원위치 반응존 공법 적용을 위한 대수층내 오염물질 및 환경영향인자의 계절 특성 평가 (Investigation of Seasonal Characteristics of Contaminants and Hydrochemical Factors in an Aquifer for Application of In Situ Reactive Zone Technology)

  • A field investigation was conducted on an aquifer contaminated with trichloroethylene (TCE) for application of in situ reactive zone treatment using nanosized zero-valent iron (NZVI). The aquifer was an unconfined aquifer with a mean hydraulic conductivity of $5.14{\times}10^{-4}cm/sec$, which would be favorable for NZVI injection. Seasonal monitoring of TCE concentration revealed a presence of non-aqueous phase liquid form of TCE near IW (injection well). The hydrochemical data characterized the site groundwater to be a $Ca-HCO_3$ type. The average value of Langelier Saturation Index of the groundwater was -1.33, which implied that the site was favorable for corrosion of NZVI. Dissolved oxygen (DO) concentration varied between 2.5~11.5 mg/L, which indicated that DO would greatly compete with TCE as an electron acceptor. The hydrogeological and hydrochemical characterization reveals that the time around November would be appropriate for NZVI injection when water level and temperature are relatively high and DO concentration is low.

  • TEPC (Tissue Equivalent Proportional Counter) was usually used for high LET radiation dosimetry. We developed a prototype TEPC for micro-dosimetry in the range of $0.2{\sim}300 keV/{\mu}m$. And, the simulated site diameter of the TEPC is $2{\mu}m$, of similar size to a cell nucleus. For purposes of characterization the response for high LET radiation of the TEPC has been investigated under 135MeV/u Carbon ions in HIMAC (Heavy Ion Medical Accelerator). We determined the gas multiplication factor and measured the lineal energy spectrum [yd(y)] of 135 MeV/u Carbon ions. The value of the gas multiplication factor was 315 at 700 V bias voltage. As a result of the experiment, we could more understand the performance of the TEPC for high LET (Linear Energy Transfer) radiation. And the procedure of high LET radiation dosimetry using TEPC is established.

Radiochemical Analysis of Filters Used During the Decommissioning of Research Reactors for Disposal

  • The decommissioning of nuclear facilities produces various types of radiologically contaminated waste. In addition, dismantlement activities, including cutting, packing, and clean-up at the facility site, result in secondary radioactive waste such as filters, resin, plastic, and clothing. Determining of the radionuclide content of this waste is an important step for the determination of a suitable management strategy including classification and disposal. In this work, we radiochemically characterized the radionuclide activities of filters used during the decommissioning of Korea Research Reactors (KRRs) 1 and 2. The results indicate that the filter samples contained mainly 3H (500-3,600 Bq·g-1), 14C (7.5-29 Bq·g-1), 55Fe (1.1- 7.1 Bq·g-1), 59Ni (0.60-1.0 Bq·g-1), 60Co (0.74-70 Bq·g-1), 63Ni (0.60-94 Bq·g-1), 90Sr (0.25-5.0 Bq·g-1), 137Cs (0.64-8.7 Bq·g-1), and 152Eu (0.19-2.9) Bq·g-1. In addition, the gross alpha radioactivity of the samples was measured to be between 0.32-1.1 Bq·g-1. The radionuclide concentrations were below the concentration limit stated in the low- and intermediatelevel waste acceptance criteria of the Nuclear Safety and Security Commission, and used for the disposal of the KRRs waste drums to a repository site.

Structural Analysis of ${\alpha}$-L-Arabinofuranosidase from Thermotoga maritima Reveals Characteristics for Thermostability and Substrate Specificity

  • An ${\alpha}$-L-arabinofuranosidase (TmAFase) from Thermotoga maritima MSB8 is a highly thermostable exo-acting hemicellulase that exhibits a relatively higher activity towards arabinan and arabinoxylan, compared with other glycoside hydrolase 51 family enzymes. In the present study, we carried out the enzymatic characterization and structural analysis of TmAFase. Tight domain associations found in TmAFase, such as an inter-domain disulfide bond (Cys306 and Cys476) in each monomer, a novel extended arm (amino acids 374-385) at the dimer interface, and total 12 salt bridges in the hexamer, may account for the thermostability of the enzyme. One of the xylan binding determinants (Trp96) was identified in the active site, and a region of amino acids (374-385) protrudes out forming an obvious wall at the substrate-binding groove to generate a cavity. The altered cavity shape with a strong negative electrostatic distribution is likely related to the unique substrate preference of TmAFase towards branched polymeric substrates.

장심도 시추공을 이용한 KURT 주변의 지질구조 연구 (Study on the Geological Structure around KURT Using a Deep Borehole Investigation)

  • 고준위방사성폐기물처분을 위한 부지특성평가 기술을 구축하고, 이를 활용하여 심부지질환경을 이해하기 위해 1997년부터 지금까지 한국원자력연구원 주변 지역을 고준위폐기물처분을 위한 연구지역으로 선정하여 다양한 지질 관련 연구를 수행해왔다. 특히, 2002년에는 고준위폐기물의 처분 대상 심도의 시추공(지하 500 m)을 굴착하였으며, 2006년에는 지하처분연구시설(KURT, KAERI Underground Research Tunnel)을 준공하여 연구지역에 대한 심부지질환경 규명을 위해 노력하고 있다. 현재, 지하처분연구터널의 좌측 연구용 모듈 내에 500 m 길이의 장심도 시추공을 굴착하여 다양한 부지특성평가 연구를 수행 중에 있다. 본 연구는 고준위폐기물의 심지층 처분을 위한 요소 기술인 심부영역의 부지특성평가기술 구축을 위해 장심도 시추공 자료를 이용하여 KURT 주변의 지질구조 분석하였다. 연구지역의 장심도 시추공에서 지구물리탐사, 시추공 지질조사 등 다양한 현장 조사를 수행하였으며, 그 결과 총 7개의 지질구조를 도출하였다. 이 연구 결과는 KURT 지역의 부지특성연구의 주요한 한 부분으로서 기존에 구축된 지질모델을 보완하는데 이용될 예정이다.

원자력이용시설 주변의 지하수 감시공의 위치와 심도 선정 (Determination of Location and Depth for Groundwater Monitoring Wells Around Nuclear Facility)

  • 원자력이용시설에서 유출된 방사성 오염물질은 지표수나 지하수의 유동에 따라 이동할 수 있다. 이 중에 지표수에 의해 이동하는 오염물질은 비교적 감시가 용이하지만, 지하수를 따라 이동하는 오염물질은 대상 매질에서의 지하수흐름에 대한 정보를 알아야 하므로 감시가 매우 어렵다. 그러므로 지하수에 의한 오염물질의 이동을 규명하기 위해서 지질환경의 특성화가 선행되어야 한다. 본 연구에서는 연구부지에 건설된 가상의 원자력이용시설에 대한 감시공의 위치를 결정하고, 감시공에서의 심도별 감시 구간을 선정하는 방법론을 제안하였다. 감시공의 위치를 결정하기 위해 지하수유동 모델링을 수행하였고, 그 결과 지하수 흐름의 하류 지역에 감시공의 위치를 선정하였으며, 감시공에서 수행한 현장조사 결과를 바탕으로 비교적 지하수의 흐름이 빠른 구간을 대상으로 감시 구간을 선정하였다. 본 연구를 통해 개발된 모니터링 방법론은 국내 원자력 발전소를 포함한 원자력이용시설 뿐만 아니라, 유류비축시설의 오염물질, 농업 관련 지하수 오염의 감시 등 다양한 분야에서 잠재적으로 지하수에 유입될 수 있는 오염물질을 조기 감시하는 데에 활용할 수 있을 것이다.

Characterization and Genetic Diversity of Benzimidazole-resistant and -sensitive Monilinia fructicola Isolates in Korea

  • To characterize benzimidazole-resistant and -sensitive Monilinia fructicola populations, the fungal isolates were obtained from peach plants showing brown rot and bloosom blight. Benzimidazole-sensitive isolates did not grow on potato dextrose agar(PDA) amended with $\geq1.0{\mu}g$ active ingredient(a.i.)/ml of the fungicides. However, benzimidazole-resistant isolates grew on PDA regardless of the tested concentrations of fungicides. Benzimidazole-resistant isolates did not grow on diethofencarb-PDA, but sensitive isolates grew on the same PDA. In the nucleotide sequences of $\beta$-tubulin gene, only codon 198(GAG: glutamic acid), a target site for benzimidazole, was replaced with GCG(alanine) in all of the resistant isolates, and this substitution seems to play an important role in the development of resistance. Other interesting codons such as 165(GCT), 200(TTC), and 241(GCT) were not changed among the isolates. Benzimidazole-resistant and -sensitive isolates were clustered clearly in random amplified polymerphic DNA analyses and the results revealed that low levels of genetic diversity between benzimidazole-sensitive and -resistant isolates of M. fructicola in the investigated regions.

Assessment of Corrosion Lifetime of a Copper Disposal Canister Based on the Finnish Posiva Methodology

  • In this paper, an approach developed by the Finnish nuclear waste management organization, Posiva, for the construction license of a geological repository was reviewed. Furthermore, a computer program based on the approach was developed. By using the computer program, the lifetime of a copper disposal canister, which was a key engineered barrier of the geological repository, was predicted under the KAERI Underground Research Tunnel (KURT) geologic conditions. The computer program was developed considering the mass transport of corroding agents, such as oxygen and sulfide, through the buffer and backfill. Shortly after the closure of the repository, the corrosion depths of a copper canister due to oxygen in the pores of the buffer and backfill were calculated. Additionally, the long-term corrosion of a copper canister due to sulfide was analyzed in two cases: intact buffer and eroded buffer. Under various conditions of the engineered barrier, the corrosion lifetimes of the copper canister due to sulfide significantly exceeded one million years. Finally, this study shows that it is necessary to carefully characterize the transmissivity of rock and sulfide concentration during site characterization to accurately predict the canister lifetime.

Crystal Structure and Biochemical Characterization of Xylose Isomerase from Piromyces sp. E2

  • Biofuel production using lignocellulosic biomass is gaining attention because it can be substituted for fossil fuels without competing with edible resources. However, because Saccharomyces cerevisiae does not have a ${\text\tiny{D}}$-xylose metabolic pathway, oxidoreductase or isomerase pathways must be introduced to utilize ${\text\tiny{D}}$-xylose from lignocellulosic biomass in S. cerevisiae. To elucidate the biochemical properties of xylose isomerase (XI) from Piromyces sp. E2 (PsXI), we determine its crystal structure in complex with substrate mimic glycerol. An amino-acid sequence comparison with other reported XIs and relative activity measurements using five kinds of divalent metal ions confirmed that PsXI belongs to class II XIs. Moreover kinetic analysis of PsXI was also performed using $Mn^{2+}$, the preferred divalent metal ion for PsXI. In addition, the substrate-binding mode of PsXI could be predicted with the substrate mimic glycerol bound to the active site. These studies may provide structural information to enhance ${\text\tiny{D}}$-xylose utilization for biofuel production.

사람에서 유래한 장독성 대장균의 이열성장독소와 관련된 Plasmid 특성 (Characterization of Plasmid Encoding a Heat-labile Enterotoxin Originated from an Human Escherichia coli Strain O15: H11.)

  • A heat-labile enterotoxin and no heat-stable enterotoxin producing($LT^+ST^-$) plasmid (110 kilobases in size) was isolated from an enterotoxigenic Escherichia coli of human strain O15:H11 and used for analysis of the $LT^+$ deoxyrionucleic acid region using recombinant DNA technology. A DNA segment containing the $LT^+$ DNA region which was one restriction endonuclease BamHl fragment(6.2 kb in size) was joind to a small multicopy plasmid, pUC9. E. coli K-12 strain, JM103 harboring the chimeric plasmid produced greater amounts of LT than did the enterotoxigenic E. coli O15:H11 strain. The BamHl fragment was further digested with various restriction endonucleases and contained no HindIll restriction site which is an essential in $LT^+ST^+$ plasmid. The detailed DNA sequencing of this $LT^+ST^-$ plasmid is required.
