Genetic Variation in Growth and Body Dimensions of Jersey and Limousin Cross Cattle. 1. Pre- Weaning Performance

  • During a 5-yr period, 1994-1998, pre-weaning and weaning data were collected on 591 calves produced by mating either straightbred Jersey, straightbred Limousin or $F_1$ (Limousin${\times}$Jersey) bulls to mature purebred Jersey or Limousin cows. Traits recorded included birth and weaning weight, height, length, girth, fat depth and a measure of muscle (ratio of stifle to hip width expressed as a percentage). All traits were analyzed assuming a model with sire and dam random effects that included effects of year and date of birth, sex, breed and year${\times}$sex interaction. Main effects were generally significant with few exceptions. Direct genetic effects were large for weight, height, girth and muscle with a breed trend from purebred Jersey (small) to purebred Limousin (large). At weaning, the maternal effect of the Jersey dam was positive for weight (10.9${\pm}$4.9 kg), girth (3.7${\pm}$1.0 cm) and muscle (6.0${\pm}$0.9%). Heterosis was highly significant and positive only for fat depth (1.5${\pm}$0.2 mm) with the $F_1$ progeny being the fattest, followed by the backcrosses, then purebred Jersey and purebred Limousin. Also, significant (p<0.001) but negative heterosis was observed for weight, girth and muscle. The change in ranking for fat depth relative to other traits is a reflection of the large heterotic effects relative to direct effects on fat depth. Epistatic effects were not significant on any trait at birth or weaning. This study has indicated the possibility of exploiting the positive heterotic and maternal effects for fat depth and muscularity to meet specific meat quality and quantity demand by consumers.

  • The occurrence of Anagyrus kamali, a solitary endoparasite of the mealybug, Maconellicoccus hirsutus has been reported for the first time from India. The parasitoid was fecund to parasitizing the field population of mealybug to the tune of 10.37 to 42.70% in different months. A comprehensive study on the development of the parasitoid on different stages of mealybug indicated that the parasitoid was able to complete its development in all the stages. Higher parasitism (67.48-78.08%) and more female progeny were observed when 3rd instar nymphs and adult female of the mealybug were exposed to the parasitoid. The biology of A. kamali studied at 25${\pm}1^{\circ}C$ temperature and 60${\pm}$$\pm$1.12 days. The duration of egg, larva (3 instars) and pupa were 2.67${\pm}$${\pm}$${\pm}$${\pm}$

Artificial gametes from stem cells

  • The generation of artificial gametes is a real challenge for the scientific community today. In vitro development of human eggs and sperm will pave the way for the understanding of the complex process of human gametogenesis and will provide with human gametes for the study of infertility and the onset of some inherited disorders. However, the great promise of artificial gametes resides in their future application on reproductive treatments for all these people wishing to have genetically related children and for which gamete donation is now their unique option of parenthood. This is the case of infertile patients devoid of suitable gametes, same sex couples, singles and those fertile couples in a high risk of transmitting serious diseases to their progeny. In the search of the best method to obtain artificial gametes, many researchers have successfully obtained human germ cell-like cells from stem cells at different stages of differentiation. In the near future, this field will evolve to new methods providing not only viable but also functional and safe artificial germ cells. These artificial sperm and eggs should be able to recapitulate all the genetic and epigenetic processes needed for the correct gametogenesis, fertilization and embryogenesis leading to the birth of a healthy and fertile newborn.

  • The life history and development of Tetrastichus sp. parasitizing fallwebworm pupa in Korea were studied. The mean lifetime fecundity was 94.7 with a maximum of 125, and the portion of female progeny averaged 90.3%. A male and a female lived for an average of 19.8${\pm}$4.5 and 21.7${\pm}$4.2 days, respectively. The duration of development for each stage was as follows: egg, 2 d; the 1st instar larva, 2${\pm}$1 d; the 2nd instar larva, 2${\pm}$1 d: the 3rd instar larva, 3${\pm}$1 d; the 4th instar larva, 6${\pm}$2 d; and pupa, 8${\pm}$2 d. Total developmental duration from hatching of the larva to adult emergence required 21.1${\pm}$3.7 d, and females took 1-2 days more than males in the incubator (28$^{\circ}C$, 70% RH, and 16:8 LD).

  • Life table studies were conducted in the laboratory for the eulophid gregarious pupal parasitoid, Tetrastichus sp., on Hyphantria cunea Drury at a constant temperature of 28.2$\pm$2$^{\circ}C$, 50-60% RH to evaluate their impact on the host and their potential biological control. Development of immature stage took 20.1$\pm$2.7 d: adult females lived for 23.2$\pm$2.2 (range, 16-27)d and produced a mean of 53.6$\pm$26.6 adult progeny per female, with a sex ratio of 1: 9.5 (M:F). The intrinsic rate of natural increase (r$_{m}$) was 0.178/ female/day; the net reproductive rate (R$_{o}$), 46.74; the capacity for increase (r$_{c}$) 0.177; the finite rate of increase (λ) 1.19/female/day; thus each female contributed 46.74 individuals to the population in a mean generation time of 21.6 d. Biological factors in determining the life history trait variation of the parasitoid were discussed.d.d.d.

  • XY 암컷에서 유도된 자성발생성 2배체 수컷 및 이의 성전환 암컷으로 부터 YY수컷(초수컷) 및 ${\Delta}YY$ 암컷(초암컷)을 선발하기 위하여 각각 정상 암컷 및 정상 수컷과 교배하여 자손 검정을 실시하였다. 교배가 이루어진 자성발생성 이배체 수컷 7마리 중 3마리가 YY 수컷으로 판정되었으며, 이들 자손의 수컷율은 $93.3\%\~100.0\%$로서 정상 교배에서 예상되는 성비인 1 : 1에 대한 $X^2$ test 결과 매우 유의하게 나타났다(P<0.01 또는 P<0.001). 자성발생성 2배체 성전환 암컷의 경우 교배가 이루어진 3마리 중 2마리가 ${\Delta}YY$ 암컷으로 판정되었으며, 이들 자손의 수컷율은 각각 $92.9\%$$86.2\%$ 그리고 $87.5\%$$95.0\%$로서 1 : 1 성비에 대해 매우 유의하였다(P<0.005). YY 수컷과 ${\Delta}YY$ 암컷간의 교배에서 전수컷 자손만이 생산되었다.

한국에서 열대거세미나방 알기생벌 Telenomus remus Nixon (가칭: 밤나방검정알벌)[검정알벌과]의 첫 보고 및 생물적 특성 연구 (First report of Telenomus remus Nixon(Scelionidae), an egg parasitoid of Spodoptera frugiperda(J.E. Smith)(Noctuidae) in Korea and its biological characteristics)

  • 본 연구에서는 국내 옥수수 포장에서 채집한 열대거세미나방 알덩어리로부터 우화한 알기생벌을 형태 및 분자동정하여 검정알벌과(Scelionidae)의 Telenomus remus Nixon (1937) [(가칭)밤나방검정알벌]로 최초로 확인되었다. T. remus 성충의 수명 및 발육기간(알-성충까지)은 기주 종류 및 성별에 따른 차이가 없었다. T. remus 암컷 성충은 우화 후 바로 산란하고, 하루에 1~3개에서부터 최고 37개까지 산란하였으며, 우화 후 1~2일째 가장 많이 산란하였다. 총 산란수는 기주로 열대거세미나방 알을 사용하였을 경우 평균 118.4 (7.0~352.0)개, 담배거세미나방의 알에서는 평균 164.9 (5.0~372.0)개로 담배거세미나방 알을 사용하였을 때 산란수가 많았으나, 유의미한 차이는 없었다. T. remus의 산란 기주 선호도는 담배거세미나방>파밤나방>열대거세미나방 순으로 높았다. 기주 알의 나이에 따라 T. remus의 선호도가 달랐는데, 1~2일 된 알을 선호하였다. T. remus 자손의 암수 비율은 기주와 상관없이 암컷의 비율이 수컷보다 높았으며, 산란 초기에는 암컷 산자의 비율이 높다가 나이가 들수록 수컷 산자의 비율이 현저하게 높았다. 교미하지 않은 T. remus 암컷이 산란하여 부화한 경우, 100% 수컷 성충으로 arrhenotoky type의 단위생식을 보여주었으며, 교미하여 산란한 경우, 암수의 비율은 8.0 : 2.0로 암컷의 비율이 높았다. T. remus는 중복기생자(gregarious egg parasitoid)가 아닌 단독기생자(solitary egg parasitoid)로 판단된다. 본 연구에서 보고한 T. remus의 생물적 특성에 관한 연구 결과는 실험실 조건에서 대량생산을 위한 정보로 활용하거나 생물적방제 프로그램을 개발할 때 활용될 수 있을 것이다.

성 감별 유전자를 도입한 다산계계통 신품종 육종에 관한 연구 I. 반생유전계통 조성 (Breeding of New Synthetic Egg Production Line in Domestic Chicken by Intlroducing Sex Linked Gene. I. Production of the Autosexing Breed)

  • 본 연구는 반성은색유전자(S:silver gene)를 도입하여 성 감별용 산란종계 계통으로 육성하여 이를 모계통으로 이용함으로서 깃털에 의한 자웅감별이 가능할 뿐만 아니라 잡종강세를 이용한 우수실용계를 작출하고자 수행되었다. 암수 감별용 계통 조성을 위하여서는 현재 시판되고 있는 백색 실용계 수컷(Z$^{s}$ Z$^{s}$ ) 및 갈색 실용계 암컷(Z$^{s}$ W)을 이용하여 이들 중 우수개체를 선발하여 교잡시킴으로서 생산된 개체중 유전적 조성이 은색유전자를 동형(homo)으로 가진 개체들을 모계통으로 육성하고, 부계통 조성을 위하여서는 갈색유전자를 동형으로 가진 개체를 선발 육성하므로서 감별의 원종계를 작출하였다. 이들 조성된 계통을 이용하여 생산능력이 우수한 계통으로 육종하기 위하여 폐쇄군 육종방법(clossed flock breeding)을 사용하였다. 합성종 계계의 산란능력 검정 결과 60주령까지의 산란기록에 근거한 주요 경제능력은 초산 일령 161일, 60주령시 산난수 219개, 산난율 84% 평균난중 61g으로 외국에서 수입되는 우수종계에 버금가는 성적을 나타내었다. 또한 깃털에 의한 자웅감별이 가능할 뿐만 아니라 산난능력도 우수한 실용성 있는 암수 감별용 합성종을 육성 할 것으로 판단된다. 합성종 계통의 유전분석 결과 이들의 유전모수가 정상집단의 유전모수와 유사한 추정치를 보임으로서 특정계통 조성을 위한 선발에 기인된 유전변이의 변화는 거의 없음을 시사한다.

Detection of Mendelian and Parent-of-origin Quantitative Trait Loci for Meat Quality in a Cross between Korean Native Pig and Landrace

  • This study was conducted to detect quantitative trait loci (QTL) affecting meat quality in an $F_2$ reference population of Korean native pig and Landrace crossbreds. The three-generation mapping population was generated with 411 progeny from 38 $F_2$ full-sib families, and 133 genetic markers were used to produce a sex-average map of the 17 autosomes. The data set was analyzed using least squares Mendelian and parent-of-origin interval-mapping models. Lack-of-fit tests between models were used to characterize the QTL for mode of gene expressions. A total of 10 (32) QTL were detected at the 5% genome (chromosome)-wise level for the analyzed traits. Of the 42 QTL detected, 13 QTL were classified as Mendelian, 10 as paternal, 14 as maternal, and 5 as partial expressed QTL, respectively. Among the QTL detected at 5% genome-wise level, four QTL had Mendelian mode of inheritance on SSCs 5, 10, 12, and 13 for cooking loss, drip loss, crude lipid and crude protein, respectively; two QTL maternal inheritance for pH at 24-h and shear force on SSC11; three QTL paternal inheritance for CIE b and Hunter b on SSC9 and for cooking loss on SSC15; and one QTL partial expression for crude ash on SSC13, respectively. Most of the Mendelian QTL (9 of 13) had a dominant mode of gene action, suggesting potential utilization of heterosis for genetic improvement of meat quality within the cross population via marker-assisted selection.

A total of 8 (39) QTL were detected at the 5% genome (chromosome)-wise level for the 17 analyzed traits. Of the 47 QTL detected, 21 QTL were classified as Mendelian expressed, 13 QTL as paternally expressed, 6 QTL as maternally expressed, and 7 QTL as partially expressed. Of the detected QTL at 5% genome-wise level, two QTL had Mendelian mode of inheritance on SSC6 and SSC9 for backfat thickness and bone weight, respectively, two QTL were maternally expressed for leather weight and front leg weight on SSC6 and SSC12, respectively, one QTL was paternally expressed for birth weight on SSC4, and three QTL were partially expressed for hot carcass weight and rear leg weight on SSC6, and bone weight on SSC13. Many of the Mendelian QTL had a dominant (complete or overdominant) mode of gene action, and only a few of the QTL were primarily additive, which reflects that heterosis for growth is appreciable in a cross between Korean native pig and Landrace. Our results indicate that alternate breed alleles of growth and body composition QTL are segregating between the two breeds, which could be utilized for genetic improvement of growth via marker-assisted selection.
