  • The reactive transport model on the biologically mediated sequential nitrate transformation and its subsequent transport was developed and tested. This model was coded as a reaction module within the RT3D framework (Clement, 1997). Transports of the reasonable six mobile solutes (dissolved organic carbon, $O_2,\;{NO_3}^-,\;{NO_2}^-,\;N_2O,\;N_2$) and two immobile microbes were simulated. The simulation results gave a reasonable match with supposed transport pattern. For the next step, the developed model will be validated against experimental data.

  • Photochemical reactions of N-methylphthalimide with silyl enol ethers have been explored. Irradiations of phthalimide (1) and cyclic silyl enol ethers (5a-b) are observed to promote formation of photoreduced phthalimides and photoaddition products by sequential SET-desilylation pathways. The photoreaction of phthalimide (1) and acyclic silyl enol ethers (5c-d) leads to produce oxetanes which arise by competitive single electron transfer (SET) and classical 2+2 photocycloaddition (Parteno-Buchi reaction) pathways.

  • Efficient double click methods for the synthesis of diblock codendrimers were developed. The synthetic strategy involved the sequential click reactions between an alkyne and an azide. The short core building block, 1,4-diazidobutane, was chosen to serve as the azide functionalities for dendrimer growth via click reactions with the alkyne-functionalized PAMAM dendrons as hydrophilic dendron and alkyne-functionalized Fr$\acute{e}$chet-type dendrons as hydrophobic dendron. The structure of diblock codendrimers was confirmed by $^1H$ and $^{13}C$ NMR spectroscopy, IR spectroscopy, mass spectrometry, and GPC analysis.

  • It is a challenging task to apply large detailed chemical mechanisms of fuel oxidation in simulation of complex combustion phenomena. There exist a few systematic methodologies to reduce detailed chemical mechanisms to smaller sizes involving less computational load. This research work concerns generation of a skeletal chemical mechanism by a directed relation graph with specified accuracy requirement. Two sequential stages for mechanism reduction are followed in a perfectly stirred reactor(PSR) for high temperature chemistry and to consider the autoignition delay time for low and high temperature chemistry. Reduction was performed for the detailed chemical mechanism of n-heptane consisting of 561 species and 2539 elementary reaction steps. Validation results show acceptable agreement for the autoignition delay time and the PSR calculation in wide parametric ranges of pressure, temperature and equivalence ratio.

Hybrid Organic-Inorganic Films Fabricated Using Atomic and Molecular Layer Deposition Techniques

  • Atomic layer deposition (ALD) and molecular layer deposition (MLD) are based on sequential, self-limiting surface reactions that produce atomic layer controlled and conformal thin film growth. ALD can deposit inorganic films and MLD can deposit films containing organics. ALD and MLD can be used together to fabricate a wide range of hybrid organic-inorganic alloy films. The relative fraction of inorganic and organic constituents can be defined by controlling the ratio of the ALD and MLD reaction cycles used to grow the film. These hybrid films can be tuned to obtain desirable mechanical, electrical and optical properties. This talk will focus on the growth and properties of metal alkoxide films grown using metal precursors and various organic alcohols that are known as "metalcones". The talk will highlight the tunable mechanical properties of alucone alloys grown using Al2O3 ALD and alucone MLD and the tunable electrical conductivity of zincone alloys grown using ZnO ALD and zincone MLD with DEZ and hydroquinone as the reactants.

Kinetic Study on Dephosphorylation of Myelin Basic Protein by Some Protein Phosphates

  • The dephosphorylation specificity of protein phosphatase 2A (PP2A), calcineurin (PP2B) and protein phosphatase 2C (PP2C) were studied in vitro using myelin basic protein (MBP) as a model substrate which was fully phosphorylated at multiple sites by protein kinase C (PKC) or cyclic AMP-dependent protein kinase (PKA). In order to determine the site specificity of phosphates in myelin basic protein, the protein was digested with trypsin and the radioactive phosphopeptide fragments were isolated by high performance liquid chromatography (HPLC) on reversed-phase column. Subsequent analysis and/or sequential manual Edman degradation of the purified phosphopeptides revealed that Thr-65 and Ser-115 were most extensively phophorylated by PKA and Ser-55 by PKC. For the dephosphorylation kinetics, the phosphorylated MBP was treated with calcineurin or PP2C with various time intervals and the reaction was terminated by direct tryptic digest. Both Thr-65 and Ser-115 residues were dephosphorylated more rapidly than any other ones by phosphatases. However it can be differentiated further by first-order kinetics that the PP2B dephosphorylated both Thr-65 and Ser-115 with almost same manner, whereas PP2C dephosphorylated somewhat preferentially the Ser-115.

  • Sequential passes through $Sephadex^{TM}$ columns were used to select phages that displays ligands for dextran ($\alpha$-1,6 linked linear chains) from a phage antibody library. Those phages that bound to the $Sephadex^{TM}$ in each iteration were replicated in E. coli. A phage preparation isolated on the third round selection produced 5.4 nephelos turbidity units (NTU) in a dextran specific immunonephelometric assay, a 2.2 fold higher value than the phage preparation from the first round selection. This phage gave $72\;{\pm}\;10$ normalized intensity (N.I.) in a dip-stick assay against high molecular size dextran (T2000, $2\;{\times}\;10^6) and significantly lower color ($30\;{\pm}\;6$ N.I.) against low molecular size dextran (T10, $10^4$). The presence of an Fab insert in each of these phages was confirmed using a $\beta-galactosidase linked assay and polymerase chain reaction.

  • This report describes the first clinical case of a transfusion-associated Mycoplasma haemocanis infection in a dog in Korea. A 6-year-old male Maltese underwent a red blood cell transfusion for idiopathic immune-mediated hemolytic anemia. Eighteen days after the blood transfusion, the recipient's packed cell volume decreased and basophilic organisms were found on erythrocytes. A polymerase chain reaction and sequential analysis showed that both the donor dog and recipient dog had M. haemocanis. Six weeks after doxycycline administration, no organisms were detected and the recipient's anemia had improved.

  • The kinetic mechanism of Hafnia alvei aspartase in the amination direction has been determined in the absence of metal. The initial velocity pattern obtained by varying the concentration of fumarate at several fixed concentrations of NH4+ , shows an intersection on the left of the ordinate at pH 7.0, indicating that the kinetic mechanism is a sequential mechanism in which substrate inhibition by fumarate is observed. The dead-end inhibition pattern by varying the concentration of NH4+ at several fixed concentration of succinate shows an intersection on the left of the ordinate. These data are consistent with random addition of NH4+, or fumarate. The Haldane relationship gives a Keq of 1.18 ${\times}$10-3 M at pH 7.0, which is in agreement with the values obtained from the direct determination of reaction concentrations at equilibrium (6.0 $\pm0.2$ ${\times}$10-3 M).

  • A rapid and sequential method was studied, which can determine nitrite, nitrate and ammonium ion in soil or water samples with flow injection analysis. Geometric factors including injection volume, length of the reaction coil and flow rate of carrier solution were investigated prior to sample measurement. Nitrite was determined at 540 nm by Griess reaction producing azo dye between N-(1-naphthylethylenediamine dihydrochloride) and sulfanilamide. Nitrate was also measured under the help of reduction mechanism toward nitrite with hydrazine. Ammonium was analyzed at 440 nm with Nessler's reagent. At the optimum condition, the detection limit(S/N=3) has been shown 0.1 ㎍/mL N(NO2-), 0.4 ㎍/mL N(NO3-) and 0.3 ㎍/mL N(NH4+) respectively. The results measured by colorimetry, ion chromatography and FIA were compared showing 80%-125% reasonable match each other. Injection throughput rate could be performed better than 30 times per hour.
