Germination Characteristics of Chinese Milk Vetch(Astragalus sinicus L.) Seeds Produced in China and Korea (수입 및 국내 채종 자운영 종자의 발아 특성)

  • Germination characteristics of Chinese milk vetch(CMV) seeds produced from Hanam Province, China and Milyang, Korea were investigated to give basic information on the stability of seedling establishment in the CMV cultivation. The germination percentage of the imported CMV seed from China varied according to importation year and seed collection site ranging from 79~95%. The germination of black colored seed coat was lower than the light green colored ones and germination by seed weight was not significantly different. Although the seed germination was lower under dark than in the presence of light, it was not significantly different. The germination of the imported CMV seed slightly declined to only less than 6% after one-year of storage under natural environment conditions but it significantly decreased after two years. However, when the seed was stored at the $5^{\circ}C$, the seed germination was the same as after two years of storage. On the other hand, fresh CMV seed produced in Milyang, Korea had only 8% germination due to seed coat dormancy but the germination increased to 73~85% after breaking seed dormancy after a year of storage. The high germination percentage of 72~82% was still maintained even after 27 months of seed storage unlike the CMV seed produced from China. These results indicate that CMV seeds do not require light for germination and the seed from China should be used within one-year after importation while the seeds produced from Korea can be used even after two years from harvest for stable CMV seedling establishment in the CMV-rice cropping system.

Response of germination rate and seed moisture contents to storage temperature and frequency of seed banking on seed soybean (Glycine max) for storage period

  • The seeds of soybean (Glycine max) were treated with different storage temperature for this study. The seeds of 3 accessions of soybean varieties in aluminum foil pack were used as materials. Storage temperature applied were $-18^{\circ}C$ and room temperature and seed banking (input after 3 days from output) frequencies were every 1, 6, and 12 month respectively for 9 years of storage period. As results seed banking frequency no affected to germination rate and seed moisture contents at $-18^{\circ}C$ storage room for seeds of soybean after 9 years. Germination rate of soybean seeds was changed from 96.2 % to 95.6 % averagely after 9 years of freezing ($-18^{\circ}C$) storage period. There were no differences in decreasing rate by number of seed banking frequency in soybean seeds. On the other hand, at room temperature germination rate of soybean seeds was decreased from 96.2 % to 27.3 % after 9 years which was decreased sharply to 55 % of initial viability after 6 years. The average rate of annual decrease of germination rate in soybean seeds was 38 % of initial viability at room temperature. Initial moisture contents of soybean seeds were 7.3 % and changed to 7.1 % at $-18^{\circ}C$ while it changed from 7.4 % to 7.0 % at room temperature after 9 years of storage period.

Seed Germination and Dormancy Breaking of Thalictrum rochebrunianum var. grandisepalum (H. Lev.) Nakai

  • This study was carried out to develop an effective seed propagation method for Thalictrum rochebrunianum var. grandisepalum (H. Lev.) Nakai by analyzing seed dormancy types and germination characteristics. Seeds were collected between September to October at Gangwon province, and well-selected seeds were used while being dry-stored at 4±1℃. The seed size ranged 4.52 × 1.58 ㎜ and the weight of thousand seeds were 1,603.5 ± 0.02 ㎎. The moisture content was 7.2%. Seeds were achene type, and morphology characters showed an elliptical shape and rough texture, and light brown in color. Moist-chilling treatment was conducted for dormancy breaking because the seeds had an undeveloped embryo of liner type. The embryo had developed during a moist-chilling period, constantly, and fully developed in 10 weeks. Consequently, it seemed to be non-deep complex or intermediate complex type of morphophysiological dormancy, and embryo dormancy was broken by wet-chilling for 10 weeks. After 10 weeks of wet-chilling treatment, seed germination began. Germination percentage was higher in dark condition raher than light condition and recorded the maximum at 25℃ in the dark (16.3%). A pre-soaking treatment with a combined plant growth hormones promoted germination and shortened T50. Specifically, seed germination of 84.5% was achieved by pre-soaking of seeds with a combined solution of 500 ㎎/L GA3 and 10 ㎎/L kinetin for 24 h after a wet-chilling treatment for 10 weeks. Thus the effect of plant growth hormones coupled with chilling temperature on seed breaking dormancy provide asubsequent growth of seedlings for successful plantation.

Effect of Seed Size and Mucilaginous Substance on Seed Germination of Peony(Paeonia lactiflora Pall.) (작약(芍藥) 종자(種子)의 형태적(形態的) 특성(特性)과 점액물(粘液物)이 발아(發芽)에 미치는 영향(影響))

  • The experiments were conducted to determine the influence of seed size and mucilaginous substance on seed germination of herbaceous peony collected from farmers' fields in Korea. No significant difference was recognized between the size of seed and the rale of germination. The maximum percentage(68%) of seed germination was obtained from the seeds with thin brown colour treated at $20^{\circ}C$. The amount of mucilaginous substances exuded from peony seeds was the highest in the seeds with black brown colour. Germination rate of peony seeds was negatively correlated with the amount of mucilaginous substance exuded from the seeds. High frequency(93%) of seed germination was obtained from the seeds which treated in the mixture(1:1) of sands and activated charcoal.

  • The morphology, seed germination, and life history of Salix nipponica were investigated in the wetland of the Nam-River, Jinju, Gyeongsangnam-do, Korea from 2000 to 2001 to provide the basic data necessary for riverine ecosystem restoration through understanding the growth characteristics of Salix nipponica. Salix nipponica had stomata on only lower side and stomata type was paracytic. Salix nipponica produced many small and light seeds. The seed number per mature ament was 1599.4, seed mass of 0.04 mg, and fertilization rate of 66.1%. Seed germination was little affected by light. Germination rate was high and mean germination time was short Under flooding condition, seeds were germinated normally and were viable after as much as 14 days of flooding. But there were no differences under various water depths on germination rates. Tree age was closely correlated with more stem diameter than tree height and there was no difference of growth rate between male and female tree. Growth rate was most rapid for 2 to 3 years after germination, and length growth was almost stopped for more than 11 years even though mass growth was done. Besides even a second-year indivisual was flowered.

Soybean Seeds Damaged by Riptortus Clavatus (Thunberg) Reduce Seed Vigor and Quality of Bean Sprout Produce

  • Riptortus clavatus, one of the many insects in major crops, damages pods and seeds, which reduces seed vigor and viability in soybeans. This study was conducted to examine the effect of diversely damaged seeds by R. clavatus on seed germination and seedling emergence and to determine the association of damaged seed with quality and yield of soybean sprouts. All seeds damaged by R. clavatus significantly (P<0.05) reduced seed vigor as measured by the rates of seed germination, germination speed, and seedling emergence. Mean seed germination rate of non-damaged seeds in sprout-soybean varieties was 97.8%, whereas the rates of seeds damaged at different levels, 31-50% and 51-80%, were 23.0 and 5.4%, respectively. The rates of seedling rot and abnormal, incomplete germination significantly (P<0.05) increased as the amount of seeds damaged by R. clavatus increased to 5, 10 and 15% against the total seeds for sprout production. Yield of soybean sprouts from seeds damaged at different levels decreased up to 13% as compared to that in normal seeds. In customer preferences on soybean sprout produce, 84% of customers participated in survey preferred to purchase sprouts from seeds with 5% of damaged seeds, but sprouts produced from seeds with 15% of damaged seeds were intended to purchase only by 22% of the customers. Areas of the seed damaged by R. clavatus were readily infected by pathogens as the seed germinated, resulted in deteriorated quality and reduced yield of sprout produce.

  • Germination and emergence habits of Chinese milk vetch (CMV) were examined to obtain the basic information for the effect of environmental conditions and cultural practices on the seedling density. Seed germination tests with different water potentials, temperatures, and soil and water depths showed the environmental effects on the characters related to seed germination in CMV. Imbibition under different temperatures reflects that initial velocity was rapid at higher temperature, however, the times to full imbibition were not different between 15 and $25^{\circ}$. The optimal germination temperature for CMV germination was ranged from 15 to 20 and the germination was highly affected by water potential of media at relatively high temperature above $20^{\circ}$. When the seeds were sown in flooded condition the germination was not proportionally affected by water depth. In addition, there was no correlation between water depth and oxygen concentration. The germination of seeds flooded by 2cm water depth were poorly germinated compared to other depths. Results indicated that the germination of submerged seeds was more highly influenced by flooding depth than the temperature, it was also affected more strongly at 10 than $20^{\circ}$. Emergence of CMV depending on the thickness of covered soil was poor when the soil layer was greater than 5cm. In the experiment with seeds collected between 22 days after flowering (OAF) and 52 DAF, the highest germination ability of CMV seeds was observed at 39 DAF and germinability was decreased subsequently as seeds became mature. The lower germinability may be due to the enhanced seed dormancy.

Changes of Protein and Lipid Composition During Germination of Perilla frutescens Seeds (들깨 종자의 발아에 따른 단백질 및 지방의 조성 변화)

  • To investigate changes in protein and total lipid contents, seed storage protein pattern, and fatty acid composition of germination perilla(Perilla frutescens) seeds. Also, the corresponding value components in cotyledons, hypocotyles and roots were measured according to germination stage. The results were summarized as follows ; During germination, pertein and total lipid contents of Yepsilldalggae and Kwangyang cultivar were decreased continuously. In particular, protein contents rapidly decreased to the 3 days after germination(DAG), and then total lipid contents rapidly decreased between 3 DAG and 10 DAG. In changes of protein and total lipid contents of cotyledons, protein contents of Yeupsildalggae was increased during the germination, but Kwangyang cultivar was decreased during the same periods. The total lipids contents of Yeupsildalggae and Kwangyang cultivar were decreased during the germination. According to SDS-PAGE analysis, there was no detectible polypeptide bands on the gel before seed germination suggesting that this may be due to the rapid degradation of the storage proteins in the mature seed by hydrolyttic enzymes during the stage. During germinatation , the polypeptide band with 27$\sim$28KD of Yeupsildalggae and Kwangyang cultivar were accumulated gradually. In changes of fatty acid composition of total lipid of Yeupsildalggae and Kwangyang cultivar , saturated fatty acids such as palmitic acid and stearic acid increased during the germination. On the other hand, unsaturated fatty acid such as linoleic acid and linolenic acid decreased during the same periods. However, oleic acid increased to the 5 DAG, and then was repidly decreased.

A Study on Characteristics of Seed Germination of Native Plants for Revegetation on the Slope of River bank (하천 제방 비탈면 녹화용 자생 지피식물의 종자발아 특성에 관한 연구)

  • To investigate the attributes of seed germination of the plants for greening of river bank slope, low-temperature treatment, chemical treatment, plant hormones treatment, and stratification were conducted using the seeds of 6 herbs growing naturally around river banks, such as Setaria glauca and faberii, Imperata cylindrica var. koenigii, Miscanthus sinensis, Pennisetum alopecuroides, Lespedeza cuneata and the seeds of 3 shrubs, such as Lespedeza bicolor., Rosa multiflora, Spiraea prunifolia for. simpliciflora. The findings are as follows. In case of herbs, Setaria glauca and faberii. have stone seeds. In the treatment with $H_2SO_4$, they were germinated through dormancy breaking, even though there were some differences depending on immersion time, and the germination rate of Setaria faberii was a bit higher than that of Setaria glauca. The germination rate of Imperata cylindrica var. koenigii was low in the treatment with $GA_3$, NAA, and the plant hormone, but the seed germination rate has improved and the germination rate of Miscanthus sinensis and Pennisetum alopecuroides showed the improved seed germination rate in low temperature treatment. In case of Lespedeza cuneata and bicolor, the leguminous plants, the treatment with $H_2SO_4$ was most effective for the improvement of seed germination and the decrease in germination time. Only Spiraea prunifolia for. simpliciflora, the rose among 3 woody plants showed the seed germination rate above 50% without any additional treatment. Thus, it was thought to be appropriate for greening. Rosa multiflora showed the attribute of germination in a dark condition at low temperature through stratification, despite its low germination rate. Based on the results as above, with respect to the greening using directly collected seeds, Miscanthus sinensis, Pennisetum alopecuroides, and Spiraea prunifolia for. simpliciflora are suitable for greening, because they do not need any additional treatment, and Lespedeza cuneata and bicolor are also suitable for greening because it is possible to improve the germination rate and reduce the germination time through the treatment on the seeds before sowing.

The Effects of Some Environmental Factors on the Seed Germination of Albizzia julibrissin Durazz. (藪種 環境要因이 자귀나무의 種子發芽에 미치는 影響)

  • Germination respone to sulfuric acid treatment, temperature, light and underwater condition were experimented for Albizzia julibrissin Durazz. seed. It took 60 minutes to break the dormancy of impermeable seed for the effective sulfuric acid treatment, and temperature sensitivity was decreased by 90 minutes' treatment. The germination rate of the seed was highest, i.e. 96% at 60 minutes acid treatment in a 21。C growth chamber. On the occasion of light sensitivity, the seed was light indifferent. The imbibition rate of seed was higher at 27。C than 21。C and in proportion to the period of acid treatment time. A. julibrissin Durazz. seed were well germinated at underwagter condition.
