대장암 세포주 SNU C2A에서 etoposide 처리에 의한 apoptosis 유도 (Induction of apoptosis by etoposide treatment in colon cancer cell line SNU C2A)

  • Background: Inactivation of tumor suppressor genes is believed to be important in the development of many human malignancies. Recently, several lines of evidence have indicated that the wild type p53 gene located at 17p13.3, may function as a tumor suppressor gene and that a mutant p53 gene could promote transformation by inactivating normal p53 function in a dominant negative fashion. These broad spectrum of p53 mutation in human cancers provide that mutant p53 and their protein may be potential targets of tumor diagnostic and therapeutic interventions. Method: Colony formation was performed to investigate growth suppressional ability. p53 expression pattern was examined by western blot and p53-mediated transactivation ability was assessed by CAT activity. SNU C2A cells were observed in apoptotic aspects induced by etoposide and $H_2O_2$ treatment, detecting sensitivity on agent, DNA fragmentation through agarose gel, chromatin condensation by fluorescence microscope, and cell cycle distribution by FACS. Result: 1) p53 mutant his179arg ($histidine{\rightarrow}arginine$) detected in SNU C2A cells lost transcriptional activity and growth suppression ability, showing dominant negative effect on its wild type p53. 2) Etoposide-treated SNU C2A cells induced apoptosis, exhibiting dramatic reduction of cell growth, DNA fragmentation, nuclear condensation formation of apoptotic body and increment of sub-G1 cell fraction. 3) Etoposide and $H_2O_2$-treated SNU C2A cells have no high increase of p53 expression and overexpressed p53 protein changed localization, from cytoplasm to nucleus. Also, p53-mediated transcriptional activity was increased by agents-treatment. Conclusion: SNU C2A cells coexpress wild-type and mutant p53 protein induced apoptosis in the condition on DNA damage, through localizational shift from cytoplasm to nucleus of p53 protein rather than the induction of p53 protein. SNU C2A cells derived mutant p53 his179arg abrogated both the growth supression ability and transactivational activity, showing inhibition effect on transcriptional activity of wild type p53, but did not repress the activity of wild type p53 in SNU C2A cells owing to dominant activity of wild type. These cell condition may provide new gene therapeutic implications leading effective antiproliferation of cell when mutant and wild-type p53 protein were co-expressed in cell.

미래학교 수학교실의 교육 방법론에 대한 탐색:비구조화된 문제에서 학생들의 질문 만들기를 중심으로 (Teaching Methodology for Future Mathematics Classroom:Focusing on Students' Generative Question in Ill-Structured Problem)

  • This paper explores students' question generation process and their study in small group discussion. The research is based on Anthropological Theory of the Didactic developed by Chevallard. He argues that the savior (knowledge) we are dealing with at school is based on a paradigm that we prevail over whether we 'learn' or 'study' socially. In other words, we haven't provided students with autonomous research and learning opportunities under 'the dominant paradigm of visiting works'. As an alternative, he suggests that we should move on to a new didactic paradigm for 'questioning the world a question', and proposes the Study and Research Courses (SRC) as its pedagogical structure. This study explores the SRC structure of small group activities in solving ill-structured problems. In order to explore the SRC structure generated in the small group discussion, one middle school teacher and 7 middle school students participated in this study. The students were divided into two groups with 4 students and 3 students. The teacher conducted the lesson with ill-structured problems provided by researchers. We collected students' presentation materials and classroom video records, and then analyzed based on SRC structure. As a result, we have identified that students were able to focus on the valuable information they needed to explore. We found that the nature of the questions generated by students focused on details more than the whole of the problem. In the SRC course, we also found pattern of a small group discussion. In other words, they generated questions relatively personally, but sought answer cooperatively. This study identified the possibility of SRC as a tool to provide a holistic learning mode of small group discussions in small class, which bring about future mathematics classrooms. This study is meaningful to investigate how students develop their own mathematical inquiry process through self-directed learning, learner-specific curriculum are emphasized and the paradigm shift is required.

항공기 계기판의 적정배열을 위한 인간공학적 연구 (A Human Factors Study in Instrument Panel Layout of the Korean Air Force Aircraft.)

  • The purpose of this thesis is to study the optimal arrangement of aircraft instrument panels through the human factors approach. Human factors engineering is the process of effectively fitting the human component to the machine component in any man-machine system. The human factors. are especially important to an aircraft pilot who must constantly shift his attention between the instrument panel within the cockpit and the surrounding area of the aircraft. The preliminary part of this study is to find the general patterns of the Korean pilot's eye movements during their various flying maneuvers, and which instruments require the most attention while in flight. It is assumed that all pilots have a general pattern of eye movement when observing the aircraft instrument panel and that an optimum arrangement would be to minimize the eye travel distance between instruments. In this thesis the arrangements of instruments is taken to be the independent variable and the eye travel distance between instruments the dependent variable. la order to compile the information necessary for this study, sixty Korean Air Force pilots were interviewed and requested to complete information forms. These information forms listed various flying maneuvers and listed each instrument used on the instrument panel. The compilation of the information on these completed forms listed the instruments most frequently used by the pilots. The second part of this study was to determine the optimum instrument arrangement. It was necessary to study the various number of possible arrangements of instruments depending upon the number of instruments involved. Therefore, these instruments are grouped by two major functions, The flight instruments were subdivided into three groups, and the engineering instruments were subdivided into six groups. With this subdivision we arrive at the possible number of arrangements of 4,320. Through the simulation method, total eye travel distance for each of these 4,320 arrangements is calculated and the arrangement which appears to be of optimum distance between the most frequently used aircraft instruments is determined. The results of this study indicate that the optimum distance between instruments would be 33,028cm and that the corresponding distance of the instrument panel now being used is 34,288cm. Therefore, an increased efficiency of $3.8\%$ would be realized if the existing aircraft instrument panel were re-arranged according to layout proposed in this thesis.

시니어의 세대별 도서관 이용행태 비교분석: 국회도서관을 중심으로 (Generational Comparative Analysis on Library Usage of Senior Groups in National Assembly Library of Korea)

  • 시니어전환 시대를 맞이하여 도서관의 주 이용군으로 부상된 시니어를 대상으로 디지털환경에서 정보접근 및 검색 편의정도, 요구사항을 파악하였다. 이를 위하여 55세 이상의 국회도서관 이용자의 이용행태 및 검색행태를 조사하였고 이러한 행태와 만족도를 시니어 세대별 즉 베이비붐세대의 액티브시니어 그룹과 노인세대의 시니어 그룹으로 나누어 비교분석하였다. 그 결과 첫째, 국회도서관을 이용하는 노인세대는 여가를 목적으로 방문하며 정치 사회, 종교 철학, 역사 지리분야를 선호하였으나, 베이비붐세대는 자기계발을 목적으로 경제 재테크, 예술 문화 분야를 선호하는 것으로 조사되었다. 둘째, 검색을 실패하였을 경우 베이비붐세대의 시니어는 스스로 결과를 제어하는 반면, 노인세대는 사서에 의존도가 높은 것으로 나타났다. 셋째, 시니어 두 그룹 모두 장시간 전자도서관을 이용하면서 자료를 브라우징하고 있음에도 불구하고 전자도서관에서 제공하고 있는 편의 정제 기능에 대한 이용률이 저조하였다. 따라서 시니어의 이용을 활성화하고 편의성을 높이기 위해서는 시니어 전담사서가 필요하고 전자도서관을 이용하는 시니어를 위해 따로 도움말, 메뉴 설명이 필요 없는 직관적인 인터페이스가 개발될 필요가 있다. 이로써 현 이용자뿐만 아니라 잠재적 시니어들의 정보 리터러시 향상과 이용성 제고를 도모할 수 있을 것으로 본다.

기혼 직장인의 성인애착이 직무동기에 미치는 영향 : 부부관계의 매개효과 (Effects of Adult Attachment of Married Employees on Job Motivation: Mediating Effect of Marital Relationship)

  • 본 연구의 목적은 기혼직장인의 성인애착, 직무동기, 부부관계 간의 관계를 파악하는 것이다. 이를 위해 충남 지역의 기혼직장인 370명을 연구 대상으로 2016년 9월 10일~10월 14일까지 설문을 실시하였다. 수집된 자료는 SPSS 23.0을 이용하여 기술통계, 독립표본 t검증 및 일원배치 분산분석, Pearson 상관관계, 구조방정식으로 분석하였다. 첫째, 연령대에 따라 애착회피, 부부관계, 직무동기, 학력에 따라 애착회피, 부부관계, 결혼기간에 따라 애착회피, 부부관계, 직책에 따라 직무동기, 가계의 연평균 소득에 따라 부부관계, 근무형태에 따라 부부관계에서 유의미한 차이가 나타났다. 둘째, 애착불안과 애착회피 간에는 유의미한 부적 상관관계가 나타났지만, 애착회피와 직무동기 간에는 유의미한 정적 상관관계가 나타났다. 애착회피와 부부관계 간에는 유의미한 부적 상관관계가 나타났지만, 직무동기와 부부관계 간에서는 유의미한 정적 상관관계가 나타났다. 셋째, 성인애착과 직무동기의 관계에서 부부관계의 매개효과를 확인하기 위해 구조방정식을 실시하였다. 그 결과 애착불안이 직무동기에 주는 간접효과가 유의미하게 나타나 완전매개효과가 검증되었다. 이러한 결과는 직장 내 직무동기, 부부관계 향상을 위한 상담 개입이나 프로그램 개발을 위해서는 개개인의 이러한 특징 또한 고려되어야 함을 시사한다.

Expression characterization and transcription regulation analysis of porcine Yip1 domain family member 3 gene

  • Objective: The Yip1 domain family (YIPF) proteins were proposed to function in endoplasmic reticulum (ER) to Golgi transport and maintenance of the morphology of the Golgi, which were homologues of yeast Yip1p and Yif1p. YIPF3, the member 3 of YIPF family was a homolog of Yif1p. The aim of present study was to investigate the expression and regulation mechanism of porcine YIPF3. Methods: Quantitative realtime polymerase chain reaction (qPCR) was used to analyze porcine YIPF3 mRNA expression pattern in different tissues and pig kidney epithelial (PK15) cells stimulated by polyinosine-polycytidylic acid (poly [I:C]). Site-directed mutations combined with dual luciferase reporter assays and electrophoretic mobility shift assay (EMSA) were employed to reveal transcription regulation mechanism of porcine YIPF3. Results: Results showed that the mRNA of porcine YIPF3 (pYIPF3) was widely expressed with the highest levels in lymph and lung followed by spleen and liver, while weak in heart and skeletal muscle. Subcellular localization results indicated that it expressed in Golgi apparatus and plasma membranes. Upon stimulation with poly (I:C), the level of this gene was dramatically up-regulated in a time- and concentration-dependent manner. pYIPF3 core promoter region harbored three cis-acting elements which were bound by ETS proto-oncogene 2 (ETS2), zinc finger and BTB domain containing 4 (ZBTB4), and zinc finger and BTB domain containing 14 (ZBTB14), respectively. In which, ETS2 and ZBTB4 both promoted pYIPF3 transcription activity while ZBTB14 inhibited it, and these three transcription factors all played important regulation roles in tumorigenesis and apoptosis. Conclusion: The pYIPF3 mRNA expression was regulated by ETS2, ZBTB4, and ZBTB14, and its higher expression in immune organs might contribute to enhancing ER to Golgi transport of proteins, thus adapting to the immune response.

수술실 간호사의 이직의도에 영향을 미치는 요인 (Factors Affecting on Turnover Intentions among Operating Room Nurses)

  • 본 연구는 수술실 간호사의 스트레스와 의료기관 인증관련 직무스트레스, 대응 및 이직의도와의 관련성을 확인하고, 이에 영향을 주는 변수들을 확인하고자 시도되었다. 구조화된 설문지를 이용한 서술적 상관관계 연구로 수술실 간호사 163명을 대상으로 하였다. 자료수집 기간은 2015년 6월 1일부터 7월 31일까지였다. 수술실 간호사의 스트레스는 연령(F=5.629, p=.004), 결혼상태(t=-2.907, p=.004), 자녀유무(t=-2.623, p=.010)에 따라 차이가 있었다. 인증관련 직무스트레스는 연령(F=11.924, p<.001), 결혼상태(t=-4.747, p<.001), 자녀유무(t=-4.434, p<.001), 수술실 근무경력(F=6.000, p=.003), 수술실 근무배경(t=-2.543, p=.012)에 따라 차이가 있었다. 이직의도는 연령(F=21.472, p<.001), 교육수준(F=4.997, p<.008), 종교유무(t=3.235, p=.001), 수술실 근무경력(F=13.599, p<.001), 직위(t=3.222, p=.002), 수술실 근무배경(t=-3.001, p=.003), 근무형태(t=-2.797, p=.006)에 따라 차이가 있었다. 이직의도는 스트레스, 인증관련 직무스트레스, 대응과 유의한 상관관계가 있었으며 이들 요인이 이직의도를 설명하는 정도는 32.6%였다. 수술실 간호사의 이직의도에 영향을 주는 요인이 스트레스, 인증관련 직무스트레스, 대응으로 나타났으므로 수술실 간호사의 이직을 예방하고 감소시키기 위해 스트레스와 인증관련 직무스트레스를 해소하고 대응 능력을 기를 수 있는 프로그램의 개발 및 중재가 필요하다.

자가 슬괵건을 이용한 전방십자인대 재건술 후 결과 판정에 있어 MRI 검사의 유용성 (The effectiveness of MRI evaluation after anterior cruciate ligament reconstruction using hamstring tendon autograft)

  • 목적: 10시 방향 대퇴 터널의 전방십자인대 재건술 후 MRI 검사의 유용성을 알아보고자 하였다. 대상 및 방법: 자가 슬괵건을 이용한 전방 십자 인대 재건술 후 MRI 검사를 시행 받았던 29명의 소견을 분석하였다. 평균 술 후 MRI 검사까지의 기간은 18.9개월($7{\sim}40$개월)이었다. 이식건의 신호강도, 형태, 연속성, 대퇴터널 시작점, 대퇴터널 위치, 과간 절흔 충돌, 교차 핀 손상 및 위치에 대해 조사하였으며 KT-2000 검사, Lysholm 점수, 축회전 검사 등과 비교하였다. 결과: 이식건의 신호강도와 술후 MRI 검사까지의 기간은 유의한 상관관계가 없었다. 이식건의 형태와 연속성은 straight, well-preserved 가 대부분이었다. 대퇴 터널의 시작점은 zone 4가 13례, zone 3가 16례였으며 과간 절흔 충돌은 관찰되지 않았다. 교차 핀의 손상은 10례였고 이중 대퇴 후방 피질골 밖으로 빠진 경우가 5례였고, 방향이 후방을 향하는 경우가 9례였다. 교차 핀의 손상 여부와 임상 결과와의 상관 관계는 통계적으로 유의하지 않았다. 결론: 전방 십자 인대 재건술 후 MRI 검사는 이식건의 상태와 위치, 교차 핀의 위치 등을 파악하는데 유용하였다. 10시 방향 대퇴터널의 전방십자인대 재건술에서는 특이 교차 핀의 위치가 중요하므로 수술 시 교차 핀의 손상을 피하기 위한 세심한 주의를 요한다.

일부 종합병원 간호사의 건강증진생활양식에 대한 실천정도와 영향요인에 관한 연구 (A Study on Health-Promoting Lifestyle and Its Affecting Factors of Hospital Nurses)

  • The chances of disease pattern increased the importance of Health-promoting Lifestyle and a large part of the Health-promoting Lifestyle is associated with individual's habit. Health-promoting Lifestyle among nurses is very important because nurses could be a role model of patients and possibly influence many patients. The purpose of this study was oat on1y to assess how hospital nurses practice their Health-promoting Lifestyle but also to identify those affecting determinants. The subjects were 392 nurses working at 3 different hospital in Seoul. These data were collected by self administered questionnaire from April 27 to May 20, 2000. This study examined Health-promoting Lifestyle using In Sook Park's model, individual characteristics. Behavior-specific Cognitions and Affect factors using Pender's model and tried to fine out their relationships. The instruments used in this study were Health-promoting Lifestyle Profile developed by In Sock Park(1995). Likert's four-point scale was used also in this research. The percentage, mean standard deviation, AVNOVA, Pearson's correlation coefficient and multiple regression in the SAS package were used to analyze the data. The results of this study were as follows; 1. 52.3% of sample were aged between 25 and 29, 67.1% were single, 55.6% were university graduates, 51.8% earned 1.5 to 2.0 million won, 57.9% slept for d to 8 hours, 74.5% stated they were healthy. 2. 32.7% of sample worked in surgical gird department, 82.4% worked in 3 shift, 26.3% have been working as nurses for 5 to 7 years. Average score of Perceived self-efficacy was 3.63, Perceived benefits of action was 3.25, Social support was 2.75, and Perceived barriers to action was 1.87 which was the lowest score. 3. The average score if the performance in Health-promoting Lifestyle variable was 2.45 which was lower than previous study. The sanitary life Was 3.18 which was the highest score, harmonious relationship 3.13, emotional support 2.90. The variable with the highest degree of performance was the sanitary life, whereas the one with the west degree was the health diet. 4. There were no significant difference in perceived benefits of action with individual's a character, but in Perceived bait his of action there were significant differences with age(p<.01), marital status(p<.05), housing(p<.05), and Perceived health status(p<.05). In Perceived self-efficacy, there were significant differences with educational level(p<05), sleeping hours(p<.05), and BMI(p<.05). In Social support here were significant difference with housing and sleeping hours.

유휴간호사 재취업 의향에 영향을 미치는 요인 (Factors influencing the intent to return to practice (work) of inactive RNs)

  • 본 연구는 유휴간호사의 재취업 의향에 영향을 미치는 요인을 규명하고자 실시하였다. 2013년 대한간호협회와 한국보건사회연구원에서 실시한 '병원간호사 이직(사직)에 대한 조사' 연구를 위한 설문조사 자료 중 최종 381명의 자료를 활용한 이차자료 분석 연구이다. 재취업 의향이 있는 경우는 70.9%로 나타났고, 이들 중 선호하는 근무형태로는 시간선택제 47.8%, 낮번전담제 43.3%, 3교대제 6.3% 이었다. 대상자의 사직의 주된 이유는 '노동 강도가 높아서' (18.8%), '밤근무가 힘들어서' (16.7%), '임금 수준이 낮아서' (15.9%), '출산 및 육아 보육' (15.9%), '직장내 인간관계의 어려움' (11.1%)이었다. 재취업 의향에 유의한 영향을 미치는 요인은 기혼, 최종 근무직장이 종합병원 또는 요양병원인 경우, 선호하는 근무형태가 3교대제인 경우가 각 기준집단에 비해 높은 것으로 나타났다. 또한 '간호직무 자체에 대한 불만족'의 사직이유인 경우로 나타났다. 이에 재취업을 활성화하기 위해 다양한 형태의 근무방식 적용과 노동강도를 고려한 인력 배치와 간호전문직 사명감을 높일 수 있는 교육 프로그램을 제안한다.
