폐가축사체 가수분해를 위한 최적 가수분해제 선정 (Selection of Optimal Degradation Agents for Hydrolysis of Animal Cadavers)

  • Many infectious diseases have emerged or re-emerged during the past 50 years in South Korea. There were three outbreaks of foot and mouth disease (FMD) in South Korea between January 2010 and March 2011. Over 3.45 million animals were slaughtered (33.3% of the existing pigs, 8.4% of dairy cows and 3.4% of cattle). To select optimal degradation agents of animal cadavers, degradation rates and fertilizer components of pig cadavers were investigated using hydrogen chloride (HCl), potassium hydroxide (KOH) and sodium hydroxide (NaOH) hydrolysis methods. Degradation rates of pig cadavers using HCl, KOH and NaOH were 81.1, 82.8 and 91.6%, respectively. Total nitrogen (T-N) concentration in degradation solution of pig cadavers using KOH hydrolysis method was higher than that in NaOH and HCl hydrolysis methods. Total phosphorus ($P_2O_5$) concentrations in degradation solution of pig cadavers in all hydrolysis methods ranged 0.14 ~ 0.28%. Total potassium ($K_2O$) concentration for KOH hydrolysis method was higher than that for other hydrolysis methods. The concentration of T-N and $K_2O$ in degradation solution of pig cadavers by KOH hydrolysis method were higher than that in NaOH and HCl hydrolysis methods. Thus, to recycle animal cadavers in agriculture, the optimal degradation agent for hydrolysis was KOH.

수정능획득 처리법이 소 난포란의 체외수정 및 분할율에 미치는 영향에 관한 연구 (Studies on the Effects of the Capacitation Methods of Spermatozoas on in-vitro Fertilization and Cleavage Rate of Bovine Follicular Oocytes)

  • The studies on the carried out to investigate the effects of capacitation method of spermatozoa on the in vitro fertilization and cleavage rate of bovine follicular oocytes. The ovaries were obtained from slaughtered Korean native cows. The follicular oocytes surrounded with cumulus cells were recovered by aspirating follicular fluids from the visible of diameter 3~5mm. The follicular oocytes were cultured in TCM-199 medium containing hormones and FCS for 24~48hrs in an incubator with 5% CO2 in air at 38.5$^{\circ}C$ and then matured oocytes were again cultured for 12~18hrs with motile capacitated sperm by preincubation of mKRB, treatment of HIS(high strength ion), Ca-IA(Inophore A), BFF(bovine follicular fluids) and heparin. The results obtained in these experiments were summarized as follows : 1. The in vitro fertilizatin and cleavage rate offollicular oocytes fertilized with capacitated spermatozoas in BO solution by preincubation of mKRB, treatment of HIS, Ca-IA, BFF and heparin method were 53.1%, 33.9%, 50.8%, 48.1%, 58.8% and 28.1%, 17.7%, 26.2%, 22.8%, 32.8%, respectively. And the fertilization and cleavage rate of heparin method was of highest of all. 2. The in vitro fertilization and cleavage rate of follicular oocytes fertilized with capacitated spermatozoas in BO solutin by both caffeine, BSA and heparin methods were 65.8%, 70.3% and 40.8%, 47.3%, respectively, and those rates were higher treatment of heparin+BSA, heparin+caffeine than treatment of heparin. 3. The in vitro fertilization and cleavage rate of follicular oocytes fertilized with capacitated spermatozoa in BO solution with heparin concentrations of 2, 5, 10, 20, 40$\mu\textrm{g}$/ml were 50.0%, 54.7%, 58.1%, 51.7% and 27.9%, 32.8%, 37.1%, 30.0%, respectively. And the fertilization and cleavage rate of follicular oocytes fertilized with capacitated spermatozoas in BO solution with 10$\mu\textrm{g}$/ml of heparin was the highest of all. 4. The in vitro fertilization and cleavage rate of follicular oocytes fertilized with capacitated spermatozoas in BO solution containing heparin with caffeine concentraton of 10, 20, 30, 40$\mu\textrm{g}$/ml were 71.4%, 74.3% and 70.6%, 70.0% and 45.7%, 47.3%, 44.1%, 41.4%, respectively. The fertilization and cleavage rate of spermatozoa fertilized in BO solution with caffeine and heparin together(70.3~74.3%) was higher than that of spermatozoa fertilized in BO solution with heparin(58.8%). 5. The in vitro fertilization and cleavage rate of follicular oocytes fertilized with capacitated spermatozoas in BO solution containing heparin with BSA concentration of 5, 10, 20, 30$\mu\textrm{g}$/ml were 63.6%, 62.9%, 66.7%, 60.3% and 44.1%, 43.5%, 48.5%, 42.7%, respectively. The fertilization and cleavage rate of spermatozoa fertilized in BO solution with BSA and heparin together(60.3~66.73%) was higher than that of spermatozoa fertilized in BO solution with heparin(58.8%).

정자미세주입술에 의하여 동결 융해 부고환 정자와 수정시킨 활성화처리 난자의 체외발생율에 관한 연구 (Studies on In Vitro Developmental Rate of Activated Bovine Oocytes by Intracytoplasmic Sperm Injection with Frozen-Thawed Epididymal Spermatozoa)

  • ICSI시 동결 융해한 부고환 정자의 이용 가능성을 알아보고자 난자의 배양시 체외성숙율과 활성화 처리를 한 난자와 동결 융해한 부고환 정자로 ICSI시 체외발생율을 조사하였으며, 결과를 요약하면 다음과 같다. 1. 난포란을 회수 후 24시간 배양하였을 때 배양 시간에 따른 GV, MI, M II로의 체외성숙율은 각각 7/60(11.7%), 5/60(8.3%), 48/60(80.0%)였고 30시간 배양 시간에 따른 GV, MI, M II로의 체외성숙율은 각각 3/60(5.0%), 4/60(6.7%), 53/60(88.3%)였고 퇴화란은 각각 2/60(3.3%)와 1/60(1.7%)였다. 2. 동결 융해한 부고환 정자를 이용하여 활성화 처리를 한 난자에 ICSI를 하였을 때 상실배와 배반포로의 체외발생율은 각각 12/46(26.1%), 22/46 (47.8%)로서 비활성화처리 난자군 5/39 (12.8%), 10/39(25.6%)에 비해 높은 체외발생율을 나타냈다. 3. 활성화 처리를 한 난자에 신선정자, 부고환 정자 및 동결 융해한 부고환 정자로 ICSI시 체외 발생율은 각각 24/45(53.3%), 15/40(37.50%), l1/43 (25.6%)로서 신선정자에 비해 동결 융해한 부고환 정자처리군은 체외발생율은 약간 낮았지만 이용 가능성이 있음을 확인하였다.

  • This studies were carried out to investigate the effects of co-culture with cumulus cells, oviduct epithelial cells and uterine endometrium cells on the in-vitro fertilization and cleavage rate of bovine follicular oocytes. The ovaries were obtained from slaughtered Korean native cows. The follicular oocytes surrounded with cumulus cells were recovered by aspirating follicular fluids from the visible follicles of diameter 3~5 mm. The follicular oocytes were cultured in TCM-199 medium containing hormones and 10% FCS for 24~48 hrs in a incubator with 5% CO2 in air at 38.5$^{\circ}C$ and then matured oocytes were again cultured for 12~18 hrs with motile capacitated sperm by preincubation. The results obtained in these experiments were summarized as follows: 1. The in vitro maturatin and fertilization rate of bovine oocytes co-cultured with cumulus cells in TCM-199 medium were 64.0~74.1% and 40.0~58.6% respectively. And in-vitro fertilization rate of cumulus-enclosed oocytes(55.4%) were significantly(p<0.05) higher than cumulus-denuded oocytes(23.1%). 2. The in-vitro maturatin and fertilization rate of bovine oocytes co-cultured with 1$\times$104 cells/ml, 1$\times$106 cells/ml, 1$\times$108 cells/ml and 1$\times$1015 cells/ml oviduct epithelial cells in TCM-199 medium were 59.3% and 40.7%, 64.0% and 48.0%, 58.3% and 37.5%, 52.0% and 32.0%, respectively. 3. The in-vitro maturation and fertilization rate of bovine oocytes co-cultured with 1$\times$104 cells/ml, 1$\times$106 cells/ml, 1$\times$108 cells/ml and 1$\times$1015 cells/ml uterine endometrium cells in TCM-199 medium were 56.0% and 36.0%, 60.7% and 42.9%, 59.3% and 37.0%, 52.0% and 36.0%, respectively. 4. When the in-vitro fertilized oocytes were co-cultured with cumulus cells, oviduct epithelial cells and uterine endometrium cells, the development rate to be blastocyst was 12.2%, 15.6% and 11.7%, respectively and rates were higher than that of control, 2.1%(P<0.05).

체외수정배양액내에 첨가된 필수 .비필수아미노산, Taurine 또는 Glycine이 소 난자의 분할 및 체외발육에 미치는 영향 (Effect of Essential and Non-essential Amino Acids, Taurine or Glycine Supplemented to Fertilization Medium on In Vitro Cleavage and Development of Bovine Oocytes Matured and Fertilized In Vitro)

  • Essential and non-essential amino acids supplemented to culture medium stimulate mammalian embryo development in vitro. Amino acids such as glycine, taurine and alanine are concentrated in the lumen of oviduct and uterus and it can he thought that these amino acids may have physiological role on fertilization and embryo development. Our aim of this experiment was to investigate the effects of essential and non-essential amino acids, taurine or glycine supplemented to fertilization medium on the cleavage and subsequent in vitro development of bovine oocytes matured and fertilized in vitro. Immature oocytes were obtained from slaughtered Holstein cows and heifers and matured in TCM199 containing 10% fetal calf serum, 2.5 $\mu$g /mL of FSH and LH and 1 $\mu$g / mL of estradiol with granulosa cells in vitro. After maturation, oocytes were coincubated with sperm in fertilization medium supplemented with Minimum Essential Medium (MEM) essential and non-essential amino acids, taurine (3.75 mM) or glycine (10 mM) for 30 hours in vitro. Inseminated oocytes were cultured in synthetic oviduct fluid medium (SOEM) containing MEM essential, non-essential amino acids and 1 mM glutarnine up to 8 days after fertilization.Supplementation of fertilization medium with MEM essential and non-essential amino acids lowered significantly (p<0.05 and p<0.001) the cleavage rate after 30 hours of IVF (53.3%) and at Day 3 (62.7%: Day 0: the day of I VF) compared to control (64.3% and 77.3%, respectively). Subsequent developmental rates to morulae (Mo) and expanding blastocysts (ExBL) also significantly decreased (p<0.001 and p<0.05 for Mo and ExBL) when oocytes were coincubated with sperm in the medium containing MEM amino acids. Taurine added to fertilization medium have not increased the cleavage rate over the control, whereas glycine showed significantly lower (p<0.01) cleavage rate at Day 3 than that of taurine, but there was no significant difference in the developmental rates to Mo and ExBL of bovine embryos irrespective of the supplementation of taurine or glycine to fertilization medium. In conclusion, supplementation of fertilization medium with essential and non-essential amino acids, taurine or glycine has no beneficial effect on in vitro cleavage and development of bovine oocytes matured and fertilization in vitro.

  • These studies were carried out ot investigate the effects of estrous cow serum(ECS), fetal calf serum(FCS), bovine follicular fluid(BFF) and matured cumulus cell(MCC) on in vitro maturation and fertilization of bovine follicular oocytes. The ovaries were obtained from slaughtered Korean native cows. The follicular oocytes surrounded with cumulus cells were recovered by aspirating follicular fluid from the visible follicles of diameter 3-5mm. The follicular oocytes were cultured in TCM-199 medium containing hormones, FCS, ECS, BFF and MCC for 24~48 hrs. in a incubator with 5% CO2 in air at 38.5$^{\circ}C$ and then matured oocytes were again cultured for 18$^{\circ}C$20 hrs. with motile capacitated sperm in the TCF(Tyrode calcium-free) solution containing 200$\mu\textrm{g}$/ml of heparin. The results obtained in these experiments were summarized as follows : 1. The oocytes classified as "A, B, C, D and Degenerative" depending morphological integrity and those were 61.4%, 12.1%, 19.2%, 4.2% and 3.0% of the total oocytes harvested, respectively. The maturation and fertilization rate of the A, B, C class follicular oocytes, cultured in the TCM-199 medium supplemented with 10% FCS were 89.1%, 78.0%, 52.6% and 78.1%, 33.3%, respectively. 2. The maturation and fertilization rate of the follicular oocytes cultured in TCM-199 medium supplemented with 5%~20% ECS and FCS were 74.0%~80.6, 26.2%~30.0% and 71.7%~76.9%, 51.9%~58.0%, and those values were higher the supplement of ECS than FCS. 3. The maturation rate(68.0%~64.6%) and fertilization rate(59.6%~60.4%) of follicular oocytes cultured in TCM-199 medium supplemented with 10% FCS and 20~30% BFF were higher than those of follicular oocytes cultured TCM-199 medium supplemented with 10% FCS and 10% and 50% BFF. 4. The maturation rate(76.5%) and fertilization rate(61.7%) of follicular oocytes cultured in TCM-199 medium supplemented with 10% FCS and 1$\times$106/ml cumulus cells were higher than those of follicular oocytes cultured in TCM-199 medium supplemented with 10% FCS and 1$\times$104~5/ml and 1$\times$108/ml cumulus cells.lus cells.

  • The studies were carried out to investigate the effects of co-culture with cumulus cells and oviduct epithelial cells on the in vitro fertilization and cleavage rate of bovine follicular cocytes and to determine the optimum thawing temperature and equilibration time on in vitro developmental rate of frozen bovine embryos. The ovaries were obtained from slaughtered Korean native cows. The follicular oocytes were cultured in TGM-199 medium containing 10 IU /ml의 PM SG, 10 IU /ml의 hCG, ip g/ml의 $\beta$-estradiol and 10% FCS for 24~48 hrs in incubator with 5% $CO_2$ in air at 38.5$^{\circ}C$. The bovine embryos following dehydration by cryoprotective agents and a various concentration of sucrose were directly plunged into liquld nitrogen and thawed in 3$0^{\circ}C$ water. Survival rate was defined as developmental rate on in vitro culture or FDA-test. The results are sunanarized as followes :1. The in vitro fertilization and in vitro developmental rates of bovine oocytes co-cultured with cumulus cells in TCM499 medium were 75.0~76.8% and 17.3~27.6%, respect-ively. And in-vitro fertilization rates of cumulus-enclosed oocytes(55.4%)were significantly(p<0.05) higher than cumulus-denuded oocytes (23.1%). 2. The in vitro fertilization and in vitro developmental rates of bovine oocytes co-cultured with l$\times$ l04cells /ml, 1 x l06cells /ml, lx l08cells /ml and 1 x l015cells /ml oviduct epithelial cells in TCM-199 medium were 74.5~77.8% and 15.7~21.20 respectively.3. The in-vitro fertilization and in vitro developmental rates of bovine oocytes cocultured in '1CM-199 media containing PMSG, hCG, PMSG+hCG. PMSG+$\beta$-estradiol, hCG+$\beta$-estradiol 0 to 40 hrs after insemination were 74.0~77.4% and l8.9~23.l%, re-spectiv ely.4.The survival rates of bovine embryos thawed after rapid freezing in the freezing medium containing a various concentration of sucrose added 1.5M and 2.OM glycerol,DMSO and propanediol were 23.5~31.4% and 20.6~34.l%, respectively. 5. The temperature thawed at 3$0^{\circ}C$ after rapid freezing of bovine embryos resulted in a significantly higher embryos survival rate than did at 2$0^{\circ}C$ and 35$^{\circ}C$.6. The equilibration time on the survival rates of bovine embryos was attained after short period of time(2.5~5 min.) in the freezing medium higher than long period of time (10~20min.). The maturation and fertilization rate of follicular oocytes, cultured in the TCM-199 medium supplemented with 5%, 10%, 15%, 20% FCS and hCG, HCG, $\beta$-estradiol were 76.0%~82.3% and 26.2%~70.0%, and those values were higher the supplementation of the hormone than the non-supplementation. 4. The fertilization and cleavage rate of the follicular oocytes, inseminated with spermatozoas of epididymis cauda, neat and frozen semen were 63.3%, 73.3%, 70.0% and 32.7%, 37.8%, 38.3, respectively. 5. The fertilization and cleavage rate of follicular oocytes, fertilized with capacitated spermatozoas by heparin, BFF and HIS methods were 70.0%, 53.8%, 34.2% and 38.3%, 23.1%, 17.1%, respectively. And the fertilization and cleavage rate were higher method of heparin than other methods.r methods.

The maturation and fertilization rate of the follicular oocytes cultured in TCM-199 medium supplemented with 5~20% ECS and FCS were 74.0~80.6%, 26.2~30.0% and 71.7~76.9%, 51.9~58.0%, and the values were higher the supplement of ECS than FCS. 7. The maturation rate (68.0~64.6%) and fertilization rate(59.6~60.4%) of follicular oocytes cultured in TCM-199 medium supplemented with 10% FCS and 20~30% BFF were higher than those of follicular oocytes cultured TCM-199 medium supplemented with 10% FCS and 10% and 50% BFF. 8. The maturation rate(76.5%) and fertilization rate (61.7%) of follicular oocytes cultured in TCM-199 medium supplemented with 10% FCS and $1{\times}10^6/ml$ MCC were higher than those of follicular oocytes cultured in TCM-199 medium supplemented with 10% FCS and $1{\times}10^{{4}{\sim}{5}}/ml$ and $1{\times}10^8/ml$ cumulus cells.

육질개량을 위한 한우번식우의 유전능력평가 (Evaluation of Genetic Ability for Meat Quality in Hanwoo Cow)

  • 본 연구는 강원도내 농촌지역 1개 군 관내 사육농가에서 사육되어 2004년 12월부터 2008년 6월까지 43개월 동안 강원도와 강원도 인근에 소재한 도축장 3개소에서 도축된 한우 거세우 6,431두의 성적과 한국종축개량협회에서 관리중인 한우 등록우 데이터베이스로부터 이들의 혈통자료 14,957두를 활용하여 도체형질에 대한 유전모수와 육종가를 추정하였다. 연구 결과를 요약하면 다음과 같다. 1. 각 도체형질에 대한 평균과 표준편차는 근내지방도, 등지방두께, 배최장근단면적, 냉도체중 및 육량지수에서 각각 $5.57{\pm}2.01$, $10.87{\pm}4.06mm$, $87.87{\pm}9.07cm^2$, $426.75{\pm}49.27kg$$65.80{\pm}3.80$으로 나타났다. 2. 단형질 분석을 통한 도체형질의 유전력 추정치는 근내지방도, 등지방두께, 배최장근단면적, 냉도체중 및 육량지수에서 각각 0.36, 0.35, 0.24, 0.29 및 0.40으로 나타났다. 3. 근내지방도는 등지방두께, 배최장근단면적, 냉도체중, 및 육량 지수와 각각 -0.21, 0.30, -0.21, 0.30의 유전상관 계수가 추정 되었으며, 등지방두께는 배최장근단면적, 냉도체중, 및 육량지수와 각각 -0.12, 0.57, -0.97의 유전 상관계수가 추정되었다. 4. 배최장근단면적은 냉도체중 및 육량지수와 각각 0.32, 0.27의 유전상관 계수가 그리고 냉도체중은 육량지수와 -0.62의 유전 상관계수가 추정되었다. 5. 근내지방도가 등지방 두께나 도체중과 음의 유전상관관계를 보이고 있는 점을 고려할 때 근내지방도가 양호한 방향으로 선발을 진행할 경우 등지방두께를 낮추는 간접선발 효과가 있을 것으로 기대된다. 그러나 근내지방도에 대한 선발은 도체중을 증가시키는데 장애를 초래할 수 있을 것으로 판단된다.
