Effect of Seed Position on Seed Size, Contents of Ginsenosides, Free Sugars and Fatty Acids in Panax ginseng (종자의 착생위치가 인삼종자의 크기, 사포닌, 유리당 및 지방산함량에 미치는 영향)

  • This study was carried out to know the effect of seed position on the size, contents of ginsenosides, free sugars, and fatty acids in ginseng seeds. Seed positions were classified by the three portions as center, middle and border in a seed cluster. Seed weight at center was light remarkably in comparison with those of seeds of at border and middle. The weight of embryo plus endosperm was in same tendency as seed weight. Percentage of single-seeded berry was smaller than that of the double-seeded, and the triple-seeded was rare. The percentage of the single-seeded increased from the border to the center. Size of the single -seeded seed was smaller than that of the double- seeded. Rate of dehiscence did not differ among different seed positions. The major ginsenosides in seed were Re, Rb$_1$, and Rb$_2$. The contents of Rb$_2$ and total saponin were highest in border, least in center, but reversed in Re and Rd. Major free sugars in seed were sucrose and glucose. The sucrose content was gradually decreased according to the seed position from border to center. Major fatty acids in the seed were oleic and linoleic acid. Contents of palmitic and linolenic acid were different according to the seed position.

Effects of Seed Size and Several Factors on Ultra-drying and Germination of Ultra-dried Seeds in Soybean

  • Ultra-drying [<5.0% seed moisture content (SMC)] storage technique is a cost-effective storage method for oily seeds. To decide proper ultra-drying condition for soybean seeds, drying rate was compared three silica gel to seed ratios, two seed sizes with varietal difference, two kinds of container, and three seed amounts per container under :t 23$\pm$1$^{\circ}C$. When the relative humidity (RH) was reduced at the rate of less than 0.1 % a day, silica gel was replaced with dry one by 47 days. Higher silica gel to seed ratios (3:1 and 2:1) dried faster than lower ratio (1:1) until 28 days, but not after 43 days of drying. Also, large seeded variety was dried faster than small seeded variety. Kinds of container and seed amounts per container didn't show differences in drying of soybean seeds. After completion of ultra-drying, percentage germination by standard germination test (SGT) was not different among silica gel to seed ratios, kinds of container, and seed amounts per container, except among seed sizes (varieties). Before SGT, soybean seeds were premoistened using saturated ${CaCl}_2$ for 48 hours and ${NH}_4$Cl for 24 hours in desiccators. To compare germinability between ordinary-dried seeds and ultra-dried seeds, the seeds of seven soybean varieties, which were varying in size from 8.1 to 34.9 g per 100 seeds, were dried using same amount of silica gel under 23$\pm$1$^{\circ}C$. After completion of 76 days of drying, SMCs were reduced to 3.13-3.45% from 7.86-8.82%. SMC after completion of drying was not correlated with 100-seed weight (r=0.556). Before germination tests, soybean seeds were premoistened using saturated salt solutions. Percentage germination was higher with ultra-dried seeds than ordinary-dried seeds in SGT and higher with ordinary-dried seeds than ultra-dried seeds in AAT at the beginning of storage and after 6 months storage, but general trend of percentage germination was not observed among varieties classified by 100-seed weight. From these results, we concluded that further studies are needed to improve ultra-drying storage method for soybean seeds.

Identification of Pelleting Materials and Effect of Nutrient Addition on the Germination of Pelleted Lettuce Seeds (상추 펠렛종자의 피복물질 탐색과 영양물질 첨가가 발아력에 미치는 영향)

  • Seedling mechanization of lettuce is known to be difficult due to the small seed size and the irregular seed shape. The purpose of seed pelleting is to make seedling mechanization possible by enlarging the seed size. After that, it can reduce seedling and thinning labours and can also save seeds. According to the results, there were significant differences on the percent germination and day to 50% final germination in accordance with the pelleting polymer. Among the pelleting polymer, gemination of seeds using polynimyl alcohol (PVA) was generally smooth, and followed by polyvinyl pyrrolidone (PVP), hydroxyethyl cellulose (HEC), methyl cellulose (MC), and wteen 80. The germination rate was also different according to the pelleting particulate matters. Generally, percent germination and speed of seed using the mixture of diatomaceous earth, talc and calcium carbonate were higher and faster than using other polleting materials, respectively. Thus, it should be suitable pelleting particulate matter for the pelleting of lettuce seeds. On the other hands germination of seeds using limestone, calcium oxide and benonite were low. The seed size of lettuce after pelleting was 33 times as large as the raw seed size. During the process of seed pelleting, netrient addition induced the decrement on the germination and the delay of germination speed. Also, there was differences in the germinability of pelleted seeds in accordance with the addition of nutrient sources. MS medium was generally lower than monosodium phosphate in inhibition of seed germination. Germination of pelleted seeds after priming was higher than the seeds without the treatment, and also showed the trend of early germination.

Effects of Seed Size on the Rate of Germination, Early Growth and Winter Survival in Four Oaks Species (종자크기에 따른 참나무 4종의 발아율, 초기 생장율과 생존율 변화)

  • In order to know the effect of seed size on the early plant life history, we measured the rate of germination, early growth and winter survival of four oak species with different acorn size (large, medium and small). The mean germination rate was higher in the larger seed species ($Q.$ $acutissima$ and ($Q.$ $variabilis$) than those of the smaller seed species ($Q.$ $aliena$ and $Q.$ $serrata$). Within $Q.$ $variabilis$, the germination rate decreased with larger acorn size class but that of rest other species was not significantly affected by the acorn size. The early mean growth rate decreased with acorn size in the order of $Q.$ $acutissima$, $Q.$ $variabilis$, $Q.$ $aliena$ and $Q.$ $serrata$. The early mean growth rate was higher in large or medium size than in small size of $Q.$ $acutissima$ and $Q.$ $variabilis$, but it showed no difference with acorn size in $Q.$ $aliena$ and $Q.$ $serrata$. Survival rate in winter was the highest in the medium acorn size among 4 oak species, the seedling of $Q.$ $serrata$ and $Q.$ $variabilis$ did not survive in both large and small acorn size. These results indicate that seed size influences a different effect on the early plant life history stage.

Diopside Crystal Glaze Using Seed (Seed를 사용한 Diopside 결정유약)

  • Currently, diopside ($MgCaSi_2O_6$) crystal glaze is used frequently for pottery works or in earthen wares, though the process is not straightforward. However, to create and control the positions and sizes of the crystals in desired amounts when making pottery is difficult. To solve this problem, a diopside crystal seed was created at a temperature of $1450^{\circ}C$. After planting this seed in the glaze, a glaze combination and firing process which allows a user to create crystals with the desired position and at the desired size were established. In addition, in order to investigate the creation process of the crystals, the growth patterns of the crystals were observed and examined using Raman spectrography and XRD and SEM analyses. As a result, the optimum synthesis condition of the diopside seed was created by mixing 1 mole of $CaCo_3$, 0.2 mole of $(MgCo_3)_4(MgCoH)_2{\cdot}5H_2O$ and 2 moles of $SiO_2$ and then applying a firing process to the mixture at $1,450^{\circ}C$ for 30 minutes. The optimum glaze content of the seed was 70 % feldspar, 20 % limestone and 10 % $MgCo_3$. For the firing process, it was confirmed that the size of crystal is larger with a longer firing time at $1100^{\circ}C$ by completing a two-hour process at $1280^{\circ}C$. In addition, the diopside crystal has columnar structure and is less than $1{\mu}m$ in size.

Composition and Content of Soyasaponins and Their Interaction with Chemical Components in Different Seed-Size Soybeans

  • Soyasaponins $A_1$, DDMP-conjugated group B soyasaponins ${\alpha}g\;and\;{\beta}g$, non-DDMP counterpart soyasaponin I, II+III, and DDMP moiety were quantified in the large-, midium-, and small-seed soybean varieties. Protein contents were ranged from 38.1% to 41.8%, and oil contents were ranged from 15.5% to 18.9%, respectively. Oil contents in the large-seed varieties were significantly higher than those of medium- and small-seed varieties. Among detected soyasaponin peaks, ${\beta}g$ was a major soyasaponin in DDMP-conjugated group B soyasaponins followed by soyasaponin I, DDMP moiety and $A_1$. Soyasaponin concentration among different seed size soybean varieties. The soyasaponin concentration of mediumseed ($4014.5{\mu}g/g$) was slightly higher than those of largeseed ($3755.0{\mu}g/g$) and small-seed varieties ($3620.3{\mu}g/g$), however, the differences was statistically not significant. The composition rates of soyasaponins in the large-size seeds were 9.4% of soyasaponin $A_1$, 26.5% of DDMP-conjugated soyasaponins, 49.9% of non-DDMP counterpart soyasaponins, and 14.2% of DDMP moiety, respectively. Similar results were observed in the composition ratios of middle- and small-size seeds. Oil content and C:N ratio showed the significant positive correlations with total soyasaponin concentration, while the 100-seed weight, fiber, and ash contents showed the negative correlations with total soyasaponin but statistically not significant. It was noted that protein contents didn't have any relationship with group A, group B, DDMP moiety, and total soyasaponin. This fact suggested that protein contents are not affects the variation of soyasaponin concentration.

Variation of β-carotene Concentration in Soybean Seed and Sprout

  • In this study, ${\beta}$-carotene concentrations was determined in soybean cultivar according to seed size, usage, seed coat color and cotyledon color as well as the process of seed germination. The total average concentration of ${\beta}$-carotene was $6.6{\mu}g/g$ in soybean seed, $33.3{\mu}g/g$ in soybean sprout. According to seed size, the total ${\beta}$-carotene concentration of soybean was $6.9{\mu}g/g$ in large soybean seed, $6.7{\mu}g/g$ in medium soybean seed, and $6.31{\mu}g/g$ in small soybean seed. In soybean sprout, the total ${\beta}$-carotene concentration was $21.4{\mu}g/g$ in large soybean sprout, $30.5{\mu}g/g$ in medium soybean sprout, and $43.5{\mu}g/g$ in small soybean sprout. According to the utilization of seed, the total ${\beta}$-carotene concentration of soybean seed was $7.2{\mu}g/g$ in cooked with rice soybean seed, $6.1{\mu}g/g$ in paste and curd soybean seed, and $6.3{\mu}g/g$ in sprout soybean seed. In soybean sprout, the total ${\beta}$-carotene concentration was $25.9{\mu}g/g$ in cooked with rice soybean sprout, $32.4{\mu}g/g$ in paste and curd soybean sprout, and $41.9{\mu}g/g$ in sprout soybean sprout. When comparison with seed coat color, the total ${\beta}$-carotene concentration of soybean with brown seed coat ($8.8{\mu}g/g$) was slightly higher than those of soybean with yellow ($6.1{\mu}g/g$). In soybean sprout, the total ${\beta}$-carotene concentration was $21.8{\mu}g/g$ in black seed coat sprout, $38.7{\mu}g/g$ in brown seed coat sprout, $34.1{\mu}g/g$ in green seed coat sprout, $39.5{\mu}g/g$ in yellow seed coat sprout, and $30.5{\mu}g/g$ in mottle seed coat sprout. The results of this study suggested the functional characteristics of soybean through quantitative analysis of ${\beta}$-carotene.

  • A large NaX type zeolite crystal of a uniform particle size of 20${\mu}{\textrm}{m}$ are grown with various H2O content by hydrothermal reaction and added seed crystal (2~3 ${\mu}{\textrm}{m}$) to reactant solution as a function of different adding seed levels from 3 to 15 %. The result that increased purity of NaX zeolite above 95% and homogeneity of crystal size by increasing adding seed levels, also decreased crystallization time. It was explained that adding seed to synthesis solution leaded out increase of surface area of physical contact reaction and directed growth of seed crystal, so more rapid consumption of reaction gel as increase seeding levels.

Quality Characteristics of Bread Prepared with the Addition of Roasted Safflower Seed Powder (볶음 홍화씨 분말첨가 식빵의 품질특성)

  • The quality characteristics of bread processed with the addition of roasted safflower seed powder were evaluated Safflower seed powder was roasted at 180$^{\circ}C$ for 20 min and pulverized below 45mesh size. Bread was processed by addition of control, 1, 2, 3 and 4% safflower seed powder at basic formulation. Dough raising power was increased as the addition of safflower seed powder increase, but Hunter's L value was decreased, From the sensory evaluation of safflower seed bread, color score was decreased significantly as the addition of roaste safflower seed powder increase, but overall acceptance had no significantly difference between control and added 4% of safflower seed.

A New Black Seed Coat Soybean Cultivar, 'Daeheug' with Lodging Tolerance, and Large Seed Size (검정콩 대립 내도복 신품종 '대흑')

  • A new black seed coat soybean cultivar, 'Daeheug', was developed at the National Institute of Crop Science (NICS) in 2007. The breeding goal for black seed coat soybean is to develop the cultivar with large seed size, high yield, lodging tolerance, and resistance to disease such as soybean mosaic virus (SMV), and bacterial pustule. 'Daeheug' was selected from the cross between 'Daehwang' which had yellow seed coat, large seed size, and late maturity, and 'Milyang 79' which was lodging tolerant, and SMV resistant with medium seed size, black seed coat, and yellow cotyledon. The preliminary, advanced and regional yield trials for evaluation and selection of this cultivar were carried out from 2003 to 2007. It has determinate growth habit, purple flower, brown pubescence, brown pod color, black seed coat, yellow cotyledon, spherical flattened seed shape, oval leaf shape and large seed size (34.3 g/100 seeds). In maturity, 'Daeheug' was 2 days later than the check cultivar 'Ilpumgeomjeongkong'. In the seed quality such as isoflavone contents ($635{\mu}g/g$), and anthocyanin contents ($11.2m{\ell}/g$ of seed coat), 'Daeheug' was better than the check cultivar. It has good adaptability for cooking with rice in ratio of water absorption and seed hardness. Specially, it has good processability for soybean tea with 2.13 absorbance at 530 nm, scavenge activity of DPPH radical was high, and contents of total phenolics was $1,263{\mu}g/m{\ell}$. The average yield of 'Daeheug' was 2.26 ton per hectare in the regional yield trials (RYT) carried out in six locations of Korea from 2005 to 2007.
