Effect of topical application with 5, 10 and 15${\mu}$g/ml thyroxine on the pre-cocoon, cocoon, postcocoon and silk improvement were studied in bivoltine silkworm NB18 breed of B. mori L. Thyroxine was applied independently on alternate days to silkworm of IV and V instar larvae. The treatment of all different concentrations resulted in a significant increase in commercial characters like larval weight, silkgland weight, cocooning percentage, female cocoon weight and its shell weight and male cocoon shell weight and its ratio, cocoon dimension, length of the filament and its weight and oviposition. However, there was no significant change in larvel weight in 5${\mu}$g/ml treated group and larval duration was significantly decrease. But moth emergence percentage and hatching percentage did not show any significant change when compared with that of carrier control.
The purpose of this treatise is to prove the presence of cystine in silk fiber through wide sampling throughout all the sericultural processes of Bombyx mori.; also to show that disulfide cross linkages exist in the silk fiber. The conclusions reached were as follows: 1. Crystalline cystine was obtained from silk fibroin using Folin's Method. 2. Analytical data showing the cystine content of silk fiber and its related materials were obtained using Sullvan's Method as follows: Material Percent Cystine A. Mulberry leaf protein 0.175 B. Silkworm egg 0.33 C. Silkworm Body, matured, fat extracted, without silk gland 0.41 D. Silk gland, matured 1.23 E. Silkworm feces none F. Silkworm pupa, fat extracted 0.30 G. Silkworm moth, fat extracted 0.60 H. Raw Silk 0.22 I. Fibroin 0.175 J. Sericin 0.30 3. The presence of cystine in the silkworm was substantiated the existence of 0.175 % methionine in mulberry leaves and 0.12% methionine in the silk gland. 4. Part of the sulfhydryl compounds in the silk gland is believed to transfer to serine and methionine, with the former being secreted into the liquid silk finally as silk fiber and the latter used for nutritive purposes in the growing of silk gland tissue. 5. The cystine content is variable by mulberry species, silkworm species, sex, breeding process, and other culturing environments. 6. Hybrid silkworms require more nutritive amino acids for effective growth than the original parents, and secrete less of them as silk fiber. 7. From such an observation, the amino acid composition of silk fiber is believed to be fairly flexible. Cystine if included in the amorphous part of the fiber, especially in sericin. 8. The result from enriching the silkworm diet with pure cystine or wool cystine did not result in any advantage, therefore it is believed that the natural cystine and methionine contents in the mulberry leafaregoodenoughforsilkwormnutrition. 9. The disulfide cross linkage in silk fiber was verified by using the Harris Method. Contraction took place following the treatment of the fiber with various salts and acids. Comparisons were made with wool fiber. 10. During these experiments, the fibrious structure of silk fiber and the net-globular liquid form were photographed microscopically. It is believed that the globules of liquid silk are net-formed by the inter attraction of the OH ion of the globular peptide and the H ion of water as shown by the hair cracking behavior of the film. The net-globular protein precipitation from the mulberry protein solution showed that mulberry is a proper diet for the formation of fibrous protein in the silk fiber. 11. The significance of the presence of cystine in silk fiber as emphasized in this paper should result in modification of the general conception that cystine is absent from this fiber. NOTICE: A part of this treatise was presented at the annual Korea Sericultural Society meeting held in 1961.
The hat excavated from the tomb of Hojeo-myon Wonsung-gun in Kangwon province was dry-cleaned and restored. The hat was made of wool felt and decorated with several strands of silk cord. It was very dirty and moth-eaten. After getting rid of the particle soils on a surface with painting brush, the hat was dry-cleaned using perchloroethylene applying the minimum force, and blotting paper was used for drying. During cleaning, it was supported by the stainless steel net. The weight loss was 7.94% after cleaning. The soils sticked on the hat was removed clearly. The texture of the hat became softer after cleaning. Color difference(∆E) was 2.57, which is a noticeable change. The color became clear and changed to the red and yellow range. After dry-cleaning, the hat was repaired. Dislocated silk cords were fixed to the right place by stitching with silk yarn. To keep the shape of the hat, it should be supported by a hat-shaped stiff net during exibition and storing.

Oral supplementation of zinc chloride to silkworm larvae in different concentrations (30, 60 and 90 ${\mu}g$/ml) to the fourth and fifth instar resulted in a significant increase in commercial traits, like larval weight, silk gland weight, cocooning percentage in lower concentrartion (30 ${\mu}g$) treated group. The female cocoon weight, shell weight and its ratio were significantly increased in 30 and 60 ${\mu}g$ treated groups, male cocoon weight, and shell weight in 30 ${\mu}g$ treated group and its ratio in 60 and 90 ${\mu}g$ treated groups, filament length, weight and moth emergence percentage in all the treated groups and egg productivity in 90 ${\mu}g$ treated group were increased significantly. The remaining groups did not show any significant changes compared with that of carrier control.

The effect of supplementation with potassium nitrate on economic parameters was analysed following treatment of last larval stadium. The treated larvae showed a significant increase in larval weight at 500$\mu\textrm{g}$/ml and silk gland weight in all the treated groups along with other enhanced larval, cocoon and adult parameters. The larval period was significantly decreased in all the treated groups with increased female and male cocoon weights its shell weights filament lengths at 500$\mu\textrm{g}$/ml. The denier was significantly decreased in all the groups. The filament weight moth emergence percentaged length of the ovariole were unaffected. However, eggs per ovariole, fecundity and hatching percentage significantly decreased compared with the corresponding parameters of the carrier control.

Filament length, weight and denier were increased significantly in all the treated groups. Moth emergence percentage, length of the ovariole, eggs per ovariole, fecundity and rind hatching percentage were unaffected when compared with that of the carrier control group. This suggests that hydrocortisone in addition to affecting larval growths also affect silk crop.

The present study aimed to determine whether a hemolymph prepared from Antheraea yamamai larvae had the same biological activities using a Bombyx mori hemolymph prior to exposure to lipopolysaccharide (LPS) in order to induce an inflammatory response. The effects of the hemolymph were determined using a reverse transcription-quantitative polymerase chain reaction to assess the expression of pro-inflammatory molecules. The A. yamamai hemolymph exerted anti-inflammatory effects on LPS-activated human monocytic leukemia cells via Toll-like receptor (TLR) 4-mediated suppression, similar to the B. mori hemocyte extract. Treatment with the A. yamamai hemolymph significantly suppressed LPS-induced upregulated inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) mRNA expression at all tested concentrations compared with the control, similar to the B. mori immune-challenged hemolymph. Finally, the A. yamamai hemolymph, like the B. mori immune-challenged hemolymph, suppressed all of these concentrations in a dose-independent manner. These results demonstrate that the hemolymph of A. yamamai exhibited important biologically active substances. Further in-depth functional studies are required to fully understand the mechanisms underlying the biological activities of wild-type silkworm hemolymphs.
The work on the 'Studies of radiosensitivity' in Korea has been initiated since 1959 by the author at various institutions in home and foreign countries, and more than 20000 of plant materials in 73 varieties and 41 kinds are treated under varied conditions for the studies of radiosensitivity in plants. However, a study of the susceptibility to radiation in insect pests as well as their biological effects has not been previously reported in this country. During the year of 1964 and 1965, near 50000 of insects at the stages of adult, egg, larva and pupa in 14 different kinds are irradiated under acute X and gamma rays to investigate the behavior of growth and lethality after irradiation, and thrir general tolerances to radiations are compared by the kind of insects, stage of growth, radiation sources and exposured dosages. Through the experiments, it is shown that the lethality in most of insects are high as several kilo roentgens above and no significant effects on pupation. emergence, and oviposition in the treated generation at the dose of 3 Kr. below, but in the following generation , the rice stem borer for instance, no oviposition or decreased laying is observed from the dosages at 3 Kr. to 1 Kr., and the hatching was possible from 1.5 Kr. below of irradiated larvae although laying and hatching are seemed in normal. In general, the lethal dose $50\%$ of tested infect materials at the larval stage can he classified some what 60-80 Kr. for Pine caterpillar, Fall webworm, Camphor silk moth and their relatives, at 40-60 Kr. for Gypsy moth, Monley prominent and some relatives and at 20-30 Kr. for House fly, Rice weevil, Pine sawfly etc. Plant parasitic nematodes such as Hirschmannia and Paratylenchus shown very high resistance to radiation and the LD-50 is observed from 100 Kr. or above. A concept to evaluate the radiosensitivity in insects by taxological family line as a practical measure is not answerable yet, but it ran he solved within the near future through the continuing experiments.
In this study we tried to transform an antimicrobial peptide gene (Shiva) under the promoter of tomato phenylalanine ammonia-lyase (tPAL5) into tobacco and potato plants. Antimicrobial peptide gene was isolated originally from giant silk moth (Hyalophora cecropia) and modified ie nucleotide sequence to increase antimicrobial activity. Transgenic tobacco plants were regenerated and their seeds were tested on the media containing kanamycin (500 mg/L). The results of PCR amplification and genomic Southern blot hybridization confirmed the integration of construct (tPAL5 promoter-Shiva-NOS-GUS-NOS) into chromosome. We observed that one of the transgenic tobacco plants showed chromosome rearrangement when integrated. In case of potato transformation, the efficiency of regeneration was maximized at the medium containing Zeatin 2mg/L, NAA 0.01mg/L, GA$_3$ 0.1mg/L. We also observed the high expression of GUS (${\beta}$-glucuronidase) enzyme which was located next to the terminator sequence of nopaline synthase gene (NOS) in the vascular tissue of stem, leaves of transgenic potatoes. This result suggested that a short sequence of Shiva gene (120 bp) and NOS terminator sequence might be served as a leader sequence of transcript when translated.

The present investigation was carried out to study the possible cause for reciprocal difference in silkworm hybrids. By utilising the polyvoltine race Pure Mysore (PM) and newly evolved breeds (CSR2, CSR5, CSR16 and CSR17), the direct and reciprocal crosses of polyvoltine ${\times}$ bivoltine and also bivoltine hybrids were studied. The hybrids of polyvoltine ${\times}$ bivoltine (direct) are superior to their reciprocal crosses in respect of cocoon yield, cocoon weight and filament length. The reciprocal crosses of polyvoltine ${\times}$ bivoltine are superior to their direct crosses in respect of fecundity and short larval duration. No significant differences were observed in the characters like cocoon shell ratio, raw silk percentage, denier, reelability and neatness in both polyvoltine ${\times}$ bivoltine direct crosses and their reciprocals. The expression of cocoon characters as a function of sex revealed that direct crosses (polyvoltine ${\times}$ bivoltine) showed higher cocoon weight, pupal weight, shell weight and longer filament length in females than the reciprocal crosses (bivoltine ${\times}$ polyvoltine), where as these characters in males were almost the same in both direct and reciprocal crosses, indicating that the sex-linked genetic factor played a more important role. it was clear that difference in cocoon yield observed in reciprocal crosses of polyvoltine ${\times}$ bivoltine was due to the low cocoon and shell weight in females which was turn due to presence of early maturity genes (Lme) linked with sex-chromosome (X) which effect on larvae period of the silkworm. In bivoltine hybrids, i.e., both direct and their reciprocals crosses, all the characters viz., hatching percentage, larval duration, survival, cocoon weight, cocoon shell weight, cocoon shell ratio, raw silk percentage, filament length, denier, reelability and neatness did not show any significant difference (except number of eggs laid by moth) which could account for presence of same maturity genes (Lm) in both direct and reciprocal crosses. it was clear that reciprocal differences occur when the hybrids are prepared from the parental strains with different voltinism.
