The effect of carbenicillin on the dedifferentiation and the regeneration efficiency of plant tissues of horseradish(Armoracia rusticana) was evaluated, Inhibition effect for callus initiation was observed when leaf blade, root and petiole segments were grown on MS medium containing 500 mg/L to 2000 mg/L carbenicillin and 0.5 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D). The regeneration of horseradish shoots from leaf blade, root and petiole explants were decreased as the addition of carbenicillin increased from 1000 mg/L to 2000 mg/L in MS medium containing 0.5 mg/L of 6-benzylaminopurine (BAP) or kinetin. Especially, 500 mg/L carbenicillin treatment significantly inhibited shoot induction when leaf blade explants were grown on hormone-free MS medium. It was suggested that the toxic effects of combinations of carbenicillin and 2,4-D may be due to high auxin activity levels.
This study was conducted to examine the position effect of axillary buds on shoot multiplication and rooting with 1-year-old seedlings of Quercus acutissima. Shoot multiplication was greatly affected by axillary bud position: Productivity of multiple shoots were decreased in the order of top, mid and basal explants respectively. The best shoot multiplication (mean 6.1 shoots per explant) was obtained on WPM medium containing $1.0mg/{\ell}$ BAP and $0.1mg/{\ell}$ NAA using basal explant after 4-week-culture. Rooting was also greatly influenced by position. Its percentage was increased in the order of top, mid and basal explant respectively. Root initiation was better and more rapid on 1/2MS medium than GD medium. High rooting percentage (100%) was obtained on 1/2MS medium containing $0.2mg/{\ell}$ IBA after 15 days culture. Sucrose concentrations did not effect on rooting. However root development and shoot growth were greatly affected by them. Root was developed shortly on 1-2% levels and shoot growth was getting retarded, whereas both of them did not show significant difference at 3-6% levels. Rotting was decreased on 7-8% levels gradually, but shoot and leaf condition was better than any other concentrations. Survival rate of rooted explants in pot was varied according to the position of explants. Seedlings of top part were survived up to about 50% but most of mid and basal part seedlings did not survive over 4 weeks even in high humidity condition. Seedlings in pots showed normal growth over 10 months but most of them showed the condition of premature leaf shedding.
Spary and vapor drift injuries of apple, pear, and peach seedling caused by soil-applied oxyfluorfen were studied in a greenhouse. Bud bursting rate of all fruit trees was reduced by both spray and vapor drifts of oxyfluorfen, but reduction in bud bursting rate of pear and peach was greater than that of apple trees. Reduction in the number of leaves per shoot of apple and peach was greater than that of pear trees. Leaf injury of pear was most severe and occurred earliest, but leaves of peach were least injured, Leaf injuries of pear and apple were caused by both spray and vapor drifts, but leaves of peach was injured largely by vapor drift. Reduction in shoot growth of and pear was greater than that of peach trees. Shoot growth of pear was more rapidly retared compared with apple trees. In the field, oxyfluorfen delayed the time of bud bursting in young apple trees. Oxyfluorfen applied between initiation and completion of bud bursting delayed bud bursting more than earlier application although ultimate number of bursted buds was similar to control. The number of leaves per shoot and total length of shoots were lower than control until 40 days after application of oxyfluorfen, and then were similar to control because of vigorous growth after May.
This study was conducted to define the effect of 2.4-D, NAA, Benzyladenine, and basic mediums on the callus induction and the organ differentiation from the meristem tips and the stem and leaf segments of the potato. Benzyladenine promoted the induction and growth of shoot from the meristem tip of potato but inhibited initiation of roots and induction of callus. At higher concentration of NAA than 0.5 ppm and of 2.4-D than 1.0 ppm the shoots were not initiated but the callus was induced from the meristem. The callus growth was significantly promoted on the medium containing NAA than 2.4-0. The initiation and growth of the shoots from the potato meristem was significantly increased in the medium containing 2.4-D and BA, or NAA and BA, compared with those containing BA, NAA or 2.4-D alone. The callus was more easily induced from the stem segments than the leaf segments of potato. And the 2.4-D was more effective for the induction and growth of the callus than the NAA. MS medium diluted its concentration to 1/2 was more suitable for the initiation and growth of the shoots from the potato meristem than the MS standard medium. For the initiation and growth of the shoots from the potato meristem, the most desirable medium was the diluted MS medium containing 1.0 ppm BA and 0.1 ppm NAA or 0.1 ppm 2.4-D.
The response of grain yield(GY) and milled-rice protein content(PC) to crop growth status and nitrogen(N) rates at panicle initiation stage(PIS) is critical information for prescribing topdress N rate at PIS(Npi) for target GY and PC. Three split-split-plot experiments including various N treatments and rice cultivars were conducted in Experimental Farm, Seoul National University, Korea in 2003-2005. Shoot N density(SND, g N in shoot $m^{-2}$) and canopy reflectance were measured before N application at PIS, and GY, PC, and SND were measured at harvest. Data from the first two years(2003-2004) were used for calibrating the predictive models for GY, PC, and SND accumulated from PIS to harvest using SND at PIS and Npi by multiple stepwise regression. After that the calibrated models were used for calculating N requirement at PIS for each of nine plots based on the target PC of 6.8% and the values of SND at PIS that was estimated by canopy reflectance method in the 2005 experiment. The result showed that SND at PIS in combination with Npi were successful to predict GY, PC, and SND from PIS to harvest in the calibration dataset with the coefficients of determination ($R^2$) of 0.87, 0.73, and 0.82 and the relative errors in prediction(REP, %) of 5.5, 4.3, and 21.1%, respectively. In general, the calibrated model equations showed a little lower performance in calculating GY, PC, and SND in the validation dataset(data from 2005) but REP ranging from 3.3% for PC and 13.9% for SND accumulated from PIS to harvest was acceptable. Nitrogen rate prescription treatment(PRT) for the target PC of 6.8% reduced the coefficient of variation in PC from 4.6% in the fixed rate treatment(FRT, 3.6g N $m^{-2}$) to 2.4% in PRT and the average PC of PRT was 6.78%, being very close to the target PC of 6.8%. In addition, PRT increased GY by 42.1 $gm^{-2}$ while Npi increased by 0.63 $gm^{-2}$ compared to the FRT, resulting in high agronomic N-use efficiency of 68.8 kg grain from additional kg N. The high agronomic N-use efficiency might have resulted from the higher response of grain yield to the applied N in the prescribed N rate treatment because N rate was prescribed based on the crop growth and N status of each plot.

Pb was first deposited on the root surface and then translocated radially in the root cap cells. During a longer Pb administration (up to 72 h) Pb penetration was nonuniform, with accumulation within the cortex or endodermis. There was drastic reduction in chlorophyll content by Pb. The Pb inhibition of chlorophyll synthesis was concentration dependent. 5-Aminolevulinic acid dehydratase (ALAD) activity exhibited distinct inhibition from control. Reduction in chlorophyll content was accompanied by proportional changes in ALAD activity. Chlorophyll content and ALAD activity were less affected by combined exposure of Pb and Al, suggesting that Al has a protective effect against the inhibiting action of Pb on photosynthetic activity.
A series of experiments were conducted to determine the effect of pH, salinity, seeding depth, and moisture stress on the germination and growth of Echinochloa colons (L.) Link. Germination significantly decreased at pH 10, but shoot lengths were not affected by the pH tested. Germination was not affected by salt concentrations of up to 0.1%, but was significantly reduced at 0.5%. A 1.0% salt concentration did not significantly reduce shoot length. Increase in seeding depth significantly reduced emergence. Irrespective of seeding depth, the coleoptilar node was always just below the soil surface. Delayed and decreased germination was obtained at -4.6 bars of simulated water potential, while no germination occurred at -9.8 bars. Soil moisture stress significantly reduced plant height, delayed panicle initiation, and reduced seed production.
This study was conducted to evaluate the effects of reduced nitrogen (N) and potassium (K) fertigation as additional fertilizer on tomato yield and nutrient contents in excessively nutrients-accumulated soil. Shoot and root dry weights (DW), dry matter rate for shoot, root and fruit and number of fruit in both AF50 and AF100 (50 and 100% levels of additional fertilizer) treatments were increased in comparison with those in AF0 (0% level of additional fertilizer) treatment. In case of nutrient uptake by tomato, nitrogen, phosphorous (P) and potassium contents in all tomato parts (leaf, stem, root and fruit) in AF50 and AF100 treatment were lower than those in AF0 treatment. On the contrary, soluble sugar and starch contents in all tomato parts in AF50 and AF100 were higher than those in AF0 treatment. There were differences between AF0 and AF50 or AF100 in tomato growth, yield, nutrient level and contents of soluble sugar and starch. In contrast, the level and initiation point of fertigation did not significantly affect the parameters. Based on our results, the application of properly reduced level of additional fertilizer is possible to maintain the productivity of tomato and alleviate the nutrient accumulation in plastic film house soils.

Somatic embryogendesis is one of good examples of the basic research for plant embryo development as well as an important technique for plant biotechnology. This paper describes the direct somatic embryogenesis from zygotic embryos of Panax ginseng is reversely related to normal axis growth of zygotic embryos by the experiment of various chemical treatments. Under the normal growth condition, the apical tips of embryo axis produced an agar-diffusible substance, which suppressed somatic embryo development from cotyledons. Although the cells of zygotic embryos were released from the restraint of embryo axis, various factors were still involved for somatic embryo development. Electron microscopic observation revealed that the ultrastructure of cells of cotyledon epidermis markedly changed before initiation of embryonic cell division, probably indicating reprogramming events into the cells embryogenically determined state. Polar accumulation of endogenous auxin or cell-cell isolation by plasmolysis pre-treatment is the strong inducer for the somatic embryo development. The cells for the process of somatic embryogenesis might be determined by the physiological conditions fo explants and medium compositions. Direct somatic embryos from cotyledons fo ginseng were originated eithrer from single or multiple cells. The different cellular origin of somatic embryos was originated either from single or multiple cell. The different cellular origin of somatic embryos was depended on various developmental stages of cotyledons. Immature meristematic cotyledons produced multiple cell-derived somatic embryos, which developed into multiple embryos. While fully mature cotyledons produced single cell-derived single embryos with independent state. Plasmolysis pretreatment of cotyledons strongly enhanced single cell-derived somatic embryogenesis. Single embryos were converted into normal plantlets with shoot and roots, while multiple embryos were converted into only multiple shoots. GA3 or a chilling treatment was prerequisite for germination and plant conversion. Low concentration of ammonium ion in medium was necessary for balanced growth of root and shoot of plantlets. Therefore, using above procedures, successful plant regeneration of ginseng was accomplished through direct single embryogenesis, which makes it possible to produce genetically transformed ginseng efficently.
