Objectives: Given current vaccinations against the hepatitis B virus and changes in dietary lifestyles, the status of liver disorders, especially in the younger population, is different than it was in the past. This study aimed to discover the prevalence of the HBsAg positive and abnormal hepatic enzyme among college students.Methods: The study analyzed the medical examinations of 2,646 college students that were performed at Semyung University from March to September 2013.Results: A total of 11 out of 2,646 students (0.4%) showed as positive for the hepatitis B antigen, while 23.8% of students had the serum HBs antibody. A total of 212 students (8.0%) showed abnormally elevated serum levels of ALT, with significant differences between men and women. The prevalence of the abnormal serum ALT and its average value significantly increased according to an increase of BMI score, especially in male students.Conclusions: This study showed the importance of obesity as a causative factor in liver disorders, especially in the young generation. This finding will be useful for the development of therapeutics using Korean medicine in the future.
Objectives : This study was carried out to research the protective effects of Sagunja-Tang(SA) through in vivo experiments, and tried to investigate the relation between oxidation of liver tissues and deficiency of Qi. Methods : Acute liver injury which initiated from free radical induced by $CCl_4$, were applied to mice and metabolic data were obtained. In order to measure the degree of liver injury, serum level of alanine aminotransferase(AST), aspartate aminotransferase(ALT), creatinine, blood urea nitrogen(BUN), total protein(TP) and glucose were measured. Lipid peroxidation of liver slice was examined by measuring malondialdehyde(MDA), a product of lipid peroxidation. Results : SA had protective effects on $CCl_4$ induced acute liver injury by decreasing serum level of ALT. Kidney injury was induced by injection of $CCl_4$ too, and SA protected kidney injury by decreasing serum level of creatinine and BUN. Conclusions : Through this study, we found that SA have healing effects on liver and kidney injury of $CCl_4$ induced oxidative stress that is similar to deficiency of Qi. And further studies have to be followed to certify the mechanisms.
Protaetia brevitarsis has been utilized as an ingredient of the description for the treatment of patients with chronic hepatic diseases in oriental medicine. This study was attempted to investigate whether Protaetia brevitarsis extract(PBE) protects or modulates the liver injuries induced by carbon tetrachloride or ethanol in Sprageue-Dawley rate. The liver injuries of rats induced by the treatment of carbon tetrachloride or ethanol were manifested by the observation of the significant changes in liver weight, serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities, serum thiobarbituric acid reactive substances (TBARS), and microsomal detocification enzymes(cytochrome P_450), cytochrome b$_{5}$, and cytochrome b$_{5}$ reductase).The effect of PBF on the liver damage induced by the chemicals was evaluated with the extent modulated in change of biochemical parameters above. Exposure to ethanol alone resulted a significant change in the ration of liver per body weight, ALT activity, and microsomal detoxification enzymes (cytochrome P_450, cytochrome b$_{5}$, and cytochrome b$_{5}$ reductase), but did not significantly changes in the levels of serum AST activity and TBARS. Pretreatment coith PBE did not modulate the alteration of the ratio of liver to body weigth, and the activities of serum aminotransferascs (AST. ALT), TBARS, and micro somal detoxification enzyme (cytochrome p_450, cytochrome b$_{5}$,and cytochrome b$_{5}$ reductase. These result suggested that PBE has not appreciable therapeutic effect on carbon tetrachloride or ethanol induced hepatotoxicity.

The antidiabetic effects of Rosa rugosa Radix were investigated in streptozotocine-induced diabetic rats. Research methods and procedure: In the present study, effects of oral administration of Rosa rugosa Radix extract(100, 250, and 500 mg/kg body wt.) for 28 days on the level of serum glucose, total cholosterol, triglycerides, cereatinine, aspartate amino transferase(AST) and alanine amino transferase(ALT) in normal and streotozotocine-induced diabetic rats were evaluated. Oral administrations of the Rosa rugosa Radix extract significantly decreased serum glucose, total cholesterol, triglyceride, AST, and ALT levels, while increased serum insulin and HDL-C in diabetic rats(p<0.05). The hypoglycemic effect of the Rosa rugosa Radix extract was more effective than normal group. It is concluded that the Rosa rugosa Radix must be considered as excellent candidate for future studies on diabetes mellitus.
In this study, the ameliorating effect of soybean embryos on the impact of alcohol consumption was investigated on rat hepatocytes and in reducing total serum cholesterol levels and total serum lipid levels. Liver histology and two clinically important enzyme markers, aspartate aminotransferase (AST) and alanine aminotransferase (ALT), of rats administered with both alcohol and soybean embryo were compared with a control group. The treatment regimen of soybean embryo significantly reduced the serum ALT and AST levels of the subjects, demonstrating the hepato-protective effects of soybean embryo. Electron microscopy indicated that the administration of soybean embryo preserved the important hepatocyte structures and prevented the presence of lipid droplets and secondary lysosomes. Furthermore, total cholesterol and total lipid levels were significantly reduced. These results indicate that treatment with soybean embryo can positively mediate the effects of alcohol on hepatocytes and general liver functions.
The herbicide, paraquat, has caused numerous poisonings in man and farm animals since its interoduction. Althought the lungs are the primary sites of toxicological effect, other organs are also damaged, such as liver, heart, and kidney. The biochemical mechanisms responsible for the toxicity of the herbicide are not clearly understood. The purpose of the present study was to determine the hepatotoxity of paraquat in rats and the effects of glycyrrhizin on the paraquat toxicity. The experimental results are the followings. 1. The values of serum AST, ALT, LDH, ALP, glucose, and cholesterol were significantly increased in the paraquat administered group. 2. The TBA values in serum and liver were also increased in the treated group. 3. The biochemical parameters of serum, such as AST, ALT, LDH and TBA values, were significantly low in the paraquat plus glycyrrhizin group in comparison with that of the paraquat only. The above results suggest that the glycyrrhizin, somehow, detoxify the hepa to toxicity of paraquat in the experimenta-animals.
Background: To investigate tumor inhibition effects and mechanisms of Angelica sinensis and Sophorae flavescentis ait decoction (ASSF) combined with diamine-dichloroplatinum (DDP). Materials and Methods: Bodyweight, tumor inhibition rate and q value were calculated for single ASSF or ASSF combined with DDP on H22 carcinoma xenograft KM mice. Biochemical methods for serum LDH, AST, ALT, and AKP, ELISA method for serum HIF-$1{\alpha}$, pathological assessemnt of thymus, immunohistochemistry detection of tumor tissue caspase3 and mutant p53 protein, and qRT-PCR detection of bax/ bcl-2 mRNA were applied. Results: Compared with DDP control group, the bodyweight increased in ASSF-DDP group (p<0.01). Tumor inhibition rates for DDP, ASSF, ASSF-DDP were 62.7%. 43.7% and 71.0% respectively, with a q value of 0.90. Compared with other groups, thymus of DDP control group had obvious pathological injury (p<0.01), serum LDH, AST, ALT, AKP increased significantly in DDP control group (p<0.01), while serum HIF-$1{\alpha}$ was increased in the model control group. Compared with this latter, the expression of mutant p53 protein and bcl-2 mRNA were decreased in all treatment groups (p<0.01), but there were no statistical difference between DDP control p and ASSF-DDP groups. The expression of caspase3 protein and bax mRNA was increased in all treatment groups, with statistical differences between the DDP and ASSF-DDP groups (p<0.01). Conclusions: ASSF can inhibit bodyweight decrease caused by DDP, can inhibit tumor growth synergistically with DDP mainly through increasing serum HIF-$1{\alpha}$ and pro-apoptotic molecules such as caspase 3 and bax, rather than through decreasing anti-apoptotic mutant p53 and bcl-2. ASSF can reduce DDP toxicity due to decreasing the release of LDH, AST, ALT, AKP into blood and enhancing thymus protection.
Cisplatin (CDDP) is a widely used anticancer drug, but at high dose, it can produce undesirable side effects such as hepatotoxicity. Because silymrin has been used to treat liver disorders, the protective effect of silymarin on CDDP -induced hepatotoxicity was evaluated in rats. Hepatotoxicity was determined by changes in serum alanine aminotransferase [ALT] and aspartate aminotransferase [AST], nitric oxide [NO] levels, albumin and calcium levels, and superoxide dismutase [SOD], glutathione peroxidase [GSHPx] activities, glutathione content, malondialdehyde [MDA] and nitric oxide [NO] levels in liver tissue of rats. Male albino rats were divided into four groups, 10 rats in each. In the control group, rats were injected i.p. with 0.2 ml of propylene glycol in saline 75/25 (v/v) for 5 consecutive days [Silymarin was dissolved in 0.2 ml of propylene glycol in saline 75/25 v/v]. The second group were injected with CDDP (7.5 mg /kg, I.P.), whereas animals in the third group were i.p. injected with silymarin at a dose of 100 mg/kg/day for 5 consecutive days. The Fourth group received a daily i.p. injection of silymarin (100 mg/kg/day for 5 days) 1 hr before a single i.p. injection of CDDP (7.5 mg/kg). CDDP hepatotoxicity was manifested biochemically by an increase in serum ALT and AST, elevation of MDA and NO in liver tissues as well as a decrease in GSH and the activities of antioxidant enzymes, including SOD, GSHPx in liver tissues. In addition, marked decrease in serum NO, albumin and calcium levels were observed. Serum ALT, AST, liver NO level, MDA was found to decreased in the combination group in comparison with the CDDP group. The activities of SOD, GSHPx, GSH and serum NO were lower in CDDP group than both the control and CDDP pretreated with silymarin groups. The results obtained suggested that silymarin significantly attenuated the hepatotoxicity as an indirect target of CDDP in an animal model of CDDP-induced nephrotoxicity.
Objectives : This study was done to investigate the protective effects of Samgiinjin-tang on liver injury of rats induced by CCI4 and d-galactosamine. Methods: All animals were divided into .5 groups, those were normal group(untreated), control group(treated with 0.9% Saline solution), sample I group(2,250mg/kg administrated), sample II group(4,500mg/kg administrated), Silymarin 200mg/kg administrated group. Liver injury of rats were induced by CCI4 and d-galactosamine, and then the serum transaminases(ALT&AST) alkaline phosphatase(ALP), lactic dehydrogenase(LDH) for enzyme activities, liver weight, lipid peroxidation and catalase, glutathione S-transferase(GST) for enzyme activities were measured. Results : The inhibitory effects on the serum ALT, AST activities in liver injury of rats induced by CCI4 were noted in both sample I and sample II group. The inhibitory effects on the serum ALP, LDH activities and the Lipid peroxidation of Mitochondria & Cytosol were noted in only sample II group. The decreased effects on the GST activities of Homogenate & Cytosol were inhibited in both sample I and sample II groups. The decreased effects on the GST activities of Mitochondria & Microsome were inhibited in sample II group. The inhibitory effects of the serum ALT, AST, LDH activities in liver injury of rats induced by d-galactosamine were noted in both sample I and sample II groups. In serum AST activities, sample II group. Conclusions : Samgiinjin-tang has protective effects against liver injury of rats induced by CCI4 and d-galactosamine. So it is required to study about the actions of mutual relation of medicines and patho-mechanism by experiment.
Objectives: Oxidative stress plays a central role in diabetes-induced complications. In the present study, the protevtive effect of Artemisia turanica (A. turanica) was evaluated against diabetes-induced liver oxidative stress and dysfunction. Methods: Fifty male Wistar rats were randomly divided into five groups: control, diabetic, diabetic + metformin, diabetic + A. turanica extract, and diabetic + A. turanica extract + metformin. Experimental diabetes was induced by a single-dose (55 mg/kg, intraperitoneally (ip)) injection of streptozotocin (STZ). Metformin (300 mg/kg) and A. turanica extract (70 mg/kg) were orally administrated three days after STZ injection for four weeks. The levels of malondialdehyde (MDA), total thiol content and superoxide dismutase (SOD) and catalase activities were measured in the liver tissue. Serum glucose concentration, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities were also determined. Results: In the diabetic group, serum glucose concentration, serum AST and ALT activities and liver MDA level were significantly higher while tissue total thiol content as well as catalase and SOD activities were lower, compared to the control group. Serum glucose in diabetic rats treated with metformin + A. turanica extract showed a significant decrease compared with the diabetic group. In all the A. turanica extract and metformin treated groups, serum ALT, tissue MDA level, total thiol content and SOD activity significantly improved compared with the diabetic rats. However, treatment of the diabetic rats only with metformin could not significantly change the activities of catalase and AST compared with the diabetic group. Conclusion: These findings suggested that A. turanica extract had a therapeutic effect on liver dysfuncyion and oxidative stress induced by diabetes, that may be probably due to its antioxidant and antiinflammatory effects.
