Experimental Infection of Different Tomato Genotypes with Tomato mosaic virus Led to a Low Viral Population Heterogeneity in the Capsid Protein Encoding Region

  • The complete genome sequence of a Slovak SL-1 isolate of Tomato mosaic virus (ToMV) was determined from the next generation sequencing (NGS) data, further confirming a limited sequence divergence in this tobamovirus species. Tomato genotypes Monalbo, Mobaci and Moperou, respectively carrying the susceptible tm-2 allele or the Tm-1 and Tm-2 resistant alleles, were tested for their susceptibility to ToMV SL-1. Although the three tomato genotypes accumulated ToMV SL-1 to similar amounts as judged by semiquantitative DAS-ELISA, they showed variations in the rate of infection and symptomatology. Possible differences in the intra-isolate variability and polymorphism between viral populations propagating in these tomato genotypes were evaluated by analysis of the capsid protein (CP) encoding region. Irrespective of genotype infected, the intra-isolate haplotype structure showed the presence of the same highly dominant CP sequence and the low level of population diversity (0.08-0.19%). Our results suggest that ToMV CP encoding sequence is relatively stable in the viral population during its replication in vivo and provides further demonstration that RNA viruses may show high sequence stability, probably as a result of purifying selection.

숙지황(熟地黃)이 Bleomycin에 의한 폐섬유화(肺纖維化)에 미치는 영향(影響) (The Effect of Rehmanniae Radix on Bleomycin-induced Lung Fibrosis)

  • Objective : The main purpose of this study was to determine if Rehmanniae Radix has significant effects on lung fibrosis. Materials and Methods : C57BL/6J mice were devided into three groups. These were the normal group, which were not treated, the control group, given Intratracheal instillation(IT) of Bleomycin, the sample group, given IT of bleomycin and water-extracted Rehmanniae Radix. Animals were sacrificed 14 days after IT treatment. Lung fibrosis was evaluated by analysis of bronchoalveolar larvage(BAL) total WBC, percentage of macrophage, lymphocyte and neutrophil. This was done histologically by semiquantitative histological index(SHT) of lung tissue. Results : The sample group in coparison with control group showed a decrease in the BAL, total WBC, lower percentage of lymphocyte and neutrophil(p<0.05) and correspondingly a lower percentage of macrophage(p<0.01). The Sample group showed a significant decrease of collagen accumulation with respect to the control group in SHI of lung tissue(p<0.01). INF-${\gamma}$ and IL-4 levels in BALF of mice significantly decreased in the control group(p<0.05). Conclusions : Results suggest that Rehmanniae Radix has an anti-imflamatory effect and anti-fibrotic effect on the lungs through decrease of IL-4 and total WBC count for not only macrophage, but also lymphocyte and neutrophil. The degradation of INF-${\gamma}$ calls for research beyond the scope of this study.

고콜레스테롤혈증 환자에서 식사요법이 혈청 지질농도에 미치는 영향 (Effect of Dietary Therapy on Blood Lipid in Outpatients with Hypercholesterolemia)

  • High serum cholesterol level is a major risk factor for coronary heart disease(CHD). Nutrition therapy of hypocholesterolemic diets with increased physical activity is the essential step in the treatment of the hypercholesterolemic patients. The purpose of this study was to evaluate the short-term effect of intensive dietary therapy combined with regular exercise in lowering serum cholesterol level. Seventy three hypercholesterolemic outpatients(mean: 268.0$\pm$24.7mg/dL), aged 34 to 73(mean: 56$\pm$9.8yrs), who visited cardiology OPD of Yonsei Cardiovascular Center from April through October, 1998 were studied. Anthropometric measurements, usual nutrient intake survey using semiquantitative food frequency questionnaire, and daily nutrient intake analysis were performed. The nutrition counseling for the hypocholesterolemic diet and exercise therapy was performed individually. After a 4 week intensive dietary therapy, comparisons of body weight, nutrient intakes, and blood lipid concentrations were evaluated for the hypocholesterolemic effect of intensive diet therapy. After 4 weeks of intensive diet therapy, significant reductions in percent ideal body weight(p<0.01), saturated fat intake(p<0.01), and cholesterol intake(p<0.01) were observed. There were significant reductions in serum total cholesterol and LDL-cholesterol levels after 4 weeks of therapy by 8.1%(p<0.01) and 9.7%(p<0.01), respectively. Our results showed that intensive dietary therapy was effective in reductions of serum total and LDL-cholesterol levels. The goal for this dietary modification should not be temporary but rather be permanent in eating behavior accompanied by appropriately increased physical activity.

농업인의 인지된 건강상태와 영양소 섭취와의 관련성 연구 (The Relation of Consciousness of Health Status and Nutrient Intakes in Korean Farmers)

  • The purpose of this study was to compare the consciousness of health status and nutrient intake of farmers in Korea. Eight hundred were surveyed by rural leaders of Rural Development Administration(RDA), composed of 35.1 % male and 64.9% female. Data collection included personal & family situation by Interview method for influencial factors of health status. Food intake was determined by semiquantitative food frequency method with 65 kinds of foods. 32.5% of the subjects were elementary school graduates, younger age groups and males had higher levels of education. Mean family numbers was 4.2 persons. In the concept of disease prevalence, gender difference existed. Liver and heart ailments were frequent in males and waist pain and urinary complaints in females, and schoulder pain and nervesness were frequently prevalent to all farmers. The energy intake of farmers was 2000Kca1/day, 19:16:65 of PFC(protein, fat, and carbohydrate) ratio was approached at recommended composition of energy. But calcium and iron were lower than Korean Recommended Dietary Allowences(KRDA), Vitamin A, thiamin, niacin, and vitamin C were over of KRDA, but riboflavin, pyridoxin, and vitamin I were lower than KRDA. The nutrient intakes of poor health groups were significantly lower than those of healthy. In fatty acid composition, monounsaturated fatty acids(FA) intake was higher than that of saturated FA. The CMI (Cornell Medical Index) and Farmers' syndrome were significantly correlated with personal factors, such as height, number in family, education level, and working level. But the correlation of health status with nutrient intakes were only weakly significant. Depending on the regression analysis, Farmers' syndrome explanation about nutrient intake was low(R2 was only 0.01 more or less), but some nutrients (energy, niacin, pyridoxine, vitamin E, and lipids) could explain this significantly. It was concluded that body complaints of farmers could ameliorate with good nutrition.

Functional Investigation on Aromatase in Endometrial Hyperplasia in Polycystic Ovary Syndrome Cases

  • Objective: To explore the possible significance of aromatase P450 in endometrial hyperplasia with a background of polycystic ovary syndrome (PCOS). Methods: Immunohistochemistry was used to determine the expression of aromatase P450 in endometrium of PCOS patients. Semiquantitative analysis of aromatase P450 expression of mRNA and protein level wasalso carried out by real-time quantitative RT-PCR method. After endometrial cells were stimulated by testosterone and letrozole in vitro, the estradiol ($E_2$) level was determined, and the expression of cell aromatase P450 mRNA was assessed. Results: The aromatase P450 mRNA level was increased in endometria of PCOS patients. When endometrial cells were cultured with $10^{-6}M$ testosterone, the $E_2$ level in the culture medium increased. An inhibitory effect on $E_2$ generation and expression of aromatase P450 mRNA was observed when the endometrial cells were treated with $10^{-5}M$ letrozole. Conclusions: There is an increased expression of aromatase P450 in PCOS patient endometrium. Androgen stimulation could enhance the synthesis of aromatase P450 mRNA and the production of $E_2$ in endometrial cells in vitro while letrozole could do the reverse.

Suppression of Cellular Apoptosis Susceptibility (CSE1L) Inhibits Proliferation and Induces Apoptosis in Colorectal Cancer Cells

  • The cellular apoptosis susceptibility (CSE1L) gene has been demonstrated to regulate multiple cellular mechanisms including the mitotic spindle check point as well as proliferation and apoptosis. However, the importance of CSE1L in human colon cancer is largely unknown. In the present study, we examined expression levels of CSE1L mRNA by semiquantitative RT-PCR. A lentivirus-mediated small interfering RNA (siRNA) was used to knock down CSE1L expression in the human colon cancer cell line RKO. Changes in CSE1L target gene expression were determined by RT-PCR. Cell proliferation was examined by a high content screening assay. In vitro tumorigenesis was measured by colony-formation assay. Cell cycle distribution and apoptosis were detected by flow cytometric analysis. We found CSE1L mRNA to be expressed in human colon cancer cells. Using a lentivirus based RNAi approach, CSE1L expression was significantly inhibited in RKO cells, causing cell cycle arrest in the G2/M and S phases and a delay in cell proliferation, as well as induction of apoptosis and an inhibition of colony growth capacity. Collectively, the results suggest that silencing of CSE1L may be a potential therapeutic approach for colon cancer.

Difference in Severity of Acute Rejection Grading between Superfical Cortex and Deep Cortex in Renal Allograft Biopsies

  • 목 적 : 이식신 생검은 이식신 기능 이상의 원인, 거부반응의 정도, 예후 등을 확인하는데 도움이 된다. 그러나 이식신의 조직학적 변화가 신피질에 고르게 분포하지 않는 경우를 흔히 보게 된다. 따라서 본 연구는 이러한 이식신 생검에서의 잠재적인 표본추출의 오류를 평가하기 위하여 시행되었다. 방법 : 569개의 이식신 생검 표본 중에서 Banff criteria에 준하는 급성 거부반응을 보이고 있으며, 신피막부터 수질까지의 전 층을 포함하고 있는 신생검 표본 26개를 조사하였다. Banff criteria를 변형하여 조직의 변화를 간질성 염증(0-3+), 부종(0-3+), 요세관간질염(0-3+)으로 구분하여 급성거부반응의 등급을 표면 피질, 깊은 피질과 피질에 근접한 수질층 각각을 비교하여 조직학적 분석을 시행하였다. 결과 : 간질성 염증(P=0.019), 부종(P=0.023), 요세관 거부등급(P=0.019)에서 깊은 피질에서 표면 피질에 비해 급성 거부반응의 정도가 심하였다. 결론 : 이식신의 급성 거부반응을 진단하기 위하여 신생검을 실시 할 경우 깊은 피질이 포함되지 않으면 급성 거부반응을 과소 평가 할 수 있으므로 주의해야 할 것으로 사료된다.

낙동강 북부 배수유역의 토양 점토광물 분포 (Distribution of Clay Minerals in Soils on the Northern Drainage Basin of the Nakdong River)

  • 경북 북부 지역, 낙동강 상류 배수유역의 기반암별 토양 점토광물의 분포 특성을 이해하기 위하여, 준정량광물학적 분석을 실시하였다. 화강암류 분포지역에는 캐올리나이트와 스멕타이트가 다른 기반암 지역에 비하여 상대적으로 많이 함유되어 있었다. 퇴적암 지역 토양에는 일라이트의 함량이 다른 점토광물에 비해 월등히 높고, 캐올리나이트, 스멕타이트, 전이형(녹니석-스멕타이트 혼합층 및 층간수산기 질석 등) 광물들이 소량 수반된다. 변성암류와 화산암류 지역은 일라이트와 캐올리나이트 함량이 화강암류와 퇴적암류 지역의 중간 정도이나, 전이형 광물의 함량이 조금 더 높았다. 토양 점토광물의 조성은 기반암의 광물조성 및 각 광물의 풍화도 차이와 관련성이 있다. 화강암류 지역 토양의 높은 캐올리나이트 함량은 사장석의 풍화작용에 기인하나, 국지적으로 높은 스멕타이트 함량은 모암의 부분적인 열수변질작용과 관련된 것으로 추정된다. 퇴적암 지역 토양에 특히 풍부한 일라이트는 대부분 모암에서 잔류한 것이다.

Paricalcitol attenuates indoxyl sulfate-induced apoptosis through the inhibition of MAPK, Akt, and NF-κB activation in HK-2 cells

  • Background/Aims: Indoxyl sulfate (IS) is a uremic toxin and an important causative factor in the progression of chronic kidney disease. Recently, paricalcitol (19-nor-1,25-dihydroxyvitamin D2) was shown to exhibit protective effects in kidney injury. Here, we investigated the effects of paricalcitol treatment on IS-induced renal tubular injury. Methods: The fluorescent dye 2',7'-dichlorofluorescein diacetate was used to measure intracellular reactive oxygen species (ROS) following IS administration in human renal proximal tubular epithelial (HK-2) cells. The effects of IS on cell viability were determined using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays and levels of apoptosis-related proteins (Bcl-2-associated protein X [Bax] and B-cell lymphoma 2 [Bcl-2]), nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) p65, and phosphorylation of mitogen-activated protein kinase (MAPK) and protein kinase B (Akt) were determined by semiquantitative immunoblotting. The promoter activity of $NF-{\kappa}B$ was measured by luciferase assays and apoptosis was determined by f low cytometry of cells stained with f luorescein isothiocyanate-conjugated Annexin V protein. Results: IS treatment increased ROS production, decreased cell viability and induced apoptosis in HK-2 cells. IS treatment increased the expression of apoptosis-related protein Bax, decreased Bcl-2 expression, and activated phosphorylation of MAPK, $NF-{\kappa}B$ p65, and Akt. In contrast, paricalcitol treatment decreased Bax expression, increased Bcl-2 expression, and inhibited phosphorylation of MAPK, $NF-{\kappa}B$ p65, and Akt in HK-2 cells. $NF-{\kappa}B$ promoter activity was increased following IS, administration and was counteracted by pretreatment with paricalcitol. Additionally, flow cytometry analysis revealed that IS-induced apoptosis was attenuated by paricalcitol treatment, which resulted in decreased numbers of fluorescein isothiocyanate-conjugated Annexin V positive cells. Conclusions: Treatment with paricalcitol inhibited IS-induced apoptosis by regulating MAPK, $NF-{\kappa}B$, and Akt signaling pathway in HK-2 cells.

Compound K, a ginsenoside metabolite, plays an antiinflammatory role in macrophages by targeting the AKT1-mediated signaling pathway

  • Background: Compound K (CK) is an active metabolite of ginseng saponin, ginsenoside Rb1, that has been shown to have ameliorative properties in various diseases. However, its role in inflammation and the underlying mechanisms are poorly understood. In this report, the antiinflammatory role of CK was investigated in macrophage-like cells. Methods: The CK-mediated antiinflammatory mechanism was explored in RAW264.7 and HEK293 cells that were activated by lipopolysaccharide (LPS) or exhibited overexpression of known activation proteins. The mRNA levels of inflammatory genes and the activation levels of target proteins were identified by quantitative and semiquantitative reverse transcription polymerase chain reaction and Western blot analysis. Results: CK significantly inhibited the mRNA expression of inducible nitric oxide synthase and tumor necrosis factor-${\alpha}$ and morphological changes in LPS-activated RAW264.7 cells under noncytotoxic concentrations. CK downregulated the phosphorylation of AKT1, but not AKT2, in LPS-activated RAW264.7 cells. Similarly, CK reduced the AKT1 overexpression-induced expression of aldehyde oxidase 1, interleukin-$1{\beta}$, interferon-${\beta}$, and tumor necrosis factor-${\alpha}$ in a dose-dependent manner. Conclusion: Our results suggest that CK plays an antiinflammatory role during macrophage-mediated inflammatory actions by specifically targeting the AKT1-mediated signaling pathway.
