Extractives from the Allium victorials (산마늘(Allium victorialis)의 추출성분)

  • Allium victorials var. platyphyllum were ground, extracted with 95% EtOH, concentrated, and fractionated with a series of n-hexane, methylene chloride, ethyl acetate, butanol and water on a separatory funnel. A portion of ethyl acetate soluble fraction was chromatographed on a Sephadex LH-20 and silica gel column using various solvent system as eluents. The isolated compounds were identified by TLC, $^1H-$, $^{13}C$-NMR, HMBC and EI-MS. Astragalin (compound 1), kaempferol (compound 2), quercetin (compound 3), ferulic acid (compound 4) were isolated from the ethyl acetate soluble fraction of A. victorials.

Isolation and Identification of Antifungal Compounds from Eugenia caryophyllata Extracts (정향 추출물로부터 항진균성 물질의 분리 및 동정)

  • Antifungal properties of clove(Eugenia caryophyllata) against food spoilage microorganism, Penicillium rugullosum IFO 4683 was investigated. Antifungal activity of the essential oil was as equivalent as potassium metabisulfite and myconazole. The clove extracts was fractionated to hexane, chloroform, ethyl acetate, butanol and water fraction. Hexane fraction showed the highest inhibitory effect on the Penicillium rugullosum IFO 4683. Hexane fraction was further fractionated by silica gel column chromatography and thin layer chromatography(TLC). The antifungal compound was isolated from their fractions and their chemical structures were identified as eugenol, eugenol acetate and chavicol by EI-MS, $^1H$-NMR and $^{13}C$-NMR.

Sterols Isolated from Nuruk (Rhizopus oryzae KSD-815) Inhibit the Migration of Cancer Cells

  • An activity-guided fractionation method was used to isolate anticancer components from Nuruk (Rhizopus oryzae KSD-815:KSD-815). Dried powder of KSD-815 was extracted with 80% methanol and partitioned successively using n-hexane, ethyl acetate, n-butanol, and water. The n-hexane and n-butanol fractions showed a strong antimigratory effect on human cancer cells. Both of these fractions were subjected to separation and purification procedures using silica gel, octadecyl silica gel, and Sephadex LH-20 column chromatographies to afford four purified compounds. These were identified as ergosterol peroxide (1), stigmast-5-en-$3\beta$,$7\beta$-diol (2), ergosta-7,22-dien-$3\beta$,$5\alpha$,$6\beta$,$9\alpha$-tetraol (3), and daucosterol (4), respectively, by spectroscopic methods such as nuclear magnetic resonance spectrometry, mass spectrometry, and infrared spectroscopy, and comparison with those in the literature. Compounds 1-4 were isolated from KSD-815 for the first time. Compounds 1 and 4 inhibited the migration of MDA-MB-231 cells at concentrations lower than $20\;{\mu}M$.

  • Sulfonated poly(arylene ether sulfone) (SPAES)-3-mercaptopropyl silica gel (3MPTSG) composite membranes with improved oxidative stability were prepared for polymer electrolyte fuel cell application. It has been reported that ether part of main chain of aromatic hydrocarbon based membranes were weak to radical attack to decrease membrane durability. In this study, the hydrophilic inorganic particles were introduced by minimizing a decrease in ion conductivity and increasing an oxidative stability. The composite membranes were investigated in terms of ionic conductivity, ion exchange capacity (IEC), FT-IR, TGA and contact angle, etc. As a result, increasing amount of the 3MPTSG resulted in decrease in proton conductivities and water uptakes at 100% R.H. but enhanced thermal and oxidative stabilities.

  • A new gas-chromatographic method for determining carbon and hydrogen in organic compounds has been developed. After sample combustion was performed in a regular analytical combustion tube with an internal oxidant (a mixture of silver oxide and manganese dioxide) under a helium flow, the water produced was converted to acetylene by passing through a calcium carbide tube. The carbon dioxide and acetylene were trapped by a molecular sieve 5A column at room temperature. The trapped gases were released under programmed temperature raise up to $340^{\circ}C$ and the released gases were passed through a silica gel column. The adsorption of $CO_2$ and $C_2H_2$ in the molecular sieve 5A trapping column were found to be quantitative and the silica gel column showed an excellent resolution of $CO_2$ and $C_2H_2$ for analytical purpose. The analytical results for various known compounds based on the out-put of the thermal conductivity cell calibrated for the amounts of carbon and hydrogen contents in benzoic acid, showed average errors ${\pm}0.5%$ and ${\pm}0.33%$ for carbon and hydrogen, respectively.

  • From water extract of red ginseng, two new-type glucosides, $A:\;C_{12}H_{16}O_8\;and\;B:\;C_9H_{16}\;O_7,$ were isolated by fractionation with highly porous polymer ollowed by chromatography on silica gel in yields of 0.04 and $0.16\%,$ respectivley. The structures of A and B were elucidated by means of NMR and MS. Neither glucosides were detected in the extract of white ginseng and the mechanism of formation of A and B during the process of steaming will be discussed. The isolation and identification of saponins of corms (head of the root) of red ginseng are also reported.

Diels-Alder type adducts from the fruits of Morus alba L. (오디(Morus alba fruit)로 부터 Diels-Alder형 부가체 화합물의 분리 및 동정)

  • The fruits of Morus alba L. were extracted with 80 % aqueous MeOH, and the concentrated extract was partitioned into EtOAc, n-butyl alcohol, and water fractions. The repeated silica gel ($SiO_2$) and octadecyl silica gel column chromatographies for the EtOAc and n-butyl alcohol fractions led to isolation of two phenolic compounds. The chemical structures of the compounds were determined as Diels-Alder type adducts, mulberrofuran E (1) and chalcomoracin (2) based on spectroscopic data analyses including nuclear magnetic resonance, mass spectrometry, and infrared spectrometry. Compounds 1 and 2 were isolated for the first time from the fruits of M. alba L. in this study.

Phenolic compounds from the leaves of eggplant (Solanum melongena L.) (가지(Solanum melongena L.) 잎으로부터 페놀 화합물의 분리 및 동정)

  • The leaves of eggplant (Solanum melongena L.) were extracted with 80 % aqueous MeOH, and the concentrated extract was partitioned with n-hexane, EtOAc, n-BuOH, and water fractions. From the n-BuOH fraction, five compounds were isolated through the repeated silica gel, octadecyl silica gel, and Sephadex LH-20 column chromatographies. On the basis of physic-chemical and spectroscopic data including mass spectrometry, infrared spectroscopy, and nuclear magnetic resonance, they were identified to be caffeic acid (1), chlorogenic acid (2), cryptochlorogenic acid (3), panasenoside (4), and (6R,7E,9R)-4,7-megastigmadien-3-one-9-${\beta}$-${\small{D}}$-glucopyranoside (5). Compounds 3 and 4 were isolated for the first time from the leaves of S. melongena L. in this study.

Isolation and Identification of Active Antimicrobial Substance against Listeria monocytogenes from Ruta graveolens Linne (운향으로부터 Listeria monocytogenes에 대한 항균 활성 물질의 분리 및 구조동정)

  • Ethanol extracts from Ruta graveolens Linne exhibited strong antimicrobial activities by disc diffusion method against 5 strains of Listeria monocytogenes (ATCC 19111, ATCC 19112, ATCC 19113, ATCC 19114 and ATCC 15313). Ethanol extract from Ruta graveolens Linne was subsequently fractionated by n-hexane, chloroform, ethyl acetate and water. Chloroform fraction of Ruta graveolens Linne showed strong growth inhibition at concentrations as low as 40 ppm level in broth culture medium against 5 strains of L. monocytogenes for 72 hr at $30^{\circ}C$. Single substance(RTG1-1) was isolated by silica gel column chromatography from chloroform fraction of Ruta graveolens Linne. RTG1-1 showed a strong bactericidal activity against L. monocytogenes at a concentration of 20 ppm level. Purified RTG1-1 was identified as gravacridonechlorine by analyses of EI-Mass, $^1H-NMR$ and $^{13}C-NMR$.

The Antimutagenic Activity and the Growth Inhibition Effect of Cancer Cells on Methanol Extracts from Small Water Dropwort (돌미나리 메탄올 추출물의 항돌연변이 작용과 암세포증식 억제효과)

  • The study was carried out to evaluate the antimutagenic and anticancer effects of small water dropwort. The methanol extracts from small water dropwort significantly reduced the mutagenicity induced by aflatoxin $B_1\;(AFB_1)$ and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Salmonella typhimutium TA 100. Also, the methanol extracts inhibited the growth of AZ-521 human gastric cancer cells and HT-29 colon cancer cells. The chloroform fraction from methanol extracts of small water dropwort inhibited $40\;to\;80\%$ of the mutagenicity by $AFB_1$ in Sal. typhimurium TA 100 by the addition of 2.5 to $10\%$. To separate active compounds, the chloroform fraction was subjected to column chromatography on a silica gel and separated into five fractions. Among the five fractions, fraction 4 showed the highest antimutagenic effect against $AFB_1$ and an anticancer effect in the HT-29 colon cancer cell. As the result of the analysis in GC-MS, 1-napthalene carbonitrile, 5,6,7,8-tetrahydrol and benzene, 1,1'-(1,4-pentadiene-1,5-diyl) bis-,(E,E) were identified potentially from fraction 4.
