ISSR marker-assisted selection of male and female plants in a promising dioecious crop: jojoba (Simmondsia chinensis)

  • Simmondsia chinensis (Link) Schneider, a multipurpose and monogeneric dioecious shrub from arid zones, has emerged as a cash crop all over the globe. Its seed propagation poses severe problems due to its male-biased population: the male:female ratio is 5:1. Investigations have been carried out to generate a sex-specific Inter-simple sequence repeat (ISSR) marker for the early detection of male and female plants. Of the 42 primers analysed with a bulk sample of pooled male DNA and a bulk sample of pooled female DNA, only one primer, UBC-807, produced a unique ~1,200 base-pair fragment in the male DNA. To validate this observation, this primer was re-tested with individual male and female samples from eight cultivars. A similar unique ~1,200 bp fragment was present in the male individuals of all eight cultivars and completely absent in the female individuals tested. This is the first report of the use of ISSR markers to ascertain sex in physiologically mature S. chinensis plants.

  • Allium속에 포함된 6종의 122점을 수집하고, 이 종들의 유전적 관계는 ISSR 마커를 이용하여 확인하였다. 형태적 분석은 6개의 양적 형질을 측정하고 1개의 질적 형질은 수치화하였다. SSR 분석은 17개의 primer를 사용하여 총 370개의 다형성 밴드를 얻었다. 형태적 특성 분석은 유전적 거리로 구분할 경우 3개의 그룹으로 분류되었으나, 부분적으로 몇 몇 종들은 분류에 어려움이 있었다. ISSR 결과를 바탕으로 Allium 속의 군집분석은 5개의 그룹으로 분리되었다. 형태적 분석과 SSR분석 간의 상관 관계는 유의성이 매우 낮았다(r = 0.036). 따라서 본 연구에서 개발한 ISSR 마커는 달래와 산달래의 분류와 교배 육종에 유용하게 이용될 수 있을 것이다.

ISSR 분석으로 살펴본 애기등의 유전적 다양성 (Genetic diversity of Millettia japonica in Korea as revealed by ISSR analysis)

  • 최근 환경부 멸종위기종 급에서 해제된 애기등(Millettia japonica) 집단의 유전적 다양성 분석을 위해 10개 집단(한국 9집단, 일본 1집단) 189개체에 대한 ISSR (Inter-Simple-Sequence-Repeat) 분석을 수행하였다. 조사된 애기등의 유전적 다양성은 같은 과내의 멸종위기종보다 더 높은 것으로 조사되었다(Shannon's information index: I = 0.2689). 집단별 유전적 다양성은 전북 고창(I = 0.2968) 집단과 경남 남해(I = 0.2951), 경북 토함산(I = 0.2823) 집단이 높았으며, 일본 큐슈(I = 0.2487) 집단이 가장 낮았다. 애기등 10개 집단이 공유하는 유전변이의 양은 전체 유전변이의 86.49%로 나타났고, 전체의 13.51%가 집단간 유전적 차이에 의한 것으로 나타났다. 또한 조사된 애기등 집단간 교류를 나타내는 Nm 값(1.8446)이 비교적 높은 것으로 나타났으며, 이에 따라 집단간 유전적 분화가 크게 일어나지 않았음을 알 수 있었다. 본 연구 결과 애기등의 유전자원 보존을 위해서는 유전다양도가 높은 집단을 중심으로 지속적인 자생지 모니터링이 요구되며, 현지외 보존을 위해서는 더 높은 유전적 다양도를 지닌 전북 고창, 경남 남해, 경북 토함산 집단에서 다수 개체를 선발하는 보존 전략이 적절할 것으로 판단된다.

SSR을 이용한 꽃치자와 열매치자의 유전적 관계 (Genetic Relationships between Gardenia jasminoides var. radicans and G. jasminoides for. grandiflora Using ISSR Markers)

  • 꽃치자(Gardenia jasminoides var. radicans)와 열매치자(C. jasminoides for. grandiflora)는 Gardenia속으로는 우리나라에 두 분류군밖에 없으며 열매치자는 약용, 식용, 꽃치자는 방향제로 쓰인다. 그런데 꽃이 지고 나면 형태적으로 구분이 거의 되지 않는다 ISSR분석으로 이들 종의 유전적 다양도와 집단구조를 실시하였다. 88개의 DNA 분절에서 한 분류군에만 나타나는 특이밴드가 탐지되었다. 한국의 세 야생 집단은 분리되어 있고 패치 분포를 보이지만 재배종에 비해 높은 유전적 다양성을 유지하친 있었다. 열매치자가 꽃치자보다 유전적 다양성이 높았으며 ISSR 마커로 이들 분류군이 잘 분리되었다. 또한 야생 집단이 재배 집단보다 다양성이 약간 높으나 유의성은 없었다. 이는 재배화과정에서 유전적 다양성의 일부 상실이 있었으나 인위적인 채취와 식재로 야생 집단과 재배 집단의 유전적 교류가 존재하였음을 시사한다.

Genetic Relationships of Panax Species by RAPD and ISSR Analyses

  • This study was carried out to develop convenient and reproducible methods for identifying the genetic relationship among germplasms of Panax species based on molecular genetics. Using random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) analyses, genetic polymorphism of the Panax species was investigated with following cultivars and accessions, such as Chunpoong, Yunpoong, Kopoong, Sunpoong, and Kumpoong in domestic cultivars, Hwangsuk, Jakyung and Suckju in domestic accessions, and Panax quinquefolius L. and Panax japonicus C.A. Meyer in foreign introduced accessions, respectively. Specific DNA fragments ranging from 200 to 3,000 base pairs in size could be obtained with various ISSR and RAPD primers under the optimized PCR conditions. The dissimilarity coefficients among the genetic polymorphisms of ginseng cultivars and accessions were calculated from 0.26 to 0.90 in RAPD and from 0.12 to 0.89 in ISSR analysis, respectively. Eleven plant samples were grouped siblings together with cultivars and parents based on cluster analysis of genetic distance depending on genetic property such as origin of the species. In results, both RAPD and ISSR analyses were useful for identifying the genetic relationship among cultivars and accessions of Panax species at DNA level.

한반도 아고산대 특산·희귀식물 설앵초의 유전적 다양성과 지리적 분화 (Genetic diversity and geographic differentiation in the endangered Primula farinosa subsp. modesta, a subalpine endemic to Korea)

  • 북방계 식물들의 잔존적 피난처인 아고산대에 분포하는 식물들은 기후변화에 따른 자생지 환경변화로 인해 멸종 및 멸절 위험에 크게 직면해 있는 실정이다. 본 연구는 한반도의 남부지역 아고산대에 제한적으로 분포하고 있는 특산식물이며 희귀식물인 설앵초(Primula farinosa subsp. modesta (Bisset & Moore) Pax)에 대한 유전적 다양성과 지리적 분화 양상을 구명을 통한 보존전략을 수립하기 위해 3지역 6집단 165개체를 대상으로 ISSR (Inter Simple Sequence Repeat) 분석을 수행하였다. 그 결과 집단 수준의 유전 다양성의 평균은(P = 60.62, SI = 0.299, h = 0.190) 유사한 생활사를 갖는 다년생 초본류의 평균보다 낮은 수준이었으며, 이형화주의 특징을 갖는Primula속의 근연 분류군들에 비해서도 다소 낮은 결과를 보였다. 유전적 분화도 구명을 위해 AMOVA(Analysis of molecular variance) 분석 결과 전체 유전변이 중 약 50%가 지역 간에 분포하는 결과를 보여 종내 지역간 분화도가 매우 높은 수준으로 판단되며, Bayesian cluster 분석 결과에서도 조사된 세 지역이 각각 독특한 유전적 cluster를 형성함으로써 개체군간 유전자 이동이 매우 제한적임을 암시하였다. 따라서 설앵초 집단의 급속한 분획화와 낮은 수준의 유전적 다양성, 지역간 높은 분화도 등의 특성들을 고려하여 설앵초 집단의 개체군 감소 및 멸절 방지를 위한 현지 내 외 보전 전략 수립이 시급한 것으로 판단된다.

Genetic characterization of microsporidians infecting Indian non-mulberry silkworms (Antheraea assamensis and Samia cynthia ricini) by using PCR based ISSR and RAPD markers assay

  • This study established the genetic characterisation of 10 microsporidian isolates infecting non-mulberry silkworms (Antheraea assamensis and Samia cynthia ricini) collected from biogeographical forest locations in the State of Assam, India, using PCR-based markers assays: inter simple sequence repeat (ISSR) and random amplified polymorphic DNA (RAPD). A Nosema type species (NIK-1s_mys) was used as control for comparison. The shape of mature microsporidian spores were observed oval to elongated, measuring 3.80 to $4.90{\mu}m$ in length and 2.60 to $3.05{\mu}m$ in width. Fourteen ISSR primers generated reproducible profiles and yielded 178 fragments, of which 175 were polymorphic (98%), while 16 RAPD primers generated reproducible profiles with 198 amplified fragments displaying 95% of polymorphism. Estimation of genetic distance coefficients based on dice coefficients method and clustering with un-weighted pair group method using arithmetic average (UPGMA) analysis was done to unravel the genetic diversity of microsporidians infecting Indian muga and eri silkworm. The similarity coefficients varied from 0.385 to 0.941 in ISSR and 0.083 to 0.938 in RAPD data. UPGMA analysis generated dendrograms with two microsporidian groups, which appear to be different from each other. Based on Euclidean distance matrix method, 2-dimensional distribution also revealed considerable variability among different identified microsporidians. Clustering of these microsporidian isolates was in accordance with their host and biogeographic origin. Both techniques represent a useful and efficient tool for taxonomical grouping as well as for phylogenetic classification of different microsporidians in general and genotyping of these pathogens in particular.

Morphological and Genetic Stability of Dormant Apple Winter Buds After Cryopreservation

  • Twenty apple germplasm accessions from the Korean Genebank were successfully cryopreserved using two-step freezing to back up genetic resources maintained by field collections. This study examined the morphological and genetic stability of cryopreserved dormant apple buds that were stored in liquid nitrogen, and then rewarmed and regrown. Whole plants were regenerated directly from dormant buds through budding without an intermediary callus phase. The cryopreserved buds produced high levels of shoot formation (76.2-100%), similar to those of noncryopreserved buds (91.3-100%), with no observed differences between cryopreserved and noncryopreserved materials. Three of the twenty cryopreserved apple germplasm accessions were used to assess morphological and genetic stability. No differences in morphological characteristics including shoot length, leaf shape, leaf width/length ratio, and root length were observed between controls (fresh control and noncryopreserved) and cryopreserved plantlets. The genetic stability of regenerants (before and after cryopreservation) was investigated using inter simple sequence repeat (ISSR) markers. The ISSR markers produced 253 bands using four primers, ISSR 810, SSR 835, ISSR 864, and ISSR 899. These markers showed monomorphic banding patterns and revealed no polymorphism between the mother plant and regenerants before and after cryopreservation, suggesting that cryopreservation using two-step freezing does not affect the genetic stability of apple germplasm. These results show that two-step freezing cryopreservation is a practical method for long-term storage of apple germplasms.

Assessment of Genetic Diversity in Different Populations of Raily Ecorace of Indian Tasar Silkworm, Antheraea Mylitta Drury Using ISSR Markers

  • Raily ecorace of Indian tasar silkworm is wild in nature and distributed abundantly in dense deciduous forest on Shorea robusta (Sal) in Bastar ($17^{\circ}4'$ and $20^{\circ}34'$ N, $80^{\circ}15'$ and $82^{\circ}15'$ E and altitude ranging from 150 to 1200 mMSL) forest ranges of Chhattisgarh, India. It is represented by about 20 populations. Out of those, eleven populations showed intra- as well as inter- population variability based on phenotypic expression and also in major economic traits viz. cocoon weight, shell weight, filament length and denier. Genetic diversity in these eleven populations was studied using Inter-Simple Sequence Repeat (ISSR) markers. The band profiles generated with eight ISSR primers have depicted variation in band size. All the primers exhibited polymorphism which is an indicative of the genetic variation in individual Raily silkworm. Among the populations, total polymorphism recorded was 76%. The population genetic aspects assessed through POPGENE software package are discussed in the paper. Nei's gene diversity (h) ranged from 0.194 to 0.337 exhibiting high heterozygosity. Relevance of the present study is of high significance in formulating conservation strategies and sustainable utilization of the economically important Raily ecorace of Antheraea mylitta.

Assessment of genetic diversity of Prangos fedtschenkoi (Apiaceae) and its conservation status based on ISSR markers

  • Prangos fedtschenkoi (Regel et Schmalh.) Korovin (Apiaceae) is an endemic species for mountainous Middle Asia, which is both a rare and useful plant. Organic extractions from this species are being used in pharmaceutics and cosmetology. In recent years, P. fedtschenkoi distribution area has considerably decreased, presumably, resulting from human activities such as agriculture, construction works, overgrazing and collection from wild for pharmaceutic purposes. Six populations were found in Uzbekistan and their genetic divergence and differentiation were studied with 10 inter-simple sequence repeat (ISSR) markers, selected out of 101. Totally 166 amplified ISSR fragments (loci) were revealed, of which 164 were polymorphic. Relatively moderate level of polymorphism was found at population level with polymorphic bands ranging from 27.71% to 47.59%. Mean P = 39.05%, $N_a=1.40$, $N_e=1.25$, S.I. = 0.21, and $H_e=0.14$ were revealed for all loci across six populations. AMOVA showed higher variation among populations (62%) than within them (38%). The Bayesian model determined 5 clusters, or genetic groups. The posteriori distribution of the Theta II estimator detected full model identifying high inbreeding, intensified by low gene flow (Nm = 0.3954). Mantel test confined population 6 as distinct cluster corresponding to geographic remoteness (R = 0.5137, $p{\leq}0.005$). Results were used as the bases for developing conserve measures to restore populations.
