Heat Stability and Glucose-Lowering Effect of 1-Deoxynojirimycin from Silkworm (Bombyx mori) extract Powder

  • Silkworm powder, which contains 1-deoxynojirimycin (DNJ), is a promising complementary and alternative medicine (CAM) in Korea. Silkworm powder was produced from Yeonnokjam pupae at d 3 of the 5th instar at the National Academy of Agricultural Science. The powder was derivatized with 9-fluorenylmethyl chloroformate (FMOC-Cl), and the DNJ-FMOC content was measured by HPLC. We investigated the content of 1-DNJ in the silkworm powder and its glucose-lowering effect when it was treated at different temperatures. The content of 1-DNJ was the lowest at $150^{\circ}C$, while it was constant at other temperatures. The silkworm extract powder was orally administered to diabetic mice (20 mg/kg/d) for 4 wk. Water intake did not significantly change when compared with the control group (T0). The blood glucose levels significantly decreased when mice where administered silkworm powder treated at $60^{\circ}C$ (T60) compared to the control group, but no difference was observed between the groups T100 and T150. Moreover, the blood levels of TG significantly decreased compared with the control group. Based on these results, we surmise that the properties of the silkworm extract powder were stable upon heating at $100^{\circ}C$ but not at $150^{\circ}C$.

Evaluation of General Toxicity and Genotoxicity of the Silkworm Extract Powder

  • The silkworm extract powder contain 1-deoxynojirimycin (DNJ), a potent ${\alpha}$-glycosidase inhibitor, has therapeutic potency against diabetes mellitus. Therefore, natural products containing DNJ from mulberry leaves and silkworm are consumed as health functional food. The present study was performed to evaluate the safety of the silkworm extract powder, a health food which containing the DNJ. The repeated toxicity studies and gentic toxicity studies of the silkworm extract powder were performed to obtain the data for new functional food approval in MFDS. The safety was evaluated by a single-dose oral toxicity study and a 90 day repeated-dose oral toxicity study in Sprague-Dawley rats. The silkworm extract powder was also evaluated for its mutagenic potential in a battery of genetic toxicity test: in vitro bacterial reverse mutation assay, in vitro chromosomal aberration test, and in vivo mouse bone marrow micronucleus assay. The results of the genetic toxicology assays were negative in all of the assays. The approximate lethal dose in single oral dose toxicity study was considered to be higher than 5000 mg/kg in rats. In the 90 day study, the dose levels were wet at 0, 500, 1000, 2000 mg/kg/day, and 10 animals/sex/dose were treated with oral gavage. The parameters that were monitored were clinical signs, body weights, food and water consumptions, ophthalmic examination, urinalysis, hematology, serum biochemistry, necropsy findings, organ weights, and histopathological examination. No adverse effects were observed after the 90 day administration of the silkworm extract powder. The No-Observed-Adverse-Effect-Level (NOAEL) of silkworm extract powder in the 90 day study was 2000 mg/kg/day in both sexes, and no target organ was identified.

1-Deoxynojirimycin Content and Blood Glucose-Lowering Effect of Silkworm (Bombyx mori) Extract Powder

  • We investigated the 1-deoxynojirimycin (1-DNJ) content of extracts from silkworm larvae at each developmental stage within three silkworm varieties. We also compared the content of the following polyhydroxylated alkaloids in the silkworm extracts: 1-DNJ, fagomine, and 1,4-dideoxy-1,4-imino-d-arabinitol (DAB). In addition, we evaluated the glucose-lowering effects of silkworm extract powder in db/db mice. The 1-DNJ content was the highest in Yeonnokjam $5^{th}$ instar $3^{rd}$ d larvae and Hansaengjam $5^{th}$ instar $3^{rd}$ d larvae, which contained 18.4 mg/100 g dry weight and 18.3 mg/100 g dry weight, respectively. The $5^{th}$ instar $3^{rd}$ d larvae exhibited a higher content of 1-DNJ than that of $5^{th}$ instar $5^{th}$ d larvae among all varieties. The glucose-lowering effects of silkworm extracts and Yeonnokjam powder were tested on db/db mice, and the blood glucose levels were found to decrease significantly in the YR70 group. Silkworm extracts (180 mg/kg, 90 mg/kg, 45 mg/kg, and 22.5 mg/kg) and acarbose (50 mg/kg) were administered orally for 4 wk. Changes in water intake were not statistically significant between control and silkworm extract-treated groups. Compared to the control group, blood glucose levels in the silkworm extract powder-treated group decreased in the 22.5 mg/kg/d group after being administered for 4 wk. This decrease was statistically significant. Furthermore, biochemical changes in the AST(Aspartate aminotransferase), ALT(Alanine aminotransferase), TCHO(Total Cholesterol), TG(Triglyceride), LDL(Low density lipoprotein), and HDL(High density lipoprotein) levels in blood were not observed. However, statistically significant decreases in blood GLU in the 22.5 mg/kg/d group compared to that of the control group occurred. In addition, the epididymal fat weight of the silkworm extract powder-treated group decreased significantly in both the 22.5 mg/kg/d group and 180 mg/kg/d group compared to that of the control group, but there were no statistically significant changes in perirenal fat weight. These results demonstrate that silkworm extracts inhibit changes in blood glucose levels in model diabetic mice.

누에분말 추출물의 이당류 경구투여에 대한 동력학적 연구 (Pharmacodynamic Study of Silkworm Powder in Mice Administered to Maltose, Sucrose and Lactose)

  • 누에분말 투여에 따른 시간별 당의 흡수패턴 및 당분해 억제로 당의 손실여부를 알아보기 위하여 12시간동안 절식된 마우스에 이당류(Maltose, Sucrose, Lactose)와 누에분말 메탄올 추출물을 동시에 투여하여 240분동안에 혈당 변화를 측정하였다. 1. Maltose와 누에분말을 투여한 군에서는 투여 30분 후에 69.0%의 혈당상승억제효과를 보였으며, 60분이후에도 당이 서서히 흡수되고 있어 지연흡수가 잘 유도되고 있었다. Maltose 군의 투여 240분까지의 당흡수는 560.7 mg/dl이고, 누에분말군은 534.7 mg/dl로써 Maltose 투여대비 95.4%로 누에분말이 당의 손실없이 당의 흡수를 지연시켰다. 2. Sucrose와 누에분말 투여군은 투여 30분 후 혈당상승억제효과가 59.9%로 maltose과 거의 같은 수준이며, 60분 이후의 당흡수 패턴 또한 maltose과 유사하였다. Sucrose 군의 투여 240분까지의 당흡수는 508.9 mg/dl이고, 누에분말군은 468.8 mg/dl로 Sucrose 투여대비 92.1%로서 누에분말 투여가 당을 지연 흡수시켰다. 3. Lactose와 누에분말 투여군은 투여 30분 후 약간의 혈당상승을 억제하였으나, lactose 투여군과 비슷한 당흡수 패턴을 유지하여 누에분말은 lactose의 흡수를 억제하지 못하였다. 이상의 결과로 누에분말은 ${\alpha}$-glucose를 억제하여 투여 30분 후의 일시적인 혈당상승을 유발하지 않고 또한 당의 손실이 거의 없이 당의 지연흡수를 유도하고 있음이 입증되었다.

키위, 파파야, 파인애플 및 배 과즙 처리에 의한 누에분말의 이화학적 특성과 생리활성 (Bioactive and Chemical Properties by Silkworm (Bombyx mori L.) Powder Degradation with Kiwifruit, Papaya, Pineapple and Pear Juice)

  • 누에분말을 과일(키위, 파파야, 파인애플 및 배) 단백질 분해 효소로 반응시켜 생리활성작용 및 이화학적 특성을 조사하기 위하여 pH, 산도, 단백질 함량, 미네랄 함량, 지방산 조성, 단백질 패턴, 항산화 및 혈전용해 활성을 측정하였다. 누에분말은 각 과일 단백질 분해 효소로 $60^{\circ}C$에서 24시간 반응시켰다. 단백질 농도는 과즙액 처리 누에분말에서 약간 높은 것으로 나타났다. 누에분말의 주요 미네랄은 칼륨, 마그네슘, 칼슘 및 아연이였으며, 주요 지방산 조성은 linolenic acid, oleic acid 및 palmitic acid 였다. SDS-PAGE상의 단백질 패턴 분석에서 66-97 kDa 정도 크기의 누에 단백질 밴드가 파인애플, 파파야 및 배 반응에 의해서는 대부분 분해가 일어났지만, 키위 반응에 의해서는 거의 분해가 일어나지 않은 것으로 관찰되었다. 혈전용해 활성은 파파야 및 배 반응에 의한 누에분말에서만 나타났다. 항산화 활성은 0.1% 처리 농도에서 과일 단백질 분해 효소 반응에 의한 누에분말에서 미반응 누에분말 보다 증가하였으나, 시판 항산화제 BHT 처리보다는 활성이 많이 낮았다. 따라서 과즙액 처리 누에분말의 경우 반응 전 누에분말 보다 생리활성작용 및 이화학적 특성이 강화됨으로써 건강기능식품 소재로서의 가치가 증가된 것으로 사료되어 진다.

Effects and Mechanisms of Silkworm Powder as a Blood Glucose-Lowerinly Agent

  • Cocoon production, which is a representative of traditional sericulture shifted into silkworm powder production in the spring of 1995. This, infect, signifies the change from the dress-centered textile business to the bio-industry and the functional resource industry. One of the most outstanding shifting is utilization of silkworm larvae for anti-diabetic agent. In Asian countries including Korea, silkworm powder derived from the domestic silkworm (Bombyx mori L.) has long been favored for anti-diabetic agent, but its efficacy was not tested until last decade by modern scientific methods. In this article, we reviewed the major researches on the silkworm powder as a blood glucose-lowering substance. After the beginning test of the efficacy of silkworm powder by a cooperative research between Department of Sericulture and Entomlogy, NIAST, RDA and Kyung Hee University, substantial data have been accumulated so far, In a serial experiment to select best condition, the fifth instar larvae prepared by freeze dry method turned out to have the best blood glucose-lowering effect. In the pharmacological experiment to understand the mechanism of silkworm powder in small intestine, the silkworm powder turned out to inhibit the activity of ${\alpha}$-glucosidase, by competitively binding to $\alpha$-type disaccharides. The animal experiment showed that the extract of silkworm powder prevents a rapid increase of blood glucose level after meal and prevents hunger and law blood glucose level during empty stomach. In the experiment to isolate the major component of silkworm powder, which exerts blood glucose-lowering effect, 1-deoxynojirimy-cin (DNJ) was eventually mass-purified, and it turned out that DNJ isolated from silkworm powder was excellent in its blood glucose-lowering effect. In the experiment to understand the personal difference of the efficacy of the silkworm powder, clinical candidates were divided on the basis of the criterion of traditional Chinese medicine: Tae-Yang, Tae-Um, So-yang, and So-Um. The result showed that silkworm powder has a tendency to reduce blood glucose level at fasting and at 2 hours after meal, and this trend was somewhat obvious in the Tae-Um body type. In summary, we reviewed scientific papers on the efficacy of silkworm powder and its purified DNJ as a blood glucose-lowering agent. These suggest that silkworm powder truly possesses blood glucose-lowering effect as documented in the traditional Chinese medicine, although further researches will be required to develop them as "medical" resource instead of functional food.

A comparative study on the phytochemical and anti-oxidant activity differences in HongJam prepared with various silkworm varieties

  • The aim of this study was to elucidate the difference in the amount of phytochemicals and anti-oxidant activities of the freeze-dried 5th instar 3rd day silkworm powder (FDSP) and 7 HongJams (HJs). Three primary colors of light of 8 silkworm powders revealed why each sample had unique color. The visual and absorption spectrum analyses of 80% methanol (MeOH) extracts of 8 silkworm powders revealed that white jade (WJ)-FDSP extracts showing strong forest-green colors had different absorption spectrum from 7 HJs. Among 7 HJs, the absorption spectrum of light green silkworm (LGS)-HJ extract was different from those of other HJ extracts. The amounts of total flavonoids and phenolic compounds of WJ-FDSP were 423.0±9.21% and 252.3±12.86%, respectively, more than those of WJ-HJ. Among 7 HJs, the amounts of total flavonoids and phenolic compounds in LGS-and red silkorm (RS)-HJ extracts were significantly more than those of WJ-HJ extract. Consistent with the amounts of phytochemicals, radical scavenging activity and reducing ability of WJ-FDSP extract were significantly higher that other HJ extracts. Among 7 silkworm powder extracts, LGS-HJ extract showed significantly higher radical scavenging activity and reducing ability than those of WJ-HJ extract. But, RS-HJ extract only showed significantly higher radical scavenging activity than that of WJ-HJ extract. Taken together, the amounts of phytochemicals in 8 silkworm powders contributed on their colors and anti-oxidant activities.

누에 추출 분말이 흰쥐의 혈장지질 및 혈당농도에 미치는 영향 (Effects of Silkworm Extract Powder on Plasma Lipids and Glucose in Rats)

  • 누에 추출 분말 투여에 의한 혈중 중성 지질, 총 콜레스테롤 및 인지질 함량의 변화 양상은 투여량 $3\~5\%$에 따른 각 성분의 감소율이 각각 중성 지질은 $5.16\~9.15\%$, 총 콜레스테롤은 $1.63\~2.76\%$였으며, 인지질은 $1.37\~2.00\%$였다. 이러한 결과로 볼 때, 중성 지질에서 가장 뛰어난 효과를 보인 반면 총 콜레스테롤 및 인지질 함량 감소에는 상대적으로 그 효과가 낮은 것으로 판명되었다. 혈장 포도당 농도의 감소는 $3\%$ 첨가군의 경우 대조군에 비하여 약$4.62\%$의 감소율을 나타내었으나 $5\%$첨가군의 경우는 대조군에 비하여 약 $6.46\%$를 나타내었다. 한편, 혈장, 췌장, 근육조직의 인슐린 농도의 변화는 혈장 포도당 농도 변화와 거의 유사한 경향을 보였는데, 결과적으로 누에 추출 분말의 첨가량과 인슐린 분비량 사이에 어떠한 정의 상관관계는 도출되지 않았다. 이와 같은 결과들로 볼 때, $3\%$내외의 누에 추출 분말처리가 당뇨병 환자에게 있어 예방 또는 치료 효과를 기대할 수 있을 것으로 판단된다.

천연추출물이 구강미생물에 미치는 항균 효과 (Antibacterial effect of natural extracts on oral microorganisms)

  • 본 연구에서는 누에 추출물과 여주 추출물이 S. mutans와 F. nucleatum에 미치는 항균효과를 알아보고자 하였다. BHI 액체배지에 S. mutans 또는 F. nucleatum과 0%, 2%, 4%, 8% 농도의 누에가루 또는 여주가루 추출물을 첨가하여 배양한 뒤 광학밀도 600nm에서 시료를 측정하였다, 그 결과 S. mutans의 흡광도는 누에 추출물에서는 8%에서만 유의적으로 흡광도의 수치가 감소하였고 여주 추출물에서는 모든 농도에서 유의적으로 흡광도의 수치가 감소하였다. F. nucleatum의 흡광도는 누에 추출물과 여주 추출물에 모두 농도 의존적으로 유의적으로 감소하였다. 이는 누에 추출물과 여주 추출물이 구강질병의 예방 및 치료들 위한 소재로 사용 할 수 있는 가능성을 시사한다.

오디와 누에 섭취가 rats의 저항성 운동에 따른 근육 증가에 미치는 영향 (The effects of the mulberry and silkworm intake on muscle increase of rats for resistance exercise)

  • The purpose of this study is to investigate the effects of supplementation of mulberry powder, mulberry extract and silkworm powder during the 8 weeks of resistance exercise on muscle increase of rats. Fifty males, Sprague-Dawley rat, were randomly divided into 5 groups: CON(control group, n = 10), REG(resistance exercise group, n = 10), MP REG(mulberry powder intake and resistance exercise group, n = 10), ME REG(mulberry extract intake and resistance exercise group, n = 10) and SP REG(silkworm powder intake and resistance exercise group, n = 10). After climbing the ladder without weights during the 1 week of adaptation period, the rats in the resistance exercise group were trained to climb a 0.98-m vertical(80 degree incline) ladder with weights in their tail during 7 weeks(10 times each day, 2 days per week). After exercise, the skeletal muscle was extracted from the flexor hallucis longus. The absolute weight of skeletal muscle was measured by the electronic balance and the relative weight of skeletal muscle about the weight was calculated. The strength and density of legs muscle were analyzed by the computed tomography scan. After 8 weeks of resistance exercise, the absolute weight of skeletal muscle wasn't significant, but it increased in MP REG 8%, ME REG 8% and SP REG 10% compared to REG. The relative weight of skeletal muscle increased significantly in SP REG compared to REG (p < 0.05). Additionally, there was a significant increase of density and strength of the forelegs in SP REG compared to CON (p < 0.05). However, there was no significant increase of density and strength of the hind legs. In conclusion, regular resistance exercise made rats muscle increase and there was more muscle increase effected by providing silkworm.
