Effect of Grapefruit Seed Extracts and Acid Regulation Agents on the Qualities of Topokkidduk (떡볶이떡 품질 개선에 자몽종자 추출물과 산도 조절제의 효과)

  • This study investigated methods to improve the quality of Topokkidduk using grapefruit seed extracts and acid regulation agents. Topokkidduk was investigated in terms of its moisture, color, pH, texture, microbial composition, and sensory properties. The moisture content of Topokkidduk was highest for Topokkidduk supplemented with grapefruit seed extracts. The color of Topokkidduk (the L value) decreased, whereas the "a" value increased with the addition of grapefruit seed extracts. The texture of Topokkidduk was affected more by additives than storage temperature. The addition of grapefruit seed extracts and acid regulation agents were effective for the inhibition of microbial activity in Topokkidduk during storage. The overall acceptability values of Topokkidduk (without supplements) were highest among the samples. These results suggest that grapefruit seed extracts and acid regulation agents can be used to control microbial load and moisture levels in Topokkidduk.

Effects of Grape Extracts on Free Radical Scavenging Activity and Inhibition of Pro-Inflammatory Mediator Production in Mouse Macrophage Cells (포도 추출물들의 자유 라디칼 소거 작용 마우스 대식세포주의 염증 발현 매개 인자들에 대한 생성 억제 효과)

  • Antioxidant and anti-inflammatory potentials of various grape extracts were evaluated. Extracts from Kyho seed, Kyho stem, and Campbell seed showed potent 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities compared to resveratrol $(IC_{50}=16.9,\;21.5,\;21.9,\;34.6\;{\mu}g/mL,\;respectively)$, among which, antioxidant effect of Kyho seed extract were similar to that of vitamin C $(IC_{50}=12.2\;{\mu}g/mL)$. These extracts also exhibited inhibitory activities on lipopolysaccharide (LPS)-induced prostaglandin $E_2$ production and nitrite formation in mouse macrophage RAW 264.7 cells at $50\;{\mu}g/mL$. Kyho stem and seed extracts showed growth inhibitory activities in human lung and colon cancer cells. These results suggest the potential roles of grape extracts as antioxidants and anti-inflammatory agents.

Effect of Paeonia lactiflora Pall. Seed Extracts and Forsythia viridissima Lindl. Extracts on Antioxidative System and Lipid Peroxidation in Erythrocytes of Rats Fed High-Cholesterol Diet

  • This study was conducted to investigate the antioxidative effects of Paeonia lactiftora Pall. (PL) seed extracts and Forsythia viridissima Lindl. (FVL) extracts on the antioxidative defense system and lipid peroxidation in the erythrocytes of rats fed a high-cholesterol diet. Sprague-Dawley male rats weighing 100 10 g were randomly assigned to nine experimental groups and fed 0.5 % cholesterol. The HC group did not receive any supplement, while the MP group was supplemented with 0.1 % methanol extract of PL seed, the MP2 group with 0.2 % methanol extract of PL seed, the EP1 group with 0.05 % ether-soluble fraction of PL seed, the EP2 group with 0.1 % ether-soluble fraction of PL seed, the MS1 group with 0.05 % methanol extract of FVL, the MS2 group with 0.1 % methanol extract of FVL, the ES group with 0.025% ethyl acetate-soluble fraction of FVL, and the ES2 group with 0.05 % ethyl acetate-soluble fraction of FVL. The experimental diets were fed ad libitum for 3 weeks. The erythrocyte SOD activity in the EP1 and EP2 groups increased 38% and 59%, respectively, when compared with the HC group, while the erythrocyte GSHpx activity in the EP1, EP2, and ES2 groups increased 30%, 31 %, and 29%, respectively, when compared with the HC group. The level of erythrocyte TBARS was significantly lower in the MP2, EP1, and EP2 groups than in the HC group, yet the level of serum TBARS was significantly lower in the all supplemented groups than in the HC group. The level of serum HDL- TBARS was significantly lower in the EP1 and EP2 groups than in the HC group, while the level of serum LDL- TBARS was significantly lower in the all the supplemented groups than in the HC group. Accordingly, the results indicated that the PL seed extracts and FVL extracts reduced oxidative damage by activating the antioxidative defense system in the erythrocytes of rats fed a high-cholesterol diet.

The Effect of Opuntia humifusa Seed Extracts on Platelet Aggregation and Serum Lipid Level in Ovariectomized Rats (천년초 씨 추출물이 난소를 절제한 흰쥐의 혈소판 응집과 혈청 내 지질 조성에 미치는 영향)

  • Postmenopausal women are at an increased risk of developing coronary artery disease, primarily due to dyslipidemia that accompanies the loss of estrogen secretion. This study was performed to investigate the effects of Opuntia humifusa seed extracts on blood flow and serum lipid level in ovariectomized rats. Twenty-eight 9-week old female Sprague-Dawley rats were randomly assigned to four groups as sham-operated rats (SHAM), ovariectomized rats (OVX-CON), and ovariectomized rats that were treated with Opuntia humifusa seed extracts (OVX-OHS 2% and OVX-OHS 6%). The diets were fed to the rats for 7 weeks after operation. Serum total cholesterol and triglyceride contents decreased in the SHAM group compared to the OVX-CON group. Seven weeks of feeding of Opuntia humifusa seed extracts resulted in significant (p<0.05) lowering of serum triglyceride and a decreasing tendency in total cholesterol levels. The level of HDL-cholesterol in serum increased significantly by feeding diets containing the 2% and 6% Opuntia humifusa seed hot-water extract (p<0.05). Blood passage times were shorter in the Opuntia humifusa seed extract-supplemented groups than in the untreated group (OVX-CON). The platelet aggregation ability was lower in groups treated with Opuntia humifusa seed extracts than in the OVX-CON group. These results suggest that Opuntia humifusa seed extracts may have benefits in improving metabolic syndrome in menopausal women.

Determination of polyphenolic compounds in grape seed extracts using reverse-phase high performance liquid chromatography

  • Oxidative stress is associated with many kinds of chronic diseases. Antioxidants such as polyphenols are compounds that protect cells against the damaging effects of reactive oxygen species. Grape seeds are considered good resources of polyphenols, and grape seed extracts have a very strong antioxidant effect. In the present study, we established a simple gradient reverse-phase high performance liquid chromatography method to determine polyphenol content from three different grape seed resources. An ODS (2), $150\;{\times}\;3.2\;mm$ column has been employed, and six polyphenols have been determined: gallic acid, protochatechuic acid, (+)-catechin, (-)-epicatechin, procyanidin B2, and epicatechin gallate. Catechin and epicatechin were the main polyphenol compounds in all three extracts. The amount of procyanidin B2 was higher in Extract 1 (from a company of China), while Extract 2 (extracted in our lab) and Extract 3 (from a company of USA) contained higher proportions of epicatechin gallate. For the total polyphenol content, Extract 1 was much higher than that of Extract 2 and 3. The results suggest that the dietary dose of grape seed extracts from different resources should be adjusted according to polyphenol content.

Effects of Safflower Seed Extracts and Bovine Bone on Regeneration of Bone Defects in Mongrel Dogs (홍화씨 추출물 및 우골유도합성골이 성견골 결손부 재생에 미치는 영향)

  • Many natural medicines have been studied for their capacity and effects of antibacterial, anti-inflammatory and regenerative potential in periodontal tissues. Safflower seed has been traditionally used as a drug for treatment of bone fracture in oriental medicine. The purpose of the present study was to compare the effects of safflower seed extract and bone substitute on bone formation and regeneration in artificial defects in mongrel dogs. The bony defects were made with round bur at mandible and tibia. Extracts of safflower seed and bovine bone were placed directly at each defect for experimental group, and the defect of control group was sutured without any other treatment. Experimental animals were sacrificed at 8 weeks. And then histopathologic reading and histomorphometric study was done. There was not significant differences between control and experimental groups in osteoclastic activity and infiltration of inflammatory cells. However, new capillary proliferation, fibrosis and new bone formation were prominent in safflower seed extract group. The mandibular defects of safflower seed extract group were healed with dense connective and bony tissues, and endochondral bone formation was observed in tibial defect of safflower seed extract group only. New bone area of safflower seed extract group was more significantly increased than that of control and that of bone substitute group. These results indicate that direct local application of safflower seed extracts on bony defects seems to reduces the early inflammatory response and to promotes the bone regeneration.

Efficacy of Different Seed Kernels against Root Knot Nematode Meloidogyne incognita in Mulberry

  • Five seed kernel namely, Neem (Azadirachta indica A. Juss.), Pongamia (Pongamia glabra L. Pierre), Tamarind (Tamarindus indica L.), Mahua (Madhuca indica Gmel.) and Shikakai (Acacia cancinna De.) were tested against hatching of eggs and larval mortality of Meloidogyne incognita causing root knot disease in mulberry along with Furadan (Carbofuran) and Bionema (a bioformulation developed from Verticillium chlamydosporium) for comparison. Results revealed that highest hush-up of hatching was observed in Neem (77.40%) and Pongamia (75.99%) seed kernel extracts at 100% concentration over the check. Similarly, highest larval mortality was observed in Neem and Pongamia by 76.00% and 74.50%, respectively at 100% concentration after 72 hrs of exposure period. Pot culture studies revealed that pre-application of seed kernel powders (20 days before inoculation of nematode) found to be more effective in controlling the root knot disease than post application. In pre application of seed kernel powders, maximum reduction of root knots was observed in case of Neem seed kernel powder (54.85%) followed by Pongamia (51.9%). Similar trend was also observed in reduction of egg masses/plant and nematode population /250 cc soil. Rest of the seed kernel extracts was found to be less effective in suppression of hatching, enhancing the larval mortality and controlling the root knot disease. However, application of Furadan and Bionema tested for the comparison were found to be more effective than seed kernel powders. The generated information seems to be useful in developing an ecofriendly integrated approach for the control of root knot nematode disease in mulberry.

Effect of Alfalfa Plant Extracts on Germination and Early Seedling Growth of Forages

  • Alfalfa (Medicago sativa L.) plants have been reported to contain water-soluble substances that are autotoxic as well as allelopathic. Laboratory experiment through a petri-dish assay with imbibed seeds was conducted to evaluate both autotoxic and allelopathic effects of alfalfa leaf extracts on the germination and early seedling growth of alfalfa, red clover, crested wheatgrass, and Russian wildrye. Alfalfa seed germination was delayed dependent on extract concentration, with no difference in final germination at 72 hours. Root growth of alfalfa was stimulated up to 14% above control at very low concentrations of both leaf and stem extracts of alfalfa and was significantly reduced at extract concentration of more than 0.5g dry tissue/L (${gL}_{-1}$). Leaf extracts were generally more autotoxic for root growth than were stem extracts. Hypocotyl growth was not affected by all the concentrations of both leaf and stem extracts. Root length of legumes was more sensitive to the autotoxic chemicals from leaf extracts than was germination or shoot length. Hypocotyl growth of two legume plants and plant height of two grasses were not influenced by extracts. Seed germination and root growth of legumes were more inhibited by aqueous extracts of alfalfa leaf than were those of grasses. This result indicates autotoxic effect of alfalfa leaf extracts seems to be greater than allelopathic effect.

Mucolytic Effects of Various Parts of FRUCTUS BENINCASAE Extracts in the Rat Trachea (백동과(白冬瓜)와 청동과(靑冬瓜)의 부위별(部位別) 추출물(抽出物)에 의(依)한 거담효과(祛痰效果)의 비교연구(比較硏究))

  • The in vivo effects of Fructus Benincasae (FB-Baekdongkwa(B) and FB- on the expectoration (decrease in sputum viscoelasticity) by their sorts and using rats (Sp. D. male, $150{\sim}160g$). FB was divided by seed, flesh, and bark, extracted by 95% ethanol for 3 hr. The extracts were given to rats administration and the following results were obtained: 1. When FB Recens-C extract was administered at the concentration of 300 mg/kg b.w., mucus secretion effect in the trachea was desirably stimulated. 2. The secretion of phenol red was increased in the FB-treated tracheas in the order of Semen Benincasae(SB)-B $(153{\pm}8\;%)$, FB-B $(149{\pm}10\;%)$, and FBR-C $(117{\pm}26\;%)$. In general, the effect of FB-B extract on phenol red secretion was stronger than that of FB-C. 3. When tracheobronchial lavage fluid was analyzed, the mucus secretion was relatively high $(111{\pm}14\;%)$ in FB-B compared with other extracts. 4. Microscopic analysis after direct treatment of the FB extracts to the rat tracheal tissue showed that all the FB extracts possessed no effects for the activity of the ciliary movement. 5. Glycoprotein content secreted by the seed extract of FB-B was increased compared with the control group, which represents the highest secretion effect of mucus. From the above results. we could conclude that the seed of SB-B possesses better activity for mucus secretion from trachea than the extracts of any other parts. Therefore, it is expected that the seed of SB-B may be available for the purpose of expectorant activity in the prescription of traditional medicine.

Effects of Added Supercritical Carbon Dioxide (SC-CO2) Extracts from Sweet Potatoes (Ipomoea batatas L.) and Watermelon (Citrullus lantus L.) on Oxidative Stability of Perilla Seed Oil (황색고구마 및 수박 초임계이산화탄소 추출물이 들기름의 산화안정성에 미치는 영향)

  • This study was carried out to investigate the effects of supercritical carbon dioxide (SC-CO2) extracts from sweet potatoes (SP) and watermelon (WM) on the oxidative stability of perilla seed oils (POs) over the existing ones. A comparison was done between the oxidative stability of perilla oil (PO) after the addition of 0.1% of SP, and WM extracts and PO without extract. The oxidative stability was measured based on the viscosity, acid value (AV), peroxide value (POV), antioxidant (DPPH) activity, p-anisidine value (p-AV), and fatty acid composition. The viscosities ranges were: PO without extract, from 53.99±0.99 to 74.38±1.61 cps, PO with SP extract, from 53.99±0.10 to 58.73±0.8 cps, and PO with WM extract, from 53.98±0.10 to 56.00±0.70 cps. While the PO containing the SC-CO2 extracts had significantly lower AV, POV, and p-AV, their antioxidant activity was approximately 10 times higher than that of the PO without extract. There were no significant differences in fatty acid composition between SC-CO2 extracts added groups and PO without extract (p<0.05). The findings of this study confirmed that the SC-CO2 extracts from sweet potatoes and watermelon enhanced the oxidative stability of perilla seed oils, and are potential natural antioxidants for use in the food industry.
