Achaete-scute like 2 (ASCL2) gene encodes a member of the basic helix-loop-helix transcription factor which is essential for the maintenance of proliferating trophoblasts during placental development. ASCL2 gene preferentially expresses the maternal allele in the mouse. However, it escapes genomic imprinting in the human. In this study, the complete open reading frame consisting of 193 amino acids of ASCL2 gene was obtained. Sequence analysis indicated that a C-G mutation existed in the 3' region between Meishan and Large White pigs. The polymorphism was used to determine the monoallelic or biallelic expression with RT-PCR-RFLP in pigs of Large $White{\times}Meishan$$F_1$ hybrids. Imprinting analysis indicated that the ASCL2 gene expression was biallelic in all the tested tissues (heart, liver, spleen, lung, kidney, stomach, small intestine, skeletal muscle, fat, uterus, ovary and pituitary). PCR-RFLP was used to detect the polymorphism in 270 pigs of the "$Large\;White{\times}Meishan$" $F_2$ resource population. The statistical results showed highly significant associations of the genotypes and fat meat percentage (FMP), lean meat percentage (LMP) and ratio of lean to fat (RLF) (p<0.01), and significant associations of the genotypes and loin eye area (LEA) and internal fat rate (IFR) (p<0.05).

This study aims to investigate the patterns and incidence rates of morphological deformities during artificial fingerling production of blackhead seabream (BSB, Acanthopagrus schlegeli) juveniles by visual and soft X-ray observations of BSB juveniles (n=881) collected from four industrial hatcheries. Using both methods, we observed seven types of external deformity ($11.2{\pm}4.4%$) and eight types of skeletal deformity ($53.4{\pm}5.1%$). Lordosis occurred in all hatcheries with the highest frequency and the mean frequency was $13.1{\pm}4.4%$ (24.5% in total abnormal) by soft X-ray. Although they had a similar standard length (SL), juveniles with lordosis had a significantly lower condition factor and body weight than normally developed juveniles (P<0.05); normal distribution curves of histogram of body proportions, i.e., body height (BH)/head length (HL) and trunk (=SL-HL)/BH, indicated differences between normally developed juveniles and those with lordosis. In addition, 59% of juveniles with lordosis had vertebral curvature of less than $20^{\circ}$, generally at the $17^{th}$ vertebrae. It is expected that the above results will be used as basic data necessary for determining the cause of lordosis and for developing techniques to prevent this symptom in the fingering production industry of BSB juveniles.

Purpose: Alagille syndrome is a complex hereditary disorder that is associated with cardiac, hepatic, skeletal, ocular, and facial abnormalities. Mutations in the Notch signaling pathway, such as in JAG1 and NOTCH2, play a key role in embryonic development. A cardiac or hepatic presentation is a critical factor for determining the prognosis. Methods: We conducted a retrospective study of 41 patients with Alagille syndrome or a JAG1 mutation between 1983 and 2013. Results: The first presentations were jaundice, murmur, cyanosis, and small bowel obstruction at a median age of 1.0 months (range, 0-24 months). The JAG1 mutation was found in 27 of the 28 genetically-tested patients. Cardiovascular anomalies were identified in 36 patients, chronic cholestasis was identified in 34, and liver transplantation was performed in 9. There was no significant correlation between the severity of the liver and cardiac diseases. The most common cardiovascular anomaly was peripheral pulmonary stenosis (83.3%), with 13 patients having significant hemodynamic derangement and 12 undergoing surgical repair. A total bilirubin level of >15 mg/dL with a complex surgical procedure increased the surgical mortality (P=0.022). Eight patients died after a median period of 2.67 years (range, 0.33-15 years). The groups with fetal presentation and with combined severe liver and heart disease had the poorest survival (P<0.001). Conclusion: The group with combined severe liver and heart disease had the poorest survival, and a multidisciplinary approach is necessary to improve the outcome.

Dietary supplements derived from natural sources are an essential factor in optimizing athletic performance. It has been proposed that the extract of Momordica charantia (M. charantia) that is known as a bitter melon can be potentially used as a novel supplement for health promotion. This pilot study aimed to examine the effects of the M. charantia extract when administered in the form of a sports drink, and we evaluated changes in body composition and metabolic factors in tennis players after 4-week consumption of the extract. Eight male college tennis players were instructed to consume an M. charantia extract 6 times per day (3 in the morning and 3 in the afternoon, and the total daily intake was 600 ml). Collected data were analyzed using paired t-tests to examine the changes over time after consumption of the M. charantia extract. The results revealed a significant increase in the trunk muscle mass, basal metabolic rate, and daily calorie intake (p < 0.05). Levels of protein, minerals, and total body water showed an increased tendency (not statistically significant), whereas intracellular water and extracellular water showed a decreased trend. Furthermore, fat-free mass, skeletal muscle mass, and muscle mass showed an increased tendency. In conclusion, consumption of the M. charantia extract caused an increase in parameters related to protein, muscle mass, and metabolism. It seems that follow-up studies related to fatigue, inflammation, and stress hormones related to the M. charantia extract consumption would be needed.
Myotonic Dystrophies type 1 and 2 (DM1/2) are neuromuscular disorders which belong to a group of genetic diseases caused by unstable CTG triplet repeat (DM1) and CCTG tetranucleotide repeat (DM2) expansions. In DM1, CTG repeats are located within the 3' untranslated region of myotonin protein kinase (DMPK) gene on chromosome 19q. DM2 is caused by expansion of CCTG repeats located in the first intron of a gene coding for zinc finger factor 9 on chromosome 3q. The CTG and CCTG expansions are located in untranslated regions and are expressed as pre-mRNAs in nuclei (DM1 and DM2) and as mRNA in cytoplasm (DM1). Investigations of molecular alterations in DM1 discovered a new molecular mechanism responsible for this disease. Expansion of un-translated CUG repeats in the mutant DMPK mRNA disrupts biological functions of two CUG-binding proteins, CUGBP and MNBL. These proteins regulate translation and splicing of mRNAs coding for proteins which play a key role in skeletal muscle function. Expansion of CUG repeats alters these two stages of RNA metabolism in DM1 by titrating CUGBP1 and MNBL into mutant DMPK mRNA-protein complexes. Mouse models, in which levels of CUGBP1 and MNBL were modulated to mimic DM1, showed several symptoms of DM1 disease including muscular dystrophy, cataracts and myotonia. Mis-regulated levels of CUGBP1 in newborn mice cause a delay of muscle development mimicking muscle symptoms of congenital form of DM1 disease. Since expansion of CCTG repeats in DM2 is also located in untranslated region, it is predicted that DM2 mechanisms might be similar to those observed in DM1. However, differences in clinical phenotypes of DM1 and DM2 suggest some specific features in molecular pathways in both diseases. Recent publications suggest that number of pathways affected by RNA CUG and CCUG repeats could be larger than initially thought. Detailed studies of these pathways will help in developing therapy for patients affected with DM1 and DM2.
The relationship between bone mineral density and the environmental factors were investigated from the view point of preventing osteoporosis in Korean pubescent girls. The effects of calcium, nutrient intake, physical activity on total bone mineral density, lumbar spine and femoral bone mineral density and total bone mineral content were evaluated 33 healthy pubescent girls aged 14∼16y. A convenient method was used to assess nutritional and energy intake and calcium index was used together. Calcium intake in childhood was estimated by asking whether subjects usually drank milk as children. Eating habits data and history of menstruation were obtained by questionnaire and interview. Average energy expenditure was calculated. Bone mineral density and content were measured by dual energy x-ray absorptiometry using a Lunar DPX+Scanner (Lunar, Madison, WI). The lumbar spine(L2∼L4) and three sites in the proximal femur (femoral neck, trochanteric region, and Ward's triangle)were measured. Height and weight were measured, and the body mass index(BMI) was derived from the formula : BMI=kg/㎡ Statistical analysis was performed by simple correlation using the SAS package. The mean calcium intake (736mg) was below the RDA of 800mg/d. Twelve percent of the total subjects did not drink milk at all because they did not like the taste. Skipping meals, low calcium intake and low energy intake were significantly correlated with the low BMD. Also the data indicate that girls who reported drinking milk with every meal during childhood had significantly higher bone densities than girls who reported drinking milk less frequently. The results suggest that milk consumption in childhood appears to be needed not only for growth and development, but possibly also to assure an optimal peak of bone mass and thus greater latitude for the maintenance or skeletal integrity in the face of bone losses. There was a highly significant correlation between the total BMD and overall level of physical activity. Body weight was a better predictor of total BMD than was and other factor. Simple mechanical loading may explain why body weight, but total BMC was positively relatd to height. Conclusively, increasing calcium intake and physical activity in the pubescent girls could influence BMD.
UCP2 and UCP3 are members of the uncoupling protein family, which may play roles in energy homeostasis. In order to determine the regulation of the predominant expression of UCP3 in skeletal muscle, the effects of differentiation and myogenic regulatory factors on the promoter activities of the mouse UCP2 and UCP3 genes were studied. Reporter plasmids, containing approximately 3 kb of the 5'-upstream region of the mouse UCP2 and UCP3 genes, were transfected into C2C12 myoblasts, which were then induced to differentiate. Differentiation positively induced the reporter expression about 20-fold via the UCP3 promoter, but by only 2-fold via the UCP2 promoter. C2C12 myoblasts were cotransfected with expression vectors for myogenin and/or MyoD as well as reporter constructs. The simultaneous expression of myogenin and MyoD caused an additional 20-fold increase in the reporter expression via the UCP3 promoter, but only a weak effect via the UCP2 promoter. In L6 myoblasts, only MyoD activated the UCP3 promoter, but in 3T3-L1 cells neither factor activated the UCP3 promoter, indicating that additional cofactors are required, which are present only in C2C12 myoblasts. The expression of UCP2 and UCP3 is differentially regulated during muscle differentiation due to the different responsiveness of their promoter regions to myogenin and MyoD.
A high prevalence of farmer's work-related musculo-skeletal disorders (MSDs) have been reported in precedent studies. Risk factors - physical, chemical, biological, ergonomic risk factor etc. - in farm-work were identified by previous studies. To prevent farmers from exposing to them and reduce, Agricultural Safety Technology were introduced and used on farm. The purpose of this study is to review and summarize the agricultural safety technology on farm in Korea. According to strategy principles, this technology is classified as four criteria - workload reduction by reducing awkward posture, workload reduction by using manual material handling aids, Comfort improvement by protecting worker from hot, cold environment and work efficiency improvement by workplace redesign and cultivation modification. This technology includes transporting tools, working cloth, personal protective equipment, and pesticide-proof Clothing. Anthropometric study of Korean farmers was also carried out to support the development of equipment and clothes. In long terms, it needs to precaution activity from risk factors by using cultivation modification and working improvement. Although working condition gets better and workload reduce by this technology, it required customized safety technology in each crops - rice cultivation, horticulture and livestock farming. And farmers should use the assistive technology and participate voluntarily in ergonomic intervention program.

Rapidly progressive interstitial renal fibrosis has recently been reported in young women who have been on a slimming regimen including chinese herbs. Aristolochic acid, suspected as the causal factor of this renal disease, is a well known carcinogen. It has been known that Madouling (Aristolochiae fructus) contains aristolochic acid. The objective of this study was to investigate the effects of Madouling, Madouling-tang, which are the extract mixture from 10 different chinese herbs including Madouling, and aristolochic acid on reproductive and developmental toxicity. Female rats were administered orally with the extracts of Madouling, madouling-tang, and aristolochic acid from 14 days before mating to day 17 of gestation. Madouling (8mg/kg) decreased fertility in the 8mg/kg group, but Madouling-tang and aristolochic acids did not. Significant decrease of mean fetal body weights were observed in the 16mg/kg group of aristolochic acids. External, visceral and skeletal malformation of fetuses were not observed with treatment. Histopathological examination showed the discrete damage of kidney in the 8mg/kg group of Madouling and 16mg/kg groups of aristolochic acid. In whole embryo culture, Madouling and Madouling-tang caused the retardation of growth and development of embryo in the dose of 1 $\mu$g/ml and 0.02 $\mu\textrm{g}$/kg, respectively while aristolochic acids showed the similar effect in the dose of 300 $\mu\textrm{g}$/kg. These results indicate that Madouling, up to 0.05mg/kg (prescription dose to human) has no adverse effects on the fertility, reproduction and development of Sprague-Dawley rats.
Therapeutic angiogenesis is the controlled induction or stimulation of new blood vessel formation to reduce unfavourable tissue effects caused by local hypoxia and to enhance tissue repair. Therapeutic ultrasound can be considered as a physical agent to deliver therapeutic angiogenesis. The purpose of this study was to evaluate the effect of therapeutic ultrasound after muscle contusion injury by observed immunoreactivity of vascular endothelial growth factor(VEGF) that plays an important role in angiogenesis and substance-P in pain transmission. Ultrasound irradiation(1MHz, $1W/cm^2$, continuous mode, treatment time 5 min) was applied through water submersion technique to 1 limb daily by kept off 5cm from muscle belly of gastrocnemius. The result of this study were as follows. 1. In morphological observation, there were no significant changes excepts of 7 days. At 7 days, granular tissue viewed abundantly in control group. In other groups, general feature were increased interspace of muscle fiber; centronucleated muscle fiber; collapsed of muscle and nerve tissue; appeared inflammatory cell. 2. The VEGF was expressed in interspace of muscle fiber. Especially, at 7 days in experimental group, VEGF was showed in connective tissue surrounding gastrocnemius muscle. 3. The VEGF was higher expressed in experimental group at 2 and 3 days, but in control group at 7 days. These data suggest therapeutic ultrasound enhanced production of VEGF in the early day relatively, therefore stimulated angiogenesis in the skeletal muscle induced contusion injury. Also therapeutic ultrasound may stimulate pain relief by diminish of substance-P in dorsal horn of spinal cord.
