As ciarette smoking adversely affects vitamin C metabolism in humans, smokers need substantially more vitamin C intake than do non-smokers to achieve similar serum vitamin C concentration. To provide the basic information currently available for the determination of vitamin C requirement for Korean smokers, we investigated the differences the serum vitamin C values between smokers(n=53) and non-smokers(n=62) in relation to their intake of the vitamin through diet in 115 male college students, who had not been using vitamin C supplements. Dietary intakes of vitamin C were determined by a 24-hour recall, and serum vitamin C was determined using the 2, 4-dinitrophenylhydrazine method. The mean vitamin C intakes of smokers and non-smokers were consuming less than 75% of the Koeran RDA for vitamin c in their diet. Smokers consumed yellow and green leafy vegetables more often (P=0.02) and fresh fruits less often(P=0.006) than non-smokers. The mean serum vitamin C concentration of smokers consuming the same amounts of vitamine C as non-smokers, 64.3 umol/1, were 20% lower than for non-smokers, 80.1 umol/l(P<0.05). The risk of low serum vitamin C concentration (LoC) among smokers were 3.8% compared with 1.6% in non-smokers, and the odd ratio for LoC risk was 2.43. There were no correlations between dietary and serum vitamin C for smokers and non-smokers. It was concluded that smokers might require at least 20% more vitamin C to reach the same concentration comparable to non-smokers.
For the investigation of the effects of G. lucidum on prevention and attention of hypercholeste-rolemia and atherosclerosis dietary hypercholesterolemic rats were fed with 2.0g% G. lucidum extracts for 4 weeks. And then concentrations of total cholesterol triglyceride phospholipid in serum and liver and those of HDL-cholesterol lipid peroxide glucose in serum were analyzed. Concentration of total cholesterol in serum was the lowest in the control group(basal diet+ wa-ter) and HDL-cholesterol in serum were significantly higher in groups of control 2(hyperchole-sterolemic ratslongrightarrowbasal+water) and 4(hypercholesterolemic ratslongrightarrowbasal+G.l. extract) The concentration of triglyceride in serum were signficantly lower in groups 4 and 5(hypercholeste-rolemic ratslongrightarrowcholesterol+G. l. extract) than in the control group. Phospholipid content in serum were not significantly different among all groups. Ttal cholesterol in liver was significa-ntly higher in hypercholesterolemic rats than in the control group and triglyceride concentration were signficantly higher in groups of 3(hypercholesterolemic ratslongrightarrowcholesterol+water) and 5 than others. Phospholipid in liver was significantly higher in group 3 than in the control group but groups of 4 and 5 were lower. As for lipid peroxide in serum was the lowest in group 4 and glucose concentration was lowest in group 5 than in other groups.
Subclinical vitamin $B_{12}$ deficiency is common in the elderly worldwide. We investigated the change of serum vitamin $B_{12}$ concentration with aging and compared anthropometric data and clinical health indicators between normal (${\geq}$ 340 pg/mL) and low (< 340 pg/mL) serum vitamin $B_{12}$ groups in 470 Korean women aged 65 years and over living in a rural area. Serum vitamin $B_{12}$ concentration showed inverse correlation with age (r = -0.0992, p < 0.05). The normal $B_{12}$ group showed significantly (p < 0.05) higher red blood cell count, hemoglobin, and hematocrit compared to the low $B_{12}$ group, however, no difference in mean corpuscular volume was observed between the two groups. The normal $B_{12}$ group showed significantly lower serum homocysteine concentration (p < 0.01) and prevalence of vitamin D (p < 0.01) or folate deficiency (p < 0.001). Bone mineral density (T-score) was significantly higher (p < 0.05) in the normal $B_{12}$ group, compared with that in the low $B_{12}$ group, and showed positive correlation (r = 0.1490, p < 0.01) with serum vitamin $B_{12}$ concentration after adjusting for age, body weight, and body mass index. No differences in anthropometric data, physical activity, and smoking and drinking habits were observed between the two groups. In conclusion, it could be suggested that older female adults with normal serum vitamin $B_{12}$ level would be less anemic and osteoporotic and more resistant to hyperhomocysteinemia associated chronic diseases than those with low serum vitamin $B_{12}$ level.
Four out of 15 dogs were successfully induced diabetes mellitus with intravenous iqiection of 30 mg of streptozotgin and 50 mg of alloxan monohydrate per kilogram body weight and maintained more than 9 weeks without iqiection of insulin or oral hypoglycemic sgent Histopathologicallyi these four dogs have typical diabetic lesions such as degeneration and vacuolation of pancreatic islet cells, and fatty change of liver at necropsy in the end of study. Serum glucose level increased dramatically at 24 hours post-injection but serum fructosamine level increased gradually and reached plateau at 31-41 days post-injection of streptozotocin and alloxan. Serum fructosamine concert%lion correlated very well with serum glucose concentration of preceding 4-7 weeks in experimentally induced diabetic dogs. Our data suggest that serum fructosamine reflects mean glucose concentration of preceding 4-7 weeks in experimentally induced diabetic dogs.
A $3{\times}3$ (Fe$\times$VA) experiment with repeats was designed to study the interaction between iron and vitamin A in broilers. 504 broilers were divided into 9 groups (50% males, 50% femals), each group with 4 repeats. Iron supplemental amount was 0, 30 and 60 mg/kg; Vitamin A supplemental amount was 750, 1,500 and 2,700 IU/kg. Iron concentration in liver, serum, tibia and duodenum and vitamin A concentration in liver and serum were measured, and erythrocyte count was also observed. Results showed with the increase of dietary supplemental iron levels, vitamin A concentration in liver significantly decreased lineally (p<0.05) (0.861, 0.671, 0.639 mg/100 g at the end of 4th week; 0.900, 0.765, 0.739 mg/100 g at the end of 7th week), and vitamin A concentration in serum significantly increased lineally (p<0.05) (82.725, 97.842, $109.475{\mu}g/100mL$ at the end of 4th week; 62.288, 91.900, $95.117{\mu}g/100mL$ at the end of 7th week), meaning iron could promote the mobilization of vitamin A from liver to serum. With the increase of dietary supplemental vitamin A levels, liver iron concentration decreased and serum iron concentration increased, vitamin A could promote the mobilization of iron from liver to blood. Iron concentration in Duodenum and tibia and erythrocyte count increased significantly with higher dietary vitamin A supplementation (p<0.01), vitamin A could promote iron absorption, iron mobilization from liver to target tissues and erythropoiesis. Effects of the interaction between iron and vitamin A on vitamin A concentrations in liver and serum, iron concentration in tibia and erythrocyte count were significant (p<0.05).
We investigated differences between effects of natural- and bucket-suckling methods on basal serum oxytocin (OT) and cortisol concentrations, and the effect of OT concentration on affiliative and investigative behavior of calves to a novel object. Ten Japanese Black calves, balanced with birth order, were allocated evenly to natural-suckling (NS) and bucket suckling (BS) groups. Blood samples were collected at the ages of 1 and 2 months (1 week after weaning) calves, and serum OT and cortisol concentrations were measured using enzyme-linked immunosorbent assay and enzymeimmunoassay tests, respectively. Each calf at the age of 2 months (2 weeks after weaning) was released into an open-field with a calf decoy, and its investigative and affiliative behaviors were recorded for 20 minutes. In 1-month-old calves, the basal serum OT concentration ($25.5{\pm}4.9$ [mean${\pm}$standard deviation, pg/mL]) of NS was significantly higher than that of BS ($16.9{\pm}6.7$) (p<0.05), whereas the basal cortisol concentration ($5.8{\pm}2.5$ [mean${\pm}$standard deviation, ng/mL]) of NS was significantly lower than that in BS ($10.0{\pm}2.8$) (p<0.05). Additionally, a negative correlation was noted between serum OT and cortisol concentrations in 1-month-old calves (p = 0.06). Further, the higher serum OT concentration the calves had at 1 month old, the more investigative the calves were at 2 months old but not affiliative in the open-field with a calf decoy. Thus, we concluded that the natural suckling method from a dam elevates the basal serum OT concentration in calves, and high serum OT concentrations induce investigative behavior and attenuate cortisol concentrations.
The aim of this study was to assess the association between total vitamin A, retinol and carotene intakes and serum concentration of total carotenoids in Korean male smokers and nonsmokers. Food frequency and nutrient intakes of 64 healthy college students were estimated by the 24 hour recall method and food frequency questionnaire and analyzed by smoking status. Serum total cartenoids and lipid profiles were measured in 28 smokers and 36 nonsmokers. The mean total vitamin A, retinol and carotene intakes of smokers were not significantly different from nonsmokers. However, carotene intakes of the smokers who have smoked more than 5 packyears were significantly lower than those of nonsmokers. Smokers consumed green yellow leafy vegetables more often(P=0.005) and fresh fruits less often(P=0.017) than nonsmokers. The serum total carotenoids of smokers consuming the same amount of retinol and carotene as nonsmokers, 1.79 g/ml, were 17% lower than nonsmokers, 2.15 g/ml(P<0.05). The lipid profiles including total cholesterol, LDL cholesterol, VLDL cholesterol and triglycerides of smokers were not significantly different from nonsmokers, however, the concentration of HDL cholesterol of smokers were significantly higher than that of nonsmokers. The serum carotenoids concentration was positively associated with fruit consumption frequency in smokers, and also was positively associated with green yellow leafy vegetable consumption frequency in non smokers. There were no correlations between dietary and serum carotenoids for smokers as well as nonsmokers. It was concluded that smoking could be inversely related to the serum concentration of carotenoids at constant carotene intakes and smokers might require more carotene to reach the same concentration comparable to nonsmokers.
As a simple and economical method for in vitro produced embryos, we have used BSA instead of serum for the production and embryo transfer of Hanwoo in vitro fertilized (IVF) embryos and obtained the following results: 1) When using serum (FBS; fetal bovine serum) or BSA-containing culture media as the initial culture media for immature oocytes, it is regarded as inappropriate to add only BSA to the culture solutions from maturation of the immature oocytes to development stage culture, but serum still needs be added though there is no significant difference in the concentration, with a change from 5% to 10%. 2) The results of culturing IVF embryos after development (4 cell stage) in the Medium199 solutions containing BSA instead of serum (FBS) showed that 0.3% BSA concentration is not optimal and 0.5% or higher BSA concentration has no significant difference among 0.5%, 0.7%, 1% and 2% (p > 0.05). 3) The post-freezing survival ratio after development in 5% FBS-Medium199 showed that 1% BSA concentration of the culture solution is the most suitable in the BSA concentrations of 0.3% (51%), 0.5% (67%), 0.7% (69%), 1% (77%) and 2% (75%). 4) The pregnancy rates of the transplanted fresh(not frozen) blastocyst had no significant concentration dependency (p > 0.5), and the average pregnancy rate was 63.8%. 14% of overweight calves were found among the calves given birth to by the transfer of IVF blastocysts cultured in the serum-added culture solution, but none was found in the experimental groups in which BSA was added instead of serum.
In order to study the relationship between life-style and serum lipids level in young adults, the author measured the concentration of serum total cholesterol (TC), triglyceride (TG) and high density lipoprotein cholesterol (HDL-C) and investigated age, relative weight, weekly alcohol intake and daily cigarette smoking through questionnaire in 310 male workers aged 20 to 39, in Ulsan area. The results obtained were as follows: 1. In the factors being able to influence serum lipids level there was negative correlation between age and alcohol intake, and the positive correlation between age and relative weight, alcohol intake and relative weight, alcohol intake and smoking with the strongest correlation between alcohol intake and smoking. 2. In univariate analysis, mean total cholesterol concentration were significantly different according to age, smoking and relative weight: mean triglyceride concentration were significantly different according to relative weight only; mean HCL-C concentration were significantly different according to alcohol intake alone. 3. In non-drinkers, HDL-C concentration of smokers were significantly lower than that of non-smokers but triglyceride concentration of smokers were significant higher. And in drinkers, total cholesterol concentration of smokers was significantly higher than that of non-smokers. 4. In multiple regression analysis, significant independent variables were relative weight, age and smoking in the total cholesterol concentration, and relative weight, age and alcohol intake in the triglyceride concentration, and alcohol intake, relative weight and smoking in the HDL-C concentration. By these independent variables, total variation in each dependent variable was explained 7.9%, 17.6% and 7.4% respectively.
