Sesamin and Sesamol Contents of Korean Sesame Oils Determined by HPLC Method and Villavecchia-suarez Color Reaction of the Oils (국내 산지별 참기름의 HPLC에 의한 Sesamin, Sesamol의 함양과 Villavecchia-suarez 정색반응)

  • In the present study the sesamin and sesamol contents of the sesame extracts prepared from nine provinces in Korea were determined HPLC method. A comparative test was also carried out using the Villavecchia-suarez test, the red colored reaction for the sesamol and sesamol derivatives. The contents of sesamin and sesamol of the sesame oils from each area by the HPLC method were 0.57-0.78% and 0.010-0.023%, respectively, and the paralled results were obtained by the Villavecchia-suarez test and the HPLC method. The average content of the sesamin was $0.68{\pm}0.074%$ by the HPLC method and the average absorbance of the Villavecchia-suarez test was $0.56{\pm}0.034$. The contents of sesamol from sesame oil by the HPLC method and the Villavecchia-suarez test were so low that it was not possible to correlate with the sesamin contents. The contents of sesamol from the sesame oil produced in Kyeong-gi and Jeon-nam provinces were $0.010{\pm}0.002%$ and $0.023{\pm}0.004%$, respectively.

Effects of Lignan Compound of Sesame on LPS-induced Nitric Oxide Generation in Murine Macrophage RAW 264.7 Cells (참깨의 리그난 화합물의 항염증 효과)

  • Sesame (Sesamum indicum L.), one of the oldest oilseed crops, has been known to posses antioxidative and inflammatory effects. This seed contains lignan compounds such as sesamin, sesamol, sesaminol, sesaminol diglucosides (SDG), and sesaminol triglucosides (STG). Sesamin, a major lignan in sesame, displayed several biological activities including a protective effects against oxidative damage in the skin. In the present study, we investigated the effect of sesamin, sesamol, sesaminol, SDG, and STG, on nitric oxide (NO) induction and inducible nitric oxide synthane (iNOS) and cyclooxygenases-2 (COX-2) expression in lipopolysaccharides (LPS)-treated RAW 264.7 cells. The results showed that sesamol and sesaminol significantly inhibited NO generation but they were also cytotoxicity however, sesamin effectively inhibited NO production ($IC_{50}: 64{\mu}M$) without my cytotoxic effect in LPS-stimulated macrophage RAW 264.7 cells. In further study, it was founded that sesamin inhibited the expression of inducible nitric oxide synthase but not COX-2 expression. These results suggest that sesamin may be useful for improvements of the inflammatory diseases.

Amino-β-cyclodextrin Complex Assisted Ionization for Labile Sesamins and their Ion-mobility Separation in ESI Q-TOF MS

  • Sesamin, one of the lignans in sesame seed, was a labile compound in MS and it was reported that the protonated molecule of sesamin decomposed easily in ES ionization process and it cannot be detected (G. Yan, et al., Rapid Commun Mass Spectrom. 2007, 21, 3613-3620). To protect labile compounds, an amino-cyclodextrin (NCyD) was added to the sample to promote the host-guest interaction complex in ESI-MS. As a result, sesamin was ionized as the NCyD-sesamin-NCyD (1:2) complex without undesired decomposition, suggesting that the amino-CyDs assist the ionization of the labile molecules capped with CyDs by host-guest interaction and these compounds were ionized without their decomposition, those are like amino-CyD complex-assisted ionization. The amino-CyD complexes of sesamin and sesamolin were also analyzed by their ion-mobility MS.

Varietal Difference of Lignan Contents and Fatty Acids Composition in Korean Sesame Cultivars (국내 참깨 품종의 리그난 함량 및 지방산 조성)

  • Although lignans of sesame seed, sesamolin and sesamin have been known as possessing an antioxidant activity, it is less known about their contents of the sesame cultivated in Korea. Collections of sesame cultivated in Korea were used for studies on their lignans content of the seed and fatty acids composition of the oil. The sesamin content of sesame seed with white-coat were 370.29 mg/100g seed, while that of sesame seed with black-coat were 246.58mg/100g seed. Also, the sesamolin contents of sesame seed were 202.22 mg/100g seed in white-coat cultivars and 132.68 mg/100g seed in black-coat sesames. Hence, the lignan content of white-coat sesame cultivars was significantly hi임or than that of black-coat ones. Korean sesame cultivars also showed considerably higher sesamin content than sesamolin content in seeds. The correlation between sesamin and sesamolin contents was not recognized in Korean sesame cultivars. The stearic acid of white-coat sesame was significantly higher than that of black-coat one (p<0.05).

Comparison of Major Components of Sesame Oil Extracted from Korean and Chinese Sesames (한국산 및 중국산 참깨로부터 착유한 참기름의 주요성분 비교)

  • Korean and Chinese sesames were subjected to microscopic observation and instrumental determination of fatty acid composition and sesamolin/sesamin ratio to obtain basic data for discriminating each other. The overall appearance of both samples was differently observed by stereo microscope (X8). Fatty acid composition of sesame oils, extracted from both samples with different roasting degrees, showed a similar pattern although Chinese samples cointained about 6% higher content of stearic acid and 47% lower content of linolenic acid then Korean ones. The sesamolin/sesamin ratio was remarkably lower(.039) in Chinese samples than Korean (0.67∼0.72). showing a variation depending on producing districts. Roasting degrees of raw sesames little influenced their composition of fatty acid and sesamolin/sesamin ratio. Based on the above results, it is considered that the comparison between domestic and Chinese sesames in view of their stearic and linolenic acid contents and sesamolin/sesamin ratio might be one of the potential criteria in discriminating their production origins.

  • Oxidative stress is believed to play an important role in the development of vascular complications associated with diabetes mellitus. We examined the antioxidative effect of sesamin and sesamolin on the preventing the development of insulin dependent diabetes mellitus using streptozotocin-induced Spraque Dawley diabetic rats. From 48 hours after injection of streptozotocin (60 mg/kg body weight), a portion of diabetic rats were fed with 0.2% sesamin and sesamolin containing diet for 3 weeks. There were significant differences of blood glucose and kidney weight between diabetic ports and control. Sesamin and sesamolin increased glutathione-S-transferase activity in kidney. The concentration of the thiobarbituric acid-reactive substances in the serum, liver, and kidney of diabetic rats administered sesamin and sesamolin decreased significantly as compared with that of the non-treated diabetic group. Dietary sesamin and sesamolin suppressed the oxidative stress in the diabetic rats. These results demonstrated that sesamin and sesamolin are potential and effective antioxidants that can protect the complications associated with diabetes.

Determination of the Adulteration of Sesame Oils Sold in Markets by Gas Chromatography (시중 참기름에 혼입된 이종기름에 관하여)

  • This study primarily attempted to establish the method for the determination of the adulteration in the sesame oil. First of all, extensive experiment was conducted to determine the composition of genuine sesame oil prepared from Korean, Japanese, Taiwanese and Chinese sesame seed. Sesamin and sterols in unsaponfiable matter were examined along with fatty acid in saponifiable fraction by Gc. There was no significant difference in the composition of sesamin and sterols in sesame oils prepared from Korean and foreign seeds. The ranges of sesamin and ${\beta}-sitosterol$ against campesterol were 3.32~5.46 and 2.39~2.99 respectively in all samples. Similiar composition of fatty acids was showed in all pure sesame oils, in which the contents were 8.37~lO.09% palmitic acid, 4.61~5.50% stearic acid, 35.24~39.97% oleic acid, 43.04~49.76% linoleic acid, O.21~O.31% linolenic acid and 0.40~O.69% arachidic acid. Among the commercial sesame oils sold in Markets, three sesame oils from Japan revealed low sesamin, high linoleic acid and linolenic acid, and low oleic acid and stearic acid, suggesting the adulteration with soybean oil.

Determination of Sesamin and Sesamolin in Sesame (Sesamum indicum L.) Seeds Using UV Spectrophotometer and HPLC

  • Sesamin and sesamolin, antioxidant lipidsoluble lignan compounds, are abundant in sesame (Sesamum indicum L.) seed oil and provide oxidative stability of oil related to sesame quality. The sesamin and sesamolin contents of 403 sesame land races of Korea were determined by HPLC analysis of methanol extract (HPLC value), and their total lignan content was compared with those by using UV-Vis spectrophotometric analysis (UV method) of methanol (UV-MeOH value) and hexane (UV-Hexane value) extracts. HPLC values of total lignan content were strongly associated with UV-Hexane (r=0.705**) and UV-MeOH (r=0.811**) values. The UV values from both the extracts were 3.8-4.7 times higher than those of HPLC values. Lignan content was overestimated by UV method because total compounds in the mixture solution were quantified by absorbing at the same ultraviolet wavelength as in HPLC method. UV method could more rapidly analyze small amount of sample with higher sensitivity of detection than HPLC method. Average contents of lignans in sesame germplasm evaluated in this study were $2.09{\pm}1.02mg/g$ of sesamin, and $1.65{\pm}0.61mg/g$ of sesamolin, respectively, showing significant variation for lignan components. The results showed that UV method for the determination of sesamin and sesamolin could be practically used as a faster and easier method than HPLC by using the regression equations developed in this study.

Effect of Temperature and Day-length on Antioxidants in Sesame (온도와 일장이 참깨의 항산화성분에 미치는 영향)

  • Sesame cultivars were treated with temperature and daylength to determine the environmental variance of Antioxidant content The experiment was performed in phytotron with 11, 13, 15hours of daylength and 20/15, 25 120, 30/25$^{\circ}C (day / night) of temperature treatment. Varietal differences of antioxidant content were significant by the temperature and daylength treatment. Antioxidant content was incrased at higher temperature(25/2$0^{\circ}C$, 30/25$^{\circ}C) and daylength(13hours) but decreased at lower temperature (20/15$^{\circ}C), short daylength (l1hours) and long day length (15hours) condition. Sesamin content were showed 0.06% difference as maxium by the temperature treatment, while 0.03% by the daylength treatment. Sesamolin content were showed 0.03% difference as maxium by the temperature treatment, while 0.01% by the daylength treatment. The antioxidant content was influenced by the increased temperature than daylength.
