The response of dissolved oxygen (DO) concentrations caused significant change in root number, root length, coleoptile length, shoot length and leaf age of seedlings. The genotypic difference in the effect of DO also highly significant (P<0.01) for all of the seedling traits. The number and length of root were extremely inhibited at the condition of $0.39\pm0.09$ DO concentration. While the coleoptile elongated markedly in the lowest DO concentrations, the shoot did not develop. The root growth was improved slightly at the $1.39\pm0.27mg L^{-1}$, however, there were no difference among genotypes at these two low DO concentrations. As the DO concentration become higher, the growth of root and shoot was improved remarkably. Root number, root length and shoot length was significantly different between $20\;and\;30^{\circ}C$ in DO rich and normal conditions, the development of those traits were apparently accelerated in high water temperature, however those traits of seedlings in DO deficiency were not different between the two temperatures except for shoot length. On the other hand the coleoptile length was not affected by the stagnant water temperature; it was stimulated by the low DO concentration. The competition of DO was greater as the seedling density was increased in the stagnant water, therefore the seedlings grown under high density have long and white coleoptiles, and the growth of roots and shoots was retarded severely.

The development and productivity of maize (Zea mays L.) is frequently impacted by water scarcity, and consequently to increased drought tolerance in a priority target in maize breeding programs. To elucidate the molecular mechanisms of resistance to drought stress in maize, RNA-seq of the public database was used for transcriptome profiling of the seedling stage exposed to drought stress of three levels, such as moderate, severe drought stress and re-watering. In silico analysis of differentially expressed genes (DEGs), 176 up-regulated and 166 down-regulated DEGs was detected at moderated stress in tolerance type. These DEGs was increasing degradation of amino acid metabolism in biological pathways. Six modules based on a total of 4,771 DEGs responses to drought stress by the analysis of co-expression network between tolerance and susceptible type was constructed and showed to similar module types. These modules were discriminated yellow, greenyellow, turquoise, royalblue, brown4 and plum1 with 318, 2433, 375, 183, 1405 and 56 DEGs, respectively. This study was selected 30 DEGs to predicted drought stress response gene and was evaluated expression levels using drought stress treated sample and re-watering sample by quantitative Real-Time Polymerase Chain Reaction (qRT-PCR). 23 genes was shown increasing with drought stress and decreasing with re-watering. This study contribute to a better understanding of the molecular mechanisms of maize seedling stage responses to drought stress and could be useful for developing maize cultivar resistant to drought stress.
The research was aimed to improve pre-harvest methods to maintain marketability in postharvest coniferous lettuce. Non-woven pots or plug plates were used to generate seedlings. No difference was found in growth characteristics of lettuce grown in non-woven pots vs plug plates. Since, seedling method with non-woven pots was convenient, lettuce harvested from non-woven pots used for water dipping treatment. The quality parameters, such as fresh weight loss, SPAD value, and appearance of lettuce were investigated after harvest. The lettuce with dipping treatment inside the plastic box container showed lower weight loss, higher SPAD value and better appearance compared to those exposed to the control (non-packaging) during the storage at 2℃. The treated plant showed higher SPAD and hue angle values 21.9 and 113.8°, respectively, compared to that of 18.8 and 108.3°, in non-treated plants. Therefore, it seems that the water dipping treatment is effective for storage method to maintain freshness of the lettuce. We showed the non-woven pot growing as a convenient seedling method for packaging treatment. Further studies will be continued to improve freshness postharvest of other horticultural crops.

To find the optimal propagation condition for endangered Cypripedium macranthos Sw., also known as lady's slipper orchid, the effect of various organic additives on in vitro germination, protocorm formation and seedling growth was investigated during asymbiotic seed culture. When $100ml{\cdot}L^{-1}$ coconut water was added to the basal medium, the highest germination rate and protocorm formation rate were achieved, with 70.8% and 74.2% respectively. Supplementation of phloem sap from birch tree or maple tree also showed a facilitating effect to improve the germination and protocorm development. With $100ml{\cdot}L^{-1}$ birch sap or maple sap, both the germination and protocorm formation rates were roughly more than 65% and 68%. The roots and buds of the seedlings grew vigorously in the medium containing $100ml{\cdot}L^{-1}$ coconut water or phloem sap, in particular, their bud formation rates increased by more than 70%. Addition of banana powder and peptone could not create a more significantly favorable culture condition, and non-addition had the worst results. Our results demonstrated that proper organic amendments such as coconut water and phloem sap might be preferred to in vitro germination and the growth of seedlings developed from the protocorm of C. macranthos Sw. during asymbiotic seed culture.

Individual differences of ozone ($O_3$) resistance for seed production, seed germination and seedling development were examined in this study. Five in each healthy and damaged trees of Pinus thunbergii growing in air polluted area for 12 years were chosen based on visible foliar injury and growth. The cones of P. thunbergii, which were collected from healthy and damaged trees, were analyzed for physical characteristics and seeds from the cones were used to test germination percentage under $O_3$ treatment. The germinated seeds were continuously exposed to $O_3$ treatment and the lipid peroxidation and activities of antioxidative enzymes were determined for both seeds and seedlings. The $O_3$ treatment for seed germination and seedling development were conducted at three conditions: control, 150 ppb and 300 ppb of $O_3$. The non-treated seeds from the damaged trees showed 21.6% lower germination than those from the healthy ones. On the $O_3$ treatment of 300 ppb, seed germination decreased approximately 10% for the healthy trees and 19% for the damaged trees compared to that on the control. The seeds from the healthy trees showed significantly higher activities of superoxide dismutase (SOD), glutathione reductase (GR), and catalase (CAT) than those from the damaged trees. The activities of GR, ascorbate peroxidase (APX), and CAT decreased along with the increasing $O_3$ concentration in two tree grades. Malondialdehyde (MDA) content of seeds was not influenced by $O_3$ treatment for two tree grades. In seedling development, there were no significant differences for length and biomass of needle and root of two tree grades at both the control and 150 ppb of $O_3$. At 300 ppb of $O_3$ treatment, however, the length and biomass of needle and stem decreased for two tree grades but no significant differences was detected in root. The seedlings from the damaged trees were more sensitive to the $O_3$ treatment, showing higher activities of SOD, APX, and CAT and content of MDA compared to those from the healthy tree seedlings. Our results indicate that seed germination and seedling development are vulnerable to increasing $O_3$ concentrations and that attention must be paid to the individual selection of tree species for reforestation.

This study was performed in order to explore the growth characteristics by growing stage in the containerized seedling and the bare root seedling of Chamaecyparis obtusa Sieb. et Zucc., which is the major plantation species of Korean southland. The height growth of 1-year-old bare root seedlings was better than containerized seedlings, which grew in the containers with capacity of 31.2 mL and 300 mL. The root collar diameter growth was the best in the containerized seedlings of container with 300 mL. The 2-year-old bare root seedlings were surveyed to be better in the height and root collar diameter growth than the containerized seedlings of container with 300 mL. In the comparison of dry mass production, it was the highest in 32 mL containerized seedlings as for 1-year-old above ground and in 300 mL containerized seedlings as for below-ground and total dry mass production. In case of 2-year-old seedlings, it was indicated to be high in bare root seedling as for the part above ground and in dry mass production of the containerized seedling as for the part below ground. In the comparison on root morphological traits of seedlings such as the total root length, the root project area, the root surface area, the average diameter in root, and the root volume, all of 1 and 2-year -old containerized seedlings showed the higher growth than the bare root seedlings except the average diameter in root. Synthesizing the results of this experiment, the containerized seedlings are relatively excellent in root development compared to bare root seedlings in the process of producing seedlings of C. obtusa, thereby being judged to have grown into excellent seedlings.
Dried top and root extracts of seven different weed species, fresh top and root extracts, and various concentrations of extract (0, 5, 10 and 15%, W/V) and residue rate (0.0, 0.25. 0.5, 0.75 and 1.0%) of velvetleaf (Abutilon theophrasti Medic.) were used to study their allelopathic effects on alfalfa in the laboratory and greenhouse. Top and root aqueous extracts of common lambsquarter (Chenopodium album L.), giant foxtail (Setaria faberii Herrm), redroot pig-weed (Amaranthus retroflexus L), velvetleaf, large crabgrass (Digitaria sanguinalis L), canada thistle (Cirsium arvense L.) and prostrate knotweed (Polygonium aviculare L.) significantly inhibited germination, seedling length, weight, and vigor in alfalfa. Top growth extracts of weeds exhibited greater allelopathic effects than root extracts. Alfalfa test species, WL-320, responded significantly different to the various weed species extracts in terms of allelopathic effect. The regression slopes of various top extracts showed significant variation with respect to germination percentage. Velvetleaf (b=3.69) extracts were the most inhibitory, while large crabgrass (b=2.39) extracts had the least allelopathic effect on alfalfa. When compared the activity of fresh velvetleaf extract to that of dry velvetleaf extract, dry extract was more inhibitory to alfalfa germination and seedling growth. Germination, seedling length and weight of alfalfa were inversely proportional to the concentration of dried velvetleaf extracts. Seedling emergence and survival percentage was inhibited by velvetleaf residue mixture treatment. Also, more of the toxic effects were observed from the dried top extracts, as compared to extracts from fresh top and root. These results demonstrate the allelopathic activity of different weed species extracts and suggest that weeds may affect crop growth and development due to the inhibitory effects of allelochemicals present in weed tissue.
This experiment was conducted to determine the effect of nursery soils and seeding density on seedling growth characteristics in automatic facility for raising of rice seedlings. The seedling characteristics were evaluated for the 10-day and 15-day old seedlings grown in six different nursery soils including farm-made soils. Two levels of seeding amount, 250 g and 300 g/tray ($60{\times}30{\times}2.5$ cm), were applied to compare the seedling characteristics according to seeding density on different nursery soils. There were wide difference in soil components among the nursery soils tested. In acidity of nursery soils, 'Bunoog 2' was the highest, pH 5.1, and 'Samkyung' was the lowest, pH 8.6. The content of available phosphate was the highest value, 843 ppm, in 'Bunong 1' and the lowest (74 ppm) in farm-made soils. The total nitrogen content of 'Bunong 1, 2, 3' and 'Weonjo' soils was higher than there of 'Samkyung' and farm-made soils. There was no difference in plant height among three types of 'Bunong' soils (Bunong 1, 2, 3), but the seedling height grown in farm-made soils was shorter than there in other nursery soils. The plant heigh was slightly taller as the increase of seeding amount from 250 g to 300 g/tray, and the difference in plant height was larger in 15-day old seedlings as compared with 10-day old seedlings. Dry weight of seedlings grown for 15 days in three 'Bunong' soils was heavier than those in other nursery soils. Based on the growing characteristics of seedlings grown in different nursery soils, the heat result was obtained from a nursery soil, 'Bunoog 2', among six marketing nursery soils tested.
Greenhouse and laboratory studies were conducted to investigate allelopathic potential of some weed species on alfalfa(Medicago sativa L.) germination and seedling growth. In the comparison between top(leaves+stems) and root extracts, top extract exhibited greater allelopathic effects on alfalfa germination than that of root. The various weed species extract differently responded to alfalfa test species, WL-320, in terms of allelopathic effect. Germination, seedling length and weight of alfalfa were inversely proportional to the concentration of dried velvetleaf extracts. Also, more of the toxic effects were observed from the dried extracts compared to the fresh extracts. Residue of velvetleaf inhibited significantly alfalfa emergence and survival percentage compared to the control. The emergence and survival percentage of alfalfa were 44%, 57% at 1.0% residue treatment, respectively. When weed residues were mixed with silica sand with incubation time, velvetleaf residue most inhibited alfalfa growth. The degree of inhibition increased as incubation time increased. An incubation for 72h caused the greatest inhibition of alfalfa growth. These results demonstrate the different allelopathic activity of weed species extracts on alfalfa and suggest that weed may affect alfalfa growth and development through the inhibitory effects of allelochemicals present in weed tissue.
