Olibanum (Boswellia serrata) has been shown to have anti-inflammatory, anti-arthritic and anticancer effects. This study determined the role of a water extract of olibanum in platelet-derived growth factor (PDGF)-stimulated proliferation and migration of rat aortic smooth muscle cells (RASMCs). PDGF-BB induced the migration and proliferation of RASMCs that were inhibited by olibanum extract in a dose-dependent manner. The PDGF-BB-increased phosphorylation of p38 mitogen-activated protein kinase (MAPK); the heat shock protein (Hsp) 27 was significantly inhibited by the olibanum extract. The effects of PDGF-BB-induced extracellular signal-regulated kinase1/2 was not altered by the olibanum extract. Treatment with olibanum extract inhibited PDGF-BB-stimulated sprout out growth of aortic rings. These results suggest that the water extract of olibanum inhibits PDGF-BB-stimulated migration and proliferation in RASMCs as well as sprout out growth, which may be mediated by the inhibition of the p38 MAPK and Hsp27 pathways.
Objectives: The purpose of this study was to investigate the anti-inflammatory effects of aqueous extract from Cicadidae Periostracum(CP) on the RAW 264.7 cells. Method: We examined the cytokine productions including nitric oxide(NO), interleukin(IL)-1b, IL-6 and tumor necrosis factor-a(TNF-a) in lipopolysaccharide (LPS)-induced RAW 264.7 cells and also inhibitory mechanisms such as mitogen -activated protein kinases(MAPKs) and nuclear factor kappa BNF-kB) using Western blot. Results: CP inhibited LPS-induced production of NO, IL-1b and TNF-a but not of IL-6 in RAW 264.7 cells. CP respectively inhibited the activation of MAPKs such as extracelluar signal-regulated kinase(ERK 1/2), c-Jun $NH_2$-terminal kinase(JNK), p38 and NF-kB in the LPS-stimulated RAW 264.7 cells. Also oral administration of CP inhibited CLP - induced endotoxin shock. Conclusion: our results showed that CP down-regulated LPS-induced NO, IL-1b and TNF-a productions mainly through ERK, JNK, p38 MAPK and NF-kB pathway, which suggest the anti-inflammatory effects of CP.

The presented paper gives an overview of several projects addressing the experimental characterization and control of the buffet phenomenon on 3D turbulent wings in transonic flow conditions. This aerodynamic instability induces strong wall pressure fluctuations and therefore limits flight domain. Consequently, to enlarge the latter but also to provide more flexibility during the design phase, it is interesting to try to delay the buffet onset. This paper summarizes the main investigations leading to the achievement of open and closed-loop buffet control and its experimental demonstration. Several wind tunnel tests campaigns, performed on a 3D half wing/fuselage body, enabled to characterize the buffet aerodynamic instability and to study the efficiency of innovative fluidic control devices designed and manufactured by ONERA. The analysis of the open-loop databases demonstrated the effects on the usual buffet characteristics, especially on the shock location and the separation areas on the wing suction side. Using these results, a closed-loop control methodology based on a quasi-steady approach was defined and several architectures were tested for various parameters such as the input signal, the objective function, the tuning of the feedback gain. All closed-loop methods were implemented on a dSPACE device able to estimate in real time the fluidic actuators command calculated mainly from the unsteady pressure sensors data. The efficiency of delaying the buffet onset or limiting its effects was demonstrated using the quasi-steady closed-loop approach and tested in both research and industrial wind tunnel environments.

Background: Hypothermia is a crucial environmental factor that elevates the risk of cardiovascular disease, but the underlying effect is unclear. Objectives: This study examined the role of cold stress (CS) in cardiac injury and its underlying mechanisms. Methods: In this study, a chronic CS-induced myocardial injury model was used; mice were subjected to chronic CS (4℃) for three hours per day for three weeks. Results: CS could result in myocardial injury by inducing the levels of heat shock proteins 70 (HSP70), enhancing the generation of creatine phosphokinase-isoenzyme (CKMB) and malondialdehyde (MDA), increasing the contents of tumor necrosis factor-α (TNF-α), high mobility group box 1 (HMGB1) interleukin1b (IL-1β), IL-18, IL-6, and triggering the depletion of superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH). Multiple signaling pathways were activated by cold exposure, including pyroptosis-associated NOD-like receptor 3 (NLRP3)-regulated caspase-1-dependent/Gasdermin D (GSDMD), inflammation-related toll-like receptor 4 (TLR4)/myeloid differentiation factor 88 (MyD88)-mediated nuclear factor kappa B (NF-κB), and mitogen-activated protein kinase (MAPK), as well as oxidative stressinvolved thioredoxin-1/thioredoxin-interacting protein (Txnip) signaling pathways, which play a pivotal role in myocardial injury resulting from hypothermia. Conclusions: These findings provide new insights into the increased risk of cardiovascular disease at extremely low temperatures.
단백질 번역 후 O-GlcNAc 수식은 단백질 조절의 새로운 기전으로 대두되고 있다. 전통적인 당수식과 달리 O-GlcNAc 수식은 단 한번의 O-GlcNAc 전달로 이루어지며, 핵 및 세포질단백질 모두에 수식될 수 있다. O-GlcNAc은 이 분자를 끝으로 하는 최종수식으로 생각되어 왔으나, 최근의 논문(J Proteome Res. 2011 10:2725-2733)은 AP180 단백질에 O-GlcNAc-P가 존재함을 보고하였다. 이 논문은 O-GlcNAc-P가 일반적인 단백질수식인지에 대한 중요한 질문을 던진다. 이에 답하고자 저자들은 HEK293T 세포에 O-GlcNAc 인산화효소 NAGK를 DsRed2에 연결한 DsRed2-$NAGK_{WT}$ 혹은 효소활성이 없는 돌연변이 NAGK를 표현하는 DsRed2-$NAGK_{D107A}$를 표현시키고, 단백질 추출물을 얻어 2D-PAGE로 분리한 후 인산화 정도를 측정하여, $NAGK_{WT}$에 의하여 인산화가 증가되는 15개의 단백질 스폿을 선별하였다. 이 가운데 7개 스팟을 동정한 결과 2개의 스폿은 O-GlcNAc 수식 단백질인 $HSP90{\beta}$, 다른 2개의 스폿도 O-GlcNAc 수식 단백질인 ENO1로 동정되었으며, 나머지(dUTP nucleotidohydrolase mitochondrial isoform 2, glutathione S-transferase P, grp94)는 O-GlcNAc 수식 여부를 아직 모르는 단백질이였다. NAGK에 의하여 O-GlcNAc 단백질의 인산화가 증가된다는 사실은 O-GlcNAc이 인산화되어 O-GlcNAc-P로 수식됨을 시사하며, 따라서 본 연구의 결과는 O-GlcNAc이 최종 수식이 아님을 지지한다.
식물에서 전이인자를 이용한 삽입 변이체의 유전자 기능분석 연구가 최근 가장 활발하게 이루어지고 있다. 본 연구에서는 동진벼의 Ac/Ds 삽입 변이체인 F2 세대 30,000 계통을 이용하여 고염과 저온에 민감한 계통과 내성이 있는 계통을 대량 스크리닝을 통해 선발하였다. 첫 번째 스크리닝에서 선발한 212 계통을 Southern blot 분석을 통해 Ds의 삽입여부 및 copy 수를 꽉인하고 표현형과 비교하여 고염과 저온에서 총 19 계통을 선발하였고, 이 중 copy 수가 하나인 계통은 13 계통이었다. 선발한 계통을 FSTs 분석을 통해 Ds의 삽입위치 및 knock-out유전자를 확인하고 염기서열 정보를 이용하여 벼 전체 염기서열 정보와 상동성 비교분석 결과 세포의 신호전달 과정과 조절 관여하는 유전자 그룹인 transpoter, protease family protein and apical meristem family protein, 삼투압조절에 관여하는 유전자 그룹인 heat shock potein, O-methyltransferase, glyceraldehyde-3-phosphate dehydrogenase and drought stress Induce protein 그리고 식물의 소포유통(vesicle trafficking)에 관여하는 유전자 SYP 5 family protein로 구분할 수 있었다. 선발된 19개 유전자의 발현 분석을 위해 9종류 비생물학적 스트레스 하에서 RT-PCR을 수행한 결과 이들 knock-out 유전자는 비생물학적 스트레스에 각각 다른 발현 패턴을 보였다. 이 연구의 결과는 삽입 변이체를 통한 유전자의 기능분석에 있어서 비생물학적인 스트레스의 응답 반응계에 관여하는 유전자를 연구하는데 유용할 것이라고 생각된다.
Intensive Monitoring Survey of Nearby Galaxies (IMSNG) is a high cadence observation program monitoring nearby galaxies with high probabilities of hosting supernovae (SNe). IMSNG aims to constrain the SN explosion mechanism by inferring sizes of SN progenitor systems through the detection of the shock-heated emission that lasts less than a few days after the SN explosion. To catch the signal, IMSNG utilizes a network of 0.5-m to 1-m class telescopes around the world and monitors the images of 60 nearby galaxies at distances D < 50 Mpc to a cadence as short as a few hours. The target galaxies are bright in near-ultraviolet (NUV) with $M_{NUV}$ < -18.4 AB mag and have high probabilities of hosting SNe ($0.06SN\;yr^{-1}$ per galaxy). With this strategy, we expect to detect the early light curves of 3.4 SNe per year to a depth of R ~ 19.5 mag, enabling us to detect the shock-heated emission from a progenitor star with a radius as small as $0.1R_{\odot}$. The accumulated data will be also useful for studying faint features around the target galaxies and other science projects. So far, 18 SNe have occurred in our target fields (16 in IMSNG galaxies) over 5 years, confirming our SN rate estimate of $0.06SN\;yr^{-1}$ per galaxy.
Global cerebral ischemia occurs commonly in patients who have a variety of clinical conditions including cardiac arrest and shock. Cerebral ischemia results in a rapid depletion of energy stores that triggers resulting in excitotoxic death. Imaging studies of the brain with computed tomography(CT) or magnetic resonance imaging(MRI) are necessary to confirm the clinical neurolocalization, identify any associated mass effect, and rule out other causes of focal brain disorders. Cardiopulmonary arrest was occurred by propofol anesthesia in a 1 year old, intact female Beagle dog. After successful cardiopulmonary resuscitation was performed within 5 minutes, clinical signs such as vocalization, paddling, opisthotonus and seizure were represented. At the 12th day, CT and MRI examinations of the brain were performed to evaluate the brain. After euthanasia, histopathologic examination was performed. On transverse image of CT, lesions appeared as a hypodense in the right dorsal surface of the frontal lobe and level of optic canal, and dorsomedial surface of occipital lobe of cerebrum. No contrast enhancement was represented following intravenous contrast administration. On MR images of brain, the lesions were seen as a hyperintense on T2-weighted(T2W) images and a isointense or mild hypointense on T1-weighted(T1W) images. Hyperintense lesions both T2W and T1W images were observed at the surrounding cerebral sulcus. There was no significant signal changes on contrast T1WI. Histopathologic examination after euthanasia revealed that the lesion was necrosis of the cerebral cortex caused by cerebral ischemia.
The NISS (Near-infrared Imaging Spectrometer for Star formation history) is the near-infrared instrument optimized to the Next Generation of small satellite series (NEXTSat). The capability of both imaging and low spectral resolution spectroscopy in the near-infrared range is a unique function of the NISS. The major scientific mission is to study the cosmic star formation history in local and distant universe. For those purposes, the main observational targets are nearby galaxies, galaxy clusters, star-forming regions and low background regions. The off-axis optical design is optimized to have a wide field of view ($2deg.{\times}2deg.$) as well as the wide wavelength range from 0.95 to $3.8{\mu}m$. Two linear variable filters are used to realize the imaging spectroscopy with the spectral resolution of ~20. The mechanical structure is considered to endure the launching condition as well as the space environment. The compact dewar is confirmed to operate the infrared detector as well as filters at 80K stage. The electronics is tested to obtain and process the signal from infrared sensor and to communicate with the satellite. After the test and calibration of the engineering qualification model (EQM), the flight model of the NSS is assembled and integrated into the satellite. To verify operations of the satellite in space, the space environment tests such as the vibration, shock and thermal-vacuum test were performed. Here, we report the test results of the flight model of the NISS.
대기 압력 변동을 측정하는 인프라사운드 관측 기술을 통하여 원거리 지표폭발 사고를 분석하였다. 2019년 12월 24일 전남 광양시에서 발생한 2차례 폭발 사고에서 발생한 인프라사운드 신호가 151-435 km 거리에 위치하는 12개 음파 관측소에 기록되었다. 당시 인프라사운드는 북북서 방향의 성층권 바람에 의해 약 40 km 고도에서 굴절되어 같은 방향에 분포하는 관측소에 도달하였다. 반면, 약 10 km 고도에서는 강한 서풍의 영향으로 대류권 굴절 신호가 북동 및 동쪽 방향에 위치하는 관측소에 도달하는 등 방향에 따라 상이한 전파 경로를 보였다. 대기 유효음파속도구조와 포물선 방정식 모델링을 통해 전파 경로상의 투과손실을 계산하고 폭발 지점으로부터 기준거리에서의 초과압력을 추정하였다. 추정된 초과압력은 초과압력-폭발량 관계식에 적용함으로써, 두 차례의 폭발은 각각 14, 65 kg TNT 폭발 에너지에 상응하는 것으로 계산되었다. 1차 폭발 당시에 폭발 충격으로 부속물이 대기 중으로 비산하는 현상이 관측되었고, 폭발충격에 의한 파편 운동과 초과압력 간의 관계식으로 1차 폭발의 에너지는 약 49 kg 이하 TNT 폭발에 상응하는 것으로 계산되었다. 본 연구에서 제안한 폭발 에너지 추정 방법은 향후 다양한 원거리 폭발 에너지 계산에 활용이 가능하리라 본다. 향후 계산 결과의 신뢰도를 높이기 위해서는 대기 속도구조 불확실성에 대한 연구와 다양한 발파 자료를 통한 검증 연구가 필요하다.
