A laboratory experiment was conducted to determine the content of phenolics and flavonoids, antioxidant activity and antioxidant enzyme activity for the extracts from cowpea seed and sprouts. Plant length and weight of cowpea sprouts were significantly increased until 7 days after seeding. Total phenolics level [mg chlorogenic acid equivalents (CAE) $kg^{-1}$ DW] was highest in dry seed (DS) extracts of cowpea ($63.9mg\;kg^{-1}$), followed by imbibed seed (IS) ($56.8mg\;kg^{-1}$) and 1-day-old sprout (1DOS) extracts ($46.4mg\;kg^{-1}$), and significantly reduced with increase of sprout age (p < 0.05). The antioxidant activity of the methanol extracts from all the samples showed same tendency to the results of total phenolics level, and dose-dependently increased. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was higher in DS (87.3%) and IS (41.2%) than in cowpea sprouts from 1DOS to 7DOS, ranging from 17.1 to 30.4%. Antioxidant enzymes, APX, POX, and POX activities were highest in 7DOS and lowest in DS. SOD activity showed much higher activity in sprouts and in seeds. Correlation coefficient between physiological-active substance and the activity was highest between APX and CAT activities ($r^2$= 0.9574). Especially, total phenolics content was more highly correlated with antioxidant or with antioxidant enzyme activities than was total flavonoid level.
Field performance and morphological characterization was conducted on seven transgenic lines of Codonopsis lanceolata expressing ${\gamma}-TMT$ gene. The shoots were obtained from leaf explants after co-cultivation with Agrobacterium tume-faciens strain LBA 4404 harboring a binary vector pYBI 121 that carried genes encoding ${\gamma}-Tocopherol$ methyltransferase gene (${\gamma}-TMT$) and a neomycin phosphotransferase II gene (npt II) for kanamycin resistance. The transgenic plants were transferred to a green house for acclimation. Integration of T-DNA into the $T_0\;and\;T_1$ generation of transgenic Codonopsis lanceolata genome was confirmed by the polymerase chain reaction and southern blot analysis. The progenies of transgenic plants showed phenotypic differences within the different lines and with relative to control plants. When grown in field, the transgenic plants in general exhibited increased fertility, significant improvement in the shoot weight, root weight, shoot height and rachis length with relation to the control plants. However, all seven independently derived transgenic lines produced normal flower with respect to its shape, size, color and seeds number at its maturity. Indicating that the addition of a selectable marker gene in the plant genome does not effect on seed germination and agronomic performance of transgenic Codonopsis lanceolata. $T_1$ progenies of these plants were obtained and evaluated together with control plant in a field experiment. Overall, the agronomic performance of $T_1$ progenies of transgenic Codonopsis lanceolata showed superior to that of the seed derived non-transgenic plant. In this study, we report on the morphological variation and agronomic performance of transgenic Codonopsis lanceolata developed by Agrobacterium transformation.
Four varieties of perilla(Perilla ocimoids L.) were tested to investigate the ecological adaptation of the crop to variations in sowing time extending from April 25 to July 25 with an interval of 30 days between each of the four sowing. As sowing time was delayed, the plant height, the number of first branches and cluster, the weight of fresh and dried stems, the day to germination and flowering decreased, showing a highly negative correlation between the showing dates and these characteristics, but a highly positive correlation between days to flowering and the required accumulated temperature. With early sowing under low temperature and long day conditions, it took about 132 days from sowing to flowering due to the long period of vegetative growth, but as sowing was delayed, the days to flowering decreased with a minimum period of 57 days. The yield of seeds in each case varied with each variety. Type A(Nonsan var.) and type B(Jinchon var.) had higher yields when sowed earlier. But type C(Namyang var. and Wooljin var.) had its highest yield in the plot sowed on May 25 and the 1000 grain weight showed a tendency to increase as sowing was delayed. In conclusion, the perilla was preyed to be a short-day plant that flowers from the begining to the middle of Sep. regardless of sowing time, so that the sowing time should be decided after due consideration of the length of the vegetative growth periods of the varieties.

This study was carried out to investigate the effect of potassium silicate on the growth and leaf epidermal characteristics of horticultural crops viz., begonia (Begonia semperflorens Link et Otto) 'Super Olympia Red' and 'Super Olympia Rose' and pansy (Viola ${\times}$ wittrockiana Hort.) 'Matrix White Blotch' and 'Matrix Yellow Blotch' in vitro. Seeds after germination were grown on a quarter strength MS medium supplemented with potassium silicate ($K_2SiO_3$) at 0, 100, 200, or $300mg{\cdot}L^{-1}$ and were maintained under a photoperiod of 16 hours at $25^{\circ}C$. Growth parameters such as plant height, root length, chlorophyll content, fresh, and dry weights have been recorded after a growth period of 58 days for begonia and 94 days for pansy. In begonia, fresh weight was significantly greatest in the $200mg{\cdot}L^{-1}$$K_2SiO_3$ treatment in both 'Super Olympia Red' and 'Super Olympia Rose'. In both pansy cultivars, fresh weight was the greatest in the $200mg{\cdot}L^{-1}$$K_2SiO_3$ treatment than other treatments. Chlorophyll content was significantly greater in the $100mg{\cdot}L^{-1}$$K_2SiO_3$ treatment for both the cultivars of begonia. Leaf area significantly increased with the higher concentrations of $K_2SiO_3$ treatment in both cultivars of pansy. Stomatal structures on the leaf epidermis were observed with scanning electron microscopy (SEM). In begonia 'Super Olympia Rose', the structure of stomata were more compact in size in the $300mg{\cdot}L^{-1}$$K_2SiO_3$ treatment than in the control. Similarly, in pansy 'Matrix White Blotch' the surface of stomata appeared to be smoother in the $300mg{\cdot}L^{-1}$$K_2SiO_3$ treatment than those wrinkled appearance in the control. The surface of the leaf epidermis appeared to be compact due to Si deposition, and thus results indicated that Si positively affected the growth and biomass production of these species. Our data show that the effect of Si on growth parameters is strongly dependent on cultivar of the plant species tested.
Seedlot disinfection techniques to control mung bean sprout rot caused by Colletoricum acutatum and C. gloeosporioides were evaluated for commercial production scheme. Soaking seedlots in propolis (100 X) and ethanol (20% for 30 min) appeared promising with control values of 85.5 and 80.8 respectively, but still resulted in up to 20% rot incidence. None of the C. acutatum conidia survived through hot water immersion treatment (HWT) for 10 min at temperatures of 55, 60 and $65^{\circ}C$, whereas the effective range of the dry heat treatment (DHT) was $60-65^{\circ}C$. Tolerance of mung bean seedlot, as estimated by hypocotyl elongation and root growth, was lower for HWT than for DHT. Germination and growth of sprouts were excellent over the range of $55-65^{\circ}C\;at\;5^{\circ}C$ intervals, except for HWT at $65^{\circ}C$ for 5 min. At this marginal condition, heat damage appeared so that approximately 2% of seeds failed to sprout to normal germling and retarded sprouts were less than 5% with coarse wrinkled hypocotyls. These results suggested that DHT would be more feasible to disinfect mung bean seedlots for commercial sprout production. Heat treatment at above ranges was highly effective in eliminating the epiphytic bacterial strains associated with marketed sprout rot samples. HWT of seedlot at 55 and $60^{\circ}C$ for 5 min resulted in successful control of mung bean sprout rot incidence with marketable sprout quality. DHT at 60 and $65^{\circ}C$ for 30 min also gave good results through the small-scale sprouting system. Therefore, we optimized DHT scheme at 60 and $65^{\circ}C$ for 30 min, considering the practical value of seedlot disinfection with high precision and accuracy. This was further proved to be a feasible and reliable method against anthracnose incidence and those bacterial strains associated with marketed sprout rot samples as well, through factory scale mung bean sprout production system.
Direct seeding is a cultivation method of Korean ginseng which can cut down production cost and increase productivity. This study was conducted to obtain detailed information about this method and to aid the development of it in Korea. Major pre-crops before ginseng cultivation were rice, corn and rye, and organic matters, such as rice straw, rye straw and poultry feces were used for basal fertilization. In direct seeding of the ginseng field, seeding density was 196 to 210 seeds per $3.3m^2$ and germination ratio was 67%. Survival ratio of 4-year-old ginsengs which were products of direct seeding was 51%, and more than 100 plants per $3.3m^2$ survived. The most critical diseases in the directly seeded ginseng field were grey mold, damping off, and stem diseases including stem spot disease. Plant growth of the ginseng cultivated by the direct seeding method was not different from the ginseng cultivated by transplanting method. But the root of the ginseng cultivated by the direct seeding method took the shape of a shorter main root compared to the ginseng cultivated by the transplanting method. Root yield per $3.3m^2$ of the direct seeding ginseng field was larger compared to the field of the transplanting cultivation.
This study was carried out to determined the optimum seeding rate and number of seedling stands per unit area for high yield with two varieties of rice, Nakdongbyeo (Japonica type) and Samgangbyeo (Indica x Japonica type), in direct water-seeded rice culture at Crop Experiment Station. Seeds coated with $CaO_2$ after germination were sowed at May 4 in 7 levels, such as 1.6, 1.8, 2.1, 2.7, 3.5, 4.0, 4.3kg /10a, and the number of seedling stands per square meter was adjusted to be 30, 40, 60, 80, 100, 120, 140 plants, respectively. The maximum number of tillers increased as the seeding rates increased, but the ratio of effective tillers decreased. The critical dates of effective tillering of tested varieties were delayed by the lower seeding rates, and the date was June 18 at the plot of seeding rate with 3.5kg /10a. The number of panicles per unit area increased in accordance with the increased seeding rates. The number of spikelets per panicle in Nakdongbyeo was not affected by the seeding rates, but in Samgangbyeo it decreased as the seeding rate increased. The optimum number of seedling stand per square meter appeared to be 78 in Nakdongbyeo and 120 in Samgangbyeo, respectively. It meant that the optimum seeding rates for Nakdongbyeo and Samgangbyeo were about 2.7 and 4.0kg /10a, respectively.
In this study, a propagation characteristics of green kyllinga(Kyllinga brevifolia var. leiolepsis H.) was investigated. Two to three rhizomes(1st rhizome) arised from the basal part of main shoot at 4th leaf stage and were grown to extend on the soil surface. Then new shoots(lst tillers) emerged from every node of the 1st rhizome. Second rhizomes also arised from the basal part of lst tiller at the time of 4th leaf openning. With such a regularity, 5th rhizomes and about 688 shoots were produced from one plant during one year-cultivation without competition under the natural condition. The degree of vegetative growth and seed formation was 3 times and 2.5 times higher in rhizomeoriginated plant than in that from seed, respectively. The amount of seed formation and the 1000 seeds weight was highest in one planted on June 1 and Aug. 1, respectively. Short-day treatment of less than 14hr appeared to be necessary for the induction of flowering and it was effective as treated not during germination but since at least 2 leaf stage of green kyllinga. Each shoot individually responded to short-day. When plants were exposed to short-day (9hr, day / 15hr, night) treatment at the stage of 2-3 leaves, more than 7 cycles were required for flowering induction. Bolting in main shoot occurred after emergence of 4 leaves under the short-day condition. Exogenous $GA_3$ slightly accelerated the velocity of bolting only in short-day condition.
The potential of juglone a plant naphthoquinone as a natural herbicide on new target, 7-keto-8-amino pelargonic acid synthetase (KAPAS) in the early step of biotin biosynthesis pathway, was performed in vitro and in vivo. Juglone effectively inhibited KAPAS activities in vitro and the $IC_{50}$ was $9.5{\mu}M$. Foliar application of juglone showed very good herbicidal activity to the eight-tested weed species. Among them, Solanum nigrum was completely controlled at a concentration of $250{\mu}g\;mL^{-1}$ with main symptoms of desiccation or burndown. Digitaria sanguinalis and Aeschynomene indica were also sensitive to juglone treatment. All eight weed species were controlled by 90~100% at a concentration of $500{\mu}g\;mL^{-1}$. However, soil application of juglone to Digitaria sanguinalis did not show any herbicidal symptoms. Cellular leakage from cucumber leaf squares treated with juglone increased depending on the concentrations increased from 6.25 to $100{\mu}M$ after 24 hours incubation with or without light. However, chlorophyll loss in cucumber leaf squares was negligible. Biotin supplements significantly rescued the inhibition of germination rate of Arabidopsis thaliana seeds previously inhibited by the juglone. Our results suggest that the juglone is a possible environmental friendly herbicide candidate with a new target KAPAS inhibiting activity.
For the enzymatic production of $2-O-{\alpha}-D-glucopyranosyl-L-ascorbic$ acid (AA-2G) from ascorbic acid, rice seed was used as the source of ${\alpha}-glucosidase$ having transglucosylation activity. Among six rice varieties, cultivated in Korea, ${\alpha}-glucosidase$ activity of Oryza savita L. cv. Ilpumbyeo was the highest with 125.03 unit/ml and it had maximum specific activity with 8.52 unit/mg protein when rice seeds were grown for 3 days after germination. For the production of AA-2G using crude extract of O. savita L. cv. Ilpumbyeo, maltose was most effective glucose donor. The optimum concentration of maltose and ascorbic acid were 125 mM and 175 mM, respectively. The optimum concentration of ${\alpha}-glucosidase$ was 100 unit. The most effective buffer was 100 mM sodium citrate. The optimum pH and temperature were 5.0 and $60^{\circ}C$, respectively. Under the optimum condition, $108.43\;{\mu}M/unit$ of AA-2G was produced from ascorbic acid after 35 minutes of reaction, which corresponds to 6.2% of conversion ratio based on the amount of ascorbic acid used.
