of the TDS sample were higher than those of TDM. The "L" values of the TDB sample increased the aging period was decreased. In contrast, the "L" and "$\alpha$" values of TDM did not show any significant differences. The main organic acids detected were malic acid > lactic acid > tartaric acid in the TDS and TDM samples. Glutamic acid contents were 2.83 g/100 g for the TDS sample and 2.15 g/100 g for the TDM. The levels of K, Mg, and Ca of the TDS sample were higher than those of TDM. No significant difference occurred between TDS and TDM regarding their quality characteristics after 80 days of aging. Further studies needed include sensory evaluation and functionality of soybean paste made from mixed soybean materials.

제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
