Sterols of sewage indicators in marine sediments of Jinhae Bay, Korea

  • The ${\Delta}^5$ sterols were measured in be sediments of Jinhae Bay surrounded by industrial cities to assess the sewage-derived contamination. The total concentrations of total sterols ranged from 2.03 to 19.56 ${\mu}g/g$ dry wt. The principal sterol was cholesterol with a contribution of more than 50% to total sterols. Coprostanol, providing an indication of long-term sewage loads, was found in all sediment samples and the concentrations were 0.03-3.86 ${\mu}g/g$ dry wt, accounting for 1-26% to total sterols. The cluster analysis of sampling stations indicated that the sewage-derived contamination was localized in inner Masan Bay.

  • Surface sediments and seawater were sampled at Masan and Haengam Bays of Korea, to evaluate contamination by sewage-derived organic matter using fecal sterols, Six stream-water samples into Masan and Haengam Bays were also sampled. Total concentrations of eight sterols (coprostanol, epichloestanol, epicoprostanol, cholesterol, cholestanol, brassicasterol, stigmasterol and $\beta-sitosterol$) were in the range of $1,274\~4,768{\mu}g/g$ dry weight in suspended particulate from the stream-water, $292\~2,244{\mu}g/g$ dry weight in suspended particulate from the seawater and $4.5\~27.2{\mu}g/g$ dry weight in the sediments. Although sterol compositions in sediments, seawater and stream-water were different, cholesterol was the predominant sterol in all samples. The proportion and concentration of coprostanol, a sewage tracer, in stream-water was much higher than those in sediment and seawater. The sterol levels including coprostanol in the sediments and seawater were higher at inner basin than at the outer bay. Some molecular indices and multivariate statistical analysis were used to assess the origin of these sterols and sewage contamination in the study area. The sterol composition patterns in stream-water were mainly associated with contamination by sewage-derived organic matters and those of seawater were associated with the activities of marine-originated organisms. Sterol levels in the sediments were both from the sewage input through stream-water and the marine- originated organisms. This survey suggests that the main source/route of sewage-derived organic matters in Masan Bay is the input of stream-water into the bay.

  • Organophosphorus pesticides (OPPs) contents in the sewage sludge derived from the residential and industrial areas were determined in order to characterize contemporary organic contamination levels as a part of the ongoing development of the environmentally sound sewage sludge management strategy in Korea. OPPs were extracted from freeze-dried sludges in a ultrasonic extractor. The extracts were cleaned-up by florisil column and subsequently fed into gas chromatograph/nitrogen phosphorus detector (GC/NPD) for determining OPP contents. Diazinon, dimethoate, disulfoton, EPN, malathion, methyl parathion, parathion, phorate and sulfotep were present in the appreciable amount in the domestic sewage sludges.   • Surface sediments (0$\~$4 cm) were sampled at 40 stations from Ulsan Bay and its adjacent areas of Korea, to evaluate the contamination by sewage-derived organic matters using fecal sterols. Some sterols were quantified by gas chromatography coupled to mass spectrometry (GC/MS). Total concentrations of eight sterols (co­prostanol, cholesterol, cholestanol, epicholestanol, epicoprostanol, ${\beta}-sitosterol,$ brassicasterol, and stigmasterol) in the sediments from Ulsan Bay and its vicinity varied from 929 to 23,444 ng/g dry weight. The most predom­inant sterols were cholesterol and coprostanol, accounting for $33\~72{\%}$ of total sterols. The concentration of coprostanol known as the indicator of human feces ranged from 141 to 8,257 ng/g dry weight. In particular, the coprostanol concentrations in the sediments from Jansaengpo Harbor and Taehwa River exceeded the value of 1,000 ng/g dry weight, suggesting that these areas could be considered as the hot-spot zones by municipal sewage contamination. Some molecular indices and multivariate data analysis were used to assess the origin of these sterols in the sediments. Results showed that the major routes of sewage contamination in Ulsan Bay and its vicinity were the input through Jansaengpo Harbor and Taehwa River.

  • Eight fecal sterols were analyzed in surface sediments collected from shellfish farming areas in Yeoja Bay and Gangjin Bay, Korea, to evaluate sewage-derived fecal pollution. The concentrations of coprostanol, a good marker of sewage-derived organic contamination, in sediments were in the range of 10-530 ng/g-dry in Yeoja Bay, and 10-190 ng/g-dry in Gangjin Bay. Coprostanol levels were markedly higher in the inner bay than in the outer bay. These levels were lower than those reported in urbanized bays in Korea, however, they were comparable to levels in other shellfish farming areas including Gamak Bay. A multivariate analysis of the ratios of other sterols suggested that the sterols originated from sewage and plankton/benthos. Sewage was the dominant source at stations located close to the river mouth and wastewater treatment plant (WWTP) outfalls, and plankton/benthos was the primary source in the outer bay. These results suggest that management of point sources, e.g., WWTP as well as non-point sources, e.g., riverine inputs is important for improving the water quality in Yeoja Bay and Gangjin Bay.

퇴적물내 Coprostanol을 이용한 진해만의 도시하수오염 평가 (Evaluation of Sewage Pollution by Coprostanol in the Sediments from Jinhae Bay, Korea)

  • Surface sediment samples from 31 stations in Jinhae Bay were analyzed to evaluate the pollution by sewage from the spatial distribution of the fecal sterol, coprostanol. The sums of eight sterols (coprostanol, epicholestanol, epicoprostanol, cholesterol, cholestanol, brassicasterol, stigmasterol and $\beta$-sitosterol) were in the range of $2,703-27,154{\cal}ng/{\cal}g$ dry weight. The concentrations of the coprostanol in the sediments, which is a good tracer of sewage-derived organic contamination, were in the range of $76-3,964{\cal}ng/{\cal}g$ dry weight. The levels of coprostanol were much higher (almost one order of magnitude) at stations close to the big cities such as Masan and Jinhae. However, those levels were comparable to or lower than those previously reported in other foreign locations. Some ratios of coprostanol to other sterols and multivariate analysis could provide us with the information on the origin of the sterols being from sewage, plankton, and/or benthos. Those showed sewage dominance at the stations near the cities Masan and Jinhae, plankton dominance at the center of Jinhae Bay and mainly marine fauna origin at the mouth of Jinhae Bay. These results suggest that the contents of sterols and some ratios of these components are quite powerful tracer for the detection of the origin of the organic contaminants in the coastal environments.

서울시 도로변의 빗물받이에서 체취한 하수슬러지의 중금속 오염 평가 (The Assesment of Heavy metal Pollution in Sewage Sludeges from Gully Pots Alongside Some Main Roads in Seoul, Korea)

  • Gully pot is a part of urban darainage system to butter the runoff water fluxes from road to rivers and to minimize environmental pollution by prevending over the surrounding area. A series of studies have been carried out concerning the physicochemical characteristics of the sewage sludge sediments sttling down in such a gully pot in order to over to evaluate the contamination for heavy metals such as pb, Zn, Cu and Cd. The roadside soil and sewage sludge samples from gully pots were characterized by XRD analyses and sequential extraction : Zn 2595.7$\mu$g/g; Cd9.8$\mu$/g; Cu602.5$\mu$g/g; Pb260.0$\mu$g/g),because of a long-term accumulation of vehicle- and industrial-related pollutants. Mean Zn concentration in Yeouido (3873$\mu$g/g) and Junggu(3262$\mu$g/g)areas are 4-5 times higher than those in Dobonggu area, suggesting that Zn may be derived from automobile traffic (including the rubber of automobile tires). The mean concentrations of Cu and Cr are very significantly high in Junggu and Gurogu areas, possibly due to the industrial activities in these areas. The low Pb levels throughtotut the whole study areas in Seoul can be accounted for the use of unleaded gasoline since 1987. Sequential extraction experiments illustrate that a major part of Zn is bound to FII and FIII, representing about 88% of the total Zn concentration. Fraction IV, related to orgnic matter, is mostly significant for Cu accounting for 60% of the total Cu, and FII is next in importance. The main carriers of Pb are the fractions III, II and FIV, and in that order. The behavior of Cd is quite different from other elements (Zn, Cu, Pb), and most of the Cd is associated with FV. Changes in the physicochemical environments (such as acidification) may result in severe environmental pollution of surface water and rivers with respect to heavy metals (especially Zn and Cu).

L-글루타민산 생산균 Brevibacterium lactofermentum의 Bacteriophag에 관한 연구 (Studies on the Bacteriophages of Brevibacterium lactofermentum)

  • Many industrial processes those employ bacteria are subjected to phage infestations. In L-glutamic acid fermentions using acetic acid, the phage infestations of the organisms have been recently recognized. In efforts to elucidate the sources of phage contamination involved in the abnormal fermentation, a series of study was conducted to isolate the phages both from the contents of abnormally fermented tanks and the soil or sewage samples from the surroundings of a fermentation factory, to define major charateristics of the phage isolates, and finally to determine the correlation between the phage isolates and temperate phages originating from the miscellaneous bacterial species isolated from the soil or sewage samples. The results are summarized as follows; 1) All phages were isolated from the irregular fermentation tanks and soil or sewage samples, and they were designated as phage PR-1, PR-2, PR-3, PR-4, PR-5, PR-6, and PR-7, in the order of isolation. These PR-series phages were proved to be highly specific for the variant strains of Br. lactofermentum only, namely, phage PR-1 and PR-2 for Br. lactofermentum No. 468-5 and phage PR-3~PR-7 for Br. lactofemrentum No. 2256. By cross-neutralization test, the 7 phagescould be subdivided into 3 groups, i. e., phage PR-I and PR-2 the first, phage PR-3, PR-4, PR-5, PR-6 the second, and the phage PR-7 the third. 2) The 7 phages were virulent under the experimental conditions. They produced plaques with clear and relatively sharp margins without distinct halo. The mean sizes of plaques were 1.5mm in diameter for phage PR-1 and PR-2, and 1. Omm for phages PR-3~PR-7. Double layer technique modified by Hongo and described by Adams, was applied to assay of the PR-series phages. The factors influencing the plaques were as follows;young age cells of host bacteria cultured for 3-6 hours represented the largest number and size, optimum was pH 7.0, incubation temperature was $30^{\circ}C$, and agar concentration and amount of overlayer medium were 0.6% and 0.2ml, respectively. 3) PR-series phages were stable in 0.05M tris buffer and 0.1M ammonium acetate buffer solution. The addition of $5{\times}10^{-3}M$ magnesium ion effectively increased the stability. Thermostability experiments indicated that PR-series phages were stable at the teinperture between $50^{\circ}{\sim}55^{\circ}C$ in nutrient medium, $45^{\circ}{\sim}50^{\circ}C$ in buffer solution. However, the phages mere completely inactivated at 603C and 65$^{\circ}$C within 10 minutes. The phages were stable at the range of pH6~9 in nutrient medium and of pH 8-9 in buffer solution, respectively. Exposure of the phages to UV for 25, 60 and 100 seconds resulted in the complete loss of infectivily, respectively. 4) Electron microscopy showed that PR-series phage particles exhibited rather similar morphology, differing in the size All of PR-series phages had a multilateral head and had a simple long tiil about three to five times long as compared with head. By the size, phage PR-1 and PR-2, PR-3, PR-4, PR-5, and PR-6 and PR-7 were classified into same groups, respectively. The head and tail size of phage PR-1, PR-5, PR-5(T) and PR-7 were 85nm, 74nm and 235nm and 350mm, and 72nm and 210nm, respectively. 5) Nucleic acids of PR-series phages were double stranded DNA. The G+C contents of phage PR-1, PR-5 and PR-7 were 56.1, 52.9 and 53.7, respectively. The values of G+C contents derived from the $T_m$ were in agreement with the chemically determined values. 6) PR-series phages effectively adsorbed on their host bacteria at the rate of more than 90% during 5 min. K value for phage PR-1, PR-5 and PR-7 were calculated to be $6{\times}10^9 ml$ per minute, respectiveky. The pH of the medium did effect adsorption rate, but both temperature and age of host cells did not. Generally, optimum adsorption condition of phages seemed to be almost same as optimum growth conditions of host bacteria. 7) In one-step growth experiments, the latent periods at $30^{\circ}C$ for PR-1, and PR-7 were about 70, 50 and 55 min, respectively. The corresponding average burst size was 200, 70 and 90, respectively. Lpsis period according to the multiplicity of infection and a phage series. In case of m. o. i. 100, strain No. 2256 (PR-5) and No. 468-5(PR-1) failed to grow and turbidity decreased after 50 and 70min, respectively. 8) In the lysate of a plaque purified phage PR-5 infected bacteria, there observed 2 types ofphage particles, i. e., phage PR-5 and PR-5 (T) of similar morphology but differing at the length of phage tail, and phage tail like particles. The phage taillike particles could be divided into 4 types by the length. Induction experiments of Br. lactofermentum with UV irradiation, mitomycin C or bacitracin treatment produced neither phage PR-5 (T) or phage tail-like particles. 9) No lysis occured when the growth of 7 strains of miscellaneous bacteria, isolated from soil and sewage samples, were inoculated with either phage PR-5 (T) or phage tail-like particles the inoculation of phage PR-5 pellet resulted in the growth inhibition of the orgainsms in the spot test. The lysates obtained from 3 miscellaneous soil derived bacteria following mitomycin C treatment the growth of Br. lactofermentum, but did not lyze the bacterium.
