Practical seismic assessment of unreinforced masonry historical buildings

  • Rehabilitation of historical unreinforced masonry (URM) buildings is a priority in many parts of the world, since those buildings are a living part of history and a testament of human achievement of the era of their construction. Many of these buildings are still operational; comprising brittle materials with no reinforcements, with spatially distributed mass and stiffness, they are not encompassed by current seismic assessment procedures that have been developed for other structural types. To facilitate the difficult task of selecting a proper rehabilitation strategy - often restricted by international treaties for non-invasiveness and reversibility of the intervention - and given the practical requirements for the buildings' intended reuse, this paper presents a practical procedure for assessment of seismic demands of URM buildings - mainly historical constructions that lack a well-defined diaphragm action. A key ingredient of the method is approximation of the spatial shape of lateral translation, ${\Phi}$, that the building assumes when subjected to a uniform field of lateral acceleration. Using ${\Phi}$ as a 3-D shape function, the dynamic response of the system is evaluated, using the concepts of SDOF approximation of continuous systems. This enables determination of the envelope of the developed deformations and the tendency for deformation and damage localization throughout the examined building for a given design earthquake scenario. Deformation demands are specified in terms of relative drift ratios referring to the in-plane and the out-of-plane seismic response of the building's structural elements. Drift ratio demands are compared with drift capacities associated with predefined performance limits. The accuracy of the introduced procedure is evaluated through (a) comparison of the response profiles with those obtained from detailed time-history dynamic analysis using a suite of ten strong ground motion records, five of which with near-field characteristics, and (b) evaluation of the performance assessment results with observations reported in reconnaissance reports of the field performance of two neoclassical torsionally-sensitive historical buildings, located in Thessaloniki, Greece, which survived a major earthquake in the past.

Crack location in beams by data fusion of fractal dimension features of laser-measured operating deflection shapes

  • The objective of this study is to develop a reliable method for locating cracks in a beam using data fusion of fractal dimension features of operating deflection shapes. The Katz's fractal dimension curve of an operating deflection shape is used as a basic feature of damage. Like most available damage features, the Katz's fractal dimension curve has a notable limitation in characterizing damage: it is unresponsive to damage near the nodes of structural deformation responses, e.g., operating deflection shapes. To address this limitation, data fusion of Katz's fractal dimension curves of various operating deflection shapes is used to create a sophisticated fractal damage feature, the 'overall Katz's fractal dimension curve'. This overall Katz's fractal dimension curve has the distinctive capability of overcoming the nodal effect of operating deflection shapes so that it maximizes responsiveness to damage and reliability of damage localization. The method is applied to the detection of damage in numerical and experimental cases of cantilever beams with single/multiple cracks, with high-resolution operating deflection shapes acquired by a scanning laser vibrometer. Results show that the overall Katz's fractal dimension curve can locate single/multiple cracks in beams with significantly improved accuracy and reliability in comparison to the existing method. Data fusion of fractal dimension features of operating deflection shapes provides a viable strategy for identifying damage in beam-type structures, with robustness against node effects.

고유진동수 이용 손상추정법과 모드형상 이용 손상추정법에 의한 PSC 보의 비파괴 손상검색 (Nondestructive Damage Detection in PSC Beams : Frequency-Based Method Versus Mode-Shape-Based Method)

  • PSC 보의 비파괴 손상검색을 위한 고유진동수 이용 손상추정법과 모드형상 이용 손상추정법을 제시하였다. 먼저, 고유진동수의 변화를 사용하여 손상의 위치를 예측하는 알고리즘과 고유진동수 1차 섭동 이론에 근거하여 균열크기를 예측하는 알고리즘을 요약하였다 다음으로, 모드형상의 변화로부터 모드민감도의 변화를 감지하고 이를 통해 손상의 위치와 크기를 추정하는 손상지수 알고리즘을 요약하였다. PSC 보의 유한요소모델을 사용하는 수치실험을 통해 고유 진동수 이용 손상추정법과 모드형상 이용 손상추정 법의 정확성을 검증하였다. 분석결과 두 방법 모두 실험 대상 구조에 도입된 균열의 위치를 정확하게 예측하였으며 균열의 크기를 비교적 근사하게 예측하였다.

Regulated Expression of Nebulin by Transfection of Green Fluorescent Protein-Tagged Nebulin Fragments in Cultured Chicken Myoblast

  • Nebulin is an approximately 700 kDa filamentous protein in vertebrate skeletal muscle. It binds to the Z line and also binds side-by-side to the entire thin actin filament in a sarcomere. The correlation of nebulin size with thin filament length have led to the suggestion that nebulin acts as a molecular ruler for the length of thin filaments. The C-terminal part of human nebulin is anchored in the sarcomeric Z-disk and contains an SH3 domain. SH3 domains have been identified in an ever-increasing number of proteins important for a wide range of cellular processes, from signal transduction to cytoskeleton assembly and membrane localization. However, the exact physiological role of SH3 domains remains, in many cases, unclear. To explore the role of nebulin SH3 in the cytoskeletal rearrangement that accompanies myoblast differentiation, we transfected sense and antisense nebulin SH3 domain fused to enhanced green fluorescent protein in myoblast. Cells expressing nebulin SH3 fragment showed decrease of cell-cell adhesion, and cells transfected with antisense nebulin SH3 gene showed a rounded cell morphology and loss of cell-matrix adhesion. No alteration in cell shape and differentiation were observed in control cells expressing enhanced green fluorescent protein. Perturbation of nebulin altered the cell shape and disrupted cell adhesion in myoblast, demonstrating that nebulin can affect cytoskeleton rearrangement.

Free vibrations of a two-cable network inter-supported by cross-links extended to ground

  • Using cross-ties to connect cables together when forming a cable network is regarded as an efficient method of mitigating cable vibrations. Cross-ties have been extended and fixed on bridge decks or towers in some engineering applications. However, the dynamics of this kind of system need to be further studied, and the effects of extending cross-links to bridge decks/towers on the modal response of the system should be assessed in detail. In this paper, a system of two cables connected by an inter-supported cross-link with another lower cross-link extended to the ground is proposed and analyzed. The characteristic equation of the system is derived, and some limiting solutions in closed form of the system are derived. Roots of cable system with special configurations are also discussed, attention being given to the case when the two cables are identical. A predictable mode behavior was found when the stiffness of inter-connection cross-link and the cross-link extended to the ground were the same. The vector of mode energy distribution and the degree of mode localization index are proposed so as to distinguish global and local modes. The change of mode behaviors is further discussed in the case when the two cables are not identical. Effects of cross-link stiffness, cross-link location, mass-tension ratio, cable length ratio and frequency ratio on $1^{st}$ mode frequency and mode shape are addressed.

사용자 참여형 연구 기반의 한국형 경기용 핸드사이클 개발과 사용성평가 - 국가대표 대상으로 - (A User Participatory Study on the Development of Korean Road Racing Hand Cycle and Usability Assessment: Targeting on National Players)

  • 본 연구의 배경 및 목적은 한국형 경기용 핸드사이클 국산화를 통해 국내 장애인의 스포츠 활성화에 기여하는 것이다. 현재 국내에서 생산하는 핸드사이클은 전무하며 모든 선수들이 외국에서 제작한 제품을 사용하고 있다. 외산제품의 경우 외국인 선수들의 체형에 맞게 제작이 되었기 때문에 국내 선수들이 사용할 경우 자신의 체형에 맞추기 위해 부품을 추가적으로 구성하여 사용하는 경우가 많다. 이는 비용적인 측면에서 부담을 가중할 뿐만 아니라 선수들의 경기력을 저하시키는 원인 중 하나이다. 이러한 문제점을 보완하기 위해 이전 연구에서 한국인 체형을 고려한 경기용 핸드사이클 개발을 진행하였고 개발된 시제품의 성능평가를 위해 외산제품과 비교 사용성평가를 실시하였다. 이렇게 제작된 경기용 핸드사이클 시제품의 정량적, 정성적 평가 결과를 분석하고 개선 사항을 도출하여 국내선수들의 체형을 고려하면서 경기력을 향상시킬 수 있는 한국형 경기용 핸드사이클 개발을 진행하였다. 본 연구는 1차 시제품에서 도출한 문제점을 토대로 크랭크와 공기흡입부 및 배출부, 뒷 축 흔들림 방지장치 등을 추가적으로 구성하였고 개발된 제품의 사용성을 평가하기 위해 현재 핸드사이클 국가대표 상비군 선수들이 사용하는 외산제품과 비교 사용성평가를 실시하였다. 사용성을 평가하기 위해 효과성, 효율성, 만족도 영역에서 결과를 측정하였고 효과성을 제외한 효율성, 만족도에서 연구를 통해 개발된 시제품이 외산제품보다 높은 것으로 평가되었다. 본 연구를 통해 외국산 핸드사이클의 수입을 재치할 수 있는 저가의 핸드사이클을 상용화하고 외국상품의 수입대체 효과와 수출로 인한 국제 경쟁력 증진에 기여할 것으로 여겨진다.

Bearing Estimation of Narrow Band Acoustic Signals Using Cardioid Beamforming Algorithm in Shallow Water

  • This paper suggests the Cardioid beamforming algorithm of the doublet sensors employing DIFAR (directional frequency analysis and recording) sensor signals in the frequency domain. The algorithm enables target bearing estimation using the signals from directional sensors. The algorithm verifies its applicability by successfully estimating bearings of a target projecting ten narrow-band signals in shallow water. The estimated bearings agree very well with those from GPS (global positioning system) data. Assuming the bearings from GPS data to be real values, the estimation errors are analyzed statistically. The histogram of estimation errors in each frequency have Gaussian shape, the mean and standard deviation dropping in the ranges -1.1°∼ 6.7°and 13.3∼43.6°, respectively. Estimation errors are caused by SNR (signal to noise ratio) degradation due to propagation loss between the source and receiver, daily fluctuating geo-magnetic fields, and non-stationary background noises. If multiple DIFAR systems are employed, in addition to bearing, range information could be estimated and finally localization or tracking of a target is possible.

Improvement of Antigen Blotting in a Tissue Blot Immunobinding Assay for the Detection of Two Chili Pepper Viruses

  • The tissue blot immunobinding assay (TBIA) is widely used for the detection and localization of plant viruses in various plant tissues. The basic experimental procedures of TBIA sampling and blotting were simplified using commercially available micropipette tips. This method was termed the ring-blot immunobinding assay (R-BIA), as the blot on the membrane forms a ring shape. The detection efficacy of R-BIA was tested for two chili pepper viruses, pepper mild mottle tobamovirus (PMMoV) and pepper mottle potyvirus (PepMoV), following the optimized serological procedures of TBIA (length of the incubation period and BSA concentration, and primary and secondary antibodies). Sensitivity of the R-BIA was about 1 ng/ml of purified PMMoV in pepper leaf sap from a healthy pepper plant. R-BIA also showed high specificity in the detection of PMMoV and PepMoV. Moreover, the modified sampling and blotting procedures were simpler and more reliable than other TBIA methods (such as whole-leaf blotting and crushed-leaf blotting), suggesting that the R-BIA may be used for medium- to large-scale detection of plant viruses in laboratories with minimal facilities.

The Localization of Cytokeratin 19 and Vimentin in Sprague Dawley Albino Rat Skin Tissue

  • Cytokeratin 19 (CK19) expressed in epidermis of skin, bulge region of hair follicle, outermost layer of outer root sheath and proximal and distal to bulge. Vimentin is a fibrous protein that localized in cytoplasm of fibroblast and forms cytoskeleton to maintain shape of cell and nucleus. In this study, CK19 and vimentin in skin were confirmed with light, fluorescence and transmission electron microscope. As a result, CK19 was localized epidermis, hair follicles, outer root sheath and nucleus of Merkel's cell. However, vimentin was localized some epidermis, dermis, hypodermis and nucleus of Merkel's cell. The role of CK19 is self-renewal and homeostasis in skin. Also, hair follicle regeneration and hair growth is known to be related. It is supposed that required of structural proteins that make up cytoskeleton is increased. Thereby, expression of CK19 is increased. It is considered that vimentin localized in order to stabilize structure of cell and cytoskeleton of fibroblasts. Also, CK19 and vimentin present in nuclei of Merkel's cell, and to act as a fibrous protein that make up end of a nerve fiber present in Merkel's cell and paracrine function of Merkel's cell.

개의 종괴형 간세포암종에 대한 진단 영상 소견에 대한 고찰 (Diagnostic Imaging of Massive Hepatocellular Carcinoma in 5 Dogs)

  • Hepatocellular carcinoma (HCC) is a primary hepatic neoplasms in dogs. Three types of HCC such as massive, nodular and diffuse form were reported. Massive HCC is most common and has relatively better prognosis than other forms because this type of HCC can be removed surgically and has low frequency of relapse or metastasis. Diagnostic image can provide useful information for shape and internal structure of the hepatic mass as well as the location and adhesion or invasiveness of the mass to establish surgical plan to remove the mass safely. In this study, we investigated diagnostic features of massive HCC in 5 dogs. Radiography showed soft tissue mass in cranial abdomen in 3 dogs. On ultrasonography, all dogs had a solitary hepatic mass with mixed echo pattern with anechoic cysts, which represented necrosis and hemorrhage. The radiographic and ultrasonographic findings of affected hepatic lobe were compared with the macroscopic findings through laparotomy. Computed tomography was performed to check metastasis and figure out the origin of the mass in two dogs. Ultrasonography is useful and relatively sensitive examination for diagnosis and planning for surgery in canine massive HCC.
