From surface flow visualization on a MIRA notchback reference model using oil flow technique, the topology of the singular points of the skin friction lines are delineated. Separation and reattachment lines at the front screen, at the A-pillar, at the C-pillar and on the rear side of the car including the trunk have been identified. It is worth to mention that two vortices emerging from the top of the trunk coil in the opposite direction as that vortices starting from the C-pillar edge. The positions of the singular points and the separation and reattachment lines and the foci of the vortices provide a sensitive database for validation of CFD-codes

Microspore-derived cell lines resistant to 5-methyltryptophan (5MT, a tryptophan analog) or S-(2-aminoethyl)-L-cysteine (AEC, a Iysine analog) were selected in rice by in vitro mutagenesis. For selection of 5MT or AEC resistant cell lines, suspension-cultured cells were irradiated with gamma rays. Thirteen 5MT resistant cell lines were selected and they were able to grow stably at 2 times higher 5MT concentration. A feedback insensitive form of anthranilate synthesis, the pathway specific control enzyme for tryptophan synthesis, was detected from the 5MT resistant lines. Contents of the free amino acids in five resistant lines (MR12-1 to MR12-5) showed a 7.4 to 46.6 times greater level than that in the control culture. Tryptophan, phenylalanine, and tyrosine levels in the shikimate pathway were 28.1 and 22.5 times higher in MR12-3 and MR12 4, respectively, than that measured in the control cells. Four AEC resistant cell lines were isolated from cultures grown on medium containing 1 mM AEC, They were able to grow stably with 2 mM AEC, while sensitive calli were inhibited by 0.5 mM AEC. Aspartate kinase activities of the resistant lines were insensitive to the natural inhibitor, Iysine, and accumulated 2.2 to 12.9-fold higher levels of free Iysine than that of the control cells. Especially, the levels of aspartate, asparagine, and methionine in the aspartate pathway showed higher accumulation in the AEC resistant lines than that in the control cells.

These compounds were designed to explore the effect of substituent on the nitrogen of carbamate. The target compounds were prepared by hetero Diels-Alder reaction as a key step followed by functionalization of benzylic methyl to the desired substituents. Growth inhibitory studies of the azaanthraquinones were conducted in vitro against human cancer cell lines (SNU-354; liver and MCF7; breast) and human epidermoid carcinoma cells that are sensitive (KB-3-1) and multidrug-resistant (KB-V-1). The compounds were less potent than doxorubicin against sensitive cell lines. However, the most active compound 12 was not cross-resistant with doxorubicin against KB-V-1.

The derivatives were 10 to 100-fold less potent than doxorubicin against sensitive cell lines. However, they were marginally cross-resistant with doxorubicin against KB-V-1.
Objective: In the current work, we investigated the cytotoxic and apoptotic effects of Thymoquinone (TQ), an active compound of Nigella sativa (N. sativa) and Cis-platinum, on normal renal epithelial (GP-293) and human renal adenocarcinoma cell lines (ACHN). Methods: GP-293 and ACHN cell lines were cultured in Dulbecco's modified Eagle's medium (DMEM) with 10% Fetal bovine serum (FBS) and 1% penicillin plus streptomycin antibiotic. The MTT assay was used for cellular viability assessment. Viability of cells was observed using inverted light microscope 24, 48 and 72 h after exposure of the cells to various concentrations of TQ (1, 2.5, 5, 10, 50 and $100{\mu}g/ml$) and Cis-platinum (0.5, 1, 1.5, 2, 3, 6 and $12.5{\mu}g/ml$). Moreover, apoptosis was analyzed with a flow-cytometry method. The untreated cells were considered as control group. Results: Morphological changes such as reduced cell number and increased intercellular distance and reduced cell viability in ACHN and GP-293cell lines were observed in both TQ and Cis- platinum groups; however, Cis-platinum had greater effect on ACHN cell line than GP-293 cell line. In addition, GP-293 cell line was more sensitive to TQ compared to ACHN cell line. Furthermore, TQ and Cis-platinum had apoptotic effects on both ACHN and GP-293 cell lines. Conclusion: Our findings demonstrated that TQ and Cis-platinum had cytotoxic and apoptotic effects on both cell lines, However, GP-293 cell line was more sensitive to TQ. Additionally, Cis-platinum was more effective on ACHN cell line than on GP-293 cell line.
In this paper, a new design method for coupled-line filters with tapped input/output has been developed. The design equations for this tapped filter have been obtained using a new equivalent circuit model of tapped lines. From an edge coupled-line filter, tapped lines replaces the input/output coupled lines which tend to have very narrow gaps (few mils). Therefore, tapped coupled-line filters tend to be less sensitive to filter fabrication tolerances and to be easily fabricated using milling tools. The new filter design algorithm allows very accurate filter design for frequencies less than 20 GHz and bandwidth less than 20%. Above 20 GHz, the filter performance can be optimized starting from the filter design algorithm in this paper. Simulation problems with 2-D EM tools to characterize filter performance at high frequencies have shown to be solved by providing a channel for the filter to eliminate higher order modes.

As part of the research for the natural products about prostate-related disease, this study screened 159 plant species from 46 families, which included a total of 213 different kinds of local native plants and these plants were tested for the ability to inhibit LNCaP proliferation, an androgen-sensitive prostate cancer cell line, and DU145 proliferation, which is a more aggressive androgen-insensitive prostate cancer cell line. The results indicated that nineteen of 213 types of plants exhibited antiproliferative activity (cell viability < 30%, $500{\mu}g/mL$) on the growth of androgen-sensitive LNCaP cell lines, and five of them exhibited DU145 cell antiproliferative activity (cell viability < 30%, $500{\mu}g/mL$). The methanol extracts of Eurya emarginata (stems), Gleditsia japonica var. koraiensis (leaves), Photinia glabra (leaves) and Elaeagnus macrophylla (leaves) showed antiproliferative activity on both the androgen-sensitive LNCaP cells (cell viability < 30%) and androgen-insensitive DU145 cells (cell viability > 100%). The study also found that the methanol extracts of Styrax japonica (fruits), Aralia continentalis (leaves), Fagus crenata var. multinervis (stems), Thuja orientalis (stems) and Poncirus trifoliate (branches) presented the strongest activity and demonstrated potent antiproliferative activity on both cell lines (LNCaP and DU145 cell viability < 30%).

Comet assay, single cell gel electrophoresis has been known as useful, rapid, simple, visual, and sensitive technique for measuring the DNA breakage in mammalian ce1ls. For evaluation of DNA damage using comet assay, early studies reported a change in comet length and intensity with DNA damage using simple visual technique, such as fluorescence microscopy with eyespiece. In recent, some workers are observing and analyzing nucleotide of comets using quantitative fluorescence image analysis system to estimate 'tail moment', which is defined as the product of the tail length and the fraction of total DNA in tail. Our laboratory also adopted the image analysis software for qualification. In addition, many of the practical features of comet assay render it potentially attractive as useful tool for molecular toxicology and carcinogenesis, because the system is already showing considerable promise as rapid predictor in both in vitro and in vivo experimental designs. Recently, the comet assay becomes a attractive technique to study of apoptosis, because apoptotic fragmentation of nuclear DNA into nucleosomal sizes can be evaluated by the comet assay. So, we attempted to apply the comet assay to studying the effect of various stress on the apoptosis-sensitive cell lines. Particularly, focusing on the hyperthermic apoptosis, we could find that heat shock(44˚C for 60 minutes) was sufficient to induced apoptosis in these cell lines. But using the highly sensitive comet assay, we could not detect DNA breaks immediately after heat shock.
In the present study, in order to make an in vitro model of gastric mucosa for physiological and pharmacological studies, we established two immortalized gastric surface mucous cell lines (GSM06 and GSM10), which produce periodic acid-Schiff (PAS)-and concanavalin A (Con A)-positive glycoproteins, from a primary culture of gastric fundic mucosal cells of adult transgenic mice harboring a temperature-sensitive simian virus 40 large T-antigen gene 〔1]. Gastric fundic mucosal cells were isolated as a modification of a previously described method for rats by Schepp et al. (2). The isolated gastric fundic mucosal cells were cultured in DME/F12 medium supplemented with 2% fetal bovine serum (FBS), 1% ITES (consisting of 2 mg/1 insulin, 2 mgg/1 transferrin, 0.122 mg/1 ethanolamine and 0.00914 mg/1 sodium selenite) and 10 ng/ml recombinant epidermal growth factor (EGF) in a collagen-coated culture dish. To remove fibroblastic cells from the culture, gastric mucosal cells were incubated in the culture medium containing dispase (25 U/ml) for 24 h. The cells, uncontaminated with fibroblastic cells, were then cloned by colony formation. In our series of three attempts, two cell lines (GSM06 and GSM10) have been established at last. The cells proliferated, attached to the dish ana grew until confluent monolayers were formed, and maintained tight contact with neighboring cells. Voltage instability is a phenomenon that could occur in power systems due to stressed conditions. The result would be an occurrence of voltage collapse leading to total blackout of the system. Therefore, voltage collapse prediction is an important part of power system planning and operation, and can help ensure that voltage collapse due to voltage instability is avoided. Line outages in power systems may also cause voltage collapse, thereby implying the contingency in the system. Contingency problems caused by line outages have been identified as one of the main causes of voltage instability in power systems. This paper presents a new technique for contingency ranking based on voltage stability conditions in power systems. A new line stability index was formulated and used to identify the critical line outages and sensitive lines in the system. Line outage contingency ranking was performed on several loading conditions in order to identify the effect of an increase in loading to critical line outages. Correlation studies on the results obtained from contingency ranking and voltage stability analysis were also conducted, and it was found that line outages in weak lines would cause voltage instability conditions in a system. Subsequently, using the results from the contingency ranking, weak areas in the system can be identified. The proposed contingency ranking technique was tested on the IEEE reliability test system.
