Biochemical Changes in Muscle Protein of Squid Sikhae during Fermentation -Effects of Temperature and Moisture Content- (오징어 식해 숙성중 단백질 화학적 변화 -온도 및 수분함량의 영향-)

  • In this study, biochemical changes of muscle protein in squid Sikhae prepared at various temperatures and moisture content were investigated. The pH values in squid Sikhae decreased rapidly with increase in titratable acidity during fermentation at high temperature and moisture content. Growth rate of lactic acid bacteria increased rapidly during fermentation at high temperature and moisture content. Changes in muscle protein subunit were analyzed by SDS-PAGE; myosin heavy chain component disappeared completely during preparation of squid Sikhae and actin component decreased slowly during fermentation. Therefore, it was suggested that myosin heavy chain component in squid muscle protein was autolyzed at the incipient for-mentation time and actin component was degraded by acidic protease in muscle and protease produced from microorganisms during fermentation.

  • Northern Kyungbuk around Andong is a mountainous and more or less dry region geomorphogically. Since this area located in the valley was less invaded by foreign countries, the folk traditions have been maintained for a long time. Also as the center of Confucian culture, this region respects ceremonies and so, the simple and noble ceremonial-foods such as Ddok, Hankwas, and Buchiki have been firmly preserved in many distinguished families. And besides Andong-Soju, many traditional liquors produced in different methods can be found in this region. As Gogi-Sikhae(a fermented beverage utilizing rice and fish) is still preserved in eastern coastal area, Sosikhae(Sikhae without fish) is maintained in this region and furthermore the unique Andong Sikhae developed by modifying Sosikhae is being consumed currently. Since it is difficult to get fish here geographically, the soybean foods were developed for protein intake, for example, various fermented soybean pastes, Andong-Songuksu(a hand-made noodle using raw soybean flour), and Siragiguk(a vegetable soup).

Anti-Helicobacter pylori Activity of Lactobacillus spp. Isolated from Gajami Sikhae (가자미식해에서 분리한 Lactobacillus spp.의 항헬리코박터 활성 평가)

  • Helicobacter pylori infects the mucosa, induces chronic inflammation and ulcers, and is known as a biological carcinogen. Antibiotics are used as therapeutic agents for H. pylori, but there are problems such as resistance. Thus, research is being conducted on the use of lactic acid bacteria (LAB) as an alternative therapeutic agent. There have been many studies on LAB related to kimchi. However, studies related to Gajami Sikhae, a traditional fermented seafood in Korea, are insufficient. In this study, we investigated the inhibitory effect of LAB isolated from Gajami Sikhae on H. pylori and its use as a probiotic. Forty species of LAB isolated from Gajami Sikhae were identified as Lactobacillus plantarum, Lactobacillus brevis, Leuconostoc mesenteroides, and Weisella paramesenteroides, and 10 strains of 40 species were selected through liquid inhibition assay of H. pylori. The selected LAB supernatant at 1%, 5%, and 10% had a growth inhibitory effect on H. pylori 52, 51, e-53, and 309. The adjusted pH of 7.0 was used for the LAB culture supernatant, in reference to a study that the growth of H. pylori is affected by acid. All 10 strains of LAB at 5% and 10% concentration suppressed the growth of H. pylori 52, and 7 strains of LAB at 10% concentration suppressed the growth of H. pylori e-53. LAB also had the effect of suppressing the activity of urease. Finally, LAB isolated from Gajami Sikhae is expected to be useful for eradicating and preventing H. pylori.

Studies on the Storage Stability of Traditional Andong Sickhae (전통 안동식혜의 저장 안정성에 관한 연구)

  • The effect of stabilizers on the sedimentation, sensory evaluation and viscosity were investigated during the fermentation of traditional Andong sikhae and storage of the product at $4^{\circ}C$ and morphological characteristics were reviewed by scanning electromicrograph. The number of acid producing bacteria was increased for the first 3 days and then slightly decreased. Among the commercial stabilizers 0.3% of Carrageenan was the most effective for proventing sedimentation of sikhae. The interaction effects between the amount of food stabilizer added and treatment groups on taste turned out to be statistically significant.

Availability of Sikhae Factory Wastewater as a Submerged Culture Medium for Lentinula edodes

  • Sikhae is a Korean traditional beverage of saccharified rice. Its factory waste(SFW) is usually thrown away instead of being used. We developed a cheap substrate of SFW for use in liquid spawn that is known for its higher fruit body yields than grain spawn in sawdust cultivation. Mycelia of Lentinula edodes ASI 3046, which is regarded as the most suitable strain for sawdust cultivation, were cultured on six kinds of previous known media and SFW. As the seven kinds of media were applied, a Sikhae Factory Waste(SFW) was most excellent in growth. The dried mycelial weight in SFW was almost four times as much as that in the other media. In the flask culture, optimum culture conditions for the mycelial growth were obtained after 13 days of cultivation at media volume of 100 ml, 100 rpm, initial pH 4.5, and $25^{\circ}C$. The best mycelial growth was observed when $MgSO_4{\cdot}7H_2O$ and D-sucrose were added as a supplement in SFW. SWM must be a remarkable medium for L. edodes because of its simple preparation and low cost.

Microbiological and Enzymatic Characteristics in Alaska Pollack Sikhae during Fermentation (전통 명태식해 숙성중의 미생물 및 효소학적 특성)

  • The changes of microflora and enzyme activities in Alaska pollack sikhae were evaluated in 3 different temperature conditions, 5$^{\circ}C$, 2$0^{\circ}C$ and alternating temperature (stored at 5$^{\circ}C$ after 10 days of fermentation at 2$0^{\circ}C$), respectively. The number of proteolytic bacteria and 2 lactic acid bacteria including Lactobacillus sp. and Pediococcus sp. increased rapidly up to 10 days and composed major portion of total viable cell (TVC) in sikhae fermented at 2$0^{\circ}C$, whereas those of TVC were occupied by Lactobacillus sp., Pediococcus sp. and yeast after 10 days of fermentation. The major species of microflora in sikhae fermented at alternating temperature were, composed of Lactobacillus, Pediococcus and Streptococcus after 10 days of fermentation. Especially, Leuconostoc sp. was kept up to 27 days at 5$^{\circ}C$ than other temperature conditions (16 days). The activities of protease and lipase in acidic region (pH 3.0) were higher at 2$0^{\circ}C$ than at 5$^{\circ}C$ due to sensitivity of temperature, although those of protease and lipase in neutral region (pH 7.0) were not found any differences in both temperatures. Changing temperature condition from 2$0^{\circ}C$ to 5$^{\circ}C$ in alternating temperature inactivated protease activity, whereas lipase activity was still maintained during fermentation.

Functional and Volatile Flavor Components in Myungtae(Alaska pollack) sikhae (마른명태 식해의 향기성분과 기능성)

  • The volatile compounds of Myungtae (Alaska pollack) sikhae obtained by simultaneous steam distillation and extraction(SDE) apparatus were separated by gas chromatography(GC) and gas chromatography mass spectrometry(GC/MS). The totals of 155 volatile flavor components was identified in traditional Kyungsangdo Myungtae (Alaska pollack) sikhae, respectively. ${\alpha}$-Zingihirene(11.03%) (E)-di-2-propenyl disulfide(7.95%) ${\beta}$-cironellol(6.02%), methyl allyl disulfide(3.58%), cryptone(3.39%), camphene(3.23%), pentanol(3.21%), penadecanal(2.66%) and ${\beta}$-phellandrene(2.06%) were contained as the main compounds of Myungtae shikae. The fraction obtained from sikhae were tested for electron donating ability, angiotensin converting enzyme and xanthine oxidase inhibitory activity. There was no electron donating abilities$(SC_{50})$ of hexane and water fraction. On the other hand, the abilities of ethylacetate fraction and butanol fraction showed $310.64\;{\mu}g/mL,\;1096.49\;{\mu}g/mL$, respectively. Angiotensin converting enzyme inhibitory activities$(IC_{50})$ of ethylacetate fraction and butanol fraction were 1.623 mg/mL, 1.303 mg/mL, respectively. Xanthine oxidase inhibitory activities$(IC_{50})$ of ethylacetate fraction and butanol fraction were 3.591 mg/mL, 2.083 mg/mL, respectively.

Effect of Reaction Time on the Rheological Properties of Dextran Formed Solution Produced by Crude Dextransucrase from Leuconostoc rnesenteroides Sikhae (Leuconostoc mesenteroides Sikhae의 Crude Dextransucrase 반응시간에 따른 Dextran 생성액의 유변성)

  • Studies on the changes in rheological properties, molecular weight distribution and dextran yield after being reacted in lO%(w/w) sucrose concentration were performed with crude dextransucrase produced from Leuconostoc mesenteroides isolated from Sikhae. The reaction rate of dextran formation was monitored by sugar analysis with HPLC and by the changes in apparent viscosity. According to the periodate oxidation test, the dextran produced in this experiment was estimated to have 89% $\alpha$-(1->6) main linkages and 11% $\alpha$-(1->) side linkages. The rheological properties of the dextran solution formed changed with reaction time, and it was related to the changes in molecular weight distribution of dextran as determined by GPC analysis. As the reaction proceeded, the rheological behavior changed from Newtonian to non-Newtonian, showing Binghampseudoplastic and thixothropic flow behavior. The apparent viscosity of dextran formed solution increased with increasing reaction time, reached a maximum value of 2680 cP ($\gamma$=$33.75s^{-1}$, $25^{\circ}C$) by enzyme reaction for 8 hours, and then decreased. The temperature dependency of dextran formed solutions was well expressed by the Arrhenius equation and the activation energy reached a maximum value of 1.69 kcal/mole by enzyme reaction for 8 hours.

Probiotic Properties of Lactobacillus spp. Isolated from Gajami Sikhae (가자미식해에서 분리한 유산균의 프로바이오틱스 특성)

  • Forty species of lactic acid bacteria isolated from gajami sikhae were identified as Lactobacillus plantarum, Leuconostoc mesenteroides, Lactobacillus brevis, and Weissella paramesenteroides. 10 of the 40 strains were selected and used for the test. In this study, experiments such as those using acid and artificial gastric juice resistance, bile acid resistance, autoaggregation, coaggregation, and cell surface hydrophobicity were conducted to utilize lactic acid bacteria separated from gajami sikhae as probiotics. The separated lactic acid strains showed high survival rates through displaying resistance to acidic and artificial gastric juices; L. plantarum GS11 showed the best resistance. Also, as a result of a measurement of bile acid resistance, all lactic acid bacteria stocks showed survival of more than 100% with a probiotic number of 108 to 109 log CFU/ml. After evaluating cohesion to indirectly measure cell surface adhesion, autoaggregation ability was shown to be more than 46%. Measurement of xylene adhesion for cell surface hydrophobicity evaluation revealed better cell adhesion than B. subtilis, which has 32.2% hydrophobicity in isolated lactic acid strains. Antibacterial force measurement found antibacterial activity in lactic acid bacteria, excluding L. plantarum GS12 and L. plantarum GS13. Therefore, it was judged that lactic acid bacteria separated from gajami sikhae could be used as probiotics with various probiotic properties.

  • Fermented fish (jeotkal) has been used as a method of preserving perishable fish and marine products. Recently, however, high salted jeotkal (20% and more) has avoided from consumer by reasons of organoleptic quality (high salinity), health value (degenerative chronic disease), and hygienic safety. Among these jeotkals, meanwhile, sikhae (lactic acid fermented fishes) has been traditionally favored in the eastern coastal area of Korea, which contain below 10% of salt content with acidic range (pH4∼5).
