Loop-Mediated Isothermal Amplification Targeting 18S Ribosomal DNA for Rapid Detection of Acanthamoeba

  • Amoebic keratitis (AK) caused by Acanthamoeba is one of the most serious corneal infections. AK is frequently misdiagnosed initially as viral, bacterial, or fungal keratitis, thus ensuring treatment delays. Accordingly, the early detection of Acanthamoeba would contribute significantly to disease management and selection of an appropriate anti-amoebic therapy. Recently, the loop-mediated isothermal amplification (LAMP) method has been applied to the clinical diagnosis of a range of infectious diseases. Here, we describe a rapid and efficient LAMP-based method targeting Acanthamoeba 18S rDNA gene for the detection of Acanthamoeba using clinical ocular specimens in the diagnosis of AK. Acanthamoeba LAMP assays detected 11 different strains including all AK-associated species. The copy number detection limit for a positive signal was 10 DNA copies of 18S rDNA per reaction. No cross-reactivity with the DNA of fungi or other protozoa was observed. The sensitivity of LAMP assay was higher than those of Nelson primer PCR and JDP primer PCR. In the present study, LAMP assay based on directly heat-treated samples was found to be as efficient at detecting Acanthamoeba as DNA extracted using a commercial kit, whereas PCR was only effective when commercial kit-extracted DNA was used. This study showed that the devised Acanthamoeba LAMP assay could be used to diagnose AK in a simple, sensitive, and specific manner.

The related factors with experience and intention of dental implant among some rural elderly (일부 농촌 지역 노인의 임플란트 치료 경험 및 의향 관련 요인)

  • Objectives: Dental implants are one treatment method for tooth loss. The purpose of this study is to investigate the status of experience and intention for dental implants and related factors among elderly. Methods: The participants were 244 elderly residing in rural areas with age over 65 in Samhoeup, Yeongamgun and Jeollanamdo, Korea. Chi-square tests, t-tests, and a multiple logistic regression analysis were performed to investigate the factors influencing the experience and intention of implants. Results: Many elderlies residing in rural areas rural elderly had experienced tooth loss (83.6%). The proportion of dental implant experience was 30.9% and intention to receive implants was 57.8%. The odds ratio (OR) for experience of implants was significantly higher for participants with a lower number of chronic diseases (0: OR=12.48, 95% CI=2.12-73.38, 1: OR=4.95, 95% CI=1.66-14.72), they have dental treatment experience during the past 1 year (OR=6.61, 95% CI=1.83-23.89), they have surrounding person who have experience of implant (OR=18.06, 95% CI=2.81-115.99), higher dental implant recognition (OR=3.97, 95% CI=1.92-8.23). The odds ratio (OR) for dental implant intention was significantly higher for participants with a lower age (65-69: OR=8.18, 95% CI=2.38-278.08, 70-79: OR=3.17 95% CI=1.04-9.68), lower number of chronic diseases (0: OR=4.15, 95% CI=1.00-17.29), they have drink alcohol (OR=5.03, Cl=1.31-19.34), they have surrounding person who has experience of implant (OR=3.22, 95% CI=1.30-8.02), they have not experience of tooth loss (OR=4.65, Cl=1.22-17.70), higher dental implant recognition (OR=2.69, 95% CI=1.55-4.67). Conclusions:In the future, it is necessary to utilize the results of this study to address loss of teeth and to support dental implant treatment selection through improved increased awareness of the advantages and disadvantages of dental implants.

A Study on the Planting and Use of Landscaping Plants in Chuncheon (춘천시의 조경식물의 식재와 이용 현황)

  • This study was executed to find out how to improve the planting and use of landscaping plants in Chuncheon. The number of street trees was 17,877 in 2003. The major species of street trees were Ginkgo biloba(52.7%), Prunus sargentii(15.3%), Zelkova serrata(8.9%), Platanus occidentalis(7.3%). Salix babylonica and Platanus occidentalis were decreased, while Prunus sargentii and Zelkova serrata were increased. Salix babylonica is a good species for a lakeside city, but its seeds cause allergies to some people. Most of the trees have been removed in Chuncheon. The planting of Salix babylonica(♂) resulted in no problems, and it should be propagated to plant. The soil of several areas was analysed to investigate soil conditions. The outskirts of city had good conditions in pH, organic matter and minerals, but the downtown areas had high alkalic, due to alkali from footpath concrete blocks. Appropriate pruning adds to the aesthetic and prolongs the useful life; it also requires less managing of insects and diseases to maintain good healthy of street trees. Street trees were not properly pruned due to electric lines and shortage of pruning information. The pruning was controlled by Korea Electric Power Co, which has no pruning information. Pruning must be maintained by a professional landscape company to maintain good shape such as with Bonsai. The Shrubs planting zone between street trees and other trees, and preservation plates were established for healthy of street trees. They have to be repaired and maintained well to keep better environmental conditions. The number of tree and flower species of 68 schools in Chuncheon were 12 and 16, respectively. The species that showed high preference were Juniperus chinensis, Pinus koraiensis, Pinus densiflora, Rosa centifolia, Forsythia koreana, and Rhododendron schlippenbachii. The high frequency of the protection trees designated by Chuncheon were Pinus densiflora, Ginko biloba, Zelkova serrata, Quercus species. It was thought that the diversification of street tree species, the selection of street trees suitable to each space, the generalization of use of native species, the appropriate pruning and proper fertilization, the control of pests and diseases, and the opinions of citizens concerning landscape plants were needed to improve the planting, use and maintenance of landscaping plants in Chuncheon.

Selection of Optimal Degradation Agents for Hydrolysis of Animal Cadavers (폐가축사체 가수분해를 위한 최적 가수분해제 선정)

  • Many infectious diseases have emerged or re-emerged during the past 50 years in South Korea. There were three outbreaks of foot and mouth disease (FMD) in South Korea between January 2010 and March 2011. Over 3.45 million animals were slaughtered (33.3% of the existing pigs, 8.4% of dairy cows and 3.4% of cattle). To select optimal degradation agents of animal cadavers, degradation rates and fertilizer components of pig cadavers were investigated using hydrogen chloride (HCl), potassium hydroxide (KOH) and sodium hydroxide (NaOH) hydrolysis methods. Degradation rates of pig cadavers using HCl, KOH and NaOH were 81.1, 82.8 and 91.6%, respectively. Total nitrogen (T-N) concentration in degradation solution of pig cadavers using KOH hydrolysis method was higher than that in NaOH and HCl hydrolysis methods. Total phosphorus ($P_2O_5$) concentrations in degradation solution of pig cadavers in all hydrolysis methods ranged 0.14 ~ 0.28%. Total potassium ($K_2O$) concentration for KOH hydrolysis method was higher than that for other hydrolysis methods. The concentration of T-N and $K_2O$ in degradation solution of pig cadavers by KOH hydrolysis method were higher than that in NaOH and HCl hydrolysis methods. Thus, to recycle animal cadavers in agriculture, the optimal degradation agent for hydrolysis was KOH.

Selection of Beneficial Microbial Agents for Control of Fungal Diseases in the Phyllosphere of Cucumber Plant (오이 지상부의 주요 곰팡이 병해의 생물적 방제용 유용미생물의 선발)

  • Bacillus subtilis B29, B. subtilis M10 and Streptomyces sp. CC19 obtained from phyllosphere of cucumber plants were selected for biological control of fungal air-borne diseases. For the downy mildew, diseased area of B. subtilis B29, B. subtilis M10 and Streptomyces sp. CC19 showed 0.5%, 20.2% and 42.0%, but that of control was 82.0% respectively, in cucumber seedling test. Incidence of powdery mildew by once application of B. subtilis B29, B. subtilis M10 and Streptomyces sp. CC19 was 2.8%, 3.6% and 12.3%, respectively, whereas that of control was 65.6%. On the gray mold, diseased area of B. subtilis B29, B. subtilis M10 and Streptomyces sp. CC19 was 8.0%, 30.8% and 5.2%, respectively, compared to 81.2% for the control. Therefore, B. subtilis B29 could be a prospective antagonist for biological control of powdery mildew, downy mildew and gray mold of cucumber plant.

Analysis of Factors that will Ensure Effective Health Care Delivery System (효율적인 의료전달체계 확보를 위한 요인 분석)

  • In order to analyze factors that support effective health care delivery system, this study aimed to research general public's perception on the functions of medical institutions(based on the levels of treatment: primary care, secondary care, and tertiary care), choices of medical institution when contracted with an unfamiliar disease, and recognition of diseases based on their severities. We collected data using self-administered survey from 400 general public living in Seoul, S. Korea from April 25 to May 18, 2011. The analysis was conducted using frequency analysis, chi-square test, and t-test; we analyzed the data to see if there are differences based on gender, age, and level of education. The result of both recognition of functional differences of medical institutions and selection of medical institutions when contracted with unfamiliar diseases showed that there were no significant differences based on the gender; however there were significant differences when considering the age and education. Looking at the result of the knowledge of the disease classification based on its severity, there were significant differences in age, gender, and education. In order to provide sustainable and effective health care delivery system, utilization of primary care as well as education and promotion regarding the functional differences of medical institutions and classification of disease based on its severity need to be encouraged.

Survey on Pesticide Use by Ginseng Growers at Gangwon Farmland in Korea (강원도 인삼재배지 농약 사용 실태)

  • This study aims to investigate if the properly use of pesticide in Ginseng farmland at Gangwon-do, Korea. Questionnaire included 36 questions such as control method for plant diseases, pests and weeds and pattern of pesticide use was answered by 271 Ginseng farmers lived in 4 cities and 8 counties at Gangwon-do. Ginseng farmers have noxious plant diseases, insects and weeds such as leaf spot, anthracnose, snail, stem-mining fly, horseweed and hairy crabgrass. To control of these, 35% of farmers relied on various type of pesticides and 31.5% of farmers used pesticides mingling with natural enemy, microorganism and organic materials. Farmers selected the pesticide based on the their own experiences or by recommendation of market dealers and neighbors, resulting in the use of inappropriate pesticides in the Ginseng farmland(78.8 % ). They followed standard dosage of the pesticide from Handbook of Pesticide Application(96.3%). They, however, used the same pesticides from 2 to 5 years(44.1 %). This consecutive use of pesticide could be induced resistance. This survey resulted that study for pesticide resistance and systematic educational program for proper use and selection of pesticide to Gingseng farmers should be conducted in farmland at Gangwon-do, Korea.

Application and utilization of marker assisted selection for biotic stress resistance in hybrid rice (Oryza sativa L.)

  • Development of disease resistant plant is one of the important objectives in rice breeding programs because biotic stresses can adversely affect rice growth and yield losses. This study was conducted to identify lines with multiple-resistance genes to biotic stress among 173 hybrid rice breeding lines and germplasms using DNA-based markers. Our results showed that one hybrid rice line [IR98161-2-1-1-k1-3 (IR86409-3-1-1-1-1-1/IRBB66)] possessed 5 bacterial blight resistance genes (Xa4, xa5, Xa7, Xa13 and Xa21) while two hybrid rice lines [IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66) and 7292s (IR75589-31-27-8-33S(S1)/IR102758B)] possessed 3 bacterial blight resistance genes (Xa4, Xa7 and Xa21, and Xa3, Xa4 and xa5). Molecular survey on rice blast disease revealed that most of these lines had two different resistant genes. Only 11 lines possessed Pib, Pi-5, and Pi-ta. In addition, we further surveyed the distribution of insect resistant genes, such as Bph1, Bph18(t), and Wbph. Three hybrid breeding lines [IR98161-2-1-1-k1-3 (IR86409-3-1-1-1-1-1/IRBB66), IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66), and 7292s (IR75589-31-27-8-33S(S1) /IR102758B)] contained all three resistance genes. Finally, we obtained four hybrid rice breeding lines and germplasms [IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66), Damm-Noeub Khmau, 7290s, and 7292s (IR75589-31-27-8-33S(S1)/IR102758B)] possessing six-gene combination. They are expected to provide higher level of multiple resistance to biotic stress. This study is important for genotyping hybrid rice with resistance to diverse diseases and pests. Results obtained in this study suggest that identification of pyramided resistance genes is very important for screening hybrid rice breeding lines and germplasms accurately for disease and pest resistance. We will expand their cultivation safely through bioassays against diseases, pests, and disaster in its main export countries.

Relationship between Periodontal Disease and Chronic Kidney Disease: A Systematic Review of Cohort Studies (치주질환과 만성신장질환의 연관성: 코호트 연구에 대한 체계적 고찰)

  • The purpose of this systematic review was to investigate the association between periodontal disease and chronic kidney disease. A search of Embase, PubMed and The Cochrane Library databases was performed up to April 17, 2016. Article selection was based on cohort study design and the study subjects were patients with periodontal disease or severe periodontal disease. The final result was development of chronic kidney disease and kidney function decrease based on the estimated glomerular filtration rate values. The quantitative synthesis of the final selected articles was assessed using Review Manager statistical analysis software. A fixed-effects model meta-analysis was performed to estimate the degree of association between periodontal disease and chronic kidney disease. The search strategy identified 3,018 potentially eligible articles, of these, four studies were finally selected for meta-analysis, revealing that periodontal disease was significantly associated with the risk of developing chronic kidney disease (odds ratio, 1.65; 95% confidence interval, 1.44~1.90; p<0.001). In order to prevent the development of chronic kidney disease and kidney function decrease it is important to prevent periodontal disease, as well as minimizing the traditional risk factors known to reduce the quality of life of patients and increase disease burden.

Transplanted and many other machines were developed in the early 1990s. In ginseng breeding only pure line selection was of practical significance several verities were at the stage of seed propagation at ginseng plantations. Mutation breeding (${\gamma}$-ray. X-ray chemicals) was not successful. The research on plantlet formation through tissue culture was a little progressed but still far behind to vegetative propagation. Disease control research was concentrated in the isolation and identification of pathogans. their ecological charactelistics and biological control and soil humigation. Weed control was less important due to the development of mulching method of ridge and ditch. Quality factors of raw ginseng in relation to red ginseng process were extensively studied. Traditional quality measures were elucidated in accordance with modern analytical chemistry resulting in the importance of peptides in the centrat part rather than ginsenosides For large root production growth promoting rootzone micrcorganisms (PGPRM) were isolated and active compounds were identified. Field test on PGPRM was on going. Varictus methods formality improvement through cultivation were developed. Management research of ginseng production was rare Extension was active throuch official and private organizations and through workshop for the extension specialists, and direct lectures to grower's. Extension services made the researcher to understand the existing problems at grower's fields. Research environment for ginseng production was in prime time only for three years when Korea Ginseng Research Institute was established then gradually aggravated.
