REACTION TIME, SEMEN OUTPUT AND SEMEN QUALITY OF BUFFALO BULLS AFTER PRE-COLLECTION INJECTION OF PROSTAGLANDIN F2 ALPHA (PGF2 ALPHA)

  • A study was conducted to determine the effect of prostaglandin $F_2$ alpha ($PGF_2$ alpha) on the reaction time and seminal characteristics of buffalo bulls. Semen was collected from three Murrah bulls in three periods: pre-treatment, treatment and post-treatment. During the treatment period each bull was administered 2 ml $PGF_2$ alpha (Synchrocept, Fenprostalene) im, 1 hour prior to semen collection. In the post-treatment, semen was collected 7 days after the last injection of $PGF_2$. Semen samples were evaluated immediately after collection. Pre-collection injection of $PGF_2$ alpha has no significant effect on reaction time, semen volume, percentage motility, sperm concentration and total number of sperms per ejaculate. Fluctuations in semen color and consistency were observed. There is a significant (p<0.05) increase in the mean percentage of normal spermatozoa during the treatment and post treatment periods. Likewise, administration of PG results into a significant (p<0.05) rise on the average percentage of live sperms but this effect was not manifested in the post-treatment period. Improvement in mass activity was observed during the treatment and post-treatment periods.

  • Fatty acids such as n-3 and n-6 polyunsaturated fatty acids (PUFA) are critical nutrients, used to improve male reproductive performance through modification of fatty acid profile and maintenance of sperm membrane integrity, especially under cold shock or cryopreservation condition. Also, PUFA provide the precursors for prostaglandin synthesis and can modulate the expression patterns of many key enzymes involved in both prostaglandin and steroid metabolism. Many studies carried out on diets supplemented with PUFA have demonstrated their capability to sustain sperm motility, viability and fertility during chilling and freezing as well as improving testis development and spermatogenesis in a variety of livestock species. In addition to the type and quantity of dietary fatty acids, ways of addition of PUFA to diet or semen extender is very crucial as it has different effects on semen quality in male ruminants. Limitation of PUFA added to ruminant ration is due to biohydrogenation by rumen microorganisms, which causes conversion of unsaturated fatty acids to saturated fatty acids, leading to loss of PUFA quantity. Thus, many strategies for protecting PUFA from biohydrogenation in rumen have been developed over the years. This paper reviews four aspects of PUFA in light of previous research including rumen metabolism, biological roles, influence on reproduction, and strategies to use in male ruminants.

Testicular and epididymal ultrasonography for the assessment of semen quality in the indigenous ram

  • The aims of this study were to measure the ultrasonographic biometry of genitalia of the indigenous rams and observe the relationship of biometry on semen parameters. The epididymal volume was significantly reduced (p < 0.01) after semen collection compared with before collection for both left and right part in all rams. The cumulative results showed that although there was no significant difference in length, width and volume of epididymis between before and after semen collection, however the values were lower after collection. The epididymal length was significantly correlated with epididymal volume (p < 0.01), semen motility (p < 0.05) and semen morphology (p < 0.01). Epididymal width was only significantly correlated with epididymal volume (p < 0.01) not with the semen parameters. Epididymal volume had a significant correlation only with semen morphology (p < 0.01).The scrotal circumference had the significant correlation with semen density, mass activity, concentration and motility (p < 0.01). The epididymis had the similar or slightly increased echogenicity as compared to the normal testis. During whole study, some white spots were found on testis which did not affect the semen quantity and quality. Significant variation was observed only for semen concentration and motility among the rams (p < 0.05). The overall normal morphology was 90.5 ± 4.6% with highest percentage of coiled tail abnormalities.

칡소 동결 정액 생산을 위한 스티로폼상자와 액체질소 이용 방법 (The Use of Styrofoam Box for Chikso (Korean Brindled Cattle) Semen Cryopreservation with Liquid Nitrogen)

  • 가축유전자원으로서 동결정액을 생산하는 가장 쉬운 방법은 스티로폼박스를 이용한 간이동결법으로 알려져 있다. 본 연구에서는 스티로폼 동결박스를 제작하여 가축의 동결정액 생산에 활용하는 방법을 검토하였으며 칡소 동결정액 생산을 최적화 할 수 있는 방법을 제시하고자 박스의 크기, 액체질소와 노출된 정액 스트로와 거리, 노출시간 및 생산량을 검토하였다. 2가지 동결박스를 비교하여 자료를 확보하였으며 내부 크기는 세로×가로×높이가 23.5×30.5×22.5 cm와 25.5×46.5×26.5 cm로 측정되었다. 액체질소를 5cm 높이로 채우고 액체질소 위 2, 5 및 8cm 높이에서 동결하여 융해 후 생존성을 조사하였다. 칡소 정액을 동결할 경우, 액체질소와의 노출시간은 모두 10분이 적절하였으며 25.5×46.5×26.5 cm 크기의 상자가 높은 생존율을 보여주었다(60.4±5.3% 대 67.2±3.1%). 동결 상자의 최적화 공간은 정자 동결에 가장 중요한 요소로 판단되며 1회 동결 시 최대 생산 가능한 칡소 동결정액은 60개 이상으로 증가시킬 수 있었다. 이러한 정보를 활용하면, 축종에 따라 동결 정액 생산량 결정하고 목적에 맞는 용기를 활용하여 효율적인 동결정액 생산이 가능할 것으로 판단된다.

돼지 액상 정액의 보관일수에 따른 오염 정도가 체외 수정란 생산 효율에 미치는 영향 (Effects of Bacterial Contamination of Extended Boar Semen Preservation Periods on Embryo Production In Vitro)

  • 본 연구는 항생제가 첨가되지 않은 돼지 혼합액상 정액을 17$^{\circ}C$ 정액 보관고에 보관하면서 보관일수의 증가가 따라 정자의 운동성, 정액 내 세균의 증식 여부 및 체외 수정란 생산 효율에 미치는 영향을 조사하고자 하였다. 정자의 운동성은 1일 (78.7$\pm$2.4%)에 비하여 3일(78.7$\pm$2.4%)과 5일째(64.8$\pm$2.4%)는 유의적으로(p<0.05) 낮은 운동성을 나타내었다. 보관일수에 따른 정액 내 세균수의 변화는 보관 5일이 $57.8\pm105.2\times10^4$ Cfu로 0일과 3일의 $32.1\pm76.8\times10^4$ Cfu와 $26.9\pm46.6\times10^4$ Cfu에 비하여 유의적인(p<0.05)증가를 나타내었다. 보관된 정액을 이용하여 체외 수정한 결과, 정상적인 수정(2PN)은 1일과 3일째의 66.0$\pm$2.7%와 64.0$\pm$2.7%에 비하여 5일째에는 56.0$\pm$2.6%로 유의적인(p<0.05) 차이를 나타내었다. 체외 수정란의 발달율에서 난할율은 1일째의 75.0$\pm$l.4%에 비하여 3일과 5일째는 70.0$\pm$0.3%와 71.0$\pm$0.3%로 유의적으로(p<0.05) 낮게 나타났으며, 상실배로의 발달율에 있어서 1일째는 32.0$\pm$1.4%의 발달율을 보였으나 3일과 5일째에는 28.0$\pm$1.3%와 24.0$\pm$1.3%로 유의적인(p<0.05) 감소치를 나타내었고, 배반포로의 발달율에 있어서도 1일에 15.0$\pm$1.0%에 비하여 3일과 5일은 11.0$\pm$0.9%와 8.0$\pm$0.9%로 유의적으로(p<0.05) 낮은 발달율을 나타내었다. 이상의 결과를 종합할 때 항생제가 첨가되지 않은 돼지 혼합 정액은 보관일수 3일째부터 정자의 운동성이 감소하고 세균수는 증가하였다. 또한 보존된 정액을 이용하여 체외 수정을 실시할 경우, 보관일수가 증가할수록 정상 수정율과 체외 발달율이 감소함으로 항생제를 첨가하지 않는 경우 3일 이상 정액을 보관하여 사용하지 않는 것이 바람직 하다고 사료된다.
