Heat shock protein gp96 is an endoplasmic reticulum chaperone, belonging to the HSP90 family. The function of gp96 as a molecular chaperone was discovered more than 10 years ago, but its importance has been overshadowed by the brilliance of its role in immune responses. It is now clear that gp96 is instrumental in the initiation of both the innate and adaptive immunity. Recently, the roles of gp96 in protein homeostasis, as well as in cell differentiation and development, are beginning to draw more attention due to rapid development in the structural study of HSP90 and some surprising new discoveries from genetic studies of gp96. In this review, we focus on the aspect of gp96 as an ER molecular chaperone in protein maturation, peptide binding and the regulation of its activity.
The structural integrity of the reactor pressure vessel under pressurized thermal shock (PTS) is evaluated in this study. For given material properties and transient histories such as temperature and pressure, the stress distribution is found and stress intensity factors are obtained for a wide range of crack sizes. The stress intensity factors are compared with the fracture toughness to check if cracking is expected to occur during the transient. A round robin problem of the PTS during a small break loss of coolant transient has been analyzed as a part of the international comparative assessment study, and the evaluation results are discussed. The maximum allowable nil-ductility transition temperatures are determined for various crack sizes.

Notches such as slots are typical geometric features on mechanical components that promote fatigue crack initiation. Unlike for components with open hole type notches, there are no conventional treatments to enhance fatigue behavior of components with slots. In this work we evaluate the viability of applying laser shock peening (LSP) to extend the fatigue life of 6061-T6 aluminum components with slots. The feasibility of using LSP is evaluated not only on damage free notched specimens, but also on samples with previous fatigue damage. For the LSP treatment a convergent lens was used to deliver 0.85 J and 6 ns laser pulses 1.5 mm in diameter by a Q-switch Nd: YAG laser, operating at 10 Hz with 1064 nm of wavelength. Residual stress distribution was assessed by the hole drilling method. A fatigue analysis of the notched specimens was conducted using the commercial code FE-Safe and different multiaxial fatigue criteria to predict fatigue lives of samples with and without LSP. The residual stress field produced by the LSP process was estimated by a finite element simulation of the process. A good comparison of the predicted and experimental fatigue lives was observed. The beneficial effect of LSP in extending fatigue life of notched components with and without previous damage is demonstrated.
Purpose: The aim of this study was to evaluate the effect of overexpression of heat shock protein 70 (HSP70) on the expression of inducible nitric oxide synthase and on the concentration of nitric oxide and to determine the mechanism for the relationship between HSP70 and inducible nitric oxide synthase (iNOS) in sepsis. Methods: Experiments were performed on male Sprague-Dawley rats, and sepsis was induced by using cecal ligation and puncture (CLP). Glutamine (GLN) or saline was administered 1 h after initiation of sepsis. We acquired serum and lung tissues from the rats 12 h or 24 h after initiation of sepsis. We analyzed the concentration of nitric oxide, the expression of HSP70 in the lung, and the gene expression of iNOS in the lung. Results: In CLP+GLN, glutamine given after initiation of sepsis enhanced the expression of HSP70 in the lung at 12 h (CLP+GLN vs. CLP:: $47.19{\pm}10.04$ vs. $33.22{\pm}8.28$, p = 0.025) and 24 h (CLP+GLN vs. CLP: $47.06{\pm}10.60$ vs. $31.90{\pm}4.83$, p = 0.004). In CLP+GLN, glutamine attenuated the expression of iNOS mRNA in the lung at 12 h (CLP+GLN vs. CLP: $4167.17{\pm}951.59$ vs. $5513.73{\pm}1051.60$, p = 0.025) and 24 h (CLP+GLN vs. CLP: $9,437.65{\pm}2,521.07$ vs. $18,740.27{\pm}8,241.20$, p = 0.016) and reduced the concentration of nitric oxide in serum at 12 h (CLP+GLN vs. CLP: $0.86{\pm}0.48$ vs. $3.82{\pm}2.53{\mu}mol/L$, p = 0.016) and 24 h (CLP+GLN vs. CLP: $0.39{\pm}0.25$ vs. $1.85{\pm}1.70{\mu}mol/L$, p = 0.025). Conclusion: The overexpression of HSP70 induced by the administration of glutamine in sepsis attenuated the gene expression of iNOS and reduced the concentration of nitric oxide.

Despite aggressive treatment, the mortality rate of cardiogenic shock with acute myocardial infarction (AMI) is high. We performed extracorporeal membrane oxygenation (ECMO) prior to coronary reperfusion, and evaluated the early clinical results and risk factors. Materials and Methods: From May 2006 to November 2009, we reviewed the medical records of 20 patients in cardiogenic shock with AMI (mean age $67.7{\pm}11.7$ yrs, M : F 14 : 6). After initially performing ECMO using the CAPIOX emergency bypass system ($EBS^{(R)}$Terumo, Tokyo, Japan), patients underwent coronary reperfusion (coronary artery bypass grafting, 13; percutaneous coronary intervention, 7). Results: All patients were in a cardiogenic shock state, cardiopulmonary resuscitations (CPR) were performed for fourteen patients (mean CPR time $20.8{\pm}26.0$ min). The mean time from vascular access to the initiation of ECMO was $17.2{\pm}9.4$ min and mean support time was $3.8{\pm}4.0$ days. Fourteen patients were able to be weaned from ECMO and ten patients were discharged (mean admission duration $50.1{\pm}31.6$ days). Patients survived on average $476.6{\pm}374.6$ days of follow-up. Longer CPR and support time, increased cardiac enzyme, lower ejection fraction, lower albumin, and major complications were the risk factors of mortality (p<0.05). Conclusion: The early application of ECMO prior to coronary reperfusion and control of risk factors allowed for good clinical results in cardiogenic shock with AMI.
A probabilistic structural integrity assessment is performed for a reactor pressure vessel under PTS(Pressurized Thermal Shock). A semi-elliptical finite axial crack is assumed to he in the beltline region(either base metal or weld meta)1 of the reactor vessel inside surface. The selected random variables are initial crack depth, neutron fluence on the vessel inside surface, copper, nickel, and phosphorus content of the vessel material, and RT/sub NDT/. The probabilities of crack initiation or vessel failure where the crack is propagated through vessel wall are calculated. The probabilities obtained with random crack size are compared to these obtained with deterministic us. Since the failure function cannot to explicitly by selected by selected random variables, Monte Carlo Simulation is applied to perform probabilistic analysis The influence of the amount of neutron fluence is also examined to assess the structural reliability for vessel life time.
The probabilistic fracture mechanics(PFM) is a useful analytical tool to assess the integrity of reactor pressure vessel(RPV) at the event of pressurized thermal shock(PTS). In PFM, the probabilities of flaw initiation and propagation are estimated by comparing the applied stress intensity factor with the fracture toughness calculated by the simulation of various stochastic variables. It is known that the results of PFM analyses are dependent on the choice of the stochastic parameters and assumptions. Of the various variables and assumptions, we investigated the effects of the RT$_{NDT}$ shift equations, fracture toughness curves, and flaw distributions on the PFM results for the three PTS transients. The results showed that the combined effects of the RT$_{NDT}$ shift equations and fracture toughness curves are complicated and dependent on the characteristics of the transients, the chemistry of the materials, the fast neutron fluence, and so on.
The detonation combustion is a supersonic combustion process follows on shock wave oscillations in detonation tube. In this paper numerical studies are carried out combined effect of blockage ratio and spacing of obstacle on detonation wave propagation of hydrogen-air mixture in pulse detonation combustor. The deflagration to detonation transition of stoichiometric (ϕ=1)fuel-air mixture in channel has been analyzed for effect of blockage ratio (BR)=0.39, 0.51, 0.59, 0.71 with spacing of 2D and 3D. The reactive Navier-Stokes equation is used to solve the detonation wave propagation mechanism in Ansys Fluent platform. The result shows that fully developed detonation wave initiation regime is observed near smaller vortex generator ratio of BR=0.39 inside the combustor. The turbulent rate of reaction has also a great significance role for shock wave structure. However, vortices of rapid detonation wave are appears near thin boundary layer of each obstacle. Finally, detonation combustor demonstrates the superiority of pressure gain combustor with turbulent rate of reaction of 0.6 kg mol/m3 -s inside the detonation tube with obstacle spacing of 12 cm, this blockage enhanced the turbulence intensity and propulsive thrust. The successful detonation wave propagation speed is achieved in shortest possible time of 0.031s with a significance magnitude of 2349 m/s, which is higher than Chapman-Jouguet (C-J) velocity of 1848 m/s. Furthermore, stronger propulsive thrust force of 36.82 N is generated in pulse time of 0.031s.
